2The combination of chidamide with the CHOEP regimen in previously untreated patients with peripheral T-cell lymphoma: a prospective, multicenter, single arm, phase 1b/2 study显示文摘Objective:To assess the efficacy and safety of the novel histone deacetylase inhibitor,chidamide,in combination with cyclophosphamide,doxorubicin,vincristine,etoposide,and prednisone(Chi-CHOEP)for untreated peripheral T-cell lymphoma(PTCL).Methods:A prospective,multicenter,single arm,phase 1 b/2 study was conducted.A total of 128 patients with untreated PTCL(18–70 years of age)were enrolled between March 2016 and November 2019,and treated with up to 6 cycles with the Chi-CHOEP regimen.In the phase 1 b study,3 dose levels of chidamide were evaluated and the primary endpoint was determination of the maximumtolerated dose and recommended phase 2 dose(RP2 D).The primary endpoint of the phase 2 study was 2-year progression-free survival(PFS).Results:Fifteen patients were enrolled in the phase 1 b study and the RP2 D for chidamide was determined to be 20 mg,twice a week.A total of 113 patients were treated at the RP2 D in the phase 2 study,and the overall response rate was 60.2%,with a complete response rate of 40.7%.At a median follow-up of 36 months,the median PFS was 10.7 months,with 1-,2-,and 3-year PFS rates of 49.9%,38.0%,and 32.8%,respectively.The Chi-CHOEP regimen was well-tolerated,with grade 3/4 neutropenia occurring in approximately two-thirds of the patients.No unexpected adverse events(AEs)were reported and the observed AEs were manageable.Conclusions:This large cohort phase 1 b/2 study showed that Chi-CHOEP was well-tolerated with modest efficacy in previously untreated PTCL patients.Wei Zhang Liping Su Lihong Liu Yuhuan Gao Quanshun Wang Hang Su Yuhuan Song Huilai Zhang Jing Shen Hongmei Jing Shuye Wang Xinan Cen Hui Liu Aichun Liu Zengjun Li Jianmin Luo Jianxia He Jingwen Wang O.A.O’Connor Daobin Zhou 2021Cancer Biology & Medicine2021,18,3:4

7Multiomics:unraveling the panoramic landscapes of SARS-CoV2 infection显示文摘In response to emerging infectious diseases,such as the recent pandemic of coronavirus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),it is critical to quickly identify and understand responsible pathogens,risk factors,host immune responses,and pathogenic mechanisms at both the molecular and cellular levels.The recent development of multiomic technologies,including genomics,proteomics,metabolomics,and single-cell transcriptomics,has enabled a fast and panoramic grasp of the pathogen and the disease.Here,we systematically reviewed the major advances in the virology,immunology,and pathogenic mechanisms of SARS-CoV-2 infection that have been achieved via multiomic technologies.Based on well-established cohorts,omics-based methods can greatly enhance the mechanistic understanding of diseases,contributing to the development of new diagnostics,drugs,and vaccines for emerging infectious diseases,such as COVID-19.Xin Wang Gang Xu Xiaoju Liu Yang Liu Shuye Zhang Zheng Zhang 2021Cellular & Molecular Immunology2021,18,10:1

11The Establishment of Infectious Clone and Single Round Infectious Particles for Coxsackievirus A10显示文摘Coxsackievirus A10(CVA10)is one of the major etiological agents of hand,foot,and mouth disease.There are no vaccine and antiviral drugs for controlling CVA10 infection.Reverse genetic tools for CVA10 will benefit its mechanistic study and development of vaccines and antivirals.Here,two infectious clones for the prototype and a Myc-tagged CVA10 were constructed.Viable CVA10 viruses were harvested by transfecting the viral m RNA into human rhabdomyosarcoma(RD)cells.Rescued CVA10 was further confirmed by next generation sequencing and characterized experimentally.We also constructed the vectors for CVA10 subgenomic replicon with luciferase reporter and viral capsid with EGFP reporter,respectively.Co-transfection of the viral replicon RNA and capsid expresser in human embryonic kidney 293 T(HEK293 T)cells led to the production of single round infectious particles(SRIPs).Based on CVA10 replicon RNA,SRIPs with either the enterovirus A71(EVA71)capsid or the CVA10 capsid were generated.Infection by EVA71 SRIPs required SCARB2,while CVA10 SRIPs did not.Finally,we showed great improvement of the replicon activity and SRIPs production by insertion of a cis-active hammerhead ribozyme(HHRib)before the 50-untranslated region(UTR).In summary,reverse genetic tools for prototype strain of CVA10,including both the infectious clone and the SRIPs system,were successfully established.These tools will facilitate the basic and translational study of CVA10.Min Wang Jingjing Yan Liuyao Zhu Meng Wang Lizhen Liu Rui Yu Ming Chen Jingna Xun Yuling Zhang Zhigang Yi Shuye Zhang 2020Virologica Sinica2020,35,4:1

16Solid-liquid contact TENG using a melting near-field direct writing PCL nano-fiber structure显示文摘Recently,the triboelectric nanogenerator(TENG)has been widely studied for the development of flexible and wearable electronics.Among the various methods for TENG modification,melting nearfield direct writing is a new method to fabricate solid-liquid TENG.Here,the electrospun PCL was introduced with traditional polymers to fabricate the compound solid triboelectric layer,and water,dimethicone and GaIn were chosen as the liquid triboelectric layers.In this paper,the solid substrate effect,temperature gradient effect,and the liquid substrate effect were compared in terms of TENGs.A free-standing model of TENG is employed in this paper,and the charge generated by the PCL-PI composite solid triboelectrical solid layer is more than 10 times higher than the pristine one,showing the high charge generating ability to melt near-field direct written microfibers.Furthermore,detailed investigation will be discussed how we can get a high the out-circuit voltage and short-circuit current.Shuye Zhang Yingjun Chen Dayin Wang Peng He 2023International Journal of Smart and Nano Materials2023,14,1:0

18A nanonewton-scale biomimetic mechanosensor显示文摘Biomimetic mechanosensors have profound implications for various areas,including health care,prosthetics,human‒machine interfaces,and robotics.As one of the most important parameters,the sensitivity of mechanosensors is intrinsically determined by the detection resolution to mechanical force.In this manuscript,we expand the force detection resolution of current biomimetic mechanosensors from the micronewton to nanonewton scale.We develop a nanocrack-based electronic whisker-type mechanosensor that has a detection resolution of 72.2 nN.We achieve the perception of subtle mechanical stimuli,such as tiny objects and airflow,and the recognition of surface morphology down to a 30 nm height,which is the finest resolution ever reported in biomimetic mechanosensors.More importantly,we explore the use of this mechanosensor in wearable devices for sensing gravity field orientation with respect to the body,which has not been previously achieved by these types of sensors.We develop a wearable smart system for sensing the body’s posture and movements,which can be used for remote monitoring of falls in elderly people.In summary,the proposed device offers great advantages for not only improving sensing ability but also expanding functions and thus can be used in many fields not currently served by mechanosensors.Chi Zhang Mengxi Wu Ming Li Lixuan Che Zhiguang Tan Di Guo Zhan Kang Shuye Cao Siqi Zhang Yu Sui Jining Sun Liding Wang Junshan Liu 2023Microsystems & Nanoengineering2023,9,4:0
19UHRF1/DNMT1—MZF1 axis-modulated intragenic site-specific CpGI methylation confers divergent expression and opposing functions of PRSS3 isoforms in lung cancer显示文摘As confusion mounts over RNA isoforms involved in phenotypic plasticity,aberrant CpG methylation-mediated disruption of alternative splicing is increasingly recognized as a driver of intratumor heterogeneity(ITH).Protease serine 3(PRSS3),possessing four splice variants(PRSS3-SVs;PRSS3-V1—V4),is an indispensable trypsin that shows paradoxical effects on cancer development.Here,we found that PRSS3 transcripts and their isoforms were divergently expressed in lung cancer,exhibiting opposing functions and clinical outcomes,namely,oncogenic PRSS3-V1 and PRSS3-V2 versus tumorsuppressive PRSS3-V3,by targeting different downstream genes.We identified an intragenic CpG island(iCpGI)in PRSS3.Hypermethylation of iCpGI was mediated by UHRF1/DNMT1 complex interference with the binding of myeloid zinc finger 1(MZF1)to regulate PRSS3 transcription.The garlic-derived compound diallyl trisulfide cooperated with 5-aza-2'-deoxycytidine to exert antitumor effects in lung adenocarcinoma cells through site-specific iCpGI demethylation specifically allowing MZF1 to upregulate PRSS3-V3 expression.Epigenetic silencing of PRSS3-V3 via i CpGI methylation(iCpGIm)in BALF and tumor tissues was associated with early clinical progression in patients with lung cancer but not in those with squamous cell carcinoma or inflammatory disease.Thus,UHRF1/DNMT1—MZF1 axismodulated site-specific iCpGIm regulates divergent expression of PRSS3-SVs,conferring nongenetic functional ITH,with implications for early detection of lung cancer and targeted therapies.Shuye Lin Hanli Xu Lin Qin Mengdi Pang Ziyu Wang Meng Gu Lishu Zhang Cong Zhao Xuefeng Hao Zhiyun Zhang Weimin Ding Jianke Ren Jiaqiang Huang 2023Acta Pharmaceutica Sinica B2023,13,5:0
