1Tumor cell membrane-camouflaged responsive nanoparticles enable MRI-guided immuno-chemodynamic therapy of orthotopic osteosarcoma显示文摘Osteosarcoma is a refractory bone disease in young people that needs the updating and development of effective treatment.Although nanotechnology is widely applied in cancer therapy,poor targeting and inadequate effi-ciency hinder its development.In this study,we prepared alendronate(ALD)/K7M2 cell membranes-coated hollow manganese dioxide(HMnO_(2))nanoparticles as a nanocarrier to load Ginsenoside Rh2(Rh2)for Mag-netic Resonance imaging(MRI)-guided immuno-chemodynamic combination osteosarcoma therapy.Subse-quently,the ALD and K7M2 cell membranes were successively modified on the surface of HMnO_(2) and loaded with Rh2.The tumor microenvironment(TME)-activated Rh2@HMnO_(2)-AM nanoparticles have good bone tumor-targeting and tumor-homing capabilities,excellent GSH-sensitive drug release profile and MRI capability,and attractive immuno-chemodynamic combined therapeutic efficiency.The Rh2@HMnO_(2)-AM nanoparticles can effectively trigger immunogenic cell death(ICD),activate CD4^(+)/CD8^(+)T cells in vivo,and upregulate BAX,BCL-2 and Caspase-3 in cellular level.Further results revealed that Rh2@HMnO_(2)-AM enhanced the secretion of IL-6,IFN-γand TNF-αin serum and inhibited the generation of FOXP3^(+)T cells(Tregs)in tumors.Moreover,the Rh2@HMnO_(2)-AM treatment significant restricted tumor growth in-situ tumor-bearing mice.Therefore,Rh2@HMnO_(2)-AM may serve as an effective and bio-friendly nanoparticle platform combined with immuno-therapy and chemodynamic therapy to provide a novel approach to osteosarcoma therapy.Liwen Fu Weiying Zhang Xiaojun Zhou Jingzhong Fu Chuanglong He 2022Bioactive Materials2022,7,11:4
2Reduced β2-GPI is associated with increased platelet aggregation and activation in patients with prolonged isolated thrombocytopenia after allo-HSCT显示文摘We aimed to measure platelet function and its relationship with β2-GPI in prolonged isolated thrombocytopenia(PT) after allogeneic hematopoietic stem cell transplantation(allo-HSCT).Fifty-six patients with PT and 60 allo-HSCT recipients without PT(non-PT controls) were enrolled.Platelet aggregation and activation,β2-GPI and anti-β2-GPI antibody levels,vWF antigen,and vWF activity were analyzed.The effect of β2-GPI on platelet aggregation was also measured ex vivo.Results showed that ADP-induced platelet aggregation significantly increased(39%±7.5% vs.23%±8.5%,P=0.032),and the platelet expression of both CD62 p(33.6%±11.6% vs.8.5%±3.5%,P<0.001) and PAC-1(42.4%±7.6% vs.6.8%±2.2%,P<0.001) was significantly higher in patients with PT than in those without PT.Significantly lower β2-GPI levels(164.2±12 μg m L^-1 vs.234.2±16 μg mL^-1,P<0.001),higher anti-β2-GPI IgG levels(1.78±0.46 U mL^-1 vs.0.94±0.39 U mL^-1,P<0.001),and increased vWF activity(133.06%±30.50% vs.102.17%±25.90%,P<0.001) were observed in patients with PT than in those without PT.Both ADPinduced platelet aggregation(n=116,r^2=-0.5042,P<0.001) and vWF activity(n=116,r^2=-0.2872,P<0.001) were negatively correlated with β2-GPI levels.In summary,our data suggested that platelet aggregation and activation were significantly higher in patients with PT than in those without PT,which might be associated with reduced β2-GPI levels.The reduced β2-GPI levels might be due to the existence of anti-β2-GPI IgG.Haixia Fu Jingzhong Zhao Lanping Xu Kaiyan Liu Yu Wang Huan Chen Wei Han Jingzhi Wang Fengrong Wang Xiaojun Huang Xiaohui Zhang 2019Science China(Life Sciences)2019,62,7:4
3Multi-source Data-driven Identification of Urban Functional Areas:A Case of Shenyang,China显示文摘Urban functional area(UFA)is a core scientific issue affecting urban sustainability.The current knowledge gap is mainly reflected in the lack of multi-scale quantitative interpretation methods from the perspective of human-land interaction.In this paper,based on multi-source big data include 250 m×250 m resolution cell phone data,1.81×105 Points of Interest(POI)data and administrative boundary data,we built a UFA identification method and demonstrated empirically in Shenyang City,China.We argue that the method we built can effectively identify multi-scale multi-type UFAs based on human activity and further reveal the spatial correlation between urban facilities and human activity.The empirical study suggests that the employment functional zones in Shenyang City are more concentrated in central cities than other single functional zones.There are more mix functional areas in the central city areas,while the planned industrial new cities need to develop comprehensive functions in Shenyang.UFAs have scale effects and human-land interaction patterns.We suggest that city decision makers should apply multi-sources big data to measure urban functional service in a more refined manner from a supply-demand perspective.XUE Bing XIAO Xiao LI Jingzhong ZHAO Bingyu FU Bo 2023Chinese Geographical Science2023,33,1:2

5Nanofibers for the Immunoregulation in Biomedical Applications显示文摘Despite great efforts and achievement of nanomaterials in immune-associated diseases,the selection of appropriate nanomaterials and preparation technology remain some challenges and vast room for improvement.Immunotherapy has received tremendous attention throughout the medical process due to its clinical successes with the pathways of immunoactivation or immunosuppression.Recently,fibrous nanomaterials have facilitated advances in tissue repair and cancer treatments owing to the superiority of multi-channel structure,biocompatibility,tunable size and controlled surface modification.The immunoactivation-based nanofibers can potentially deliver functional agents to lesions and further actively promote immunologic intervention.On the contrary,the immunosuppression-based nanofibers prevent the immune system from overreacting through the blockage of critical pathways in vivo.This review summarizes the current application of nanofiber materials in diverse diseases,including cancer therapy,tissue regeneration(cartilage/bone,skin,tendon,nerves),myocardial infarction,psoriasis and organ defects.Some common fabrication technologies of biomedical nanofibers are also introduced.Meanwhile,the existing technical barriers and perspectives are rationally discussed,providing a constructive inspiration for the follow-up basic research and clinical transformation of nanofibers in the vibrant biomedical fields.Liwen Fu Qian Feng Yujie Chen Jingzhong Fu Xiaojun Zhou Chuanglong He 2022Advanced Fiber Materials2022,4,6:0
