1The model of cytokine release syndrome in CAR T-cell treatment for B-cell non-Hodgkin lymphoma显示文摘Chimeric antigen receptor T(CAR T)cell therapy has demonstrated efficacy in the treatment of haematologic malignancies.However,the accompanying adverse events,the most common of which is cytokine release syndrome(CRS),substantially limit its wide application.Due to its unique physiological characteristics,CRS in CAR T-cell treatment for B-cell non-Hodgkin lymphoma(BNHL)may exhibit some special features.Although existing guidelines had greatly promoted the recognition and management of CRS,many recommendations are not fully applicable to B-NHL.Therefore,it is imperative to identify responses that are specific to CRS observed following CAR T treatment for B-NHL.Based on underlying biological processes and known pathophysiological mechanisms,we tentatively propose a new model to illustrate the occurrence and evolution of CAR T-cell-therapy-related CRS in BNHL.In this model,tumour burden and bone marrow suppression are considered determinants of CRS.Novel phenomena after CAR T-cell infusion(such as local inflammatory response)are further identified.The proposed model will help us better understand the basic biology of CRS and recognize and manage it more rationally.Jianshu Wei Yang Liu Chunmeng Wang Yajing Zhang Chuan Tong Guanghai Dai Wei Wang John E.J.Rasko J.Joseph Melenhorst Wenbin Qian Aibin Liang Weidong Han 2020Signal Transduction and Targeted Therapy2020,5,1:2
2Advances in targeted therapy for malignant lymphoma显示文摘The incidence of lymphoma has gradually increased over previous decades,and it ranks among the ten most prevalent cancers worldwide.With the development of targeted therapeutic strategies,though a subset of lymphoma patients has become curable,the treatment of refractory and relapsed diseases remains challenging.Many efforts have been made to explore new targets and to develop corresponding therapies.In addition to novel antibodies targeting surface antigens and small molecular inhibitors targeting oncogenic signaling pathways and tumor suppressors,immune checkpoint inhibitors and chimeric antigen receptor T-cells have been rapidly developed to target the tumor microenvironment.Although these targeted agents have shown great success in treating lymphoma patients,adverse events should be noted.The selection of the most suitable candidates,optimal dosage,and effective combinations warrant further investigation.In this review,we systematically outlined the advances in targeted therapy for malignant lymphoma,providing a clinical rationale for mechanism-based lymphoma treatment in the era of precision medicine.Li Wang Wei Qin Yu-Jia Huo Xiao Li Qing Shi John E.J.Rasko Anne Janin Wei-Li Zhao 2020Signal Transduction and Targeted Therapy2020,5,1:1
