| 1 | Fluorescence regional integration and differential fluorescence spectroscopy for analysis of structural characteristics and proton binding properties of fulvic acid sub-fractions显示文摘Structural characteristics and proton binding properties of sub-fractions(FA3–FA13) of fulvic acid(FA), eluted stepwise by pyrophosphate buffer were examined by use of fluorescence titration combined with fluorescence regional integration(FRI) and differential fluorescence spectroscopy(DFS). Humic-like(H-L) and fulvic-like(F-L) materials, which accounted for more than 80% of fluorescence response, were dominant in five sub-fractions of FA. Based on FRI analysis, except the response of F-L materials in FA9 and FA13, maximum changes in percent fluorescence response were less than 10% as pH was increased from 2.5 to 11.5.Contents of carboxylic and phenolic groups were compared for fluorescence peaks of FA sub-fractions based on pH-dependent fluorescence derived from DFS. Static quenching was the dominant mechanism for binding of protons by FA sub-fractions. Dissociation constants(p Ka) were calculated by use of results of DFS and the modified Stern-Volmer relationship. The p Kaof H-L, F-L, tryptophan-like and tyrosine-like materials of FA subfractions exhibited ranges of 3.17–4.06, 3.12–3.97, 4.14–4.45 and 4.25–4.76, respectively, for acidic pHs. At basic pHs, values of p Ka for corresponding materials were in ranges of 9.71–10.24, 9.62–10.99, 9.67–10.31 and 9.33–10.28, respectively. At acidic pH, protein-like(P-L)materials had greater affinities for protons than did either H-L or F-L materials. The dicarboxylic and phenolic groups were likely predominant sites of protonation for both H-L and F-L materials at both acidic and basic pHs. | 2 | Design and simulation of self-powered radio frequency identification (RFID) tags for mobile temperature monitoring显示文摘This paper presents a design for a self-powered radio frequency identification (RFID) tag with a thin film bulk acoustic reso- nating piezoelectric power supply (PPS), which can be used for portable remote temperature monitoring. We call this system a PPS-RFID for short. The RFID systems have been found to have many applications in the internet of things (IOT) in the past decade. But semi-active RFID tags require an onboard battery which limits their applications in many fields. For these reasons, our research focuses on power sources for the RFID tags. This paper emphasizes the circuit design and simulation of PPS. In our tests, 0.283 mW was generated by PPS at 1 Hz vibration by a 650 N impact force. The results showed that the integrated PPS could supply sufficient power for the designed PPS-RFID tag. | 5 | Haploid Induction via In vitro Gynogenesis in Tomato(Solanum lycopersicum L.)显示文摘In order to determine the potential for haploid induction via in vitro gynogenesis in tomato, the ovules and protoplasts of embryo sacs from the hybrids Zhongza 101 and Zhongza 105 were cultured. An efficient method of ovule isolation was established in this study. Using this method, 100-150 ovules could be isolated from one ovary. Isolated ovules were cultured on three induction media to induce gynogenesis in vitro. During culture, ovules were enlarged markedly, with opaque white color. When observed microscopically, there were cell divisions and cell clumps in embryo sacs. Subsequently, the cell clumps in embryo sacs ceased growth, likely because the integument grew faster than embryo sacs did and hindered the further development of embryo sacs. Therefore, subsequent callus morphogenesis might be originated from the integument. Thousands of calli from the two tomato varieties were obtained. Five diploid plants were regenerated after 15 months of subculturing. To eliminate the hindering effect of integument on embryo sac cells, the protoplasts of embryo sacs were prepared and cultured. After 48 hours of culture, the protoplasts of embryo sacs doubled in size and gradually formed clusters of cells. | 8 | Berberine alleviates visceral hypersensitivity in rats by altering gut microbiome and suppressing spinal microglial activation显示文摘Accumulating evidence shows that agents targeting gut dysbiosis are effective for improving symptoms of irritable bowel syndrome(IBS).However,the potential mechanisms remain unclear.In this study we investigated the effects of berberine on the microbiota-gut-brain axis in two rat models of visceral hypersensitivity,i.e.,specific pathogen-free SD rats subjected to chronic water avoidance stress(WAS)and treated with berberine(200 mg·kg^(−1)·d^(−1),ig,for 10 days)as well as germ-free(GF)rats subjected to fecal microbiota transplantation(FMT)from a patient with IBS(designated IBS-FMT)and treated with berberine(200 mg·kg^(−1)·d^(−1),ig,for 2 weeks).Before the rats were sacrificed,visceral sensation and depressive behaviors were evaluated.Then colonic tryptase was measured and microglial activation in the dorsal lumbar spinal cord was assessed.The fecal microbiota was profiled using 16S rRNA sequencing,and short chain fatty acids(SCFAs)were measured.We showed that berberine treatment significantly alleviated chronic WAS-induced visceral hypersensitivity and activation of colonic mast cells and microglia in the dorsal lumbar spinal cord.Transfer of fecal samples from berberine-treated stressed donors to GF rats protected against acute WAS.FMT from a patient with IBS induced visceral hypersensitivity and pro-inflammatory phenotype in microglia,while berberine treatment reversed the microglial activation and altered microbial composition and function and SCFA profiles in stools of IBS-FMT rats.We demonstrated that berberine did not directly influence LPS-induced microglial activation in vitro.In both models,several SCFA-producing genera were enriched by berberine treatment,and positively correlated to the morphological parameters of microglia.In conclusion,activation of microglia in the dorsal lumbar spinal cord was involved in the pathogenesis of IBS caused by dysregulation of the microbiota–gut–brain axis,and the berberine-altered gut microbiome mediated the modulatory effects of the agent on microglial activation and visceral hypersensitivity,providing a potential option for the treatment of IBS. | Jin-dong Zhang Jiao Liu Shi-wei Zhu Yuan Fang Ben Wang Qiong Jia Hui-feng Hao John YKao Qi-hua He Li-jin Song Fei Liu Bao-li Zhu Chung Owyang Li-ping Duan | 2021 | Acta Pharmacologica Sinica2021,42,11: | 2 |
| 9 | Magnetic resonance studies of jets in a gas-solid fluidised bed显示文摘Magnetic resonance imaging (MRI) has been used to study the behaviour of jets at the distributor of a 50 mm diameter fluidised bed of 0.5 mm diameter poppy seeds. Two perforated-plate distributors were examined, containing either 10 or 14 holes, each 1 mm diameter. Ultra-fast MR imaging was able to show the transient nature of the upper parts of the jets, where discrete bubbles are formed. Imaging in 3D showed that the central jets were the longest for flow rates below minimum fluidisation. Above minimum fluidisation, the outer jets, nearest the wall of the fluidised bed, arched inward towards the central axis. In this latter case, interpretation of the time-averaged 3D image required the use of ultra-fast MR imaging to identify the approximate height above the distributor at which discrete bubbles were formed. The apparently continuous void extending along the central axis above this height in the time-averaged 3D image was thus identified, using ultra-fast MR imaging, as representing the averaged paths of released bubbles. Time-averaged MR velocity mapping was also used to identify dead zones of stationary particles resting on the distributor between the jets. The dead zones could be observed when the superficial velocity of the gas approached minimum fluidisation, but they were smaller than those observed at lower gas superficial velocity. Comparable images of a single jet through 1.2 mm diameter poppy seeds from MRI and electrical capacitance volume tomography (ECVT) are also demonstrated. | Meenal Pore Thusara C. | 10 | Excitation of extremely low-frequency chorus emissions: The role of background plasma density显示文摘Low-frequency chorus emissions have recently attracted much attention due to the suggestion that they may play important roles in the dynamics of the Van Allen Belts.However, the mechanism(s) generating these low-frequency chorus emissions have not been well understood..In this letter, we report an interesting case in which background plasma density lowered the lower cutoff frequency of chorus emissions from above 0.1 f_(ce)(typical ordinary chorus) to 0.02 f_(ce)(extremely low-frequency chorus).Those extremely low-frequency chorus waves were observed in a rather dense plasma, where the number density N_e was found to be several times larger than has been associated with observations of ordinary chorus waves.For suprathermal electrons whose free energy is supplied by anisotropic temperatures, linear growth rates(calculated using in-situ plasma parameters measured by the Van Allen Probes) show that whistler mode instability can occur at frequencies below 0.1 f_(ce) when the background plasma density N_e increases.Especially when N_e reaches 90 cm–3 or more, the lowest unstable frequency can extend to 0.02 f_(ce) or even less, which is consistent with satellite observations.Therefore, our results demonstrate that a dense background plasma could play an essential role in the excitation of extremely lowfrequency chorus waves by controlling the wave growth rates. | XiongDong Yu ZhiGang Yuan ShiYong Huang Fei Yao Zheng Qiao John R.Wygant Herbert O.Funsten | 2019 | Earth and Planetary Physics2019,3,1: | 2 |
