16Functional Inferences of Environmental Coccolithovirus Biodiversity显示文摘The cosmopolitan calcifying alga Emiliania huxleyi is one of the most abundant bloom forming coccolithophore species in the oceans and plays an important role in global biogeochemical cycling.Coccolithoviruses are a major cause of coccolithophore bloom termination and have been studied in laboratory,mesocosm and open ocean studies.However,little is known about the dynamic interactions between the host and its viruses,and less is known about the natural diversity and role of functionally important genes within natural coccolithovirus communities.Here,we investigate the temporal and spatial distribution of coccolithoviruses by the use of molecular fingerprinting techniques PCR,DGGE and genomic sequencing.The natural biodiversity of the virus genes encoding the major capsid protein(MCP)and serine palmitoyltransferase(SPT)were analysed in samples obtained from the Atlantic Meridional Transect(AMT),the North Sea and the L4 site in the Western Channel Observatory.We discovered nine new coccolithovirus genotypes across the AMT and L4 site,with the majority of MCP sequences observed at the deep chlorophyll maximum layer of the sampled sites on the transect.We also found four new SPT gene variations in the North Sea and at L4.Their translated fragments and the full protein sequence of SPT from laboratory strains EhV-86 and EhV-99B1 were modelled and revealed that the theoretical fold differs among strains.Variation identified in the structural distance between the two domains of the SPT protein may have an impact on the catalytic capabilities of its active site.In summary,the combined use of‘standard’markers(i.e.MCP),in combination with metabolically relevant markers(i.e.SPT)are useful in the study of the phylogeny and functional biodiversity of coccolithoviruses,and can provide an interesting intracellular insight into the evolution of these viruses and their ability to infect and replicate within their algal hosts.Jozef I Nissimov Mark Jones Johnathan A Napier Colin B Munn Susan A Kimmance Michael J Allen 2013Virologica Sinica2013,28,5:1
17Degradable magnesium-hydroxyapatite interpenetrating phase composites processed by current assisted metal infiltration in additive-manufactured porous preforms显示文摘This work explores ceramic additive manufacturing in combination with liquid metal infiltration for the production of degradable interpenetrating phase magnesium/hydroxyapatite(Mg/HA) composites. Material extrusion additive manufacturing was used to produce stoichiometric,and calcium deficient HA preforms with a well-controlled open pore network, allowing the customization of the topological relationship of the composite. Pure Mg and two different Mg alloys were used to infiltrate the preforms by means of an advanced liquid infiltration method inspired by spark plasma sintering, using a novel die design to avoid the structural collapse of the preform. Complete infiltration was achieved in 8 min, including the time for the Mg melting. The short processing time enabled to restrict the decomposition of HA due to the reducing capacity of liquid Mg. The pure Mg-base composites showed compressive yield strength above pure Mg in cast state. Mg alloy-based composites did not show higher strength than the bare alloys due to grain coarsening, but showed similar mechanical properties than other Mg/HA composites that have significantly higher fraction of metallic phase. The composites showed faster degradation rate under simulated body conditions than the bare metallic component due to the formation of galvanic pairs at microstructural level. Mg dissolved preferentially over HA leaving behind a scaffold after a prolonged degradation period. In turn, the fast production of soluble degradation products caused cell metabolic changes after 24 h of culture with not-diluted material extracts. 18Corrosion Resistance of AZ91 Magnesium Alloy after Laser Remelting Treatment显示文摘The main objective of the study was the modification of the surface layer of magnesium alloy by the CO_2 laser. The studied material was the commercial AZ91 magnesium alloy. The effectiveness of the alternations caused by the remelting process was verified on the basis of microscopic observation and corrosion investigations, i e, recording of potentiodynamic polarization curves, electrochemical noise measurements and hydrogen evolution rate measurements. For the adopted range of the treatment parameters, favourable changes were observed in the surface layer such as the refinement of structure and more uniform arrangement of individual phases. As a consequence of those favourable structural changes the improvement of the corrosion resistance of the alloy was achieved in comparison to its non-remelted equivalent. For the treated material corrosion rates expressed as corrosion current densities were at least three times lower than the appropriate values for the untreated alloy comparing them for the same period of samples immersion in the test solution.
