Methods: In this study, 205 cases of PMOP were divided into the PMOP Shen-yin deficiency group(Group A), PMOP Shen-yang deficiency group(Group B), PMOP without Shen deficiency group(Group C), and control group(Group N). Real-time polymerase chain reaction(RT-PCR) and Western blot techniques were used to observe the effects of LDP treatment on the cardiotrophin-like cytokine factor 1(CLCF1), ankyrin repeat and SOCS box containing 1(ASB1), and prokineticin 2(PROK2) genes and the Janus kinase/signal transducer and activator of transcription(JAK/STAT) signaling pathway. Results: The m RNA(P<0.05) and protein(P<0.01) expression levels of the CLCF1 gene in Group A were significantly lower than the corresponding levels in Group N. After LDP treatment for 3 months, the m RNA expression levels of the CLCF1 gene were obviously up-regulated(P<0.01). After 6-month treatment, the expression levels of CLCF1 m RNA and protein were significantly up-regulated(both P<0.01), and the average bone density of the top femur had significantly increased(P<0.05). In vitro, CLCF1 overexpression resulted in a significant increase in the total protein and phosphorylated protein levels of JAK2 and STAT3. | 5 | Spatial transmission and meteorological determinants of tuberculosis incidence in Qinghai Province,China:a spatial clustering panel analysis显示文摘Background:Tuberculosis(TB)is the notifiable infectious disease with the second highest incidence in the Qinghai province,a province with poor primary health care infrastructure.Understanding the spatial distribution of TB and related environmental factors is necessary for developing effective strategies to control and further eliminate TB.Methods:Our TB incidence data and meteorological data were extracted from the China Information System of Disease Control and Prevention and statistical yearbooks,respectively.We calculated the global and local Moran’s I by using spatial autocorrelation analysis to detect the spatial clustering of TB incidence each year.A spatial panel data model was applied to examine the associations of meteorological factors with TB incidence after adjustment of spatial individual effects and spatial autocorrelation.Results:The Local Moran’s I method detected 11 counties with a significantly high-high spatial clustering(average annual incidence:294/100000)and 17 counties with a significantly low-low spatial clustering(average annual incidence:68/100000)of TB annual incidence within the examined five-year period;the global Moran’s I values ranged from 0.40 to 0.58(all P-values<0.05).The TB incidence was positively associated with the temperature,precipitation,and wind speed(all P-values<0.05),which were confirmed by the spatial panel data model.Each 10°C,2 cm,and 1 m/s increase in temperature,precipitation,and wind speed associated with 9%and 3%decrements and a 7%increment in the TB incidence,respectively.Conclusions:High TB incidence areas were mainly concentrated in south-western Qinghai,while low TB incidence areas clustered in eastern and north-western Qinghai.Areas with low temperature and precipitation and with strong wind speeds tended to have higher TB incidences. | Hua-Xiang Rao Xi Zhang Lei Zhao Juan Yu Wen Ren Xue-Lei Zhang Yong-Cheng Ma Yan Shi Bin-Zhong Ma Xiang Wang Zhen Wei Hua-Fang Wang Li-Xia Qiu | 2016 | Infectious Diseases of Poverty2016,5,1: | 10 |

| 7 | Endothelium-Mimicking Multifunctional Coating Modified Cardiovascular Stents via a Stepwise Metal-Catechol-(Amine)Surface Engineering Strategy显示文摘Stenting is currently the major therapeutic treatment for cardiovascular diseases.However,the nonbiogenic metal stents are inclined to trigger a cascade of cellular and molecular events including inflammatory response,thrombogenic reactions,smooth muscle cell hyperproliferation accompanied by the delayed arterial healing,and poor reendothelialization,thus leading to restenosis along with late stent thrombosis.To address prevalence critical problems,we present an endothelium-mimicking coating capable of rapid regeneration of a competently functioning new endothelial layer on stents through a stepwise metal(copper)-catechol-(amine)(MCA)surface chemistry strategy,leading to combinatorial endothelium-like functions with glutathione peroxidase-like catalytic activity and surface heparinization.Apart from the stable nitric oxide(NO)generating rate at the physiological level(2:2×10^(-10) mol/cm^(2)/min lasting for 60 days),this proposed strategy could also generate abundant amine groups for allowing a high heparin conjugation efficacy up to∼1μg/cm^(2),which is considerably higher than most of the conventional heparinized surfaces.The resultant coating could create an ideal microenvironment for bringing in enhanced antithrombogenicity,anti-inflammation,anti-proliferation of smooth muscle cells,re-endothelialization by regulating relevant gene expressions,hence preventing restenosis in vivo.We envision that the stepwise MCA coating strategy would facilitate the surface endothelium-mimicking engineering of vascular stents and be therefore helpful in the clinic to reduce complications associated with stenosis. | Ying Yang Peng Gao Juan Wang Qiufen Tu Long Bai Kaiqin Xiong Hua Qiu Xin Zhao Manfred FMaitz Huaiyu Wang Xiangyang Li Qiang Zhao Yin Xiao Nan Huang Zhilu Yang | 2020 | Research2020,,1: | 8 |
| 8 | NT4(Si)-p53(N15)-antennapedia induces cell death in a human hepatocellular carcinoma cell line显示文摘AIM:To construct the recombinant lentivirus expression plasmid,pLenti6/V5-NT4 p53(N15)-antennapedia(Ant),and study its effect on HepG2 cells.METHODS:Plasmid pLenti6/V5-NT4 p53(N15)-Ant was constructed incorporating the following functional regions,including signal peptide sequence and pro-region of neurotrophin 4,N-terminal residues 12-26 of p53 and 17 amino acid drosophila carrier protein,Ant.Hepatocellular carcinoma(HepG2)cells were used for transfection.3-[4,5-dimethyl-thiazol-2yl]-2,5 diphenyl tetrazolium bromide(MTT)assay,lactate dehydrogenase(LDH)release assay,transmission electron microscopy(TEM)and flow cytometric analysis(FCM)were employed to investigate the effects of LV-NT4(Si)-p53(N15)-Ant in vitro on HepG2 cells.In vivo experiment was also performed to investigate the inhibitory effect of LV-NT4(Si)-p53(N15)-Ant on tumor growth in nude mice.RESULTS:LV-NT4(Si)-p53(N15)-Ant significantly suppressed the growth of HepG2 cells.MTT assay showed that the growth of HepG2 cells was mucj more significantly inhibited by LV-NT4(Si)-p53(N15)-Ant than by LV-EGFP.The inhibition rate for HepG2 cell growth in the two groups was 46.9% and 94.5%,respectively,48 h after infection with LV-NT4(Si)-p53(N15)-Ant,and was 33.9% and 95.8%,respectively,72 h after infection with LV-NT4(Si)-p53(N15)-Ant(P < 0.01).Light microscopy and TEM showed morphological changes in HepG2 cells infected with LV-NT4(Si)-p53(N15)-Ant,but no signif icant changes in HepG2 cells infected with LV-EGFP.Changes were observed in ultra-structure of HepG2 cells infected with LV-NT4(Si)-p53(N15)-Ant,with degraded membranes,resulting in necrosis.LDH release from HepG2 cells was analyzed at 24,48,72 and 96 h after infection with LV-NT4(Si)-p53(N15)-Ant and LV-EGFP,which showed that LDH release was signif icantly higher in LV-NT4(Si)-p53(N15)-Ant treatment group(682 IU/L)than in control group(45 IU/L,P < 0.01).The longer the time was after infection,the bigger the difference was in LDH release.FCM analysis showed that LV-NT4(Si)-p53(N15)-Ant could induce two different kinds of cell death:necrosis and apoptosis,with apoptosis being the minor type and necrosis being the main type,suggesting that LV-NT4(Si)-p53(N15)-Ant exerts its anticancer effect on HepG2 cells by inducing necrosis.The in vivo study showed that LV-NT4(Si)-p53(N15)-Ant significantly inhibited tumor growth with an inhibition rate of 66.14% in terms of tumor size and weight. | Li-Ping Song Yue-Ping Li Ning Wang Wei-Wei Li Juan Ren Shu-Dong Qiu Quan-Ying wang Guang-Xiao Yang | 2009 | World Journal of Gastroenterology2009,15,46: | 8 |
| 9 | Protective effect of Bifidobacterium infantis CGMCC313-2 on ovalbumin-induced airway asthma and b-lactoglobulininduced intestinal food allergy mouse models显示文摘AIM To determine whether oral administration of Bifidobacterium infantis CGMCC313-2(B. infantis CGMCC313-2) inhibits allergen-induced airway inflammation and food allergies in a mouse model.METHODS Ovalbumin(OVA)-induced allergic asthma and b-lactoglobulin-induced food allergy mouse models were used in this study. Following oral administration of B. infantis CGMCC313-2 during or after allergen sensitization, histopathologic changes in the lung and intestine were evaluated by hematoxylin and eosin(HE) staining. In the allergic asthma mouse model, we evaluated the proportion of lung-infiltrating inflammatory cells. OVAspecific IgE and IgG1 levels in serum and cytokine levels in bronchoalveolar lavage fluid(BALF) were also assessed. In the food allergy mouse model, the levels of total Ig E and cytokines in serum were measured.RESULTS Oral administration of B. infantis CGMCC313-2 during or after allergen sensitization suppressed allergic inflammation in lung and intestinal tissues, while the proportion of infiltrating inflammatory cells was significantly decreased in the BALF of allergic asthma mice. Moreover, B. infantis CGMCC313-2 decreased the serum levels of total Ig E in food allergy mice, and reductions in IgE and IgG1 were also observed in OVA-induced allergic asthma mice. | 13 | Comparison of clinical features and stent placement outcomes between airway stenosis caused by primary pulmonary malignancies and that caused by primary non-pulmonary malignancies显示文摘Background: Primary pulmonary malignancies (PPMs) and non-pulmonary malignancies (PNPMs) may result in airway stenosis requiring stenting. This study aimed to compare and evaluate the clinical features and stent placement outcomes of airway stenosis caused by PPMs and PNPMs. Methods: A total of 141 patients with malignant airway stenosis who underwent Micro-Tech stent placements between January 2004 and October 2017 at Department of Respiratory Medicine, Beijing Tian Tan Hospital, Capital Medical University were divided into PPM (n= 100) and PNPM groups (n = 41). Patients’ clinical features and stent placement outcomes were collected and analyzed. Chi-square test was used to compare the categorical variables, while independent- or paired-sample t test was used to compare the continuous variables. Results: There were no significant differences in age, sex, treatment history, respiratory symptoms, and incidence of obstructive pneumonia between groups. Multiple airway involvement (63.0% vs. 31.7%;χ^2=11.459, P= 0.001) and atelectasis (17.0% vs. 2.4%;χ^25.536, P= 0.019) were more common in the PPM group, while extraluminal obstruction (24.4% vs. 6.0%;χ^28.033, P= 0.005) was more common in the PNPM group. Before stenting, the American Thoracic Society Dyspnea Index (ADI) and Karnofsky Performance Scale (KPS) scores showed no significant differences between groups (all P > 0.05). After stenting, a satisfactory rate of symptom improvement was achieved in both groups (98.0% and 100.0% in the PPM and PNPM groups, respectively;χ^2 0.016, P= 0.898);ADI and KPS scores, which showed no significant differences between groups (all P > 0.05), were significantly improved in each group (all P < 0.001). Complications after stenting could be effectively managed using bronchoscopic procedures. Conclusions: Among cases of malignant airway stenosis requiring stenting, those caused by PPM are more likely to involve multiple airways and are associated with atelectasis, while those caused by PNPM are more likely to cause extraluminal obstruction. | 14 | FRUCTOKINASE-LIKE PROTEIN 1 interacts with TRXz to regulate chloroplast development in rice显示文摘Chloroplast genes are transcribed by the plastidencoded RNA polymerase(PEP) or nucleus-encoded RNA polymerase. FRUCTOKINASE-LIKE PROTEINS(FLNs) are phosphofructokinase-B(Pfk B)-type carbohydrate kinases that act as part of the PEP complex; however, the molecular mechanisms underlying FLN activity in rice remain elusive.Previously, we identified and characterized a heat-stress sensitive albino(hsa_1) mutant in rice. Map-based cloning revealed that HSA_1 encodes a putative OsFLN_2. Here, we further demonstrated that knockdown or knockout of the OsFLN_1, a close homolog of HSA_1/OsFLN_2, considerably inhibits chloroplast biogenesis and the fln_1 knockout mutants, created by clustered regularly interspaced short palindromic repeats(CRISPR) and CRISPR-associate protein_9, exhibit severe albino phenotype and seedling lethality. Moreover, OsFLN_1 localizes to the chloroplast.Yeast two-hybrid, pull-down and bimolecular fluorescencecomplementation experiments revealed that OsFLN_1 and HSA_1/OsFLN_2 interact with THIOREDOXINZ(OsTRXz) to regulate chloroplast development. In agreement with this,knockout of OsTRXz resulted in a similar albino and seedling lethality phenotype to that of the fln_1 mutants. Quantitative reverse transcription polymerase chain reaction and immunoblot analysis revealed that the transcription and translation of PEP-dependent genes were strongly inhibited in fln_1 and trxz mutants, indicating that loss of OsFLN_1, HSA_1/OsFLN_2, or OsTRXz function perturbs the stability of the transcriptionally active chromosome complex and PEP activity. | 16 | Improved stability and oral bioavailability of Ganneng dropping pills following transforming lignans of herpeto- spermum caudigerum into nanosuspensions显示文摘The present study was designed to improve storage stability and oral bioavailability of Ganneng dropping pills(GNDP) by transforming lignans of Herpetospermum caudigerum(HL) composed of herpetrione(HPE) and herpetin(HPN) into nanosuspension(HL-NS), the main active ingredient of GNDP, HL-NS was prepared by high pressure homogenization and lyophilized to transform into solid nanoparticles(HL nanoparticles), and then the formulated HL nanoparticles were perfused into matrix to obtain NS-GNDP by melting method. For a period of 3 months, the content uniformity, storage stability and pharmacokinetics test in vivo of NS-GNDP were evaluated and compared with regular GNDP at room temperature. The results demonstrated that uniformity of dosage units of NS-GNDP was acceptable according to the criteria of Chinese Pharmacopoeia 2015 J. Physical stability of NS-GNDP was investigated systemically using photon correlation spectroscopy(PCS), zeta potential measurement, and scanning electron microscopy(SEM). There was a slight increase in particles and PI of HL-NS re-dispersed from NS-GNDP after storage for 3 months, compared with new formulated NS-GNDP, which indicated a good redispersibility of the NS-GNDP containing HL-NS after storage. Besides, chemical stability of NS-GNDP was studied and the results revealed that HPE and HPN degradation was less when compared with that of GNDP, providing more than 99% of drug residue after storage for 3 months. In the dissolution test in vitro, NS-GNDP remarkably exhibited an increased dissolution velocity compared with GNDP and no distinct dissolution difference existed within 3 months. The pharmacokinetic study showed that HPE and HPN in NS-GNDP exhibited a significant increase in AUC0–t, Cmax and decrease in Tmax when compared with regular GNDP.
