| 2 | Moxibustion eases chronic inflammatory visceral pain through regulating MEK, ERK and CREB in rats显示文摘AIM To investigate the effects of herb-partitioned moxibustion(HPM) on phosphorylation of mitogen-activated extracellular signal-regulated kinase(MEK)1, extracellular signal-regulated kinase(ERK)1/2 and c AMP response element binding protein(CREB) in spinal cord of rats with chronic inflammatory visceral pain(CIVP), and to explore the central mechanism of HPM in treating CIVP.METHODS Male Sprague-Dawley rats were randomized into normal, model, HPM, sham-HPM, MEK-inhibitor and dimethyl sulfoxide(DMSO) groups. The CIVP model was established using an enema mixture of trinitrobenzene sulfonic acid and ethanol. HPM was applied at bilateral Tianshu(ST25) and Qihai(CV6) acupoints in the HPM group, while in the sham-HPM group, moxa cones and herb cakes were only placed on the same points but not ignited. The MEK-inhibitor and DMSO groups received L5-L6 intrathecal injection of U0126 and 30% DMSO, respectively. Abdominal withdrawal reflex(AWR), mechanical withdrawal threshold(MWT) and thermal withdrawal latency(TWL) were applied for the assessment of pain behavior. The colonic tissue was observed under an optical microscope after hematoxylin-eosin staining. Expression of phosphor(p)MEK1, p ERK1/2 and p CREB in rat spinal cord was detected using Western blotting. The levels of MEK, ERK and CREB m RNA in rat spinal cord were detected using real-time polymerase chain reaction. RESULTS Compared with the normal group, the AWR scores were increased significantly(P < 0.01) and the MWT and TWL scores were decreased significantly(P < 0.05) in the model, sham-HPM and DMSO groups. Compared with the model group, the AWR scores were decreased significantly(P < 0.01) and the MWT and TWL scores were increased significantly in the HPM and MEK-inhibitor groups(P < 0.05). Compared with the sham-HPM and DMSO groups, the AWR scores were decreased significantly(P < 0.01) and the MWT and TWL scores were increased significantly(P < 0.05) in the HPM and MEK-inhibitor groups. Compared with the normal group, the expression of p MEK1, p ERK1/2 and p CREB proteins and the levels of MEK, ERK and CREB m RNA in rat spinal cord were increased significantly in the model, sham-HPM and DMSO groups(P < 0.01 or < 0.05). Compared with the model group, the expression of p MEK1, p ERK1/2 and p CREB proteins and the levels of MEK, ERK and CREB m RNA in rat spinal cord were reduced significantly in the HPM and MEK-inhibitor groups(P < 0.01 or < 0.05). Compared with the sham-HPM and DMSO groups, expression of p MEK1, p ERK1/2 and p CREB proteins and the levels of MEK, ERK and CREB m RNA in rat spinal cord were reduced significantly in the HPM and MEK-inhibitor groups(P < 0.01 or < 0.05). | 3 | Herb-partitioned moxibustion alleviates colon injuries in ulcerative colitis rats显示文摘AIM To observe the effect of herb-partitioned moxibustion(HPM) on expression of colonic cytokines in ulcerative colitis(UC) rats.METHODS A UC rat model was established by protein immunization in combination with topical chemical stimulation.Rats in the HPM group(n = 8) received HPM at bilateral Tianshu(ST25) points.The gross injury and pathological scores of the colon were recorded.The expression profile of colonic cytokines was assayed using the protein microarray technique.Specific differential cytokines were selected and verified by ELISA.The corresponding Uni Prot Accessions of the differentially expressed cytokines were retrieved in the Uni Prot database.The pathways involved were analyzed with the help of the KEGG PATHWAY database.The DAVID database was used for functional cluster and pathway analysis.RESULTS HPM improved colon injuries in UC rats,manifested by accelerated repair of ulcers and alleviation of inflammation,and the gross injury and pathological scores both significantly decreased(P < 0.01).Fold change > 1.3 or < 0.77 was taken as the screening standard.There were 77 down-regulated and 9 up-regulated differentially expressed colonic cytokines in the HPM group compared with the model group,and expression of 20 differed significantly(P < 0.05).Twelve of the 20 significantly differentially expressed cytokines [β-catenin,interleukin-1 receptor 6(IL-1 R6),IL-1β,B7-1,nerve growth factor receptor,AMP-activated protein kinase-α1,neuropilin-2,orexin A,adipocyte differentiation-related protein,IL-2,Fas and Fas L] were up-regulated in the model group(n = 3,compared with the normal group) but downregulated in the HPM group(n = 3,compared with the model group).Functional cluster analysis showed that the differentially expressed colonic cytokines in the HPM group regulated apoptosis and protein phosphorylation.KEGG pathway analysis showed that 52 down-regulated and 7 up-regulated differentially expressed colonic cytokines in the HPM group had pathways.The pathways that interacted between the cytokines and their receptors accounted for the largest proportion(28 of the downregulated and 5 of the up-regulated cytokines).CONCLUSION HPM promotes the repair of colon injuries in UC rats,which is related to the regulation of several abnormally expressed cytokines. | Dan Zhang Yan-Bo Ren Kai Wei Jue Hong Yan-Ting Yang Li-Jie Wu Ji Zhang Zheng Shi Huan-Gan Wu Xiao-Peng Ma | 2018 | World Journal of Gastroenterology2018,24,30: | 11 |
| 4 | ERK1/2 Activities Are Dispensable for Oocyte Growth but Are Required for Meiotic Maturation and Pronuclear Formation in Mouse显示文摘Previous studies revealed that extracellular regulated kinase-1 and-2(ERK1/2) cascade plays pivotal roles in regulating oocyte meiotic cell cycle progression. However, most knowledge about the in vivo function of ERK1/2 in mammalian oocytes was indirectly obtained from analyzing the phenotypes of Mos knockout mice. In this study, we knocked out Erk1 and Erk2 in mouse oocytes as early as the primordial follicle stage using the well-characterized Gdf9-Cre mouse model, and for the first time directly investigated the in vivo function of ERK1/2 in mouse oocytes. In this novel mouse model, we observed that ERK1/2 activities in oocyte are dispensable for primordial follicle maintenance,activation and follicle growth. Different from the Mos null oocytes, the ERK1/2-deleted oocytes had well-assembled spindles at metaphase Ⅰ(MⅠ), extruded polar body-1(PB1) with normal sizes, and did not undergo a full parthenogenetic activation characterized for pronuclear formation. However, the ovulated ERK1/2-deleted oocytes had poorly-assembled metaphase Ⅱ(MⅡ) spindles, spontaneously released polar body-2(PB2), and were arrested at another metaphase called metaphase Ⅲ(MⅢ). In addition, ERK1/2 deletion prevented male pronuclear formation after fertilization, and caused female infertility. | 5 | Modulation Recognition in Maritime Multipath Channels:A Blind Equalization-Aided Deep Learning Approach显示文摘Modulation recognition has been long investigated in the literature,however,the performance could be severely degraded in multipath fading channels especially for high-order Quadrature Amplitude Modulation(QAM)signals.This could be a critical problem in the broadband maritime wireless communications,where various propagation paths with large differences in the time of arrival are very likely to exist.Specifically,multiple paths may stem from the direct path,the reflection paths from the rough sea surface,and the refraction paths from the atmospheric duct,respectively.To address this issue,we propose a novel blind equalization-aided deep learning(DL)approach to recognize QAM signals in the presence of multipath propagation.The proposed approach consists of two modules:A blind equalization module and a subsequent DL network which employs the structure of ResNet.With predefined searching step-sizes for the blind equalization algorithm,which are designed according to the set of modulation formats of interest,the DL network is trained and tested over various multipath channel parameter settings.It is shown that as compared to the conventional DL approaches without equalization,the proposed method can achieve an improvement in the recognition accuracy up to 30%in severe multipath scenarios,especially in the high SNR regime.Moreover,it efficiently reduces the number of training data that is required. | Xuefei Ji Jue Wang Ye Li Qiang Sun Chen Xu | 2020 | China Communications2020,17,3: | 4 |

| 7 | Cytokinesis and cancer:Polo loves ROCK'n' Rho(A)显示文摘Cytokinesis is the last step of the M (mitosis) phase, yet it is crucial for the faithful division of one cell into two. Cytokinesis failure is often associated with cancer. Cytokinesis can be morphologically divided into four steps: cleavage furrow initiation, cleavage furrow ingression, midbody formation and abscission. Molecular studies have revealed that RhoA as well as its regulators and effectors are important players to ensure a successful cytokinesis. At the same time, Polo-like kinase 1 (Plk1) is an important kinase that can target many substrates and carry out different functions during mitosis, including cytokinesis. Recent studies are beginning to unveil a closer tie between Plk1 and RhoA networks. More specifically, Plk1 phosphorylates the centralspindlin complex Cyk4 and MKLP1/CHO1, thus recruiting RhoA guanine nucleotide-exchange factor (GEF) Ect2 through its phosphopeptide-binding BRCT domains. Ect2 itself can be phosphorylated by Plk1 in vitro. Plk1 can also phosphorylate another GEF MyoGEF to regulate RhoA activity. Once activated, RhoA-GTP will activate downstream effectors, including ROCK1 and ROCK2. ROCK2 is among the proteins that associate with Plk1 Polo-binding domain (PBD) in a large proteomic screen, and Plk1 can phosphorylate ROCK2 in vitro. | 8 | Altered relationship between thickness and intrinsic activity amplitude in generalized tonic–clonic seizures显示文摘A thinner cortex has higher potential for binding GABA receptor A which is associated with larger amplitudes of intrinsic brain activity(i BA). However, the relationship between cortical thickness and i BA is unknown in intact and epileptic brains. To this end, we investigated the relationship between cortical thickness measured by highresolution MRI and surface-based i BA derived from resting-state functional MRI in normal controls(n = 82) andpatients with generalized tonic–clonic seizures(GTCS)only(n = 82). We demonstrated that the spatial distribution of cortical thickness negatively correlated with surface-based i BA amplitude at both whole-brain and within independent brain functional networks. In GTCS patients,spatial coupling between thickness and i BA amplitude decreased in the default mode, dorsal attention, and somatomotor networks. In addition, the vertex-wise acrosssubject thickness–i BA amplitude correspondence altered in the frontal and temporal lobes as well as in the precuneus in GTCS patients.
