1Paris saponin Ⅶ,a direct activator of AMPK,induces autophagy and exhibits therapeutic potential in non-small-cell lung cancer显示文摘Paris saponinⅦ(PSⅦ),a bioactive constituent extracted from Trillium tschonoskii Maxim.,is cytotoxic to several cancer types.This study was designed to explore whether PSⅦprevents non-small-cell lung cancer(NSCLC)proliferation and to investigate its molecular target.AMP-activated protein kinase(AMPK)has been implicated in the activation of autophagy in distinct tissues.In cultured human NSCLC cell lines,PSⅦinduces autophagy by activating AMPK and inhibiting m TOR signaling.Furthermore,PSⅦ-induced autophagy activation was reversed by the AMPK inhibitor compound C.Computational docking analysis showed that PSⅦdirectly interacted with the allosteric drug and metabolite site of AMPK to stabilize its activation.Microscale thermophoresis assay and drug affinity responsive target stability assay further confirmed the high affinity between PSⅦand AMPK.In summary,PSⅦacts as a direct AMPK activator to induce cell autophagy,which inhibits the growth of NSCLC cells.In the future,PSⅦtherapy should be applied to treat patients with NSCLC.XIANG Yu-Chen SHEN Jie SI Yuan LIU Xue-Wen ZHANG Liang WEN Jun ZHANG Te YU Qing-Qing LU Jun-Fei XIANG Ke LIU Ying 2021Chinese Journal of Natural Medicines2021,19,3:4

3Multifocal intraductal papillary mucinous neoplasm of the pancreas-A case report显示文摘Cystic neoplasms of the pancreas are relatively rare, comprising 10 percent of pancreatic cysts and only 1 percent of pancreatic cancers. Cystic neoplasms include mucinous cystic neoplasms, serous cystadenomas, papillary cystic tumors, cystic islet cell tumors and intraductal papillary mucinous neoplasms of the pancreas (IPMNs). IPMN was first described in 1982. It has been most commonly described in 60 to 70 years old males, and represents a relatively ''new'' but increasingly recognized disease. The improvement and widespread use of modern imaging equipments and heightened awareness of physicians contribute to the increasing incidence of IPMN. The majority of IPMNs are located in the pancreatic head (75%) while the rest involves the body/tail regions. Multifocal IPMNs have been hypothesized, but the true presence of multifocality is unknown. Here we present a 72-yearold male diagnosed with IPMN (carcinoma in situ ) in the pancreatic head and a branch duct type IPMN (duct atypia) in the pancreatic body and tail. The patient underwent a Whipple intervention and a distal pancreatectomy. 11Induction of a 55 kDa acetylcholinesterase protein during apoptosis and its negative regulation by the Akt pathway显示文摘Acetylcholinesterase(AChE)is emerging as an important contributor to apoptosis in various cell types.However,overexpression of AChE does not initiate apoptosis,and cells which express AChE at basal levels grow normally,suggesting that AChE may function differently between normal and apoptotic conditions.In this study,we determined that an AChE-derived protein(∼55 kDa)positively correlated with cellular apoptotic levels.The 55 kDa AChE protein was not a result of a novel splice variant of the AChE primary transcript.Instead,it was determined to be a cleaved fragment of the full-length 68 kDa AChE protein that could not be inhibited by cycloheximide(CHX)but could be suppressed by caspase inhibitors in apoptotic PC-12 cells.Furthermore,activation of the Akt cascade abolished the 55 kDa protein,and both AChE protein forms(68 and 55 kDa)accumulated in the nucleus during apoptosis.In a mouse model for ischemia/reperfusion(I/R)-induced acute renal failure,the 55 kDa AChE protein was detected in the impaired organs but not in the normal ones,and its levels correlated with the genotype of the mice.In summary,a 55 kDa AChE protein resulting from the cleavage of 68 kDa AChE is induced during apoptosis,and it is negatively regulated by the Akt pathway.This study suggests that an alternative form of AChE may play a role in apoptosis.Jing Xie Hua Jiang Yi-Han Wan Ai-Ying Du Kai-Jie Guo Te Liu Wei-Yuan Ye Xin Niu Jun Wu Xiao-Qin Dong Xue-Jun Zhang 2011Journal of Molecular Cell Biology2011,3,4:1
