| 1 | Whole-genome,transcriptome,and methylome analyses provide insights into the evolution of platycoside biosynthesis in Platycodon grandiflorus,a medicinal plant显示文摘Triterpenoid saponins(TSs)are common plant defense phytochemicals with potential pharmaceutical properties.Platycodon grandiflorus(Campanulaceae)has been traditionally used to treat bronchitis and asthma in East Asia.The oleanane-type TSs,platycosides,are a major component of the P.grandiflorus root extract.Recent studies show that platycosides exhibit anti-inflammatory,antiobesity,anticancer,antiviral,and antiallergy properties.However,the evolutionary history of platycoside biosynthesis genes remains unknown.In this study,we sequenced the genome of P.grandiflorus and investigated the genes involved in platycoside biosynthesis.The draft genome of P.grandiflorus is 680.1 Mb long and contains 40,017 protein-coding genes.Genomic analysis revealed that the CYP716 family genes play a major role in platycoside oxidation.The CYP716 gene family of P.grandiflorus was much larger than that of other Asterid species.Orthologous gene annotation also revealed the expansion ofβ-amyrin synthases(bASs)in P.grandiflorus,which was confirmed by tissue-specific gene expression.In these expanded gene families,we identified key genes showing preferential expression in roots and association with platycoside biosynthesis.In addition,wholegenome bisulfite sequencing showed that CYP716 and bAS genes are hypomethylated in P.grandiflorus,suggesting that epigenetic modification of these two gene families affects platycoside biosynthesis.Thus whole-genome,transcriptome,and methylome data of P.grandiflorus provide novel insights into the regulation of platycoside biosynthesis by CYP716 and bAS gene families. | Jungeun Kim Sang-Ho Kang Sin-Gi Park Tae-Jin Yang Yi Lee Ok Tae Kim Oksung Chung Jungho Lee Jae-Pil Choi Soo-Jin Kwon Keunpyo Lee Byoung-Ohg Ahn Dong Jin Lee Seung-il Yoo In-Gang Shin Yurry Um Dae Young Lee Geum-Soog Kim Chang Pyo Hong Jong Bhak Chang-Kug Kim | 2020 | Horticulture Research2020,7,1: | 2 |
| 2 | A simple, flexible and high-throughput cloning system for plant genome editing via CRISPR-Cas system显示文摘CRISPR-Cas9 system is now widely used to edit a target genome in animals and plants. Cas9 protein derived from Streptococcus pyogenes(Sp Cas9) cleaves double-stranded DNA targeted by a chimeric single-guide RNA(sg RNA). For plant genome editing, Agrobacterium-mediated T-DNA transformation has been broadly used to express Cas9 proteins and sg RNAs under the control of Ca MV 35 S and U6/U3 promoter, respectively. We here developed a simple and high-throughput binary vector system to clone a 19 20 bp of sg RNA, which binds to the reverse complement of a target locus, in a large T-DNA binary vector containing an Sp Cas9 expressing cassette. Twostep cloning procedures:(1) annealing two target-specific oligonucleotides with overhangs specific to the Aar I restriction enzyme site of the binary vector; and(2) ligating the annealed oligonucleotides into the two Aar I sites of the vector, facilitate the high-throughput production of the positive clones. In addition, Cas9-coding sequence and U6/U3 promoter can be easily exchanged via the GatewayTMsystem and unique Eco RI/Xho I sites on the vector, respectively. We examined the mutation ratio and patterns when we transformed these constructs into Arabidopsis thaliana and a wild tobacco, Nicotiana attenuata. | 3 | Enhancing Hydrophilicity of Thick Electrodes for High Energy Density Aqueous Batteries显示文摘Thick electrodes can substantially enhance the overall energy density of batteries.However,insufficient wettability of aqueous electrolytes toward electrodes with conventional hydrophobic binders severely limits utilization of active materials with increasing the thickness of electrodes for aqueous batteries,resulting in battery performance deterioration with a reduced capacity.Here,we demonstrate that controlling the hydrophilicity of the thicker electrodes is critical to enhancing the overall energy density of batteries.Hydrophilic binders are synthesized via a simple sulfonation process of conventional polyvinylidene fluoride binders,considering physicochemical properties such as mechanical properties and adhesion.The introduction of abundant sulfonate groups of binders(i)allows fast and sufficient electrolyte wetting,and(ii)improves ionic conduction in thick electrodes,enabling a significant increase in reversible capacities under various current densities.Further,the sulfonated binder effectively inhibits the dissolution of cathode materials in reactive aqueous electrolytes.Overall,our findings significantly enhance the energy density and contribute to the development of practical zinc-ion batteries. | Jungeun Lee Hyeonsoo Lee Cheol Bak Youngsun Hong Daeha Joung Jeong Beom Ko Yong Min Lee Chanhoon Kim | 2023 | Nano-Micro Letters2023,15,7: | 2 |

| 15 | Tomato expressing Arabidopsis glutaredoxin gene AtGRXS17 confers tolerance to chilling stress via modulating cold responsive components显示文摘Chilling stress is a production constraint of tomato,a tropical origin,chilling-sensitive horticultural crop.The development of chilling tolerant tomato thus has significant potential to impact tomato production.Glutaredoxins(GRXs)are ubiquitous oxidoreductases,which utilize the reducing power of glutathione to reduce disulfide bonds of substrate proteins and maintain cellular redox homeostasis.Here,we report that tomato expressing Arabidopsis GRX gene AtGRXS17 conferred tolerance to chilling stress without adverse effects on growth and development.AtGRXS17-expressing tomato plants displayed lower ion leakage,higher maximal photochemical efficiency of photosystem II(Fv/Fm)and increased accumulation of soluble sugar compared with wild-type plants after the chilling stress challenge.Furthermore,chilling tolerance was correlated with increased antioxidant enzyme activities and reduced H2O2 accumulation.At the same time,temporal expression patterns of the endogenous C-repeat/DRE-binding factor 1(SlCBF1)and CBF mediated-cold regulated genes were not altered in AtGRXS17-expressing plants when compared with wild-type plants,and proline concentrations remained unchanged relative to wild-type plants under chilling stress.Green fluorescent protein-AtGRXS17 fusion proteins,which were initially localized in the cytoplasm,migrated into the nucleus during chilling stress,reflecting a possible role of AtGRXS17 in nuclear signaling of chilling stress responses.Together,our findings demonstrate that genetically engineered tomato plants expressing AtGRXS17 can enhance chilling tolerance and suggest a genetic engineering strategy to improve chilling tolerance without yield penalty across different crop species. | Ying Hu Qingyu Wu Stuart A Sprague Jungeun Park Myungmin Oh C B Rajashekar Hisashi Koiwa Paul A Nakata Ninghui Cheng Kendal D Hirschi Frank F White Sunghun Park | 2015 | Horticulture Research2015,2,1: | 1 |
