| 1 | Phytophthora Effectors Modulate Genome-wide Alternative Splicing of Host mRNAs to Reprogram Plant Immunity显示文摘Alternative splicing(AS)of pre-mRNAs increases transcriptome and proteome diversity,regulates gene expression through multiple mechanisms,and plays important roles in plant development and stress responses.However,the prevalence of genome-wide plant AS changes during infection and the mechanisms by which pathogens modulate AS remain poorly understood.Here,we examined the global AS changes in tomato leaves infected with Phytophthora infestans,the infamous Irish famine pathogen.We show that more than 2000 genes exhibiting significant changes in AS are not differentially expressed,indicating that AS is a distinct layer of transcriptome reprogramming during plant-pathogen interactions.Furthermore,our results show that P.infestans subverts host immunity by repressing the AS of positive regulators of plant immunity and promoting the AS of susceptibility factors.To study the underlying mechanism,we established a luminescence-based AS reporter system in Nicotiana benthamiana to screen pathogen effectors modulating plant AS.We identified nine splicing regulatory effectors(SREs)from 87 P.infestans effectors.Further studies revealed that SRE3 physically binds U1-70K to manipulate the plant AS machinery and subsequently modulates AS-mediated plant immunity.Our study not only unveils genome-wide plant AS reprogramming during infection but also establishes a novel AS screening tool to identify SREs from a wide range of plant pathogens,providing opportunities to understand the splicing regulatory mechanisms through which pathogens subvert plant immunity. | Jie Huang Xinyu Lu Hongwei Wu Yuchen Xie Qian Peng Lianfeng Gu Juyou Wu Yuanchao Wang Anireddy SNReddy Suomeng Dong | 2020 | Molecular Plant2020,13,10: | 4 |

| 3 | Theoretic heat sink simulation and experimental investigation of the pyrolysis of substituted cyclohexanes显示文摘Reaxgen program for the pyrolysis mechanism of cycloalkanes was adopted to simulate the heat sink of substituted cyclohexanes. Thermal cracking of cyclohexanes was performed to examine the cracking performance, wherein the substituent effects were detailedly discussed under supercritical condition. It was found that Reaxgen program played a good part in the screening and optimization of cyclohexanes. A good agreement with the experimental data for the mono-substituted and bi-substituted cyclohexanes was demonstrated, however, some deviation for the tri-substituted cyclohexanes were observed. The experiment results indicated that the gaseous product yield increased sharply for monosubstituted cyclohexanes with short substituents containing no more than two carbons. Nevertheless, continuous increase in the alkyl chain depressed the gaseous product yield smoothly. The cyclic substituent dramatically inhibited the pyrolysis of cyclohexanes. All the substituents but cyclohexyl had no obvious effect on the yield of hydrogen and olefins(≤C4). For bi-substituted cyclohexanes, the more close the distance between the two substituents, the higher the gaseous product yield was obtained. However, opposite result on the selectivity to hydrogen and olefins(≤C4) was generally obtained except 1,3-dimethylcyclohexane. | 8 | PbrRALF2-elicited reactive oxygen species signaling is mediated by the PbrCrRLK1L13-PbrMPK18 module in pear pollen tubes显示文摘Rapid alkalinization factors(RALFs)are cysteine-rich peptides that play important roles in a variety of biological processes,such as cell elongation and immune signaling.Recent studies in Arabidopsis have shown that RALFs regulate pollen tube growth via plasma membrane receptor-like kinases(RLKs).However,the downstream signal transduction mechanisms of RLKs in pollen tubes are unknown.Here,we identified PbrRALF2,a pear(Pyrus bretschneideri)pollen RALF peptide that inhibits pollen tube growth.We found that PbrRALF2 interacts with a malectin-like domain-containing RLK,PbrCrRLK1L13.The relative affinity between PbrRALF2 and PbrCrRLK1L13 was at the submicromolar level,which is consistent with the values of ligand–receptor kinase pairs and the physiological concentration for PbrRALF2-mediated inhibition of pollen tube growth.After binding to its extracellular domain,PbrRALF2 activated the phosphorylation of PbrCrRLK1L13 in a dose-dependent manner.We further showed that the MAP kinase PbrMPK18 is a downstream target of PbrCrRLK1L13 that mediates PbrRALF2-elicited reactive oxygen species(ROS)production.The excessive accumulation of ROS inhibits pollen tube growth.We show that MPK acts as a mediator for CrRLK1L to stimulate ROS production,which might represent a general mechanism by which RALF and CrRLK1L function in signaling pathways. | Xiaobing Kou Jiangmei Sun Peng Wang Danqi Wang Peng Cao Jing Lin Youhong Chang Shaoling Zhang Juyou Wu | 2021 | Horticulture Research2021,8,1: | 0 |
