1Diarrhoea-predominant irritable bowel syndrome distinguishable by 16S rRNA gene phylotype quantification显示文摘AIM:To study whether selected bacterial 16S ribosomal RNA(rRNA)gene phylotypes are capable of disting- uishing irritable bowel syndrome(IBS). METHODS:The faecal microbiota of twenty volunteers with IBS,subdivided into eight diarrhoea-predominant (IBS-D),eight constipation-predominant(IBS-C)and four mixed symptom-subtype(IBS-M)IBS patients,and fifteen control subjects,were analysed at three time-points with a set of fourteen quantitative real-timepolymerase chain reaction assays.All assays targeted 16S rRNA gene phylotypes putatively associated with IBS,based on 16S rRNA gene library sequence analysis. The target phylotypes were affiliated with Actinobac-teria,Bacteroidetes and Firmicutes.Eight of the target phylotypes had less than 95%similarity to cultured bacterial species according to their 16S rRNA gene sequence.The data analyses were made with repeated-measures ANCOVA-type modelling of the data and principle component analysis(PCA)with linear mixed-effects models applied to the principal component scores. RESULTS:Bacterial phylotypes Clostridium cocleatum 88%,Clostridium thermosuccinogenes 85%,Coprobacillus catenaformis 91%,Ruminococcus bromii-like, Ruminococcus torques 91%,and R.torques 93%were detected from all samples analysed.A multivariate analysis of the relative quantities of all 14 bacterial 16S rRNA gene phylotypes suggested that the intestinal microbiota of the IBS-D patients differed from other sample groups.The PCA on the first principal component(PC1),explaining 30.36%of the observed variation in the IBS-D patient group,was significantly altered from all other sample groups(IBS-D vs control, P=0.01;IBS-D vs IBS-M,P=0.00;IBS-D vs IBS-C, P=0.05).Significant differences were also observed in the levels of distinct phylotypes using relative values in proportion to the total amount of bacteria.A phy- lotype with 85%similarity to C.thermosuccinogenes was quantified in significantly different quantities among the IBS-D and control subjects(-4.08±0.90 vs -3.33±1.16,P=0.04)and IBS-D and IBS-M subjects (-4.08±0.90 vs-3.08±1.38,P=0.05).Furthermore,a phylotype with 94%similarity to R.torques was more prevalent in IBS-D patients'intestinal micro- biota than in that of control subjects(-2.43±1.49 vs -4.02±1.63,P=0.01).A phylotype with 93%simi- larity to R.torques was associated with control sam- ples when compared with IBS-M(-2.41±0.53 vs -2.92±0.56,P=0.00).Additionally,a R.bromii-like phylotype was associated with IBS-C patients in com- parison to control subjects(-1.61±1.83 vs-3.69± 2.42,P=0.01).All of the above mentioned phylotype specific alterations were independent of the effect of time. 2复杂情境中的日常活动可视化与应用研究显示文摘时间地理学以时空路径为代表的符号系统由于具备抽象、简明、直观等特点,在地理学及其他社会科学领域得到了广泛传播与应用。随着GIS与地理可视化技术的不断进步、高精度时空行为数据的批量生产等,真实地理环境中个体时空运动轨迹的三维可视化技术也在不断进步。信息化社会的到来与城市社会转型等使得现代社会中的人类日常活动变得十分复杂,时间地理学由于直观刻画时空间中人类活动的时空连续性与复杂情境性而备受关注。本文首先界定了人类日常活动复杂情境的分析维度,包括日常情境、企划情境、地理情境、社会情境、技术情境等,并在此基础上将绝对时空间坐标系下的时空路径拓展到相对时空观下多维活动序列的可视化表达。其次,简单说明Daily life与VISUAL-Time PAc TS/energy use等可视化工具,介绍时空路径在复杂情境性下的可视化、模式挖掘及个案分析。最后,案例分析个体与群体尺度上活动情境可视化方法在家庭成员间分工与互动、日常活动模式差异与家庭能源消耗差异等方面的应用。Kajsa Ellegard 张艳 蒋晨 柴彦威 2016人文地理2016,31,5:16

4时间地理学的企划概念及其研究案例显示文摘企划是为完成任何意愿和目标而进行的一系列必要的简单或复杂的任务,即为达到某个个人或组织的长期或短期目标而实施的活动。企划研究旨在建立意图和实现之间的桥梁,从个体或组织主体意图的视角构建活动之间的关系,它不仅关注时空间行为的客观发生,还关注时空间行为的计划和时空间行为结果的影响反馈等主观方面。企划视角下的时间被看作将'未来'不断转换为'过去'的连续过程,一个完整的企划包括孕育计划、活动实现、影响反馈三个主要阶段,基于企划可以分析个体行为的闭环决策过程。个体企划与组织企划交织的情景分析有助于解释个体时空间行为以及个体企划与组织企划的关系。沃尔沃乌德瓦拉工厂'结构性变革'的案例为如何从组织企划与个体企划交织的视角探索组织管理变革及其综合效益评价提供了很好的启示。企划视角下的日常活动研究将极有可能推动时间地理学从主客观结合、能动被动结合的微观视角透视日常活动决策的黑箱,进而对时空间行为进行解释和预测,最终实现物质世界时空制约与人的时空意图的统一。Kajsa Ellegard 刘伯初 张艳 柴彦威 2016人文地理2016,31,5:9

6Elevated pro-inflammatory and lipotoxic mucosal lipids characterise irritable bowel syndrome显示文摘AIM:To investigate the pathophysiology of irritable bowel syndrome(IBS)by comparing the global mucosal metabolic profiles of IBS patients with those of healthy controls.METHODS:Fifteen IBS patients fulfilling the Rome Ⅱcriteria,and nine healthy volunteers were included in the study.A combined lipidomics(UPLC/MS)and metabolomics(GC×GC-TOF)approach was used to achieve global metabolic profiles of mucosal biopsies from the ascending colon.RESULTS:Overall,lipid levels were elevated in patients with IBS.The most significant upregulation was seen for pro-inflammatory lysophosphatidylcholines.Other lipid groups that were significantly upregulated in IBS patients were lipotoxic ceramides,glycosphingolipids,and di-and triacylglycerols.Among the metabolites,the cyclic ester 2(3H)-furanone was almost 14-fold upregulated in IBS patients compared to healthy subjects(P=0.03).CONCLUSION:IBS mucosa is characterised by a distinct pro-inflammatory and lipotoxic metabolic profile.Especially,there was an increase in several lipid species such as lysophospholipids and ceramides.Kajsa Kajander Eveliina Myllyluoma Sinikka Kyrnpalo Martin Rasmussen Pentti Sipponen Ismo Mattila Tuulikki Seppnen-Laakso Heikki Vapaatalo Matej Orei Riitta Korpela 2009World Journal of Gastroenterology2009,15,48:5

8Marginal zone B cells are naturally reactive to collagen type II and are involved in the initiation of the immune response in collagen-induced arthritis显示文摘Antibodies against type II collagen(CII)are essential for development of collagen-induced arthritis(CIA),but how and where the B-cell response to CII is initiated is not fully known.We show here that naive DBA/1 mice display naturally reactive IgM and IgG anti-CII producing B cells prior to immunization.The CII-reactive B cells were observed in the spleen and recognized as marginal zone(MZ)B cells.After CII immunization,CII-specific B cells expanded rapidly in the spleen,in contrast to the lymph nodes,with the initial response derived from MZ B cells and later by follicular(FO)B cells.This was evident despite that the MZ B cells were subject to stringent tolerance mechanisms by having a greater Fc gamma receptor IIb expression than the FO B cells.Further,the MZ B cells migrated to the FO areas upon immunization,possibly providing antigen and activating FO T cells and subsequently FO B cells.Thus,around CIA onset increased numbers of IgG anti-CII producing FO B cells was seen in the spleen,which was dominated by IgG2a-and IgG2b-positive cells.These data demonstrate that CII-reactive MZ B cells are present before and expand after CII immunization,suggesting an initiating role of MZ B cells in the development of CIA.Cecilia Carnrot Kajsa E Prokopec Kristina Rasbo Mikael CI Karlsson Sandra Kleinau 2011Cellular & Molecular Immunology2011,8,4:3

10Nested hollow architectures of nitrogen-doped carbon-decorated Fe,Co,Ni-based phosphides for boosting water and urea electrolysis显示文摘Tailoring the nanostructure/morphology and chemical composition is important to regulate the electronic configuration of electrocatalysts and thus enhance their performance for water and urea electrolysis.Herein,the nitrogen-doped carbon-decorated tricomponent metal phosphides of FeP4 nanotube@Ni-Co-P nanocage(NC-FNCP)with unique nested hollow architectures are fabricated by a self-sacrifice template strategy.Benefiting from the multi-component synergy,the modification of nitrogen-doped carbon,and the modulation of nested porous hollow morphology,NC-FNCP facilitates rapid electron/mass transport in water and urea electrolysis.NC-FNCP-based anode shows low potentials of 248 mV and 1.37 V(vs.reversible hydrogen electrode)to attain 10 mA/cm^(2) for oxygen evolution reaction(OER)and urea oxidation reaction(UOR),respectively.In addition,the overall urea electrolysis drives 10 mA/cm^(2) at a comparatively low voltage of 1.52 V(vs.RHE)that is 110 mV lower than that of overall water electrolysis,as well as exhibits excellent stability over 20 h.This work strategizes a multi-shell-structured electrocatalyst with multi-compositions and explores its applications in a sustainable combination of hydrogen production and sewage remediation.Jie Zhang Shoushuang Huang Ping Ning Peijun Xin Zhiwen Chen Qing Wang Kajsa Uvdal Zhangjun Hu 2022Nano Research2022,15,3:2

18Single-wavelength-excited fluorogenic nanoprobe for accurate realtime ratiometric analysis of broad pH fluctuations in mitophagy显示文摘Mitophagy has a critical role in maintaining cellular homeostasis through acidic lysosomes engulfing excess or impaired mitochondria,thereby pH fluctuation is one of the most significant indicators for tracking mitophagy.Then such precise pH tracking demands the fluorogenic probe that has tailored contemporaneous features,including mitochondrial-specificity,excellent biocompatibility,wide pH-sensitive range of 8.0–4.0,and especially quantitative ability.However,available molecular probes cannot simultaneously meet all the requirements since it is extremely difficult to integrate multiple functionalities into a single molecule.To fully address this issue,we herein integrate two fluorogenic pH sensitive units,a mitochondria-specific block,cellpenetrating facilitator,and biocompatible segments into an elegant silica nano scaffold,which greatly ensures the applicability for real-time tracking of pH fluctuations in mitophagy.Most significantly,at a single wavelength excitation,the integrated pHsensitive units have spectra-distinguishable fluorescence towards alkaline and acidic pH in a broad range that covers mitochondrial and lysosomal pH,thus enabling a ratiometric analysis of pH variations during the whole mitophagy.This work also provides constructive insights into the fabrication of advanced fluorescent nanoprobes for diverse biomedical applications.Xin Zhang Juan Chen Jiwen Hu Anna du Rietz Xiongyu Wu Ruilong Zhang Zhongping Zhang Kajsa Uvdal Zhangjun Hu 2022Nano Research2022,15,7:1
