1Fluorescent nanoparticles for chemical and biological sensing显示文摘Fluorescent nanoparticles (NPs), including quantum dots (QDs), dye-doped NPs, and rare earth-based NPs, etc., have been a major focus of research and development during the past decade. The impetus behind such endeavors can be attributed to their unique chemical and optical properties, such as bright fluorescence, high photostability, large Stocks shift and flexible processability. The introduction of fluorescent NPs into analytical chemistry has opened up new venues for fluorescent analysis. In this review, we focus on the developments and analytical applications of fluorescent NPs in the chemical and biological sensing of pH, ions, organic compounds, small biological molecules, nucleic acids, proteins, virus and bacteria. The review also points out the in vitro and in vivo imaging application of fluorescent NPs at the cell and body levels. 2Preparation of luminescent CdTe quantum dots doped core-shell nanoparticles and their application in cell recognition显示文摘Based on the reverse microemulsion technique, luminescent quantum dots doped core-shell nanoparticles have been prepared by employing silica as the shell and CdTe quantum dots as the core of the nanoparticles, which have an excellent solubility and dispersibility, especially amine and phosphonate groups have been modified on their surface synchronously. In comparison with CdTe quantum dots, these nanoparticles show superiority in chemical and photo-chemical stability. 3Structural changes in the gray matter of unmedicated patients with obsessive-compulsive disorder: a voxel-based morphometric study显示文摘The aim of the current study was to use whole brain voxel-based morphometry(VBM)to assess the gray matter(GM)changes in unmedicated patients with obsessive-compulsive disorder(OCD)compared with normal controls.We compared the GM volumes in28 patients with 22 matched healthy controls using a1.5T MRI.Three-dimensional T1-weighted magnetic resonance images were obtained from all participants.VBM was performed to detect GM volume differences between the two groups.We detected increased regional GM volumes in the bilateral middle temporal gyri,bilateral middle occipital gyri,bilateral globus pallidus,right inferior parietal gyrus,left superior parietal gyrus,right parahippocampus,right supramarginal gyrus,right medial superior frontal gyrus,and left inferior frontal opercular cortex in the OCD patients relative to controls(P<0.001,uncorrected,cluster size>100 voxels).No decreased GM volume was found in the OCD group compared with normal controls.Our findings suggest that structural changes in the GM are not limited to fronto-striato-thalamic circuits in the pathogenesis of OCD.Temporo-parietal cortex may also play an important role.Ling Tan Qing Fan Chao You Jijun Wang Zhao Dong Xuemei Wang Kemin Chen Zeping Xiao Kaida Jiang 2013Neuroscience Bulletin2013,29,5:5
4A novel fluorescent label based on biological fluores-cent nanoparticles and its application in cell recognition显示文摘Uniform-sized fluorescent nanoparticles have been prepared by employing silica as the shell and a highly luminescent dye complex of ruthenium ion and bipyridyl, tris(2,2 ’-bipyridyl) dichlororuthenium( II) hexahydrate as the core of the nanoparticles. A novel fluorescent label method is proposed, which is based on the biological fluorescent nanoparticles on the foundation of nanotechnology, biotechnology and fluorescent label technology. In comparison with the conventional fluorophores as fluorescent labels such as fluorescein isothiocyanate (FITC) label, this new label shows more superiority in photochemical stability, detection sensitivity and application scope for the biomedicine research. 5Ribozyme probe based on molecular beacon for real time monitoring of enzymatic cleavage process显示文摘Ribozyme probe based on molecular beacon (MBR) for monitoring enzymatic cleavage process in real time is designed and studied. The approach relies on ri-bozyme substrates modified at the two arms, with a fluores-cent moiety attached to the end of one arm and a non-fluorescent quenching moiety attached to the end of the other arm. MBR is employed to directly convert the cleavage information into fluorescence signal in real time. Compared with traditional approach, this method provides a no-radiolabeling, sensitive and effective way to research on the ribozyme activity, enzymatic dynamic process and ri-bozyme function during gene therapy. 6Novel protein detection method based on proximity- dependent polymerase reaction and aptamers显示文摘In recent years, specific detection of proteins is one of the hot issues about aptamers in proteomics. Here we reported a simple, sensitive and specific proximity-dependent protein assay with dual DNA aptamers. Thrombin was used as the model protein, and two aptamer probes with complementary se- quence at 3′-end were designed for the two distinct epitopes of the protein. Association of the two ap- tamers with thrombin resulted in stable hybrids due to the proximity of 3′-end, then polymerase reac- tion was induced. The amount of obtained dsDNA was indicated using the fluorescence dye Sybr Green I. The results showed that the initial velocity of polymerase reaction had a positive correlation with concentration of thrombin. The advantages of this dual-aptamer-based approach included simple and flexible design of aptamer probes, high selectivity and high sensitivity. 8Pledgee competition, strategic disclosure, and future crash risk显示文摘We investigate whether pledgee competition affects the disclosure choice of firms whose controlling shareholders pledge their shares.We find that pledgee competition is positively related to pledge firms’ annual report tone management.This positive relationship is stronger for pledge firms with lower credit quality and non-state-owned enterprise pledge firms.Further corroborating our results, higher pledgee competition increases the future crash risk of pledge firms.Collectively, our results suggest that competition pressure induces pledgees to lower their monitoring incentives to remain competitive in the marketplace, thus leading to pledge firms’ bad news hoarding behavior.Xiaoxi Li Jing Liu Kemin Wang 2019China Journal of Accounting Research2019,,3:3
9A photosensitizer-loaded zinc oxide-polydopamine core-shell nanotherapeutic agent for photodynamic and photothermal synergistic therapy of cancer cells显示文摘In clinical cancer research,it is quite promising to develop multimodal synergistic therapeutic strategies.Photodynamic and photothermal synergistic therapy is a very desirable multimodal therapy strategy.Herein,we report a facile and simple method to construct a nanotherapeutic agent for photodynamic and photothermal therapy.This nanotherapeutic agent(ZnO@Ce6-PDA)is composed of a ZnO nanoparticle core,an interlayer of photosensitizer chlorin e6(Ce6)and an outer layer of polydopamine(PDA).Due to the existence of Ce6,the ZnO@Ce6-PDA can efficiently generate singlet oxygen(1O2)under 660 nm laser irradiation.Moreover,the ZnO@Ce6-PDA can serve as a photothermal agent,because of the excellent photothermal conversion efficiency of the PDA coating layer in the presence of 780 nm laser.Experiment results demonstrated that the designed nanotherapeutic agent had outstanding phototoxicity upon the combination of laser irradiation at 660 and 780 nm.Thus,our work proves that the ZnO@Ce6-PDA is a promising photodynamic/photothermal dual-modal nanotherapeutic agent for enhanced cancer therapy.Ri Wu Huizhen Wang Luo Hai Tianzheng Wang Min Hou Dinggeng He Xiaoxiao He Kemin Wang 2020Chinese Chemical Letters2020,31,1:3
10Protein analysis based on molecular beacon probes and biofunctionalized nanoparticles显示文摘With the completion of the human genome-sequencing project, there has been a resulting change in the focus of studies from genomics to proteomics. By utilizing the inherent advantages of molecular beacon probes and biofunctionalized nanoparticles, a series of novel principles, methods and techniques have been exploited for bioanalytical and biomedical studies. This review mainly discusses the applications of molecular beacon probes and biofunctionalized nanoparticles-based technologies for realtime, in-situ, highly sensitive and highly selective protein analysis, including the nonspecific or specific protein detection and separation, protein/DNA interaction studies, cell surface protein recognition, and antigen-antibody binding process-based bacteria assays. 11Study on the effect of electrostatic interaction on core-shell nanoparticles preparation with microemulsion technique显示文摘The routine method for preparation of silica core-shell nanoparticles (NPs) is to carry out nucleation and shell coating through the hydrolysis of silane in water in oil (W/O) microemulsion to form three-dimensional netted silica shell. We found that electrostatic interaction of the core ma- terials with shell materials would determine whether the stable core-shell silica NPs formed or not. The traditional important factors such as molecular weight of core materials or the thickness of the shell have no obvious relationship with it. 12Research of the relationship of intracellular acidification and apoptosis in Hela cells based on pH nanosensors显示文摘In this paper,the relationship of intracellular acidification and apoptosis in Hela cells induced by vin-cristine sulfate has been studied by use of the ratiometric pH nanosensors that have been developed by our group,employing fluorescein isothiocyanate(FITC) doped as the pH-sensitive dye and Tris(2,2'-bipyidyl) dichlororuthenium(II) hexahydrate(RuBpy) doped as reference dye. The pH change of the Hela cells induced by vincristine sulfate has been monitored in vivo,in situ and real time by use of the ratiometric pH nanosensors. The experimental results show that the pH of the apoptotic Hela cells induced by vincristine sulfate has been acidified from 7.11 to 6.51,and the percentage of intra-cellular acidification is correlated with the induced concentration and incubation time of the vincristine sulfate. The further study of the percentage of intracellular acidification and the percentage of apop-tosis of Hela cells at the same time reveals that apoptosis of Hela cells induced by vincristine sulfate is preceded by intracellular acidification. 14The amplified fluorescence quenching of heteroatomic conjugated polymers based on the “molecular wire”effects显示文摘Tremendous interest has been evoked in the utility of the fluorescent conjugated polymers towards the creation of ultra-sensitive systems based on the molecular wire effects which can amplify the transduction events. Here, a series of new polymers pyridine substitute poly (phen- ylene ethynylene)s are synthesized by Sonogashira-Hagihara coupling reaction. The impact of structure modification on the physical properties and spectral characteristics of poly-mers has been investigated. The quenching studies are car-ried out and exhibit that recognition site (pyridyle group) highly influences the quenching efficiency. The results show that polymer PI with pyridyl group entrapped in the conju-gated backbone has the specific and sensitive response to Pd(Ⅱ). 15Bioeffects of different functionalized silica nano-particles on HaCaT cell line显示文摘The bioeffects of silica nanoparticles (SiNP), phosphorylate-terminated nanoparticles (PO4- NP) and amino-terminated nanoparticles (NH2NP) on HaCaT cell line have been studied in this paper. The effects of the three kinds of functionalized silica nanoparticles on adherence, proliferation and cycle of HaCaT cells have been investigated. And the cel- lular uptake of the three kinds of functionalized silica nanoparticles by HaCaT cells has also been exam- ined. Results indicated that the bioeffects of the three kinds of functionalized nanoparticles on HaCaT cells were concentration-dependent. And the three kinds of functionalized nanoparticles all exhibited well bio- compatibility if the concentration was below 0.2 μg/μL. While the cytotoxicities of the three kinds of function- alized nanoparticles on HaCaT cells would increase with the increasing of nanoparticles concentration, and the following order was observed: NH2NP > SiNP > PO4NP. In addition, the quantity and rapidity of cellular uptake of nanoparticles by HaCaT cells were diverse due to the different functional groups. Under the same conditions, NH2NP was most and fast internalized by HaCaT cells, followed by SiNP, and PO4NP was the least and slowest. 16Study on the specific interaction between angiogenin and aptamer by atomic force microscopy (AFM)显示文摘The specific interaction between angiogenin and aptamer has been investigated by using AFM. The specificity of the interaction is revealed by comparing the binding probability of aptamer to other ele- ments in a series of control experiments. The results have shown that there is specific interaction force between angiogenin and aptamer. Moreover, the single molecular pull-off force between angiogenin and aptamer has also been determined using the Poisson statistical method to be 133.7±11.7 pN. 17Monitoring p21 mRNA expression in living cell based on molecular beacon fluorescence increasing rate显示文摘Studying the expression level of mRNA in living cells will offer tremendous opportunities for ad-vancement in cell biology research,disease diagnostics,and drug discovery.In this paper,a molecular beacon(MB)specific for the important tumor suppressor gene p21 has been designed and synthesized.The fluorescence signal was detected in real-time after the MB entered the cytoplasm of nasopharyn-geal carcinoma cells.After injecting the p21MB into nasopharyngeal carcinoma cell and p33-trans-fected nasopharyngeal carcinoma cell,the consistent increase of fluorescent signal intensity was de-tected in both cell lines,and maximum fluorescence intensity achieved in about 15 min.In about 4 min following microinjection,the fluorescence increasing rate was significantly different between these two cell lines,which indicate the different p21 mRNA expression levels.The results obtained in the real-time detection were also validated by RT-PCR.Analysis of the initial fluorescence increasing rate can effi-ciently reduce the side effect of enzyme and improve the accuracy in living cell mRNA detection.TANG HongXing YANG XiaoHai WANG KeMin TAN WeiHong LIU Bin HE LiFang WANG Wei 2008Chinese Science Bulletin2008,53,3:2
