| 2 | Origin and phylogenetic analysis of Tibetan Mastiff based on the mitochondrial DNA sequence显示文摘At present,the Tibetan Mastiff is the oldest and most ferocious dog in the world. However,the origin of the Tibetan Mastiff and its phylogenetic relationship with other large breed dogs such as Saint Bernard are unclear. In this study,the primers were designed accord-ing to the mitochondrial genome sequence of the domestic dog,and the 2,525 bp mitochondrial sequence,containing the whole sequence of Cytochrome b,tRNA-Thr,tRNA-Pro,and control region of the Tibetan Mastiff,was obtained. Using grey wolves and coyotes as out-groups,the Tibetan Mastiff and 12 breeds of domestic dogs were analyzed in phylogenesis. Tibetan Mastiff,domestic dog breeds,and grey wolves were clustered into a group and coyotes were clustered in a group separately. This indicated that the Tibetan Mastiff and the other domestic dogs originated from the grey wolf,and the Tibetan Mastiff belonged to Carnivora,Canidae,Canis,Canis lupus,Canis lupus familiaris on the animal taxonomy. In domestic dogs,the middle and small breed dogs were clustered at first; German Sheepdog,Swedish Elkhound,and Black Russian Terrier were clustered into one group,and the Tibetan Mastiff,Old English Sheepdog,Leonberger,and Saint Bernard were clustered in another group. This confirmed the viewpoint that many of the famous large breed dogs worldwide such as Saint Bernard possibly had the blood lineage of the Tibetan Mastiff,based on the molecular data. | 3 | Estimation of the Effects of Maize Straw Return on Soil Carbon and Nutrients Using Response Surface Methodology显示文摘Straw incorporation is generally considered an effective agricultural management practice that improves nutrient cycling and maintains soil fertility. To study the interactive effects of straw returning factors on soil organic carbon and available nutrients, a17-month(May 6, 2016 to October 6, 2017) experiment was conducted on straw incorporation by using response surface methodology under a three-factor(straw length, amount, and burying depth), five-level quadratic orthogonal rotation experimental design. Weight was assigned to each indicator for soil carbon and nutrients and then a comprehensive indicator was established. Then, a second-order polynomial model of the three straw returning factors was established using response surface methodology. Results indicated that17 months after straw incorporation, straw amount and burying depth had significant effects on the comprehensive indicator of soil carbon and nutrients. Straw length and the interactions of straw amount and burying depth showed no significant effects on the comprehensive indicator of soil carbon and nutrients. | 4 | Spatial and Temporal Changes of Arable Land Driven by Urbanization and Ecological Restoration in China显示文摘Since the industrial revolution, human activities have both expanded and intensified across the globe resulting in accelerated land use change. Land use change driven by China’s development has put pressure on the limited arable land resources, which has affected grain production. Competing land use interests are a potential threat to food security in China. Therefore, studying arable land use changes is critical for ensuring future food security and maintaining the sustainable development of arable land. Based on data from several major sources, we analyzed the spatio-temporal differences of arable land among different agricultural regions in China from 2000 to 2010 and identified the drivers of arable land expansion and loss. The results revealed that arable land decreased by 5.92 million ha or 3.31%. Arable land increased in the north and decreased in the south of China. Urbanization and ecological restoration programs were the main drivers of arable land loss, while the reclamation of other land cover types(e.g., forest, grassland, and wetland) was the primary source of the increased arable land. The majority of arable land expansion occurred in the Northwest, but the centroid for grain production moved to northeast, which indicated that new arable land was of poor quality and did not significantly contribute to the grain production capacity. The results show that the GFA of Mg86.33Ni13.67-xYx(x=0, 1, 3, 6, 10) is improved successfully with the Y addition. | 7 | TRAP1 Shows Clinical Significance and Promotes Cellular Migration and Invasion through STAT3/MMP2 Pathway in Human Esophageal Squamous Cell Cancer显示文摘Tumor necrosis factor receptor-associated protein 1(TRAP1), an important member of mitochondrial heat shock protein 90 family, is involved in multiple biological processes in several types of tumors. However, its pathological role in esophageal squamous cell cancer(ESCC) remains unknown. Herein, we demonstrated the clinical value of TRAP1, and its role in apoptosis and motility in ESCC. The clinical potential of TRAP1 was investigated through immunohistochemical analysis in 328 ESCC samples, which revealed that strong TRAP1 expression was associated with increased risk of lymph node metastasis, while high TRAP1 expression correlated with poor prognosis. Expression of TRAP1 was found to be an independent prognostic factor for patients with ESCC. Additionally, the upregulation of TRAP1 antagonized cisplatin-induced apoptosis while its downregulation sensitized cells to cisplatin-induced apoptosis. As revealed by the transwell assay, TRAP1 overexpression promoted cellular migration and invasion as compared to the control groups. In contrast,silencing of endogenous TRAP1 expression attenuated the ability of migration and invasion. Finally, the molecular mechanism investigated in the present study demonstrated that TRAP1-mediated migration and invasion occurred through STAT3/MMP2 signaling pathway. | 12 | Uncompromised NK cell activation is essential for virus-specific CTL activity during acute influenza virus infection显示文摘Natural killer (NK) cells are indispensable components of both the innate and adaptive immune response. However,their precise roles in the cross-talk between innate and adaptive immunity during influenza virus infection remaincontroversial. By comparing NK cell dynamics and activity under a sub-lethal dose and high dose of influenza virusinfection, we showed that influenza virus PR8 directly infected NK cells during natural infection, which wasconsistent with our previous findings obtained from an in vitro investigation of human NK cells. The impairments incytotoxicity and IFN-γ production by spleen NK cells following high-dose infection were accompanied by decreasedvirus-specific killing mediated by cytotoxic T lymphocytes (CTLs). Importantly, the weakened CTL activity could bereversed by adoptive transfer of spleen NK cells harvested from low-dose-infected mice but not healthy donors.Taken together, our data provide direct evidence supporting the contribution of NK cells to antiviral T-cellresponses. | 14 | Analysis of IGF2 mRNA expression and its methylation status between cattle yaks and their parents显示文摘Cattle yaks are an F1 hybrid between cattle and yaks and exhibit significant hybrid vigor. However, cattle yaks are sterile males. To study the epigenetic aspects of this reproductive isolation, IGF2 mRNA expression in cattle yaks and their parents and the patterns of the IGF2 differentially methylated regions (DMR) were examined in blood and testes. The results showed that IGF2 expression in cattle yaks' testes was lower than in their parents (P < 0.01). The IGF2 DMR was highly methylated (above 90%) in blood and testes of cattle yaks, yaks and cattle, with the highest level in cattle yaks (P > 0.05). Our study showed that IGF2 plays an important role in bovine spermato-genesis and might be involved in cattle-yak male sterility. | 17 | Influence of Na^+,K^+,Mg^(2+),Ca^(2+),and Fe^(3+) on filterability and settleability of drilling sludge显示文摘Solid–liquid separation is a vital step in drilling sludge disposal, and the filterability and settleability of drilling sludge are the main evaluating indicators for the separation process. The influence of Na^+,K^+,Mg^(2+),Ca^(2+),and Fe^(3+) on drilling sludge filterability and settleability was investigated in our research. The water content,filtration rate, supernatant volume and supernatant turbidity were measured to evaluate the filterability and settleability of drilling sludge. Meanwhile, the zeta potential, specific surface area of sludge flocs, particle size distribution and Fourier-transformed infrared spectra were employed to clarify the influencing mechanism.The experimental results showed that the filterability and settleability of drilling sludge were related to concentration and types of cations. Mg^(2+),Ca^(2+),and Fe^(3+) performed better than Na^+, K^+, and the cations with smaller hydrated radius got superior solid–liquid separation behavior at same valence. Finally, the spectra indicated that no chemical adsorption occurred between inorganic cations and drilling sludge flocs. | 19 | Preparation and luminescence properties of orange-red Ba3Y4O9:Sm3+phosphors显示文摘A novel orange-red emitting Ba_3 Y_4 O_9:Sm^(3+) phosphors were prepared by a high temperature solidstate reaction in air. X-ray diffraction(XRD), photoluminescence spectra, fluorescence decay and temperature-dependent emission spectra were utilized to characterize the structure and luminescence properties. The results show that the excitation spectrum includes a series of linear peaks at350, 367, 382, 410, 424, 445, 470 and 495 nm, respectively. Under 410 nm excitation, the emission peaks were located at 574 nm(~4 G_(5/2)-~6 H_(5/2)), 608 nm(~4 G_(5/2)-~6 H_(7/2)),659 nm(~4 G_(5/2)-~6 H_(9/2)) and722 nm(~4 G_(5/2)-~6 H_(11/2)), respectively. The concentration quenching occurs when x equals 0.08 for Ba_3 Y_(4-x)O_9:xSm^(3+) phosphor and its mechanism is ascribed to the dipole-dipole interaction. The chromaticity coordinates of Ba_3 Y_(3.92)O_9:0.08 Sm^(3+) phosphor are in the orange-red region. The temperature-dependent study shows that this phosphor has excellent luminescence thermal-stability.And the luminescence intensity of Ba_3 Y_(3.92)O_9:0.08 Sm^(3+) phosphor at 473 K only declines by about25.75% of its initial intensity. | 20 | Development of Solar Spectroradiometer for Meteorological Observation显示文摘A newgeneration of solar spectroradiometer has been developed by CUST/JRSI to improve solarirradiance observation data under hyperspectral resolution. It is based on the grating spectroradiometer with a back-thinned CCD linear image sensor and is operated in a hermetically sealed enclosure. The solar spectroradiometer is designed to measure the solar spectral irradiance from300 nm to 1100 nm wavelength range with the spectral resolution of 2 nm( the full width at half maximum). The optical bench is optimized to minimize stray light.
