| 1 | SHP2 inhibition triggers anti-tumor immunity and synergizes with PD-1 blockade显示文摘Tyrosine phosphatase SHP2 is a promising drug target in cancer immunotherapy due to its bidirectional role in both tumor growth promotion and T-cell inactivation. Its allosteric inhibitor SHP099 is known to inhibit cancer cell growth both in vitro and in vivo. However, whether SHP099-mediated SHP2 inhibition retards tumor growth in vivo via anti-tumor immunity remains elusive. To address this, a CT-26 colon cancer xenograft model was established in mice since this cell line is insensitive to SHP099.Consequently, SHP099 minimally affected CT-26 tumor growth in immuno-deficient nude mice, but significantly decreased the tumor burden in CT-26 tumor-bearing mice with intact immune system.SHP099 augmented anti-tumor immunity, as shown by the elevated proportion of CD8tIFN-γtT cells and the upregulation of cytotoxic T-cell related genes including Granzyme B andPerforin, which decreased the tumor load. In addition, tumor growth in mice with SHP2-deficient T-cells was markedly slowed down because of enhanced anti-tumor responses. Finally, the combination of SHP099 and antiPD-1 antibody showed a higher therapeutic efficacy than either monotherapy in controlling tumor growthin two colon cancer xenograft models, indicating that these agents complement each other. | 3 | Danggui Buxue Tang ameliorates bleomycin-induced pulmonary fibrosis in rats through inhibiting transforming growth factor-β1/Smad3/plasminogen activator inhibitor-1 signaling pathway显示文摘OBJECTIVE: To investigate the effect of Danggui Buxue Tang(DBT), a decoction from Traditional Chinese Medicine, on bleomycin-induced pulmonary fibrosis in rats, and to propose the possible underlying mechanism.METHODS: Forty male Sprague-Dawley rats were randomly divided into sham group, model group,prednisone group and DBT group. Pulmonary fibrosis rat model was established by intratracheal injection with bleomycin. Body weight and lung index were monitored. Histopathologic examination and collagen deposition were determined using Hematoxylin and eosin(HE) and Masson's trichrome staining. Immunohistochemistry staining was applied to observe the expression of alpha-smooth muscle actin(α-SMA). m RNA expression of α-SMA,collagen Ⅰ and collagen Ⅲ were measured by realtime fluorescence quantitative PCR(RT-q PCR). Inflammatory cytokines, including tumor necrosis factor alpha(TNF-α), interleukin-6(IL-6) and IL-1β in serum were detected by Enzyme-linked immunosorbent assay. Alkali hydrolysis method was conducted to investigate the content of hydroxyproline(HYP). Transforming growth factor-β1(TGF-β1),Smad3 and plasminogen activator inhibitor-1(PAI-1) protein level were examined by Western blot assay.RESULTS: DBT significantly reduced the severity of bleomycin-induced pulmonary fibrosis and inflammation as indicated by minimizing the lost of weight, and by lowering the levels of lung index, inflammation score, Ashcroft score, collagen volume fraction(%), HYP, α-SMA, collagen Ⅰ, collagen Ⅲ,TNF-α, IL-6, IL-1β, TGF-β1, Smad3 and PAI-1, consistent with the effect of prednisone.CONCLUSION: Our findings suggest that DBT is able to ameliorate the pulmonary fibrosis, the possible mechanism may involve inhibition of pulmonary inflammation and collagen deposition, possibly via suppressing TGF-β1/Smad3/PAI-1 signaling pathway. | Wang Jiepeng Fang Chaoyi Wang Shaoxian Fang Fang Chu Xinqiao Liu Na Lu Chenxi Wang Shuo Li Wei | 2020 | Journal of Traditional Chinese Medicine2020,40,2: | 5 |

| 5 | Near-Real-Time Carbon Emission Accounting Technology Toward Carbon Neutrality显示文摘Climate change is the greatest environmental threat to humans and the planet in the 21st century.Global anthropogenic greenhouse gas emissions are one of the main causes of the increasing number of extreme climate events.Cumulative carbon dioxide(CO_(2))emissions showed a linear relationship with cumulative temperature rise since the pre-industrial stage,and this accounts for approximately 80%of the total anthropogenic greenhouse gases.Therefore,accurate and reliable carbon emission data are the foundation and scientific basis for most emission reduction policymaking and target setting.Currently,China has made clear the ambitious goal of achieving the peak of carbon emissions by 2030 and achieving carbon neutrality by 2060.The development of a finer-grained spatiotemporal carbon emission database is urgently needed to achieve more accurate carbon emission monitoring for continuous implementation and the iterative improvement of emission reduction policies.Near-real-time carbon emission monitoring is not only a major national demand but also a scientific question at the frontier of this discipline.This article reviews existing annual-based carbon accounting methods,with a focus on the newly developed real-time carbon emission technology and its current application trends.We also present a framework for the latest near-real-time carbon emission accounting technology that can be widely used.The development of relevant data and methods will provide strong database support to the policymaking for China’s“carbon neutrality”strategy.Finally,this article provides an outlook on the future of real-time carbon emission monitoring technology. | Zhu Liu Taochun Sun Ying Yu Piyu Ke Zhu Deng Chenxi Lu Da Huo Xiang Ding | 2022 | Engineering2022,8,7: | 5 |

| 7 | The long-term prognosis and predictors of epilepsy:a retrospective study in 820 patients显示文摘Background:To investigate the prognosis and predictors for seizure control in epileptic patients in China.Methods:Eight hundred and twenty patients with epilepsy who visited Xuanwu Hospital from October 2017 to January 2020,were enrolled.The clinical information of the patients was obtained by retrospectively reviewing medical records.Prognostic measures of seizure control included remission,relapse and occurrence of drug resistance.The relationship of prognosis of seizure control with factors such as demographics,clinical characteristics and initial electroencephalography(EEG)features was investigated.Results:A total of 503(61.3%)patients experienced a 1-year remission and 330(49.3%of 669)had a 2-year remission.Idiopathic type of epilepsy(P<0.001),normal EEG(P<0.05),number of antiepileptic drugs(P<0.05)and seizure frequency of<1/month(P<0.001)at the first arrival predicted a remission independently.Of the 503 patients who achieved a 1-year remission,184(36.6%)experienced a relapse,due to external reversible causes(58 patients)or with unknown reversible triggers(126 patients).No factors were found to be associated with a relapse(P>0.05).At the end of the study,322 patients(39.3%)developed drug resistance.The development of drug resistance was associated with the following factors:symptomatic aetiology of epilepsy,epileptiform abnormality in EEG,number of antiepileptic drugs and seizure frequency of≥1/month at first arrival(P<0.001).For symptomatic epilepsy,patients with meningitis/encephalitis(P=0.007)were more likely to develop drug-resistant epilepsy than these with other causes.Conclusions:Remission is a common process.The type of epilepsy(idiopathic or symptomatic),EEG features,seizure frequency and treatment history at first arrival are related to both remission and terminal drug resistance.Among various causes of symptomatic epilepsy,meningitis/encephalitis is associated with the worst prognosis of epilepsy. | Haoran Yang Jun Zhang Chenxi Yang Dan Wu Xiaoyun Liu Hongjuan Lu Wei Zhou Wei Sun | 2021 | Acta Epileptologica2021,3,1: | 2 |

| 15 | Facile fabrication of branched-chain carbohydrate chips for studying carbohydrate-protein interactions by QCM biosensor显示文摘A novel approach for fabricating branched-chain(BC) carbohydrate chips to study carbohydrate-protein interactions using Quartz Crystal Microbalance(QCM) biosensor was developed. This approach utilizes functional alkynyl-branch molecule modified chip surfaces, which through the terminal alkynyl group for covalent linking of unprotected azide-carbohydrates. The unprotected azide-carbohydrates were syhthesized in one-step using 2-azido-1,3-dimethyl-imidazolinium as catalyst, avoiding complex chemical modifications. Additionally, the branch surface modified with the carbohydrates not only supplies more specific binding site but also reveals significant cluster effect. To exemplify the sugar cluster effect on BC carbohydrate chips, BC Galactose and Mannose chips prepared in this work were used to determine carbohydrate-lectin interactions using QCM biosensor. The results clearly showed that BC chip significantly improves the detection sensitivity compared with the straight-chain(SC) chip.
