| 1 | Meteorin-β/Meteorin like/IL-41 attenuates airway inflammation in house dust mite-induced allergic asthma显示文摘We sought to examine the regulatory effect of Meteorin-β(Metrnβ)/Meteorin like(Metrnl)/IL-41 on lung inflammation in allergic asthma.We found that Metrnβwas elevated significantly in asthmatic patients and in mice with allergic asthma induced by house dust mite(HDM)extract.Upon exposure to HDM,Metrnβwas secreted predominantly by airway epithelial cells and inflammatory cells,including macrophages and eosinophils.The increased Metrnβeffectively blocked the development of airway hyperreactivity(AHR)and decreased inflammatory cell airway infiltration and type 2 cytokine production,which was associated with downregulated DC-mediated adaptive immune responses.Moreover,Metrnβimpaired the maturation and function of bone marrow-derived dendritic cells in vitro.Asthmatic mice adoptively transferred with dendritic cells isolated from Metrnβ-treated allergic mice displayed decreased AHR,airway inflammation,and lung injury.Metrnβalso displayed anti-inflammatory properties in immunodeficient SCID mice with allergic asthma and in in vitro 3D ALI airway models.Moreover,blockade of Metrnβby anti-Metrnβantibody treatment promoted the development of allergic asthma.These results revealed the unappreciated protective roles of Metrnβin alleviating DC-mediated Th2 inflammation in allergic asthma,providing the novel treatment strategy of therapeutic targeting of Metrnβin allergic asthma. | Xun Gao Ting-Fan Leung Gary Wing-Kin Wong Wing-Hung Ko Mengyun Cai Ellie Jiayi He Ida Miu-Ting Chu Miranda Sin-Man Tsang Ben Chung-Lap Chan Jiawei Ling Xiao Fan Liwei Lu Christopher Wai-Kei Lam Chun-Kwok Wong | 2022 | Cellular & Molecular Immunology2022,19,2: | 5 |
| 2 | A Detailed Study on the Equal Arrival Time Surface Effect in Gamma-Ray Burst Afterglows显示文摘Due to the relativistic motion of gamma-ray burst remnant and its deceleration in the circumburst medium, the equal arrival time surfaces at any moment are not spherical, rather, they are distorted ellipsoids. This will leave some imprints in the afterglows. We study the effect of equal arrival time surfaces numerically for various circumstances, i.e., isotropic fireballs, collimated jets, density jumps and energy injection events. For each case, a direct comparison is made between including and not including the effect. For isotropic fireballs and jets viewed on axis, the effect slightly hardens the spectra and postpones the peak time of the afterglows, but does not change the shapes of the spectra and light curves significantly. | 4 | Novel role of STAT3 in microglia-dependent neuroinflammation after experimental subarachnoid haemorrhage显示文摘Background and purpose Signal transducer and activator of transcription 3(STAT3)may contribute to the proinflammation in the central nervous system diseases by modulating the microglial responses.Thus,this study was intended to investigate the effect of STAT3 on microglia-dependent neuroinflammation and functional outcome after experimental subarachnoid haemorrhage(SAH).Methods The SAH model was established by endovascular perforation in the mouse.Real-time PCR(RtPCR)and western blot were used to examine the dynamic STAT3 signalling pathway responses after SAH.To clarify the role of the STAT3 signalling pathway in the microglia-dependent neuroinflammation after SAH,the microglia-specific STAT3 knockout(KO)mice were generated by the Cre-LoxP system.The neurological functions were assessed by Catwalk and Morris water maze tests.Neuronal loss after SAH was determined by immunohistochemistry staining.Microglial polarisation status after STAT3 KO was then examined by RtPCR and immunofluorescence.Results The STAT3 and Janus kinase-signal transducer 2 activated immediately with the upregulation and phosphorylation after SAH.Downstream factors and related mediators altered dynamically and accordingly.Microglial STAT3 deletion ameliorated the neurological impairment and alleviated the early neuronal loss after SAH.To investigate the underlying mechanism,we examined the microglial reaction after STAT3 KO.STAT3 deletion reversed the increase of microglia after SAH.Loss of STAT3 triggered the early morphological changes of microglia and primed microglia from M1 to M2 polarisation.Functionally,microglial STAT3 deletion suppressed the SAH-induced proinflammation and promoted the anti-inflammation in the early phase.Conclusions STAT3 is closely related to the microglial polarisation transition and modulation of microglia-dependent neuroinflammation.Microglial STAT3 deletion improved neurological function and neuronal survival probably through promoting M2 polarisation and anti-inflammatory responses after SAH.STAT3 may serve as a promising therapeutic target to alleviate early brain injury after SAH. | Zhiyuan Vera Zheng Junfan Chen Hao Lyu Sin Yu Erica Lam Gang Lu Wai Yee Chan George K C Wong | 2022 | Stroke & Vascular Neurology2022,7,1: | 3 |
| 5 | Comprehensive lipidomics in apoM^(-/-) mice reveals an overall state of metabolic distress and attenuated hepatic lipid secretion into the circulation显示文摘Apolipoprotein M(apoM) participates in both high-density lipoprotein and cholesterol metabolism. Little is known about how apo M affects lipid composition of the liver and serum. In this study, we systemically investigated the effects of apo M on liver and plasma lipidomes and how apo M participates in lipid cycling, via apo M knockout in mice and the human SMMC-7721 cell line. We used integrated mass spectrometry-based lipidomics approaches to semiquantify more than 600 lipid species from various lipid classes, which include free fatty acids, glycerolipids, phospholipids, sphingolipids, glycosphingolipids, cholesterol, and cholesteryl esters(CEs), in apo M^(-/-)mouse. Hepatic accumulation of neutral lipids, including CEs, triacylglycerols, and diacylglycerols, was observed in apo M^(-/-)mice;while serum lipidomic analyses showed that, in contrast to the liver, the overall levels of CEs and saturated/monounsaturated fatty acids were markedly diminished. Furthermore, the level of Apo B-100 was dramatically increased in the liver, whereas significant reductions in both Apo B-100 and low-density lipoprotein(LDL) cholesterol were observed in the serum of apo M^(-/-)mice, which indicated attenuated hepatic LDL secretion into the circulation. Lipid profiles and proinflammatory cytokine levels indicated that apo M^(-/-)leads to hepatic steatosis and an overall state of metabolic distress. | 17 | A facile design for multifunctional AIEgen based on tetraaniline derivatives显示文摘Aniline oligomers have been widely used in many fields due to their excellent physicochemical properties. Owing to strong intermolecular interactions, their emission is always weakened or quenched when they are in high concentration or aggregated state, which greatly limits their fluorescent applications. Inspired by the concept of aggregation-induced emission(AIE), herein we introduced large steric groups onto the aniline oligomer to prevent the formation of packing structure. In particular, diphenyl vinyl group was bonded with oligomeric tetraaniline by a facile synthetic procedure with high yield. The obtained aniline oligomer derivative exhibited typical AIE features, which was also confirmed by density functional theoretical calculation. More importantly, this AIE oligomer was able to detect Fe^(3+) ions selectively and quantitatively. The fluorescence intensity decreased linearly along with the increment of Fe^(3+) concentration. Moreover, we demonstrated that this AIE oligomer could stain live bacteria, such as E. coli and S. aureus efficiently.
