| 1 | Expression of gonadotropin-releasing hormone receptor and effect of gonadotropin-releasing hormone analogue on proliferation of cultured gastric smooth muscle cells of rats显示文摘AIM: To investigate the expression of gonadotropin-releasing hormone (GnRH) receptor and the effects of GnRH analog(alarelin) on proliferation of cultured gastric smooth muscle cells (GSMC) of rats. METHODS: Immunohistochemical ABC methods and in situ hybridization methods were used to dectect protein and mRNA expression of GnRH receptor in GSMC, respectively. Techniques of cell culture, OD value of MTT test, measure of ^3H-TdR incorporation, average fluorescent values of proliferating cell nuclear antigen (PCNA) and flow cytometric DNA analysis were used in the experiment. RESULTS: The cultured GSMC of rats showed immunoreactivity for GnRH receptor; positive staining was located in cytoplasm. GnRH receptor mRNA hybridized signals were also detected in cytoplasm. When alarelin (10^-9, 10^-7, 10^-5 mol/L) was administered into the medium and incubated for 24 h, OD value of MTT, ^3H-TdR incorporation and average fluorescent values of PCNA all decreased significantly as compared with the control group (P<0.05). The maximum inhibitory effect on cell proliferation was achieved a concentration of 10s mol/L and it acted in a dose-dependent manner. Flow cytometric DNA analysis revealed that alarelin could significantly enhance ratio of G1 phase and decrease ratio of S phase of GSMC of rats (P<0.05).The maximum inhibitory effect on ratio of S phase was at the concentration of 10-s mol/L and also acted in a dose-dependent manner. CONCLUSION: Our data suggest that GnRH receptor can be expressed by GSMC of rats. | 2 | Action of aluminum on high voltage-dependent calcium current and its modulation by ginkgolide B显示文摘Aim: To investigate the effect of aluminum (Al) on high voltage-dependent calcium current (IHVA) and its modulation by ginkgolide B (Gin B). Methods: The whole-cell, patch-clamp technique was used to record IHVA from acutely isolated hippocampal CA1 pyramydal neurons in rats. Results: A1 0.1mmol/L (low concentration) reduced IHVA; Al 0.75 and 1.0mmol/L (high concentrations) increased IHVA, and Al decreased and increased IHVA at intermediate concentrations of 0.25 and 0.5mmol/L. The increase of IHVA by Al 1.0mmol/L was enhanced by the adenylyl cyclase (AC) agonist forskolin and was partly abolished by the cyclic adenosine monophosphate (cAMP)-dependent protein kinase A (PKA) antagonist H-89, whereas the decrease observed with Al 0.1mmol/L was neither reversed by forskolin nor affected by H-89. Gin B had no effect on IHVA in normal neurons,but canceled the increase in IHVA by 1.0mmol/L Al. Conclusion: The results indicate that the mechanism of Al affecting IHVA differs at different concentrations,and this may be attributed to its complex actions. | 9 | Low-cycle fatigue behaviour of Mg-9Gd-4Y-2Zn-0.5Zr alloys with different structures显示文摘The strain-controlled cyclic deformation behaviour of Mg-9Gd-4Y-2Zn-0.5Zr with different structures was investigated. Alloys were prepared by solution, extrusion and pre-ageing extrusion, and the microstructures before and after the fatigue tests were characterized.Experimental results indicated that the bimodal structure owned the better performance in fatigue test, which was attributed to the higher yield strength. For the equiaxed structure, cyclic hardening induced stress concentration until the failure. Stable cyclic deformation and persistent cyclic softening played an important role at the low and high strain amplitudes, respectively. This was attributed to the formation of fine grains relieving the stress concentration during cyclic loading. | 10 | Grain refinement impact on the mechanical properties and wear behavior of Mg-9Gd-3Y-2Zn-0.5Zr alloy after decreasing temperature reciprocating upsetting-extrusion显示文摘Based on the deforming technique of severe plastic deformation(SPD), the grain refinement of a Mg-9Gd-3Y-2Zn-0.5Zr alloy treated with decreasing temperature reciprocating upsetting-extrusion(RUE) and its influence on the mechanical properties and wear behavior of the alloy were studied. The RUE process was carried out for 4 passes in total, starting at 0 ℃ and decreasing by 10 ℃ for each pass. The results showed that as the number of RUE passes increased, the grain refinement effect was obvious, and the second phase in the alloy was evenly distributed. Room temperature tensile properties of the alloy and the deepening of the RUE degree showed a positive correlation trend, which was due to the grain refinement, uniform distribution of the second phase and texture weakening. And the microhardness of the alloy showed that the microhardness of RUE is the largest in 2 passes. The change in microhardness was the result of dynamic competition between the softening effect of DRX and the work hardening effect. In addition, the wear resistance of the alloy showed a positive correlation with the degree of RUE under low load conditions. When the applied load was higher, the wear resistance of the alloy treated with RUE decreased compared to the initial state alloy. | 11 | Tonic activation of presynaptic GABAB receptors on rat pallidosubthalamic terminals显示文摘Aim: The subthalamic nucleus plays a critical role in the regulation of movement, and abnormal activity of its neurons is associated with some basal ganglia motor symptoms. We examined the presence of functional presynaptic GABAB receptors on pallidosubthalamic terminals and tested whether they were tonically active in the in vitro subthalamic slices. Methods: Whole-cell patch-clamp recordings were applied to acutely prepared rat subthalamic nucleus slices. The effects of specific GABAB agonist and antagonist on action potential-independent inhibitory postsynaptic currents (IPSCs), as well as holding current, were examined. Results: Superfusion of baclofen, a GABAa receptor agonist, significantly reduced the frequency of GABAA receptor-mediated miniature IPSCs (mIPSCs), ina Cd^2+-sensitive manner, with no effect on the amplitude, indicating presynaptic inhibition on GABA release. In addition, baclofen induced a weak outward current only in a minority of subthalamic neurons. Both the pre-and post-synaptic effects of baclofen were prevented by the specific GABAB receptor antagonist, CGP55845. Furthermore, CGP55845 alone increased the frequency of mIPSCs, but had no effect on the holding current. Conclusion: These findings suggest the functional dominance of presynaptic GABAB receptors on the pallidosubthalamic terminals over the postsynaptic GABAB receptors on subthalamic neurons.
