1New possible silver lining for pancreatic cancer therapy:Hydrogen sulfide and its donors显示文摘As one of the most lethal diseases,pancreatic cancer shows a dismal overall prognosis and high resistance to most treatment modalities.Furthermore,pancreatic cancer escapes early detection during the curable period because early symptoms rarely emerge and specific markers for this disease have not been found.Although combinations of new drugs,multimodal therapies,and adjuvants prolong survival,most patients still relapse after surgery and eventually die.Consequently,the search for more effective treatments for pancreatic cancer is highly relevant and justified.As a newly re-discovered mediator of gasotransmission,hydrogen sulfide(H2S)undertakes essential functions,encompassing various signaling complexes that occupy key processes in human biology.Accumulating evidence indicates that H2S exhibits bimodal modulation of cancer development.Thus,endogenous or low levels of exogenous H2S are thought to promote cancer,whereas high doses of exogenous H2S suppress tumor proliferation.Similarly,inhibition of endogenous H2S production also suppresses tumor proliferation.Accordingly,H2S biosynthesis inhibitors and H2S supplementation(H2S donors)are two distinct strategies for the treatment of cancer.Unfortunately,modulation of endogenous H2S on pancreatic cancer has not been studied so far.However,H2S donors and their derivatives have been extensively studied as potential therapeutic agents for pancreatic cancer therapy by inhibiting cell proliferation,inducing apoptosis,arresting cell cycle,and suppressing invasion and migration through exploiting multiple signaling pathways.As far as we know,there is no review of the effects of H2S donors on pancreatic cancer.Based on these concerns,the therapeutic effects of some H2S donors and NO-H2S dual donors on pancreatic cancer were summarized in this paper.Exogenous H2S donors may be promising compounds for pancreatic cancer treatment.Xu Hua Yan Xiao Jianan Sun Bao Ji Shanshan Luo Bo Wu Chao Zheng Peng Wang Fanxing Xu Keguang Cheng Huiming Hua Dahong Li 2021Acta Pharmaceutica Sinica B2021,11,5:4
2Synthesis of selective PAK4 inhibitors for lung metastasis of lung cancer and melanoma cells显示文摘The p21 activated kinase 4(PAK4) is serine/threonine protein kinase that is critical for cancer progression.Guided by X-ray crystallography and structure-based optimization,we report a novel subseries of C-3-substituted 6-ethynyl-1 H-indole derivatives that display high potential and specificity towards group Ⅱ PAKs.Among these inhibitors,compound 55 exhibited excellent inhibitory activity and kinase selectivity,displayed superior anti-migratory and anti-invasive properties against the lung cancer cell line A549 and the melanoma cell line B16.Compound 55 exhibited potent in vivo antitumor metastatic efficacy,with over 80% and 90% inhibition of lung metastasis in A549 or B16-BL6 lung metastasis models,respectively.Further mechanistic studies demonstrated that compound 55 mitigated TGF-β1-induced epithelial-mesenchymal transition(EMT).Peilu Song Fan Zhao Dahong Li Jiqiang Qu Miao Yao Yuan Su Hanxun Wang Miaomiao Zhou Yujie Wang Yinli Gao Feng Li Dongmei Zhao Fengjiao Zhang Yu Rao Mingyu Xia Haitao Li Jian Wang Maosheng Cheng 2022Acta Pharmaceutica Sinica B2022,12,6:2

5Thermoluminescence and fission track impact features of rocks显示文摘Results of thermoluminescence (TL) measurement of quartz sandstone before and after impact using a two-stage light gas gun pressure apparatus indicate that, the TL of quartz sandstone decreases as the impact pressure increases, there exists a TL gradient from the outside to the inside around the impacted area, and the induced TL at the same location also has a TL gradient. Impact experiments were conducted under pressures of 12.4, 28.9, 37.8, 51.8, 64.6 and 77.6 GPa, respectively, and fission track density and fission track length before and after the impact were measured. 6Effect of esculetin on bone metabolism in ovariectomized rats显示文摘OBJECTIVE: To determine the effect of an esculetin formulation(at 97.4% purity) on osteoporosis, and to investigate the potential underlying molecular mechanism(s).METHODS: Sixty specific pathogen free-grade female Wistar rats were randomly assigned to three groups: blank control(n = 12), sham(n = 12), and model(n = 36). The model group were bilaterally ovariectomized. The sham group had the tissue surrounding the ovaries removed, while the ovaries were retained. After 3 months, the model group was randomly divided into three subgroups: OVX(n = 12), positive control(n = 12), and esculetin(n =12). The positive control group and the esculetin group were intragastrically administered diethylstilbestrol(0.046 mgkd^(-1)), respec·kg^(-1)tively,·d^(-1)) or esculetin(384 mgg^(-1) once per day for 6 consecu··-tive days; medication administration was then stopped for 1 d, before being administered for another 6 consecutive days. All rats were treated for 3months. Samples were collected at the end of the treatment period. An Osteocore3 Digital 2D bone densitometer was used to test the bone mineral density, and histomorphometric analysis was performed to measure bone mass, bone formation,and bone resorption. Enzyme-linked immunosorbent assay analysis was used to measure the serum concentrations of interleukin-6(IL-6), osteoprotegerin(OPG), and receptor activator of nuclear factor-kappa B ligand(RANKL). Immunohistochemistry and in situ hybridization were performed to detect the protein and mRNA expressions of OPG and RANKL in osteoblasts and bone marrow stromal cells.RESULTS: Compared with the OVX group, the esculetin group had significantly greater femoral bone mineral density and tibial trabecular bone volume,and significantly smaller trabecular resorption surface. The percentage of trabecular formation surface, average osteoid width, trabecular bone mineralization rate, and cortical bone mineralization rate did not significantly differ between groups. Compared with the sham group, the esculetin group had significantly decreased serum levels of IL-6and RANKL, and significant downregulation of RANKL protein and mRNA expression levels in osteoblasts and bone marrow stromal cells; however, there was no significant difference between groups in OPG.CONCLUSION: Esculetin can increase bone mass by upregulating RANKL expression in osteoblasts and bone marrow stromal cells, and decreasing serum IL-6 concentration. 8Tum or Necrosis Factor Expression in ArterialWallsof Diabetic Rats显示文摘Immunohistochemistry was used to detect tumor necrosis factor (TNF α) expression in arterial wall of diabetic rats. It was found that endothelial cells were swollen and markedly proliferative in these vessels and accordingly TNF α showed strong positive immunohistochemical reaction in endothelial cells or extracellular intimal matrix of such vessels, which might be caused by the expression and release of TNF α from monocytes and arterial wall cells stimulated by AGEs. 14Twelve-week study of moxa smoke: occupational exposure in female rats显示文摘OBJECTIVE: To assess the toxicity of moxa smoke in rats.METHODS: Forty-eight female Wister rats were randomly divided into 4 groups(n = 12/group) to simulate moxa smoke exposure in Chinese medicine clinics(CMCs): the control group, and three moxa-smoke exposed groups of PM10 mass concentrations 3-5, 7-9 and 27-30 mg/m^3, respectively.These concentrations were 1 ×, 2-3 ×, and 7-9 ×fold the concentrations found in CMCs. Exposures continued for 12 weeks(200 min/d, 5 d/week).RESULTS: No deaths were noted. After the exposure, the body weights, ratios of organ weight to body weight, urinary parameters, hematological parameters, clinical chemistry parameters and microscopic examinations revealed no obvious toxicity.CONCLUSION: Moxa smoke did not induce toxic effects in female rats in the study. 16β-Carboline alkaloids from the roots of Peganum harmala L.显示文摘This study reports the isolation of four newβ-carboline alkaloids(1−4)and six previously identified alkaloids(5−10)from the roots of Peganum harmala L.Among these compounds,1 and 2 were characterized as rareβ-carboline-quinazoline dimers exhibiting axial chirality.Compound 3 possessed a unique 6/5/6/7 tetracyclic ring system with an azepine ring,and compound 4 was a novel annomontineβ-carboline.The structures of these compounds were elucidated by spectroscopic data and quantum mechanical calculations.The biosynthetic pathways of 1−3 were proposed.Additionally,the cytotoxicity of some isolates against four cancer cell lines(HL-60,A549,MDA-MB-231,and DU145)was evaluated.Notably,compound 4 exhibited significant cytotoxicity against HL-60,A549,and DU145 cells with IC_(50) values of 12.39,12.80,and 30.65μmol·L^(−1),respectively.Furthermore,compound 2 demonstrated selective cytotoxicity against HL-60 cells with an IC_(50) value of 17.32μmol·L^(−1).LI Shengge ZHANG Qin WANG Yuetong LIN Bin LI Dahong HUA Huiming HU Xu 2024Chinese Journal of Natural Medicines2024,22,2:0
