1Nonhuman primate models of focal cerebral ischemia显示文摘Rodents have been widely used in the production of cerebral ischemia models. However, successful therapies have been proven on experimental rodent stroke model, and they have often failed to be effective when tested clinically. Therefore, nonhuman primates were recommended as the ideal alternatives, owing to their similarities with the human cerebrovascular system, brain metabolism, grey to white matter ratio and even their rich behavioral repertoire. The present review is a thorough summary of ten methods that establish nonhuman primate models of focal cerebral ischemia; electrocoagulation, endothelin-1-induced occlusion, microvascular clip occlusion, autologous blood clot embolization, balloon inflation, microcatheter embolization, coil embolization, surgical suture embolization, suture, and photochemical induction methods. This review addresses the advantages and disadvantages of each method, as well as precautions for each model, compared nonhuman primates with rodents, different species of nonhuman primates and different modeling methods. Finally it discusses various factors that need to be considered when modelling and the method of evaluation after modelling. 2Frequent Mutation of rs13281615 and Its Association with PVT1 Expression and Cell Proliferation in Breast Cancer显示文摘The q24 band of chromosome 8(8q24)is frequently amplified in human cancers including breast cancer,and several SNPs(single nucleotide polymorphisms)at 8q24,including rs13281615,have been identified for their association with cancer risks.These SNPs are in a'gene desert'region,and their functions in cancer development remain to be illustrated,although several of the SNPs appear to influence the genes in the'desert'in a long-range manner,including the v-myc avian myelocytomatosis viral oncogene homolog(MYC)and the nonprotein coding plasmacytoma variant translocation 1(PVT1),both of which have been implicated in human cancers.In the current study,we examined rs13281615 for its potential role in breast cancer using normal and cancer tissues from 121 Chinese women with breast cancer.In addition to confirming the association of the GG genotype of rs13281615 with breast cancer risk,we found that germline GG genotype was significantly associated with estrogen receptor(ER)positivity,higher tumor grade and higher proliferation index.We also found frequent somatic mutations(22/121 or 18.2%)of this SNP in breast cancer.Interestingly,the majority of the mutations(17/22 or 77%)involved a G/A change,resulting in a decrease in the number of cancers with the GG risk genotype and subsequent loss of GG association with higher tumor grade and proliferation index in cancers.Furthermore,PVT1 expression was increased in cancers,and the increase was associated with the GG genotype of rs13281615.These results suggest that the GG genotype of SNP rs13281615 plays a role in breast cancer likely by influencing PVT1 expression,and that during oncogenesis,'protective'mutations could occur.Zhiqian Zhang Zhengmao Zhu Baotong Zhang Weidong Li Xin Li Xiao Wu Lijuan Wang Liya Fu Li Fu Jin-Tang Dong 2014Journal of Genetics and Genomics2014,41,4:12
3Transcriptomic landscape regulated by the 14 types of bone morphogenetic proteins(BMPs)in lineage commitment and differentiation of mesenchymal stem cells(MSCs)显示文摘Mesenchymal stem cells(MSCs)are ubiquitously-existing multipotent progenitors that can self-renew and differentiate into multiple lineages including osteocytes,chondrocytes,adipocytes,tenocytes and myocytes.MSCs represent one of the most commonly-used adult progenitors and serve as excellent progenitor cell models for investigating lineagespecific differentiation regulated by various cellular signaling pathways,such as bone morphogenetic proteins(BMPs).As members of TGFb superfamily,BMPs play diverse and important roles in development and adult tissues.At least 14 BMPs have been identified in mammals.Different BMPs exert distinct but overlapping biological functions.Through a comprehensive analysis of 14 BMPs in MSCs,we demonstrated that BMP9 is one of the most potent BMPs in inducing osteogenic differentiation of MSCs.Nonetheless,a global mechanistic view of BMP signaling in regulating the proliferation and differentiation of MSCs remains to be fully elucidated.Here,we conducted a comprehensive transcriptomic profiling in the MSCs stimulated by 14 types of BMPs.Hierarchical clustering analysis classifies 14 BMPs into three subclusters:an osteo/chondrogenic/adipogenic cluster,a tenogenic cluster,and BMP3 cluster.We also demonstrate that six BMPs(e.g.,BMP2,BMP3,BMP4,BMP7,BMP8,and BMP9)can induce ISmads effectively,while BMP2,BMP3,BMP4,BMP7,and BMP11 up-regulate Smad-independent MAP kinase pathway.Furthermore,we show that many BMPs can upregulate the expression of the signal mediators of Wnt,Notch and PI3K/AKT/mTOR pathways.While the reported transcriptomic changes need to be further validated,our expression profiling represents the first-of-its-kind to interrogate a comprehensive transcriptomic landscape regulated by the 14 types of BMPs in MSCs.Linghuan Zhang Qing Luo Yi Shu Zongyue Zeng Bo Huang Yixiao Feng Bo Zhang Xi Wang Yan Lei Zhenyu Ye Ling Zhao Daigui Cao Lijuan Yang Xian Chen Bin Liu William Wagstaff Russell R*Reid Hue H*Luu Rex C*Haydon Michael J*Lee Jennifer Moriatis Wolf Zhou Fu Tong-Chuan He Quan Kang 2019Genes & Diseases2019,6,3:11
4Dynamic channel reservation scheme based on priorities in LEO satellite systems显示文摘According to low earth orbit(LEO) satellite systems with users of different levels, a dynamic channel reservation scheme based on priorities is proposed. Dynamic calculation of the thresholds for reserved channels is the key of this strategy. In order to obtain the optimal thresholds, the traffic is predicted based on the high-speed deterministic movement property of LEO satellites firstly. Then, a channel allocation model based on Markov is established. Finally, the solution of the model is obtained based on the genetic algorithm. Without user location, this strategy effectively reduces handover failures and improves channel utilization by adjusting dynamically the thresholds according to traffic conditions. 6Genetic contribution of foreign germplasm to elite Chinese soybean (Glycine max) cultivars revealed by SSR markers显示文摘Simple sequence repeats (SSR) marker analysis, combined with pedigree analysis, was car- ried out to trace the genetic constitutes of Suinong 14 and Hefeng 25 in assessing the contribution of for- eign germplasm to those elite cultivars. The overall goal is to accumulate information for further effective utilization of foreign germplasm in Chinese soybean breeding programs. SSR clustering results indicated that the genetic base of elite Chinese soybean culti- vars, including Suinong 14 and Hefeng 25, were broadened and enhanced via introduction of Amsoy from the U.S. and Shishengchangye from Japan. The pedigree analysis showed a very high coefficient of parentage between Shishengchangye and Hefeng 25, between Shishengchangye and Suinong 14, and between Amsoy and Suinong 14. The genetic simi- larity between Suinong 14 and Hefeng 25 was 60.58%. Among 20 linkage groups, more genomic regions were transferred from Hefeng 25 to Suinong 14 in linkage group (LG) I, L and C2 than other LGs. Among the unique alleles of foreign parents, five specific loci in Amsoy were transferred to Suinong 14, and three loci from Shishengchangye to Suinong 14. Some SSR loci were proved to be correlated with phenotypes: two loci for seed size introgressed from Shishengchangye to Suinong 14, and one locus forprotein content from Amsoy to Suinong 14. 7Information acquisition system of multipoint soil surface height variation for profiling mechanism of seeding unit of precision corn planter显示文摘The emergence rate and vitality of maize are directly affected by the sowing depth,and the uniformity of this depth is an important performance indicator of a planter,while the effective soil surface height information acquisition is the prerequisite for ensuring the accuracy of sowing depth control.The soil surface height variation acquisition system of a precision corn planter often produces profiling errors when performing active profiling due to interference from ground debris.In this study,a multipoint soil surface height variation information acquisition system was investigated,which consists of a ranging sensor group and a microcontroller unit(MCU)using a data comparison and screening method.The structure and specifications of the ranging sensors were determined according to the soil surface height variation and debris size,and a nonessential profiling control program was developed.Performed tests on the information acquisition system indicated that the measurement accuracy of the system was 3 mm,and when advancing at a speed of 8 km/h,the accuracy of the profiling decision and the system stability were 97.1%and 94.1%,respectively,indicating that the system was capable of nonessential profile control.The designed ranging system could provide a reference for the design of a ground information acquisition system of precision planters with an active profiling mechanism.Bo Zhang Wei Zhang Liqiang Qi Haiba Fu Lijuan Yu Rui Li Yue Zhao Xinxian Ma 2018International Journal of Agricultural and Biological Engineering2018,11,6:4
8Surface Coverage Algorithm in Directional Sensor Networks for Three-Dimensional Complex Terrains显示文摘Coverage is an important issue in the area of wireless sensor networks, which reflects the monitoring quality of the sensor networks in scenes. Most sensor coverage research focuses on the ideal two-dimensional(2-D) plane and full three-dimensional(3-D) space. However, in many real-world applications, the target field is a3-D complex surface, which makes conventional methods unsuitable. In this paper, we study the coverage problem in directional sensor networks for complex 3-D terrains, and design a new surface coverage algorithm. Based on a 3-D directional sensing model of nodes, this algorithm employs grid division, simulated annealing, and local optimum ideas to improve the area coverage ratio by optimizing the position coordinates and the deviation angles of the nodes, which results in coverage enhancement for complex 3-D terrains. 9Robust and Passive Motion Detection with COTS WiFi Devices显示文摘Device-free Passive(DfP) detection has received increasing attention for its ability to support various pervasive applications. Instead of relying on variable Received Signal Strength(RSS), most recent studies rely on finer-grained Channel State Information(CSI). However, existing methods have some limitations, in that they are effective only in the Line-Of-Sight(LOS) or for more than one moving individual. In this paper, we analyze the human motion effect on CSI and propose a novel scheme for Robust Passive Motion Detection(R-PMD). Since traditional low-pass filtering has a number of limitations with respect to data denoising, we adopt a novel Principal Component Analysis(PCA)-based filtering technique to capture the representative signals of human motion and extract the variance profile as the sensitive metric for human detection. In addition, existing schemes simply aggregate CSI values over all the antennas in MIMO systems. Instead, we investigate the sensing quality of each antenna and aggregate the best combination of antennas to achieve more accurate and robust detection. 10Complement C3a activates osteoclasts by regulating the PI3K/PDK1/SGK3 pathway in patients with multiple myeloma显示文摘Objective:Myeloma bone disease(MBD)is the most common complication of multiple myeloma(MM).Our previous study showed that the serum levels of C3/C4 in MM patients were significantly positively correlated with the severity of bone disease.However,the mechanism of C3 a/C4 a in osteoclasts MM patients remains unclear.Methods:The formation and function of osteoclasts were analyzed after adding C3 a/C4 a in vitro.RNA-seq analysis was used to screen the potential pathways affecting osteoclasts,and the results were verified by Western blot,q RT-PCR,and pathway inhibitors.Results:The osteoclast area per view induced by 1μg/m L(mean±SD:50.828±12.984%)and 10μg/m L(53.663±12.685%)of C3 a was significantly increased compared to the control group(0μg/m L)(34.635±8.916%)(P<0.001 and P<0.001,respectively).The relative m RNA expressions of genes,OSCAR/TRAP/RANKL/cathepsin K,induced by 1μg/m L(median:5.041,3.726,1.638,and 4.752,respectively)and 10μg/m L(median:5.140,3.702,2.250,and 5.172,respectively)of C3 a was significantly increased compared to the control group(median:3.137,2.004,0.573,and 2.257,respectively)(1μg/m L P=0.001,P=0.003,P<0.001,and P=0.008,respectively;10μg/m L:P<0.001,P=0.019,P<0.001,and P=0.002,respectively).The absorption areas of the osteoclast resorption pits per view induced by 1μg/m L(mean±SD:51.464±11.983%)and 10μg/m L(50.219±12.067%)of C3 a was also significantly increased(33.845±8.331%)(P<0.001 and P<0.001,respectively)compared to the control.There was no difference between the C4 a and control groups.RNA-seq analysis showed that C3 a promoted the proliferation of osteoclasts using the phosphoinositide 3-kinase(PI3 K)signaling pathway.The relative expressions of PIK3 CA/phosphoinositide dependent kinase-1(PDK1)/serum and glucocorticoid inducible protein kinases(SGK3)genes and PI3 K/PDK1/p-SGK3 protein in the C3 a group were significantly higher than in the control group.The activation role of C3 a in osteoclasts of MM patients was reduced by the SGK inhibitor(EMD638683).Conclusions:C3 a activated osteoclasts by regulating the PI3 K/PDK1/SGK3 pathways in MM patients,which was reduced using a SGK inhibitor.Overall,our results identified potential therapeutic targets and strategies for MBD patients。Fengjuan Jiang Hui Liu Fengping Peng Zhaoyun Liu Kai Ding Jia Song Lijuan Li Jin Chen Qing Shao Siyang Yan Kim De Veirman Karin Vanderkerken Rong Fu 2021Cancer Biology & Medicine2021,18,3:3
11PHYTOCHROME-INTERACTING FACTORS Interact with the ABA Receptors PYL8 and PYL9 to Orchestrate ABA Signaling in Darkness显示文摘PHYTOCHROME-INTERACTING FACTORS(PIFs)are a group of basic helix-loop-helix transcription factors that can physically interact with photoreceptors,including phytochromes and cryptochromes.It was previously demonstrated that PIFs accumulated in darkness and repressed seedling photomorphogenesis,and that PIFs linked different photosensory and hormonal pathways to control plant growth and development.In this study,we show that PIFs positively regulate the ABA signaling pathway during the seedling stage specifically in darkness.We found that PIFs positively regulate ABI5 transcript and protein levels in darkness in response to exogenous ABA treatment by binding directly to the G-box motifs in the ABI5 promoter.Consistently,PIFs and the G-box motifs in the ABI5 promoter determine ABI5 expression in darkness,and overexpression of ABI5 could rescue the ABA-insensitive phenotypes of pifq mutants in the dark.Moreover,we discovered that PIFs can physically interact with the ABA receptors PYL8 and PYL9,and that this interaction is not regulated by ABA.Further analyses showed that PYL8 and PYL9 promote PIF4 protein accumulation in the dark and enhance PIF4 binding to the ABI5 promoter,but negatively regulate PIF4-mediated ABI5 activation.Taken together,our data demonstrate that PIFs interact with ABA receptors to orchestrate ABA signaling in darkness by controlling ABI5 expression,providing new insights into the pivotal roles of PIFs as signal integrators in regulating plant growth and development.Lijuan Qi Shan Liu Cong Li Jingying Fu Yanjun Jing Jinkui Cheng Hong Li Dun Zhang Xiaoji Wang Xiaojing Dong Run Han Bosheng Li Yu Zhang Zhen Li William Terzaghi Chun-Peng Song Rongcheng Lin Zhizhong Gong Jigang Li 2020Molecular Plant2020,13,3:3
12Application of structural curvature of coalbed floor on CBM development in the Zaoyuan block of the southern Qinshui Basin显示文摘Based on controls of structural style and the position in coalbed methane (CBM) development, we used a method of curvatures to study its relations with CBM development parameters. We calculated structural curvatures of contours of the No.3 coal seam floor of the Shanxi Formation in the Zaoyuan block of the Qinshui Basin and analyzed its relations with development parameters of coalbed methane wells. The results show that structural curvature is negatively related to coal reservoir pressure, while positively related to permeability. With an increase in structural curvature, the average production of coalbed methane wells increases at first and then decreases, reaching the highest production at 0.02 m–1 of structural curvature. Therefore, structural cur-vature can be an important index for potential evaluation of coalbed methane development and provide a basis for siting coalbed methane wells.SHEN Jian 2010Mining Science and Technology2010,20,6:2
