2USP19 suppresses inflammation and promotes M2-like macrophage polarization by manipulating NLRP3 function via autophagy显示文摘Macrophage polarization to proinflammatory M1-like or anti-inflammatory M2-like cells is critical to mount a host defense or repair tissue.The exact molecular mechanisms controlling this process are still elusive.Here,we report that ubiquitin-specific protease 19(USP19)acts as an anti-inflammatory switch that inhibits inflammatory responses and promotes M2-like macrophage polarization.USP19 inhibited NLRP3 inflammasome activation by increasing autophagy flux and decreasing the generation of mitochondrial reactive oxygen species.In addition,USP19 inhibited the proteasomal degradation of inflammasome-independent NLRP3 by cleaving its polyubiquitin chains.USP19-stabilized NLRP3 promoted M2-like macrophage polarization by direct association with interferon regulatory factor 4,thereby preventing its p62-mediated selective autophagic degradation.Consistent with these observations,compared to wild-type mice,Usp19−/−mice had decreased M2-like macrophage polarization and increased interleukin-1βsecretion,in response to alum and chitin injections.Thus,we have uncovered an unexpected mechanism by which USP19 switches the proinflammatory function of NLRP3 into an anti-inflammatory function,and suggest that USP19 is a potential therapeutic target for inflammatory interventions.Tao Liu Liqiu Wang Puping Liang Xiaojuan Wang Yukun Liu Jing Cai Yuanchu She Dan Wang Zhi Wang Zhiyong Guo Samuel Bates Xiaojun Xia Junjiu Huang Jun Cui 2021Cellular & Molecular Immunology2021,18,10:15
3Promotion of spinosad biosynthesis by chromosomal integration of the Vitreoscilla hemoglobin gene in Saccharopolyspora spinosa显示文摘To promote spinosad biosynthesis by improving the limited oxygen supply during high-density fermentation of Saccharopolyspora spinosa, the open reading frame of the Vitreoscilla hemoglobin gene was placed under the control of the promoter for the erythromycin resistance gene by splicing using overlapping extension PCR. This was cloned into the integrating vector pSET152, yielding the Vitreoscilla hemoglobin gene expression plasmid pSET152EVHB. This was then introduced into S. spinosa SP06081 by conjugal transfer, and integrated into the chromosome by site-specific recombination at the integration site ΦC31 on pSET152EVHB. The resultant conjugant, S. spinosa S078-1101, was genetically stable. The integration was further confirmed by PCR and Southern blotting analysis. A carbon monoxide differential spectrum assay showed that active Vitreoscilla hemoglobin was successfully expressed in S. spinosa S078-1101. Fermentation results revealed that expression of the Vitreoscilla hemoglobin gene significantly promoted spinosad biosynthesis under normal oxygen and moderately oxygen-limiting conditions (P<0.01). 5Effect of additive boron on type-Ib gem diamond single crystals synthesized under HPHT显示文摘The boron-doped type-Ib gem diamond crystals were synthesized successfully by adding amorphous boron into a system of graphite and Kovar catalyst under high pressure and high temperature (HPHT).The effect of additive boron on type-Ib gem diamond was extensively studied including the growth characteristic,morphology and nitrogen concentration.The synthesized boron-doped type-Ib gem diamond crystals were characterized by optical microscope (OM),scanning electron microscope (SEM) and infrared spectrometer (IR).The results show that the growth region of the {111} face becomes wide,whereas the growth region of the {100} face becomes narrow and nearly disappears as increasing additive boron in the sample.The crystal will be opaque and imperfect,and the concentration of nitrogen will be decreased when the boron atoms are incorporated into gem diamond lattice.These techniques are very important and will be widely applied.MA LiQiu MA HongAn XIAO HongYu LI ShangSheng LI Yong JIA XiaoPeng 2010Chinese Science Bulletin2010,55,8:8
6Influencing factors and degradation products of antipyrine chlorination in water with free chlorine显示文摘Owing to its low cost, free chlorine is one of the most common disinfectants for wastewater and drinking water treatment. However, the formation of disinfection byproducts has been found to occur after free chlorine disinfection in recent decades. Antipyrine (ANT), an anti-inflammatory analgesic, has been frequently detected in the aquatic environment. In this work, the removal efficiency of ANT by free chlorine oxidation in ultrapure water was investigated with batch experiments. The influencing factors on the removal of ANT were explored at initial concentrations of ANT from 0.04 to 0.64 mg/L, free chlorine dosage from 0.30 to 1.31 mg/L, and pH from 1.5 to 9.0. The main degradation products were identified by solid phase extraction-gas chromatography-mass spectrometry. The results showed that ANT reacted rapidly with free chlorine in ultrapure water systems and up to 90.6% removal efficiency of ANT was achieved after 25 sec (initial free chlorine 1 mg/L, ANT 0.5 mg/L, pH 7.0). Higher oxidant dosage, lower ANT initial concentration and low pH favor the ANT removal. The main degradation product in ANT chlorination was a monochlorine substitution product (4-chloro-1,2-dihydro- 1,5-dimethyl-2-phenyl-3H-pyrazol-3-one), which can be further chlorinated by free chlorine. 7Effects of carbon diffusing field in alloy solvent on the growth of tower-shape diamond single crystal显示文摘High-quality type-Ib tower-shape diamond single crystals were synthesized in cubic anvil high pressure apparatus (SPD-6×1200) at 5.4 GPa and 1250-1450°C. The (100) face of seed crystal was used as the growth face, and FeNiMnCo alloy was used as the solvent/catalyst. Two kinds of carbon diffusing fields (type-B and type-G) were simulated by finite element method (FEM). Using the two kinds of carbon diffusing fields, many diamond single crystals were synthesized. The effects of carbon diffusing fields on the crystal quality and β value (the ratio of height to diameter of diamond crystal) were studied. The results show that using type-B diffusing field, considerable inclusions appeared in tower-shape diamond crystals with high β values; however, using type- G diffusing field, inclusions were reduced markedly in high β values tower-shape diamond crystals, and the crystal, up to 0.6 carat in weight and 5.3 mm in size, was synthesized. 8Dependence of nitrogen concentration in type Ib diamonds on synthesis temperature显示文摘Type Ib diamonds were grown by the temperature gradient method (TGM) at 5.5 GPa and 1500-1560 K in a china-type cubic anvil high pressure apparatus using Ni70Mn25Co5 alloy as solvent/catalyst. The concentration of nitrogen (CN) in type Ib diamonds synthesized at different synthesis temperatures was measured by a Fourier transform infrared (FTIR) spectrometer. The dependence of CN in diamond on synthesis temperature was studied. 10Downregulation of long non-coding RNA MEG3 promotes proliferation, migration, and invasion of human hepatocellular carcinoma cells by upregulating TGF-β1显示文摘Hepatocellular carci no ma is a common malig nant can cer with high incide nee. And long noncoding RNAs (lncRNAs) play pivotal roles in the development of different types of cancers. In this study, we aimed to investigate the role of lncRNA maternally expressed gene 3 (MEG3) in the development and progression of hepatocellular carcinoma. Expression of MEG3 in tumor tissues and adjacent healthy tissues of hepatocellular carcinoma patients, as well as the serum of both hepatocellular carcinoma patients and healthy controls, was detected by quantitative reverse transcriptase-polymerase chain reaction. The results showed that expression level of MEG3 was significantly lower in tumor tissues than in adjacent healthy tissues. Serum level of MEG3 was also significantly lower in hepatocellular carci noma patients tha n in normal controls. The receiver operating characteristic curve analysis was used to evaluate the diagnostic value of MEG3 for hepatocellular carcinoma, and the prognostic value of MEG3 for this disease was analyzed using Kaplan-Meier method. The results indicated that serum level of MEG3 was a diagnostic and prognostic marker for hepatocellular carcinoma. We also found that MEG3 small interfering Ribonucleic Acid (siRNA) silencing promoted the proliferation, migration, and invasi on of hepatocellular carci noma cells by CCK-8 assay, transwell migration, and invasion assay, respectively, while TGF-β inhibitor treatment reduced those enhancing effects. MEG3 siRNA silencing also increased the expression level of TGF-β1. 11The role of β18-β19 loop structure in insecticidal activity of Cry1Ac toxin from Bacillus thuringiensis显示文摘The β18-β19 loop in domain III of Cry1Ac toxin is unique among Bacillus thuringiensis Cry proteins. In this study, the role of the loop structure in insecticidal activity of Cry1Ac toxin was investigated. Alanine scanning mutations within the loop were initially generated and most mutants were over-expressed and reduced toxicity at different degrees, except mutant N546A that showed almost 2 times enhanced toxicity against Helicoverpa armigera larvae. Further mutagenic analysis of N546 revealed that a charged amino acid in this position would cause very unfavorable influence on insecticidal activity. In addition, the deletion of N546 led to protein instability because of destruction of the loop integrity. Besides, mutant W544F was much more toxic than W544Y, indicating that hydrophobic nature of the position was important for maintaining the stability and activity of Cry1Ac protein. 12Long non-coding RNA LINC-PINT attenuates paclitaxel resistance in triple-negative breast cancer cells via targeting the RNA-binding protein NONO显示文摘The treatment of triple-negative breast cancer(TNBC)relies largely on chemotherapies.However,it is frequent that TNBC patients develop resistance to the chemotherapy drugs.Generation of drug-resistant cell lines facilitates the identification of drug resistance.Here,we established two paclitaxel(PTX)-resistant TNBC cancer cell lines using an intermittent and stepwise method and found that long non-coding RNA long intergenic non-protein-coding RNA p53-induced transcript(LINC-PINT)was significantly decreased in PTX-resistant cancer cells.Ectopic expression of LINC-PINT sensitized both PTX-resistant TNBC and wild-type TNBC to PTX.Moreover,RNA immunoprecipitation showed that LINC-PINT bound to RNA-binding protein NONO.Overexpression of LINC-PINT resulted in the degradation of NONO in a proteasome-dependent manner and vice versa.Knockdown of NONO with siRNA sensitized TNBC to PTX.We further analyzed the expression level of LINC-PINT and NONO in patient samples via online database and found that LINC-PINT and NONO may function antagonistically in all types of breast cancers.Taken together,our data illustrated a tumor suppressor role of LINC-PINT in sensitizing TNBC to chemotherapies via destabilizing NONO.Jinghua Chen Meiqin Zhu Liqiu Zou Junxian Xia Jiacheng Huang Quantong Deng Ruilian Xu 2020Acta Biochimica et Biophysica Sinica2020,52,8:4
13A biomass carbon material with microtubule bundling and natural O-doping derived from goldenberry calyx and its electrochemical performance in supercapacitor显示文摘A promising biomass carbon material,manufactured by the carbonation of Physalis peruviana L.calyx at 700℃,is presented in this wo rk.Morphology characterization shows that the carbon material possesses long microtubule bundling and above 30%natural O-atom component on the surface.After KOH chemical etching,the materials maintain the oxygen content but exhibit more micropores and higher specific surface area up to 1732.6 m^2/g.Using as an electrode material for supercapacitor,the active carbon material exhibits high specific capacitance up to 339.7 F/g at 0.5 A/g in 3 mol/L KOH aqueous solution through three-electrode system.The active carbon material also exhibits excellent cycling stability(97%retention)by 10,000 cycles at 10 A/g.The outstanding electrochemical performances are attributing to the unique long microtubule bundling with much more pores and the abundant Oelement on the surface.This biomass carbon material with excellent electrochemical properties could be a useful material for multiple applications.Yang Liu Mengyi Zhang Liqiu Wang Yajuan Hou Chenxiao Guo Haiying Xin Shuang Xu 2020Chinese Chemical Letters2020,31,3:4
14Performance and fouling mechanism of direct contact membrane distillation(DCMD) treating fermentation wastewater with high organic concentrations显示文摘In this study,direct contact membrane distillation(DCMD)was used for treating fermentation wastewater with high organic concentrations.DCMD performance characteristics including permeate flux,permeate water quality as well as membrane fouling were investigated systematically.Experimental results showed that,after 12 hr DCMD,the feed wastewater was concentrated by about a factor of 3.7 on a volumetric basis,with the permeate flux decreasing from the initial 8.7 L/m^2/hr to the final 4.3 L/m^2/hr due to membrane fouling;the protein concentration in the feed wastewater was increased by about 3.5 times and achieved a value of 6178 mg/L,which is suitable for reutilization.Although COD and TOC in permeate water increased continuously due to the transfer of volatile components from wastewater,organic rejection of over 95%was achieved in wastewater.GC–MS results suggested that the fermentation wastewater contained 128kinds of organics,in which 14 organics dominated.After 12 hr DCMD,not only volatile organics including trimethyl pyrazine,2-acetyl pyrrole,phenethyl alcohol and phenylacetic acid,but also non-volatile dibutyl phthalate was detected in permeate water due to membrane wetting.FT-IR and SEM–EDS results indicated that the deposits formed on the membrane inner surface mainly consisted of Ca,Mg,and amine,carboxylic acid and aromatic groups.The fouled membrane could be recovered,as most of the deposits could be removed using a HCl/Na OH chemical cleaning method.Yan Wu Yun Kang Liqiu Zhang Dan Qu Xiang Cheng Li Feng 2018Journal of Environmental Sciences2018,30,3:4
15Metagenome analysis reveals potential microbial functions in topsoil of wheat-maize rotation system with five-year application of fertilizers显示文摘Fertilization mode affects soil quality and ecological health.The effects of four fertilization regimens on lignocellulose content,readily degradable carbohydrate decomposition,and potential microbial functions in the topsoil of a wheat-maize rotation system between 2012 and 2017 were investigated.The fertilization regimens of control(control NFNB),high chemical fertilizer(HCF),high biochar plus low chemical fertilizer(HBLCF),and biochar-based fertilizer(BBF)were compared on soil fundamental properties,microbial structure,and potential function in soil carbohydrate degradation based on metagenome analysis.The diversity of carbohydrate-active enzyme genes in the topsoil microbial consortia in the four trials was primarily distributed within the ten ecologically most dominant phyla.Application of BBF was associated with the lowest decline in total nitrogen and P2O5(2012-2017:6.5%and 28.1%,respectively)and the most effective carbohydrate decomposition(2015-2017:67.0%for cellulose and 59.9%for readily degradable carbohydrate).Carbohydrate transport and metabolism accounted for 6.0%of reads assigned functional classification under the BBF regimen.These findings reveal the ecologically functional diversity of topsoil microorganisms and suggest BBF application as a promising strategy for sustainable agriculture and beneficial to soil health.Yujun Shen Lixin Zhao Haibo Meng Hongsheng Cheng Jingtao Ding Jiarui Wang Shanshan Dong Xi Zhang Liqiu Song Shengwei Zheng 2019International Journal of Agricultural and Biological Engineering2019,12,6:3
16Proliferation of Cartographic Education in the Age of Big Data显示文摘Maps have long been a part of everyday life for the general public,and even more so in today’s knowledge society.No doubt,cartography as a profession of map design is assuming a more important role in the formation of intellectual skills in terms of spatial reasoning.Since its emergence as an academic discipline about 100 years ago,cartography has undergone many paradigm shifts.Its interaction with other disciplines has also constantly unfolded.These changes have left traces in cartographic education programs.In the age of big data,however,we are facing four fundamental challenges:(1) cartographic courses are being marginalized or even disappearing from degree programs in geospatial sciences;(2) the role of cartographers is increasingly eclipsed as a side effect of participatory cartography;(3) cartographers are blamed whenever something goes wrong with map use;and(4) professional map publishers can hardly compete with online mapping platforms dominated by Internet giants.Based on a contextual analysis of this seemingly gloomy situation,the paper reveals a number of proliferation points for the design of future cartographic curricula.First,cartography,once dedicated to supporting geospatial sciences,is thriving in the soil of data science,mapping not only the earth or other celestial bodies,but literally any kind of virtual space.Second,cartography has benefited from theoretical and technological advances in cognitive sciences,especially non-intrusive user studies,so that spatial cognition is becoming an integral component of cartographic education.Third,the role of scapegoat for wrongdoing of maps has accentuated cartographer’s overarching responsibility for quality and ethical issues in the geodata value chain.Finally,the diversification of the labor market requires new approaches to prepare future talents for a coopetition-oriented ecosystem in the marketplace.Liqiu MENG 2022Journal of Geodesy and Geoinformation Science2022,5,3:3
17Synthesis and thermal conductivity of microfluidic copper nanofluids显示文摘A microfluidic chemical solution method is developed for the synthesizing Cu nanofluids. The method replaces batch-based macroreactors in the conventional chemical solution method by continuous-flow microfluidic microreactors, thereby enabling the synthesis of nanofluids with various microstructures. The Cu nanofluids synthesized by this technology show a better stability, remaining stable even after more than 100 h standing. 18Shell Model Representation as a Substitute of LoD3 for 3D Modeling in CityGML显示文摘The OGC standard for 3D city modeling is widely used in an increasing number of applications.It defines five consecutive Levels of Detail(LoD0 to LoD4 with increasing accuracy and structural complexity),in which LoD3 includes all exterior appearances and geometrical details and subsequently requires much storage space.A new LoD is introduced as shell model with the exterior shell of the LoD3 model and the opening objects like windows,doors as well as smaller faade objects are projected onto walls.In this paper,a user survey is presented.The results of this survey show that the shell model can give users almost the same visual impression as the LoD3 model.Furthermroe,algorithms are developed to extract the shell model from LoD3 model.Experiments show that this shell model can reduce up to 90% storage of the original LoD3 model.Therefore,on one hand it can be used as a substitute for a LoD3 model for the visualization on small displays.On the other hand,it can be treated as a sub-level of detail(SLoD3) in CityGML,since it retains almost the same amount of information but requires much less storage space.FAN Hongchao MAO Bo SHEN Jie MENG Liqiu 2011Geo-Spatial Information Science2011,14,2:2
