1Identification of ferroptosis as a novel mechanism for antitumor activity of natural product derivative a2 in gastric cancer显示文摘Ferroptosis is a type of cell death accompanied by iron-dependent lipid peroxidation,thus stimulating ferroptosis may be a potential strategy for treating gastric cancer,therapeutic agents against which are urgently required.Jiyuan oridonin A(JDA) is a natural compound isolated from Jiyuan Rabdosia rubescens with anti-tumor activity,unclear anti-tumor mechanisms and limited water solubility hamper its clinical application.Here,we showed a2,a new JDA derivative,inhibited the growth of gastric cancer cells.Subsequently,we discovered for the first time that a2 induced ferroptosis.Importantly,compound a2 decreased GPX4 expression and overexpressing GPX4 antagonized the anti-proliferative activity of a2.Furthermore,we demonstrated that a2 caused ferrous iron accumulation through the autophagy pathway,prevention of which rescued a2 induced ferrous iron elevation and cell growth inhibition.Moreover,a2 exhibited more potent anti-cancer activity than 5-fluorouracil in gastric canc er cell line-derived xenograft mice models.Patient-derived tumor xenograft models from different patients displayed varied sensitivity to a2,and GPX4 downregulation indicated the sensitivity of tumors to a2.Finally,a2 exhibited well pharmacokinetic characteristic s.Overall,our data suggest that inducing ferroptosis is the major mechanism mediating anti-tumor activity of a2,and a2 will hopefully serve as a promising compound for gastric cancer treatment.Ying Liu Zan Song Yajie Liu Xubin Ma Wang Wang Yu Ke Yichao Xu Dequan Yu Hongmin Liu 2021Acta Pharmaceutica Sinica B2021,11,6:10
2Data mining to effect of key alloying elements on corrosion resistance of low alloy steels in Sanya seawater environmentAlloying Elements显示文摘In this paper,the relationship model between seawater environment,chemical composition and corrosion potential of low alloy steel is established and the distribution of corrosion potential of low alloy steel with changes in key alloying elements is excavated.The research was carried out with the following steps:Firstly,the relationship model between corrosion potential of low alloy steel and its influencing factors was established by data dimension reduction and artificial neural network(ANN).Secondly,key alloying elements of experimental steels were selected out by Pearson correlation analysis,then the corrosion resistance element model was visualized to show the effect of key alloying elements on corrosion potential of low alloy steel.Finally,corrosion potential of low alloy steel with the change of key alloying elements was classified and visualized by classification method.The mining results can reflect the validity of the proposed mining methods to a certain extent and provide an intuitive data basis for the development of high-quality and low-cost low alloy steels.Xin Wei Dongmei Fu Mindong Chen Wei Wu Dequan Wu Chao Liu 2021Journal of Materials Science & Technology2021,,5:9

4A Fuzzy Set-Based Approach for Model-Based Internet-Banking System Security Risk Assessment显示文摘A fuzzy set-based evaluation approach is demonstrated to assess the security risks for Internet-banking System. The Internet-banking system is semi-formally described using Unified Modeling Language (UML) to specify the behavior and state of the system on the base of analyzing the existing qualitative risk assessment methods. And a quantitative method based on fuzzy set is used to measure security risks of the system. A case study was performed on the WEB server of the Internet-banking System using fuzzy-set based assessment algorithm to quantitatively compute the security risk severity. The numeric result also provides a method to decide the most critical component which should arouse the system administrator enough attention to take the appropriate security measure or controls to alleviate the risk severity. 5The maize late embryogenesis abundant protein Zm DHN13 positively regulates copper tolerance in transgenic yeast and tobacco显示文摘Late embryogenesis abundant (LEA) proteins accumulate in the late stage of plant seed development, and are upregulated in most plants during drought, cold, heat, or salinity stress. LEA proteins can be classified by amino-acid sequence into seven groups. Dehydrins belong to LEA protein group Ⅱ. In previous studies, the maize KS type dehydrin ZmDHN13 increased the tolerance of transgenic tobacco to oxidative stress. In the present study, ZmDHN13 was identified under copper stress conditions, and the protein was then characterized using transgenic yeast and tobacco plants to investigate its functions. ZmDHN13 bound Cu2+. Its overexpression in transgenic tobacco conferred tolerance to copper stress by binding metals and reducing the accumulation of reactive oxygen species (ROS). 6The Functional Roles of Lipid Rafts in T Cell Activation,Immune Diseases and HIV Infection and Prevention显示文摘The first appearance of lipid rafts,or lipid rafts-like structure,was occasionally observed by cryo-electronic microscopy in 1980s as cavity,such as caveolae. However,the fully understanding of lipid raft was attributed by the studies of T cell activation,virus entry/budding,and other membrane events. During the interaction of T cell and antigen presenting cell,a highly organized structure is formed at the interface of the two cells,where cholesterol and sphingolipids are enriched,and form a liquid ordered phase that facilitates the signaling proteins on and off. Lipid rafts are also involved in virus entry and assembly. In this review,we will discuss cholesterol-sphingolipid floating microdomain,the lipid raft as a unique compartment of the plasma membrane,with biological functions that ensure correct intracellular traffic of proteins and lipids,such as protein-protein interactions by concentrating certain proteins in these microdomains,while excluding others. 7Transparent,stretchable,temperature-stable and self-healing ionogel-based triboelectric nanogenerator for biomechanical energy collection显示文摘A flexible and stable power supply is essential to the rapid development of wearable electronic devices.In this work,a transparent,flexible,temperature-stable and ionogel electrode-based self-healing triboelectric nanogenerator(IS-TENG)was developed.The ionogel with excellent stretchability(1,012%),high ionic conductivity(0.3 S·m^(−1))and high-temperature stability(temperature range of−77 to 250℃)was used as the electrode of the IS-TENG.The IS-TENG exhibited excellent transparency(92.1%)and stability.The output performance did not decrease when placed in a 60℃oven for 48 h.In addition,the IS-TENG behaved like a stable output in the range of−20 to 60℃.More importantly,the IS-TENG could also achieve self-healing of electrical performance at temperatures between−20 and 60℃and its output can be restored to its original state after healing.When the single-electrode IS-TENG with an area of 3 cm×3 cm was conducted under the working frequency of 1.5 Hz,the output values for open-circuit voltage,short-circuit current,short-circuit transferred charge,and maximum peak power density were 189 V,6.2μA,57 nC,and 2.17 W·m^(−2),respectively.The IS-TENG enables to harvest biomechanical energy,and drive electronic devices.Furthermore,the application of IS-TENGs as self-driven sensors for detecting human behavior was also demonstrated,showing good application prospects in the field of wearable power technology and self-driven sensing.Weiqiang Liao Xiukun Liu Yuqi Li Xu Xu Jinxing Jiang Shaorong Lu Dequan Bao Zhen Wen Xuhui Sun 2022Nano Research2022,15,3:2
8An international multicentric phaseⅢrandomized controlled trial of time-acupoints-space acupuncture for the prevention of chemotherapy-induced fatigue in patients with early stage breast cancer:a study protocol显示文摘OBJECTIVE:To determine the difference in prevention of chemotherapy-induced fatigue between time-acupointsspace acupuncture(ATAS)administered weekly compared to sham acupuncture and non-acupuncture in patients with early breast cancer receiving adjuvant chemotherapy by epirubicine-cyclophosphamide(EC)followed by paclitaxel,as measured by the multidimensional fatigue inventory(MFI)over the previous week and Visual Analogue Scale measuring fatigue(VAS-F),and to evaluate the effects of ATAS on selfreported neuropathy pain,sleep,anxiety and depression.METHODS:In this multicenter clinical trial,we have randomized patients into 3 groups:ATAS,Sham and nonacupuncture with an unequal randomization of 2∶1∶1.A cloud related electronical clinical report form and smartphone platform was established for data entry.Patients with a history of stageⅠ-Ⅲbreast cancer scheduled to receive adjuvant chemotherapy.Acupuncture will be delivered once a week during chemotherapy with VAS-F evaluation.In order to qualify and quantify the mechanism of fatigue induced by chemotherapy with or without acupuncture,an evaluation of immune profiling was incorporate in this study.RESULTS:The presence and seriousness of chemotherapy-induced fatigue should be considered in therapeutic programs for early breast cancer treatment.Fatigue induced by adjuvant chemotherapy in patients with breast cancer remains a major concern affecting the quality of life significantly.Unfortunately,we do not have effective pharmacological interventions yet.And clinical trials of acupuncture in preventing chemotherapyinduced fatigue in patients with early breast cancer have not been reported.CONCLUSION:The findings of the trial will allow us to determine the effects of acupuncture treatment approach.We will also be able to confirm whether ATAS is better than sham acupuncture and non-acupuncture treatments.Saghachian Mahasti Barry Caroline Asselain Bernard LI Yunfen NIE Jianyun LIU Dequan CHEN Dedian CHEN Chunxin Laura Estevez LI Wenhui Jean-Pierre Armand ZHU Miansheng JU Liya 2022Journal of Traditional Chinese Medicine2022,42,2:2
9DISTRIBUTED QUANTIZED CONSENSUS FOR AGENTS ON DIRECTED NETWORKS显示文摘Communication bandwidth and network topology are two important factors that affect performance of distributed consensus in multi-agent systems.The available works about quantized average consensus assume that the adjacency matrices associated with the digraphs are doubly stochastic,which amounts to that the digital networks are balanced.However,this assumption may be unrealistic in practice.In this paper,without assuming double stochasticity,the authors revisit an existing quantized average consensus protocol with the logarithmic quantization scheme,and investigate the quantized consensus problem in general directed digital networks that are strongly connected but not necessarily balanced.The authors first derive an achievable upper bound of the quantization precision parameter to design suitable logarithmic quantizer,and this bound explicitly depends on network topology.Subsequently,by means of the matrix transformation and the Lyapunov techniques,the authors provide a testable condition under which the weighted average consensus can be achieved with the proposed quantized protocol.LI Dequan LIU Qipeng WANG Xiaofan 2013Journal of Systems Science & Complexity2013,26,4:2
10Helioscopianoids A–Q,bioactive jatrophane diterpenoid esters from Euphorbia helioscopia显示文摘The EtOH extracts of the whole plants of Euphorbia helioscopia afforded 17 new jatrophane diterpenoid esters, helioscopianoids A–Q(1–17), along with eight known compounds(18–25). Their structures were elucidated by extensive spectroscopic methods and Mo_2(OAc)_4-induced ECD analysis,and the structures of compounds 1, 2, and 7 were confirmed by X-ray crystallography. Compounds 1–17 were evaluated for inhibitory effects on P-glycoprotein(P-gp) in an adriamycin(ADM)-resistant human breast adenocarcinoma cell line(MCF-7/ADR) and neuroprotective effects against serum deprivationinduced and rotenone-induced PC12 cell damage. Compounds 8 and 16 increased the accumulation of ADM in MCF-7/ADR cells by approximately 3-fold at a concentration of 20 μmol/L. 12Varicella-zoster virus ORF7 interacts with ORF53 and plays a role in its trans-Golgi network localization显示文摘Varicella-zoster virus(VZV) is a neurotropic alphaherpesvirus that causes chickenpox and shingles. ORF7 is an important virulence determinant of VZV in both human skin and nerve tissues,however, its specific function and involved molecular mechanism in VZV pathogenesis remain largely elusive. Previous yeast two-hybrid studies on intraviral protein-protein interaction network in herpesviruses have revealed that VZV ORF7 may interact with ORF53, which is a virtually unstudied but essential viral protein. The aim of this study is to identify and characterize VZV ORF53, and to investigate its relationship with ORF7. For this purpose, we prepared monoclonal antibodies against ORF53 and, for the first time, characterized it as a ~40 k Da viral protein predominantly localizing to the trans-Golgi network of the infected host cell. Next, we further confirmed the interaction between ORF7 and ORF53 by co-immunoprecipitation and co-localization studies in both plasmid-transfected and VZV-infected cells. Moreover, interestingly, we found that ORF53 lost its trans-Golgi network localization and became dispersed in the cytoplasm of host cells infected with an ORF7-deleted recombinant VZV, and thus ORF7 seems to play a role in normal subcellular localization of ORF53.
