| 3 | The FGOALS climate system model as a modeling tool for supporting climate sciences:An overview显示文摘Climate system models are useful tools for understanding the interactions among the components of the climate system and predicting/projecting future climate change. The development of climate models has been a central focus of the State Key Laboratory of Numerical Modeling for Atmospheric Sciences and Geophysical Fluid Dynamics, Institute of Atmospheric Physics, Chinese Academy of Sciences(LASG/IAP) since the establishment of the laboratory in 1985. In China, many pioneering component models and fully coupled models of the climate system have been developed by LASG/IAP. The fully coupled climate system developed in the recent decade is named FGOALS(Flexible Global Ocean-Atmosphere-Land System Model). In this paper, an application-oriented review of the LASG/IAP FGOALS model is presented. The improved model performances are demonstrated in the context of cloud-radiation processes, Asian monsoon, ENSO phenomena, Atlantic Meridional Overturning Circulation(AMOC) and sea ice. The FGOALS model has contributed to both CMIP5(Coupled Model Intercomparison Project-phase 5) and IPCC(Intergovernmental Panel on Climate Change) AR5(the Fifth Assessment Report). The release of FGOALS data has supported the publication of nearly 500 papers around the world. The results of FGOALS are cited ~106 times in the IPCC WG1(Working Group 1) AR5. In addition to the traditional long-term simulations and projections, near-term decadal climate prediction is a new set of CMIP experiment, progress of LAGS/IAP in the development of nearterm decadal prediction system is reviewed. The FGOALS model has supported many Chinese national-level research projects and contributed to the national climate change assessment report. | 4 | Viral MIPα homologous with human MIP-1α acts on HIV co-receptor CCR5显示文摘The function and usage of vMIPα encoded by K6 gene of herpesvirus 8 (HHV8) which has homology with human macrophage protein (MIP) have not been clearly known. In the present note the K6 gene of HHV8 was cloned and transfected into NIH3T3 cells and E. coli cells. Conditional media from the 3T3-transfected cells and K6 product vMIPa from E. coli. Cells were used to perform the experiments of ligand-receptor binding and cellular adhesion with peripheral blood macrophages. The conditional media and the purified vMIPa from E. coli could compete to bind to CCR5 located on macrophages from peripheral blood with I125-hMIP-1α chemokine of human. Cellular adhesion showed that the conditional media from transfected cells and the purified vMIPa did not induce the adhesion of macrophages from peripheral blood to ICAM-1. In conclusion, vMIPα encoded by K6 gene of HHV8 can bind to CCR5 of peripheral blood macrophage cells and does not induce their adhesion. | 5 | Effect of Tangnaikang on TGF-β_1-induced transdifferentiation of human renal tubular epithelial HK-2 cells显示文摘OBJECTIVE: To explore the function of Tangnaikang (TNK) in the prevention and treatment of renal interstitial fibrosis through transdifferentiation of the human renal tubular epithelial cell line HK-2 induced by transforming growth factor-β1(TGF-β1).METHODS: HK-2 cells cultured in dulbecco's modified eagle medium/F12 (1∶1) with 10% fetal calf serum were divided into six groups: blank control group, TGF-β1 group (TGF-β1 10 ng/mL), serum control group (TGF-β1 10 ng/mL+10% serum), treatment group 1 (TGF-β1 10 ng/mL+ 5% TNK serum),treatment group 2 (TGF-β1 10 ng/mL+10% TNK serum), and treatment group 3 (TGF-β1 10 ng/mL+20% TNK serum). Cell proliferation was detected by 4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Expression of α-smooth muscle actin (α-SMA) and E-cadherin were observed by immunohistochemical assay. The contents of collagen Ⅰ (Col Ⅰ), collagen Ⅲ (Col Ⅲ), and fibronectin (FN) in the culture medium supernatant were detected by ELISA.RESULTS: E-cadherin was expressed and α-SMA was not expressed in normal HK-2 cells. In HK-2 cells cultured with TGF-β1,α-SMA expression significantly increased, HK-2 cells significantly proliferated, and secretion of Col Ⅰ, Col Ⅲ, and FN significantly increased compared with the blank control group (all P<0.05). | 9 | Observational analysis and numerical simulation of the interannual variability of the boreal winter Hadley circulation over the recent 30 years显示文摘The interannual variability of the boreal winter (DJF) Hadley Cell strength during 1979-2008 is investigated using NCEP/NCAR reanalysis data. The results of AMIP simulation of LASG/IAP AGCM GAMIL2.0 are compared against the reanalysis data. Both the reanalysis data and the simulation show that the interannual variability of the Hadley Cell strength has a non-uniform spatial distribution, as evidenced by the 1st Empirical Orthogonal Function (EOF) mode. The change of Hadley cell strength in the tropics is opposite to that in the subtropical regions. Our analysis indicates that a positive phase of EOF1 is associated with an El Ni o-like warmer equatorial central and eastern Pacific and a warmer southern Indian Ocean. Above features are also seen in the results of GAMIL2.0 simulation, indicating that the interannual variability of the Hadley Cell strength is driven by the tropical ocean variability. Our analysis also demonstrates that the contribution of the warmer central-eastern Pacific to the 1st EOF mode is larger than that of the South Indian Ocean. The SST forcing enhances the local Hadley circulation strength in the central Pacific and Africa (30°S-30°N, 150°E-90°W), while it weakens the local Hadley circulation in other regions (30°S-30°N, 90°-10°W). The western Pacific anticyclone remotely driven by the El Ni o forcing leads to a weakened local Hadley cell in the Northern Hemisphere, while the South Indian Ocean anticyclone driven by the remote El Ni o forcing and the local warmer SST anomalies in the southern Indian Ocean results in a weakened local Hadley Cell in the Southern Hemisphere. The enhancement of the Pacific local Hadley Cell is stronger (weaker) than that of the Atlantic, the western Pacific, and the southern Indian Ocean in the tropical (subtropical) part, thus for the zonal mean condition the strength of the total Hadley Cell is stronger (weaker) in the tropical (subtropical) limb. The amplitude of the Hadley Cell change in the Northern Hemisphere is stronger than that in the Southern Hemisphere. Abdominal withdrawal reflex test, open field test, and intestinal motility test were used to evaluate visceral sensitivity, anxiety,and intestinal motility in mice. The expressions of NLRP6, Mucin-2(MUC2) and E-cadherin were determined using immunofluorescence and Western blotting assays.RESULTS: EA significantly upregulated the expression of NLRP6 in the intestine of mice. | 12 | 64 × 64 GM-APD array-based readout integrated circuit for 3D imaging applications显示文摘Using the high sensitivity of the avalanche photodiode(APD) detector operated in the Geigermode(GM), an array readout integrated circuit(ROIC) comprising a two-segment time-to-digital converter(TDC) is employed for wide-dynamic time interval measurement, where a 1-bit low-segment TDC is implemented by discriminating a single-phase clock period. The proposed 64 × 64 GM-APD array ROIC fabricated using Taiwan semiconductor manufacturing company(TSMC) 0.18-μm complementary metal oxide semiconductor(CMOS) technology can operate at a maximum frequency of 500 MHz provided by an external phase-locked loop clock. The time resolution is reduced to < 1 ns along with a maximum range of4 μs; the differential non-linearity(DNL) and integral non-linearity(INL) are restricted to approximately-0.15 to 0.15 least significant bit(LSB) and-0.3 to 0.32 LSB, respectively; and the power consumption is490 mW under a frame rate of 20 kH z. | 13 | Metabolic profiling of DREB-overexpressing transgenic wheat seeds by liquid chromatographymass spectrometry显示文摘DREBs are transcription factors that regulate abiotic stress tolerance in plants.Previously,we reported that wheat transgenic lines overexpressing Gm DREB1 showed increased tolerance to drought and salt stress.However,the molecular basis of increased tolerance is still poorly understood,and whether the overexpression of DREB will cause unexpected effects is also of concern.We performed seed metabolic profiling of the genetically modified(GM)wheat T349 and three non-GM cultivars with LC-MS to identify the metabolic basis of stress tolerance and to assess the unexpected effects of exogenous gene insertion.Although we did not note the appearance of novel metabolites or the disappearance of existing metabolites,overexpression of the transcription factor Gm DREB1 in T349 wheat influenced metabolite levels in seeds.Increased levels of stress tolerance-associated metabolites were found in the stress-sensitive non-transgenic acceptor counterpart J19,while metabolites associated with cell membrane structure and stability accumulated in T349.Among these metabolites in T349,most showed levels similar to those in the non-GM wheats.Overexpression of Gm DREB1 in T349 may cause a shift in its metabolic profile leading to down-regulation of several energy-consuming processes to favor increased yield under stress conditions,which is a reasonable expectation of breeders while creating the GM wheat and Gm DREB1 overexpression did not cause unexpected effects in T349 seeds.These results may be helpful for GM crop research and risk assessment. | Fengjuan Niu Qiyan Jiang Xianjun Sun Zheng Hu Lixia Wang Hui Zhang | 2020 | The Crop Journal2020,8,6: | 2 |
| 14 | Controlling effects of the Ordovician carbonate pore structure on hydrocarbon reservoirs in the Tarim Basin, China显示文摘The Ordovician carbonate reservoirs in the Tarim Basin with secondary dissolution pores and vugs have complicated pore structures. The weathering crust reservoirs mainly consist of large cavities or vugs connected by fractures, but most of the reef-shoal reservoirs have complex and small throats among matrix pores. The pore structure can be divided into four types: big pore and big throat, big pore but small throat, small pore and small throat, and fracture type. Most of the average throat radius falls between 0.03 and 0.07 μm, close to that of unconventional reservoirs except in local areas with developed fractures. Fluid driving force analysis shows that the differentiation of fluid is mainly controlled by the throat radius in two kinds of mechanism separated by the critical throat radius about 0.1 μm. There is obvious fluid differentiation and oil/gas/water contact in fracture-cavity reservoirs with big throats.
