| 1 | Developing superior alleles of yield genes in rice by artificial mutagenesis using the CRISPR/Cas9 system显示文摘Rice yield is an important and complex agronomic trait controlled by multiple genes.In recent decades,dozens of yield-associated genes in rice have been cloned,many of which can increase production in the form of loss or degeneration of function.However,mutations occurring randomly under natural conditions have provided very limited genetic resources for yield increases.In this study,potentially yield-increasing alleles of two genes closely associated with yield were edited artificially.The recently developed CRISPR/Cas9system was used to edit two yield genes:Grain number 1a(Gn1a)and DENSE AND ERECT PANICLE1(DEP1).Several mutants were identified by a target sequence analysis.Phenotypic analysis confirmed one mutant allele of Gn1a and three of DEP1 conferring yield superior to that conferred by other natural high-yield alleles.Our results demonstrate that favorable alleles of the Gnla and DEP1 genes,which are considered key factors in rice yield increases,could be developed by artificial mutagenesis using genome editing technology. | Liyu Huang Ru Zhang Guangfu Huang Yanxia Li Getachew Melaku Shilai Zhang Haitao Chen Yanjuan Zhao Jing Zhang Yesheng Zhang Fengyi Hu | 2018 | The Crop Journal2018,6,5: | 8 |
| 2 | Identification of Key Genes for the Ultrahigh Yield of Rice Using Dynamic Cross-tissue Network Analysis显示文摘Significantly increasing crop yield is a major and worldwide challenge for food supply and security.It is well-known that rice cultivated at Taoyuan in Yunnan of China can produce the highest yield worldwide.Yet,the gene regulatory mechanism underpinning this ultrahigh yield has been a mystery.Here,we systematically collected the transcriptome data for seven key tissues at different developmental stages using rice cultivated both at Taoyuan as the case group and at another regular rice planting place Jinghong as the control group.We identified the top 24 candidate high-yield genes with their network modules from these well-designed datasets by developing a novel computational systems biology method,i.e.,dynamic cross-tissue(DCT)network analysis.We used one of the candidate genes,Os SPL4,whose function was previously unknown,for gene editing experimental validation of the high yield,and confirmed that Os SPL4 significantly affects panicle branching and increases the rice yield.This study,which included extensive field phenotyping,cross-tissue systems biology analyses,and functional validation,uncovered the key genes and gene regulatory networks underpinning the ultrahigh yield of rice.The DCT method could be applied to other plant or animal systems if different phenotypes under various environments with the common genome sequences of the examined sample.DCT can be downloaded from https://github.com/ztpub/DCT. | Jihong Hu Tao Zeng Qiongmei Xia Liyu Huang Yesheng Zhang Chuanchao Zhang Yan Zeng Hui Liu Shilai Zhang Guangfu Huang Wenting Wan Yi Ding Fengyi Hu Congdang Yang Luonan Chen Wen Wang | 2020 | Genomics, Proteomics & Bioinformatics2020,18,3: | 2 |
| 3 | Combination therapy of thiamine,vitamin C and hydrocortisone in treating patients with sepsis and septic shock:a meta-analysis and trial sequential analysis显示文摘Background:The objective of this study was to evaluate the clinical efficacy of thiamine and vitamin C with or without hydrocortisone coadministration on the treatment of sepsis and septic shock.Methods:MEDLINE,EMBASE and CENTRAL databases were searched for randomized controlled trials(RCTs)that made a comparative study between the combination therapy of vitamin C and thiamine with or without hydrocortisone and the administration of placebo in patients with sepsis or septic shock.Two reviewers independently performed study selection,data extraction and quality assessment.Both short-term mortality and change in the sequential organ failure assessment(SOFA)score from baseline(delta SOFA)were set as the primary outcomes.Secondary endpoints included intensive care unit(ICU)mortality,new onset of acute kidney injury,total adverse events,ICU and hospital length of stay,duration of vasopressor usage and ventilator-free days.Meanwhile,trial sequential analysis was conducted for primary outcomes.Results:Eight RCTs with 1428 patients were included in the current study.The results showed no significant reduction of short-term mortality in sepsis and septic shock patients who received combination therapy of vitamin C and thiamine with or without hydrocortisone compared to those with placebo{risk ratio(RR),1.02[95%confidence interval(CI),0.87 to 1.20],p=0.81,I2=0%;risk difference(RD),0[95%CI,−0.04 to 0.05]}.Nevertheless,the combination therapy was associated with significant reduction in SOFA score[mean difference(MD),−0.63,(95%CI,−0.96 to−0.29,p<0.001,I^(2)=0%]and vasopressors duration(MD,−22.11[95%CI,−30.46 to−13.77],p<0.001,I2=6%).Additionally,there were no statistical differences in the pooled estimate for other outcomes.Conclusions:In the current meta-analysis,the combination therapy of vitamin C and thiamine,with or without hydrocortisone had no impact on short-term mortality when compared with placebo,but was associated with significant reduction in SOFA score among patients with sepsis and septic shock. | Renqi Yao Yibing Zhu Yue Yu Zhixuan Li Lixue Wang Liyu Zheng Jingyan Li Huibin Huang Guosheng Wu Feng Zhu Zhaofan Xia Chao Ren Yongming Yao | 2021 | Burns & Trauma2021,9,1: | 2 |

DNA vaccine formulations of the two antigens (VRSj31 and VRSj32) were produced and injected intramuscularly twice at a 2-week interval and then recombinant proteins (rSj31 and rSj32) together with Freund Complete Adjuvant (FCA) were used to boost the goats. The experiment was repeated in different batche cercariae. A strong anamnestic antibody response was induced after boost. A significant reduction of liver egg counts and miracidial hatching was showed in both experiments. | 18 | GIGANTEA orthologs,E2 members,redundantly determine photoperiodic flowering and yield in soybean显示文摘Soybean(Glycine max L.)is a typical photoperiodsensitive crop,such that photoperiod determines its flowering time,maturity,grain yield,and phenological adaptability.During evolution,the soybean genome has undergone two duplication events,resulting in about 75%of all genes being represented by multiple copies,which is associated with rampant gene redundancy.Among duplicated genes,the important soybean maturity gene E2 has two homologs,E2-Like a(E2La)and E2-Like b(E2Lb),which encode orthologs of Arabidopsis GIGANTEA(GI).Although E2 was cloned a decade ago,we still know very little about its contribution to flowering time and even less about the function of its homologs.Here,we generated single and double mutants in E2,E2La,and E2Lb by genome editing and determined that E2 plays major roles in the regulation of flowering time and yield,with the two E2 homologs depending on E2 function.At high latitude regions,e2 single mutants showed earlier flowering and high grain yield.Remarkably,in terms of genetic relationship,genes from the legume-specific transcription factor family E1 were epistatic to E2.We established that E2 and E2-like proteins form homodimers or heterodimers to regulate the transcription of E1 family genes,with the homodimer exerting a greater function than the heterodimers.In addition,we established that the H3 haplotype of E2 is the ancestral allele and is mainly restricted to low latitude regions,from which the loss-of-function alleles of the H1 and H2haplotypes were derived.Furthermore,we demonstrated that the function of the H3 allele is stronger than that of the H1 haplotype in the regulation of flowering time,which has not been shown before.Our findings provide excellent allelic combinations for classical breeding and targeted gene disruption or editing. | Lingshuang Wang Haiyang Li Milan He Lidong Dong Zerong Huang Liyu Chen Haiyang Nan Fanjiang Kong Baohui Liu Xiaohui Zhao | 2023 | Journal of Integrative Plant Biology2023,65,1: | 0 |
