1Lanthanum-dopedα-Ni(OH)_(2)1D-2D-3D hierarchical nanostructures for robust bifunctional electro-oxidation显示文摘The development of advanced electrocatalysts for electro-oxidation reactions has attracted much attention because of the critical role of such electrocatalysts in improving the overall efficiency of coupled hydrogen production.We have developed an efficient lanthanum-dopedα-Ni(OH)_(2) bifunctional catalyst with a 1D-2D-3D hierarchical nanostructure.It shows superior activity and stability in the anodic oxygen evolution reaction(OER)and urea oxidation reaction(UOR).Enrichment of the edge sites and conducting La doping inα-Ni(OH)_(2) phase enable formation and stabilization of abundant local Ni^(3+)ions.This guarantees ultralow onset potentials in electro-oxidation reactions.The 1D-2D-3D hierarchical nanostructure significantly boosts the in situ generation of high-valence active species,which results in efficient and stable OER and UOR performances,and the synergistic merit of doping-induced facile reaction kinetics.Because of the structural benefits of a large surface area,charge-transfer promotion,good structural stability,and bifunctionality,a 1%La-dopedα-Ni(OH)_(2) hierarchical nanostructure gives superior OER and UOR performances with low overpotentials,large catalytic current densities,and excellent operational stability.It is therefore a promising catalyst for use in simultaneous alkaline wastewater treatment and hydrogen production.Zimeng Wei Wenbin Sun Shanshan Liu Jindi Qi Luyao Kang jiechen Li Shanshan Lou Junfeng Xie Bo Tanga Yi Xie 2021Particuology2021,19,4:1
2The diapycnal mixing in the upper Pacific estimated from GTSPP observations显示文摘Diapycnal mixing(DM) in the upper 600 m of the Pacific Ocean was estimated based on the huge amount of the observations from Global Temperature-Salinity Profile Programme(GTSPP), using the strain version of the finescale parameterization. It is found that DM in each season exhibits similar distribution pattern, but differs in details. The intensification of DM is related to bottom roughness, surface near-inertial energy, and proximity to the equator. The intensified DM caused by rough topography shows in the profiles near the Mendocino fracture zone in the northeast Pacific, and the heightened DM caused by wind-generated near-inertial energy appears in the westerly region of the Southern Ocean. 3Rapid and Scalable Synthesis of Prussian Blue Analogue Nanocubes for Electrocatalytic Water Oxidation显示文摘Design and fabrication of earth-abundant electrocatalysts for oxygen evolution reaction(OER)is essential in improving the overall ef-ficiency of water electrolysis.In this work,we proposed a rapid and scalable synthesis route for fabricating Prussian blue analogue(PBA)nano cubes with tun able compositi ons and uniform particle size.With the structural ben efits of abu ndant surface sites,facile charge transfer behavior and favorable Co^(2+)-to-Co^(3+)pre-oxidation reaction,fast generation and accumulation of the catalytically active Co3+sites can be achieved for the CoCo PBA nano cubes,leadi ng to remarkable OER activity with simulta neously achieved low overpotential,large anodic current density,small Tafel slope as well as outstanding intrinsic activity.Of note,by performing Iong-term OER operati on,the CoCo PBA nano cubes exhibit a remarkable 5.5-folds performs nee enhan ceme nt,and obvious surface rec on struc-tion and the accumulation of high-valence Co species can be identified,proving the crucial role of pre-oxidation process in boosting the OER catalysis.This work proposed a un iversal approach for the rapid,scalable and con trollable fabricati on of the PBA-based materials,and the elucidation of the pre-oxidation process in facilitating the OER catalysis may provide useful guidanee for designing and optimizing advanced catalysts for energy-related electro-oxidation reactions in the future.Wenbin Sun Zimeng Wei Jindi Qi Luyao Kang Jiechen Li Junfeng Xie Bo Tang Yi Xie 2021Chinese Journal of Chemistry2021,39,9:1
4Design of artificial small regulatory trans-RNA for gene knockdown in Bacillus subtilis显示文摘Bacillus subtilis as the Gram-positive model bacterium has been widely used in synthetic biology and biotechnology while the regulatory RNA tools for B.subtilis are still not fully explored.Here,a bottom-up approach is proposed for designing artificial trans-acting sRNAs.By engineering the intrinsic sRNA SR6,a minimized core scaffold structure consisting of an 8 bp stem,a 4 nt loop,and a 9 nt polyU tail was generated and proven to be sufficient for constructing sRNAs with strong repression activity(83%).Moreover,we demonstrate this artificial sRNA system functions well in an hfq-independent manner and also achieves strong repression efficiency in Escherichia coli(above 80%).A structure-based sRNA design principle was further developed for the automatic generation of custom sRNAs with this core scaffold but various sequences,which facilitates the manipulation and avoids structure disruption when fusing any base-pairing sequence.By applying these auto-designed sRNAs,we rapidly modified the cell morphology and biofilm formation,and regulated metabolic flux toward acetoin biosynthesis.This sRNA system with cross-species regulatory activities not only enriched the gene regulation toolkit in synthetic biology for B.subtilis and E.coli but also enhanced our understanding of trans-acting sRNAs.Guobin Yin Anqi Peng Luyao Zhang Yang Wang Guocheng Du Jian Chen Zhen Kang 2023Synthetic and Systems Biotechnology2023,8,1:0
5Gene knockdown by structure defined single-stem loop small non-coding RNAs with programmable regulatory activities显示文摘Gene regulation by trans-acting small RNAs(sRNAs)has considerable advantages over other gene regulation strategies.However,synthetic sRNAs mainly take natural sRNAs(MicC or SgrS)as backbones and comprise three functional elements folding into two or more stem-loop structures:an mRNA base pairing region,an Hfq-binding structure,and a rho-independent terminator.Due to limited numbers of natural sRNAs and complicated backbone structures,synthetic sRNAs suffer from low activity programmability and poor structural modularity.Moreover,natural sRNA backbone sequences may increase the possibility of unwanted recombination.Here,we present a bottom-up approach for creating structure defined single-stem loop small non-coding RNAs(ssl-sRNAs),which contain a standardized scaffold of a 7 bp-stem-4 nt-loop-polyU-tail and a 24 nt basing pairing region covering the first eight codons.Particularly,ssl-sRNA requires no independent Hfq-binding structure,as the polyU tail fulfills the roles of binding Hfq.A thermodynamic-based scoring model and a web server sslRNAD(http://www.kangzlab.cn/)were developed for automated design of ssl-sRNAs with well-defined structures and programmable activities.ssl-sRNAs displayed weak polar effects when regulating polycistronic mRNAs.The ssl-sRNA designed by sslRNAD showed regulatory activities in both Escherichia coli and Bacillus subtilis.A streamlined workflow was developed for the construction of customized ssl-sRNA and ssl-sRNA libraries.As examples,the E.coli cell morphology was easily modified and new target genes of ergothioneine biosynthesis were quickly identified with ssl-sRNAs.ssl-sRNA and its designer sslRNAD enable researchers to rapidly design sRNAs for knocking down target genes.Yang Wang Guobin Yin Huanjiao Weng Luyao Zhang Guocheng Du Jian Chen Zhen Kang 2023Synthetic and Systems Biotechnology2023,8,1:0
6Lin28a maintains a subset of adult muscle stem cells in an embryonic-like state显示文摘During homeostasis and after injury,adult muscle stem cells(MuSCs)activate to mediate muscle regeneration.However,much remains unclear regarding the heterogeneous capacity of MuSCs for self-renewal and regeneration.Here,we show that Lin28a is expressed in embryonic limb bud muscle progenitors,and that a rare reserve subset of Lin28a^(+)Pax7^(-)skeletal MuSCs can respond to injury at adult stage by replenishing the Pax7^(+)MuSC pool to drive muscle regeneration.Compared with adult Pax7^(+)MuSCs,Lin28a^(+)MuSCs displayed enhanced myogenic potency in vitro and in vivo upon transplantation.The epigenome of adult Lin28a^(+)MuSCs showed resemblance to embryonic muscle progenitors.In addition,RNA-sequencing revealed that Lin28a^(+)MuSCs co-expressed higher levels of certain embryonic limb bud transcription factors,telomerase components and the p53 inhibitor Mdm4,and lower levels of myogenic differentiation markers compared to adult Pax7^(+)MuSCs,resulting in enhanced self-renewal and stress-response signatures.Functionally,conditional ablation and induction of Lin28a^(+)MuSCs in adult mice revealed that these cells are necessary and sufficient for efficient muscle regeneration.Together,our findings connect the embryonic factor Lin28a to adult stem cell self-renewal and juvenile regeneration.Peng Wang Xupeng Liu Ziyue Yao Yu Chen Lanfang Luo Kun Liang Jun-Hao Elwin Tan Min-Wen Jason Chua Yan-Jiang Benjamin Chua Shilin Ma Liping Zhang Wenwu Ma Shuqing Liu Wenhua Cao Luyao Guo Lu Guang Yuefan Wang He Zhao Na Ai Yun Li Chunwei Li Ruiqi Rachel Wang Bin Tean Teh Lan Jiang Kang Yu Ng Shyh-Chang 2023Cell Research2023,33,9:0
