| 1 | TNF-α Induces Transient Resistance to Fas-Induced Apoptosis in Eosinophilic Acute Myeloid Leukemia Cells显示文摘Tumor necrosis factor α (TNF-α) has been recognized as an activator of nuclear factor κB (NF-κB), a factor implicated in the protection of many cell types from apoptosis. We and others have presented evidence to suggest that Fas-induced apoptosis may be an important aspect of the resolution of inflammation, and that delayed resolution of inflammation may be directly associated with NF-κB-dependent resistance to Fas. Because TNF-α activates NF-κB in many cell types including inflammatory cells such as eosinophils, we examined effects of TNF-α signaling on the Fas-mediated killing of an eosinophilic cell line AML14. While agonist anti-Fas (CH11) treatment induced apoptosis in AML14 cells, no significant cell death occurred in response to TNF-α alone. Electrophoretic mobility shift assay (EMSA) revealed that TNF-α induced NF-κB transactivation in AML14 cells in a time- and dose-dependent fashion, and subsequent supershift assays indicated that the translocated NF-κB was the heterodimer p65 (RelA)/p50. Pre-treatment of cells with TNF-α dramatically decreased the CH11-induced cell death in a transient fashion, accompanied by suppression of activation of caspase-8 and caspase-3 activation. Inhibition of NF-κB transactivation by inhibitors, BAY 11-7085 and parthenolide, reversed the suppression of Fas-mediated apoptosis by TNF-α. Furthermore, TNF-α up-regulated X-linked inhibitor of apoptosis protein (XIAP) transiently and XIAP levels were correlated with the temporal pattern of TNF-α protection against Fas-mediated apoptosis. | 3 | Impact of air drought on photosynthesis efficiency of the Siberian crabapple(Malus baccata L. Borkh.) in the forest-steppe zone of Transbaikalia, Russia显示文摘The adaption of photosynthesis, being a key metabolic process, plays an important role in plant resistance to air drought. In this study, the Siberian crabapple(Malus baccata L. Borkh.) in the forest-steppe zone of Transbaikalia region, Russia, was subjected to air drought stress and its photosynthesis characteristics were analyzed. The results show that air drought and sufficient soil moisture supply lead to the decrease in the total chlorophyll(Chl) content, while the ratio of Chls to carotenoids is constant in the Siberian crabapple tree. The function of photosystem II(PS-II) in the crabapple trees is characterized by a decrease in the fraction of absorbed light energy spent on the photochemical work and an increase in the proportion of non-photosynthetic thermal quenching. These changes indicate the photosynthetic down-regulation that acts as a universal photoprotective mechanism. During the midday hours, the combination of high air temperature and low air humidity leads to the decrease in the maximum photochemical quantum yield of photosystem II(Fv/Fm) and the efficiency of photosynthesis(PABS). The parameters of leaf gas exchange show the significant differences in these values between the control and experimental variants. During the morning hours, the Siberian crabapple, growing in the Irkutsk City,assimilates carbon dioxide more intensively. Due to the higher air humidity, the stomata are kept open and the necessary amount of carbon dioxide entries the sites of carboxylation. The low air humidity combined with wind in the experimental variants leads to the unreasonably high water loss in the crabapple leaves by more than 27% as compared to the control variant(Irkutsk City). However, water use efficiency in the morning hours increases during plant photosynthetic processes, i.e., 42% higher than that of control. | 4 | Endo-microscopy beyond the Abbe and Nyquist limits显示文摘For several centuries,far-field optical microscopy has remained a key instrument in many scientific disciplines,including physical,chemical,and biomedical research.Nonetheless,far-field imaging has many limitations:the spatial resolution is controlled by the diffraction of light,and the imaging speed follows the Nyquist–Shannon sampling theorem.The recent development of super-resolution techniques has pushed the limits of spatial resolution.However,these methods typically require complicated setups and long acquisition times and are still not applicable to deeptissue bioimaging.Here,we report imaging through an ultra-thin fibre probe with a spatial resolution beyond the Abbe limit and a temporal resolution beyond the Nyquist limit simultaneously in a simple and compact setup.We use the random nature of mode coupling in a multimode fibre,the sparsity constraint and compressive sensing reconstruction.The new approach of super-resolution endo-microscopy does not use any specific properties of the fluorescent label,such as depletion or stochastic activation of the molecular fluorescent state,and therefore can be used for label-free imaging.We demonstrate a spatial resolution more than 2 times better than the diffraction limit and an imaging speed 20 times faster than the Nyquist limit.The proposed approach can significantly expand the realm of the application of nanoscopy for bioimaging. | Lyubov V.Amitonova Johannes F.de Boer | 2020 | Light(Science & Applications)2020,9,1: | 2 |

| 18 | Karyotypes and recombination patterns of the Common Swift(Apus apus Linnaeus,1758) and Eurasian Hobby(Falco subbuteo Linnaeus, 1758)显示文摘Background: Meiotic recombination is an important source of genetic variability.Studies on mammals demonstrate a substantial interspecies variation in overall recombination rate, which is dependent mainly on chromosome(2 n) and chromosome arm number(FN).Bird karyotypes are very conservative with 2 n being about 78–82 and FN being 80–90 in most species.However, some families such as Apodidae(swifts) and Falconidae(falcons) show a substantial karyotypic variation.In this study, we describe the somatic and pachytene karyotypes of the male Common Swift(Apus apus) and the pachytene karyotype of the male Eurasian Hobby(Falco subbuteo) and estimate the overall number and distribution of recombination events along the chromosomes of these species.Methods: The somatic karyotype was examined in bone marrow cells.Pachytene chromosome spreads were prepared from spermatocytes of adult males.Synaptonemal complexes and mature recombination nodules were visualized with antibodies to SYCP3 and MLH1 proteins correspondingly.Results: The karyotype of the Common Swift consists of three metacentric, three submetacentric and two telocentric macrochromosomes and 31 telocentric microchromosomes(2 n = 78; FN = 90).It differs from the karyotypes of related Apodidae species described previously.The karyotype of the Eurasian Hobby contains one metacentric and 13 telocentric macrochromosomes and one metacentric and ten telocentric microchromosomes(2 n = 50; FN = 54) and is similar to that described previously in 2 n, but differs for macrochromosome morphology.Despite an about 40% difference in 2 n and FN, these species have almost the same number of recombination nodules per genome: 51.4 ± 4.3 in the swift and 51.1 ± 6.7 in the hobby.The distribution of the recombination nodules along the macrochromosomes was extremely polarized in the Common Swift and was rather even in the Eurasian Hobby.Conclusions: This study adds two more species to the short list of birds in which the number and distribution of recombination nodules have been examined.Our data confirm that recombination rate in birds is substantially higher than that in mammals, but shows rather a low interspecies variability.Even a substantial reduction in chromosome number does not lead to any substantial decrease in recombination rate.More data from different taxa are required to draw statistically supported conclusions about the evolution of recombination in birds. | Lyubov Malinovskaya Elena Shnaider Pavel Borodin Anna Torgasheva | 2018 | Avian Research2018,9,1: | 0 |

Pahudioxylon sp. and two unknown taxa( Dicotyloxylon sp.).The preservation of two dicotyledonous wood samples is insufficient for precise identification to family level,such that the samples are not suitable with respect to investigation of the river palaeocourse. However,these findings increase the systematic data of the fossil wood from the Mekong Basin.
