1Reducing nitrogen runoff from paddy fields with arbuscular mycorrhizal fungi under different fertilizer regimes显示文摘Nitrogen(N) runoff from paddy fields serves as one of the main sources of water pollution. Our aim was to reduce N runoff from paddy fields by fertilizer management and inoculation with arbuscular mycorrhizal fungi(AMF). In northeast China, Shuangcheng city in Heilongjiang province, a field experiment was conducted, using rice provided with 0%, 20%, 40%, 60%, 80%,and 100% of the local norm of fertilization(including N, phosphorus and potassium), with or without inoculation with Glomus mosseae. The volume, concentrations of total N(TN),dissolved N(DN) and particulate N(PN) of runoff water were measured. We found that the local norm of fertilization led to 18.9 kg/ha of N runoff during rice growing season, with DN accounting for 60%–70%. We also found that reduction in fertilization by 20% cut down TN runoff by 8.2% while AMF inoculation decreased N runoff at each fertilizer level and this effect was inhibited by high fertilization. The combination of inoculation with AMF and 80% of the local norm of fertilization was observed to reduce N runoff by 27.2%. 2Experimental study on seed-filling performance of maize bowl-tray precision seeder显示文摘According to the requirement of assorted maize transplanting machine,a maize bud-seed precise sowing device was presented for use in cold areas.Experiments were performed to optimise the parameters of a maize bowl-tray precise seeder to improve the seed-filling performance.When the diameter of brush seed wheel was 78 mm,the clearance of brushing seed was 3 mm,with the maize varieties Demeiya 1,the moisture content about 40%,the depth of shaped hole 6 mm,the two major factors,the diameter of shaped hole and the velocity of seed box,were tested at five levels for each factor using the orthogonal rotational regression experiment.The effects on the single-seed rate,multi-seed rate,empty-seed rate,broken-bud rate and damage rate were investigated.The test results indicated that the factors affecting the single-seed rate and the multi-seed rate were the diameters of shaped hole and the velocity of seeds box,and the factor affecting the empty-seed rate was primarily the diameter of shaped hole.This study determined the optimal parameters,and verified the results,which provide the basis for the design and performance improvement of the maize bud-seed bowl-tray precision seeder.Mao Xin Yi Shujuan Tao Guixiang Yang Li Liu Haiyan Ma Yongcai 2015International Journal of Agricultural and Biological Engineering2015,8,2:7
3FAT1, a direct transcriptional target of E2F1, suppresses cell proliferation, migration and invasion in esophageal squamous cell carcinoma显示文摘Objective: Growing evidence indicates that FAT atypical cadherin 1(FAT1) has aberrant genetic alterations and exhibits potential tumor suppressive function in esophageal squamous cell carcinoma(ESCC). However, the role of FAT1 in ESCC tumorigenesis remains not well elucidated. The aim of this study was to further investigate genetic alterations and biological functions of FAT1, as well as to explore its transcriptional regulation and downstream targets in ESCC.Methods: The mutations of FAT1 in ESCC were achieved by analyzing a combined study from seven published genomic data, while the copy number variants of FAT1 were obtained from an analysis of our previous data as well as of The Cancer Genome Atlas(TCGA) and Cancer Cell Line Encyclopedia(CCLE) databases using the cBioPortal. The transcriptional regulation of FAT1 expression was investigated by chromatin immunoprecipitation(ChIP) and the luciferase reporter assays. In-cell western, Western blot and reverse transcription-quantitative polymerase chain reaction(RT-qPCR) were used to assess the indicated gene expression. In addition, colony formation and Transwell migration/invasion assays were employed to test cell proliferation, migration and invasion.Finally, RNA sequencing was used to study the transcriptomes.Results: FAT1 was frequently mutated in ESCC and was deleted in multiple cancers. Furthermore, the transcription factor E2 F1 occupied the promoter region of FAT1, and depletion of E2 F1 led to a decrease in transcription activity and mRNA levels of FAT1. Moreover, we found that knockdown of FAT1 promoted KYSE30 and KYSE150 cell proliferation, migration and invasion;while overexpression of FAT1 inhibited KYSE30 and KYSE410 cell proliferation, migration and invasion. 4Is resource allocation and grain yield of rice altered by inoculation with arbuscular mycorrhizal fungi?显示文摘Aims our study quantified the combined effects of fertilization and inoculation with arbuscular mycorrhizal fungi(AMF)on grain yield and allocation of biomass and nutrients in field-grown rice(Oryza sativa l.).Methods a two-factor experiment was conducted at a field site in northeast of China(in shuangcheng,Heilongjiang Province,songhua river basin):six nitrogen-phosphorus-potassium fertilizer levels were provided(0,20,40,60,80 and 100%of the local norm of ferti-lizer supply),with or without inoculation with Glomus mosseae.at maturity,we quantified the percentage of root length colonization by AMF,grain yield,shoot:root ratios,shoot N and P contents and nutrients allocated to panicles,leaves and stems.Important Findingsas expected,inoculation resulted in greatly increased AMF colo-nization,which in turn led to higher shoot:root ratios and greater shoot N contents.shoot:root ratios of inoculated rice increased with increasing fertilization while there was a significant interaction between fertilization and inoculation on shoot:root ratio.additionally,a F inoculation increased panicle:shoot ratios,panicle N:shoot N ratios and panicle P:shoot P ratios,especially in plants grown at low fertilizer levels.Importantly,inoculated rice exhibited higher grain yield,with the maximum improvement(near 62%)at the lower fertilizer end.our results showed that(i)AMFinoculated plants conform to the functional equilibrium theory,albeit to a reduced extent compared to non-inoculated plants and(ii)AMF inoculation resulted in greater allocation of shoot biomass to panicles and increased grain yield by stimulating N and P redis-tribution to panicles.Shujuan Zhang Li Wang Fang Ma Keith J.Bloomfield Jixian Yang Owen K.Atkin 2015Journal of Plant Ecology2015,8,4:5
5Can arbuscular mycorrhiza and fertilizer management reduce phosphorus runoff from paddy fields?显示文摘Our study sought to assess how much phosphorus(P) runoff from paddy fields could be cut down by fertilizer management and inoculation with arbuscular mycorrhizal fungi. A field experiment was conducted in Lalin River basin, in the northeast China: six nitrogen–phosphorus–potassium fertilizer levels were provided(0, 20%, 40%, 60%, 80%, and 100% of the recommended fertilizer supply), with or without inoculation with Glomus mosseae. The volume and concentrations of particle P(PP) and dissolved P(DP) were measured for each runoff during the rice growing season. It was found that the seasonal P runoff, including DP and PP, under the local fertilization was 3.7 kg/ha, with PP, rather than DP, being the main form of P in runoff water. Additionally, the seasonal P runoff dropped only by 8.9% when fertilization decreased by 20%; rice yields decreased with declining fertilization. We also found that inoculation increased rice yields and decreased P runoff at each fertilizer level and these effects were lower under higher fertilization. 6Disruption of the bHLH transcription factor Abnormal Tapetum 1 causes male sterility in watermelon显示文摘Although male sterility has been identified as a useful trait for hybrid vigor utilization and hybrid seed production,its underlying molecular mechanisms in Cucurbitaceae species are still largely unclear.Here,a spontaneous male-sterile watermelon mutant,Se18,was reported to have abnormal tapetum development,which resulted in completely aborted pollen grains.Map-based cloning demonstrated that the causal gene Citrullus lanatus Abnormal Tapetum 1(ClATM1)encodes a basic helix-loop-helix(bHLH)transcription factor with a 10-bp deletion and produces a truncated protein without the bHLH interaction and functional(BIF)domain in Se18 plants.qRT–PCR and RNA in situ hybridization showed that ClATM1 is specifically expressed in the tapetum layer and in microsporocytes during stages 6–8a of anther development.The genetic function of ClATM1 in regulating anther development was verified by CRISPR/Cas9-mediated mutagenesis.Moreover,ClATM1 was significantly downregulated in the Se18 mutant,displaying a clear dose effect at the transcriptional level.Subsequent dual-luciferase reporter,β-glucuronidase(GUS)activity,and yeast one-hybrid assays indicated that ClATM1 could activate its own transcriptional expression through promoter binding.Collectively,ClATM1 is the first male sterility gene cloned from watermelon,and its self-regulatory activity provides new insights into the molecular mechanism underlying anther development in plants.Ruimin Zhang Jingjing Chang Jiayue Li Guangpu Lan Changqing Xuan Hao Li Jianxiang Ma Yong Zhang Jianqiang Yang Shujuan Tian Li Yuan Xian Zhang Chunhua Wei 2021Horticulture Research2021,8,1:3

8A near-infrared phosphorescent probe for F^- based on a cationic iridium(Ⅲ) complex with triarylboron moieties显示文摘In this work, a near-infrared (NIR) phosphorescent probe for F- based on a cationic Ir(Ⅲ) complex [Ir(Bpq)2(quqo)]PF6 (1) with dimesitylboryl (Mes2B) groups on the cyclometalated C^N ligands (Bpq) and 2-(quinolin-2-yl)quinoxaline (quqo) as N^N ligand was designed and synthesized. The excited state properties of 1 were investigated in detail using molecular orbital cal- culations and experimental methods. Upon excitation, complex 1 shows NIR phosphorescent emission around 680 nm. Inter- estingly, the complex can be excited with long wavelength around 610 nm. Such long-wavelength excitation can reduce the background emission interference and improve the signal-to-noise ratio. Furthermore, the selective binding between boron at- om and F- can give rise to the quenching of emission and realize the near-infrared phosphorescent sensing for F-. 9Stimuli-responsive photofunctional materials for green and security printing显示文摘Information recording on paper has always been the most important approach to keep records of human activity and to spread civilization.With the progress of science and technology,paper with different functions should be exploited to conform to the increasing demands in various scenarios.In one aspect,traditional paper can only be used once,and using large amounts of paper causes deforestation,additional solid waste treatment,environmental pollution,and high energy consumption.Consequently,the development of rewritable paper that is environment-friendly,low cost,and can save resources is significant for green printing.In the other aspect,information leakage brings security issues,which may lead to severe consequences,such as war outbreak,economic loss,social problems,and so on.Therefore,the development of security printing has also attracted wide interests.Stimuli-responsive photofunctional materials that have reversible variations in absorption or emission in response to changes in the external environmental have a great potential for the achievement of green and security printing.To date,much progress has been made in these research areas.This paper lists different smart materials that respond to various external stimuli,such as light,water,pH,heat,and metal ions,and summarizes the recent advances towards green and security printing.Also,we discuss the current challenges and future directions in this rapidly growing research field.It is expected that this review article will stimulate and guide future studies for the advanced green and security printing.Yun Ma Yaxin Yu Jiangang Li Shujuan Liu Wei Huang Qiang Zhao 2021InfoMat2021,3,1:2
10Human cytomegalovirus miR-US5-1 inhibits viral replication by targeting Geminin mRNA显示文摘Viruses commonly create favorable cellular conditions for their survival through multiple mechanisms. Micro RNAs(mi RNAs), which function as post-transcriptional regulators, are utilized by human cytomegalovirus(HCMV) in its infection and pathogenesis. In the present study, the DNA replication inhibitor Geminin(GMNN) was identified to be a direct target of hcmv-mi R-US5-1. Overexpression of hcmv-mi R-US5-1 could block the accumulation of GMNN during HCMV infection, and the decrease of GMNN expression caused by hcmv-mi R-US5-1 or GMNN specific si RNA reduced HCMV DNA copies in U373 cells. Meanwhile, ectopic expression of hcmv-mi R-US5-1 and consequent lower expression of GMNN influenced host cell cycle and proliferation. 20The binding epitope of sintilimab on PD-1 revealed by AbMap显示文摘PD-1 plays an important role as an immune checkpoint.Sintilimab is a newly approved PD-1 antibody for cancer immunotherapy with an unknown binding epitope on PD-1.In this study,to elucidate the molecular mechanism by which sintilimab blocks PD-1 activation,we applied Antibody binding epitope Mapping(AbMap)to identify the binding epitope of sintilimab.An epitope was successfully identified,i.e.SLAPKA,aa 127-132.By constructing a series of point mutations,the dominant residues S127,L128,A129,P130,and A132 of PD-1 were further validated by western blot analysis,biolayer interferometry,and flow cytometry.Structural analysis showed that the epitope is partially within the binding interface of PD-1 and PD-L1,and this epitope also partially overlaps with that of nivolumab and pembrolizumab.These results demonstrate that sintilimab can attenuate PD-1 activation by directly competing with the interaction between PD-1 and PD-L1 through binding with the key residues of the FG loop on PD-1.This study also demonstrates the high efficiency and accuracy of AbMap for determining the binding epitope of therapeutic antibodies.Mingliang Ma Huan Qi Chuansheng Hu Zhaowei Xu Fanlin Wu Nan Wang Danyun Lai Yang Li Hainan Zhang Hewei Jiang Qingfeng Meng Shujuan Guo Yani Kang Xiaodong Zhao Hua Li Sheng-ce Tao 2021Acta Biochimica et Biophysica Sinica2021,53,5:1
