11株红树林来源枝孢属真菌的分离鉴定及纤维素酶性质分析显示文摘利用水解圈法从三亚红沙河红树林区分离得到1株纤维素降解真菌SCSIO 43503。分子生物学鉴定表明,该菌ITS序列与菌株Ochrocladosporium frigidarii CZ549(FJ755255)的相似度为96%,β微管蛋白(β-tubulin)序列与Shiraia bambusicola(AB355003)的相似性为86%,钙调蛋白(calmodulin)与Paramyrothecium viridisporum CBS 873.85(KU846278)序列相似性为89%;综合其形态学特征,推测其为枝孢属(Gladaxporism)真菌,命名为Gladaxporism sp.SCSIO 43503(KY224732)。进一步对该菌株分泌的纤维素酶进行分析,结果显示该菌株维素酶活性的最适反应温度为50℃,在45~50℃范围内活性较高;最适pH为5.0,在3.0~7.0范围内具有较高的活性,最适发酵时间为3 d。在最优条件下,酶活力达到最高,为23.46 U。本研究发现了一株纤维素降解真菌,对纤维素酶的生产和利用具有重要价值。曾思泉 凌娟 林丽云 Manzoor Ahmad 张燕英 张颖 董俊德 2018微生物学杂志2018,38,2:16
2Antibiotic resistance and cagA gene correlation:A looming crisis of Helicobacter pylori显示文摘AIM:To determine antibiotic resistance of Helicobacter pylori(H.pylori) in Pakistan and its correlation with host and pathogen associated factors.METHODS:A total of 178 strains of H.pylori were isolated from gastric biopsies of dyspeptic patients.Susceptibility patterns against first and second-line antibiotics were determined and trends of resistance were analyzed in relation to the sampling period,gastric conditions and cagA gene carriage.The effect of cagA gene on the acquisition of resistance was investigated by mutant selection assay.RESULTS:The observations showed that monoresistant strains were prevalent with rates of 89% for metronidazole,36% for clarithromycin,37% for amoxicillin,18.5% for ofloxacin and 12% for tetracycline.Furthermore,clarithromycin resistance was on the rise from 2005 to 2008(32% vs 38%,P = 0.004) and it is significantly observed in non ulcerative dyspeptic patients compared to gastritis,gastric ulcer and duodenal ulcer cases(53% vs 20%,18% and 19%,P = 0.000).On the contrary,metronidazole and ofloxacin resistance were more common in gastritis and gastric ulcer cases.Distribution analysis and frequencies of resistant mutants in vitro correlated with the absence of cagA gene with metronidazole and ofloxacin resistance.CONCLUSION:The study confirms the alarming levels of antibiotic resistance associated with the degree of gastric inflammation and cagA gene carriage in H.pylori strains.Adnan Khan Amber Farooqui Hamid Manzoor Syed Shakeel Akhtar Muhammad Saeed Quraishy Shahana Urooj Kazmi 2012World Journal of Gastroenterology2012,18,18:15

4Trajectory tracking control of a VTOL unmanned aerial vehicle using offset-free tracking MPC显示文摘Designing a stable and robust flight control system for an Unmanned Aerial Vehicle(UAV)is an arduous task.This paper addresses the trajectory tracking control problem of a Ducted Fan UAV(DFUAV)using offset-free Model Predictive Control(MPC)technique in the presence of various uncertainties and external disturbances.The designed strategy aims to ensure adequate flight robustness and stability while overcoming the effects of time delays,parametric uncertainties,and disturbances.The six degrees of freedom DFUAV model is divided into three flight modes based on its airspeed,namely the hover,transition,and cruise mode.The Dryden wind turbulence is applied to the DFUAV in the linear and angular velocity component.Moreover,different uncertainties such as parametric,time delays in state and input,are introduced in translational and rotational components.From the previous work,the Linear Quadratic Tracker with Integrator(LQTI)is used for comparison to corroborate the performance of the designed controller.Simulations are computed to investigate the control performance for the aforementioned modes and different flight phases including the autonomous flight to validate the performance of the designed strategy.Finally,discussions are provided to demonstrate the effectiveness of the given methodology.Tayyab MANZOOR Yuanqing XIA Di-Hua ZHAI Dailiang MA 2020Chinese Journal of Aeronautics2020,33,7:10
5Hepatitis B virus therapy:What's the future holding for us?显示文摘Hepatitis B is one of the leading causes of liver cancer worldwide and unfortunately the number of people affected with hepatitis B virus(HBV) infection is still on the rise. Although the HBV has been known to cause fatal illness since decades but the population effected by this lethal virus have still only a few options for its management. The major treatment strategies include interferons and nucleos(t)ide analogues. These agents have so far produced unsatisfactory results in terms of complete virus eradication. Interferons cannot be used for long term therapy because of their potential side effects. Prolong treatment with nucleos(t)ide analogues has also been reported to cause serious side effects besides the increasing resistance by the virus. The need for new innovative solutions for treatment of HBV has been realized by global research institutes and pharmaceutical industry. Present review focuses in detail on the new ideas that are being transformed into therapeutic tools for use as future therapies in HBV infection. Modern drug designing and screening methods have made the drug discovery process shorter and more reliable. HBV therapeutics will take a new turn in coming years owing to these intelligent drug designing and screening methods. 6Hepatitis C virus NS5A promotes insulin resistance through IRS-1 serine phosphorylation and increased gluconeogenesis显示文摘AIM: To investigate the mechanisms of insulin resistance in human hepatoma cells expressing hepatitis C virus(HCV) nonstructural protein 5A(NS5A).METHODS: The human hepatoma cell lines,Huh7 and Huh7.5,were infected with HCV or transientlytransfected with a vector expressing HCV NS5 A. The effect of HCV NS5 A on the status of the critical players involved in insulin signaling was analyzed using realtime quantitative polymerase chain reaction and Western blot assays. Data were analyzed using Graph Pad Prism version 5.0.RESULTS: To investigate the effect of insulin treatment on the players involved in insulin signaling pathway,we analyzed the status of insulin receptor substrate-1(IRS-1) phosphorylation in HCV infected cells or Huh7.5 cells transfected with an HCV NS5 A expression vector. Our results indicated that there was an increased phosphorylation of IRS-1(Ser307) in HCV infected or NS5 A transfected Huh7.5 cells compared to their respective controls. Furthermore,an increased phosphorylation of Akt(Ser473) was observed in HCV infected and NS5 A transfected cells compared to their mock infected cells. In contrast,we observed decreased phosphorylation of Akt Thr308 phosphorylation in HCV NS5 A transfected cells. These results suggest that Huh7.5 cells either infected with HCV or ectopically expressing HCV NS5 A alone have the potential to induce insulin resistance by the phosphorylation of IRS-1 at serine residue(Ser307) followed by decreased phosphorylation of Akt Thr308,Fox01 Ser256 and GSK3β Ser9,the downstream players of the insulin signalingpathway. 7Deep reinforcement learning based computation offloading and resource allocation for low-latency fog radio access networks显示文摘Fog Radio Access Networks(F-RANs)have been considered a groundbreaking technique to support the services of Internet of Things by leveraging edge caching and edge computing.However,the current contributions in computation offloading and resource allocation are inefficient;moreover,they merely consider the static communication mode,and the increasing demand for low latency services and high throughput poses tremendous challenges in F-RANs.A joint problem of mode selection,resource allocation,and power allocation is formulated to minimize latency under various constraints.We propose a Deep Reinforcement Learning(DRL)based joint computation offloading and resource allocation scheme that achieves a suboptimal solution in F-RANs.The core idea of the proposal is that the DRL controller intelligently decides whether to process the generated computation task locally at the device level or offload the task to a fog access point or cloud server and allocates an optimal amount of computation and power resources on the basis of the serving tier.Simulation results show that the proposed approach significantly minimizes latency and increases throughput in the system.G.M.Shafiqur Rahman Tian Dang Manzoor Ahmed 2020Intelligent and Converged Networks2020,1,3:5

9A journey towards shared governance: status and prospects for collaborative management in the protected areas of Bangladesh显示文摘Establishment of Protected Areas (PAs), in the face of rapid deforestation, forest degradation and climate change has been one of the key efforts in conservation of biodiversity worldwide in recent times. While Bangladesh has gained a degree of prominence in the world for its successful social forestry programs, the concept of collaborative protected area management is rather new in the country, initiated in 2004 by the Bangladesh Forest Department in five PAs with financial assistance from USAID. Based on empirical evidence from three of the pilot PAs, we examined the achievements and associated challenges and prospects for co-management. Our fieldwork revealed a number of challenges faced by co-management institutions: (1) institutions were dominated by the elite group, overshadowing the voice of the community people; (2) mutual trust and collective performance are key to good governance but had not taken root in the PAs; (3) encroachment onto forest land and subsequent conversion to agriculture remained a serious problem that discouraged forest-dependent people from participating actively in co-management initiatives; (4) legal provisions (including acts, rules and policies) were not clearly and adequately disseminated and understood at the community level; (5) there remained a degree of ambiguity regarding the roles and responsibilities of forest department (FD) and co-management committees (CMC) in field operations, and this was not enhancing transparency and accountability of the overall initiative; (6) the long-term sustainability of co-management institutions was another major concern, as the local intuitional structure was still in a nascent stage, and provisioning of resources (either internally or externally) remained somewhat uncertain. 11斑衣蜡蝉若虫寄生蜂地理种群遗传差异及早期快速检测显示文摘【目的】明确不同地理种群斑衣蜡蝉Lycorma delicatula若虫寄生蜂种群遗传差异,实现若虫被寄生早期的快速准确检测,以评估其对斑衣蜡蝉种群的控制作用。【方法】利用DNA条形码技术获得不同地理种群斑衣蜡蝉若虫寄生蜂COI和28S rDNA序列,利用K2P模型计算不同地理种群间的遗传距离,以邻接(neighbor-joining,NJ)法构建系统发育树;基于COI序列设计种特异性PCR(SS-PCR)引物,利用SS-PCR对斑衣蜡蝉若虫DNA进行扩增,测定其体内是否有中华螯蜂Dryinus sinicus寄生;利用目测法观察和PCR扩增测定寄生蜂对不同采样点斑衣蜡蝉若虫的寄生率。【结果】经鉴定,不同地理种群斑衣蜡蝉若虫寄生蜂为中华螯蜂,该寄生蜂COI序列共检测到16个单倍型,28S rDNA序列共检测到4个单倍型。不同地理种群间中华螯蜂遗传距离在0.00691~0.01310之间。邻接法构建的系统发育树显示不同地理种群中华螯蜂均聚于一枝。基于COI序列设计的SS-PCR引物对中华螯蜂成虫、幼虫均具有良好的扩增效果,最低检测阈值为0.000005 ng/μL DNA。利用SS-PCR测定中华螯蜂对各采样点斑衣蜡蝉若虫的寄生率为22.54%~60.00%,显著高于目测统计的结果(5.63%~36.98%)。【结论】不同中华螯蜂地理种群间遗传差异较小;SS-PCR引物可用于不同地区中华螯蜂对斑衣蜡蝉早期寄生率的快速检测。辛蓓 Atif MANZOOR 曹亮明 王小艺 2020昆虫学报2020,63,2:3
12海草根际微生物与海草植株的互作效应显示文摘【目的】本文以三亚湾泰来草根际沉积物为主要研究对象,研究室内培养条件下泰来草根际沉积物微生物对于高温处理和海草定殖的响应。【方法】通过对培养过程中水体物理化学参数(如pH、溶解氧、磷酸盐、硝酸盐、亚硝酸盐和铵盐)的监测以分析环境因子的变化;16S rRNA扩增子测序研究微生物群落结构的动态响应;通过荧光定量分析16SrRNA基因丰度变化。【结果】研究表明高温处理组在培养35 d后海水中磷酸盐、硝酸盐、亚硝酸盐和铵盐含量以及pH均要高于模拟原位环境的对照组,高温处理组根际沉积物微生物丰度在培养过程中呈现先上升后降低的趋势,同时,高温处理组根际微生物中初始阶段由厚壁菌门(32.4%)、变形菌门(22.92%)和梭杆菌门(27.21%)占据优势,培养一段时间后,厚壁菌门和梭杆菌门大幅度减少,逐渐被蓝细菌门和放线菌门所替代,最终由变形菌门(51.1%)占据主导地位,其中,属于硫还原细菌的脱硫杆菌科(Desulfobacteraceae)和硫氧化细菌的螺杆菌科(Helicobacteraceae)的细菌丰度不断提高。【结论】揭示了海草的定殖会提高高温处理后沉积物的多样性,并塑造和改善其根际沉积物微生物群落组成。周卫国 丁德文 凌娟 林显程 杨清松 张颖 Manzoor Ahmad 张燕英 董俊德 2019微生物学报2019,59,11:3
13一株海草沉积物菲降解菌的筛选、鉴定和降解特性显示文摘【背景】多环芳烃(Polycyclic Aromatic Hydrocarbons,PAHs)是一类高毒性的有机污染物,在海洋环境尤其是沿海环境中广泛分布。海草床生态系统作为沿海环境的重要组成部分,深受环境污染等人类活动的影响而处于严重衰退的状态。微生物修复是修复环境中多环芳烃污染的重要途径,具有经济简便、环境友好和无二次污染等特点。【目的】从深圳市大亚湾的海草床沉积物中筛选获得高效多环芳烃降解菌,并分析其降解特性,从而探究海草床生态系统中多环芳烃污染物的微生物修复可行性。【方法】以多环芳烃菲为唯一碳源从海草床沉积物样品中筛选菌株,再通过形态学观察、生理生化实验和16SrRNA基因序列对筛选的菌株进行鉴定,并利用特定引物扩增多环芳烃降解的功能基因——双加氧酶(nidA)基因,最后通过培养实验分析该菌株对菲的降解特性。【结果】筛选出一株高效降解菲的菌株SCSIO 43702,经鉴定为玫瑰杆菌属(Roseovarius)的潜在新菌,并成功扩增得到双加氧酶相似(nidA like)基因;培养实验结果表明,玫瑰杆菌SCSIO 43702在10 d内对100 mg/L菲的降解率最高可达96%,而且其对菲的最适降解条件为:温度30℃、pH值7.5和8.0、盐度3%。【结论】玫瑰杆菌SCSIO 43702凭借其良好的菲降解能力和较强的环境适应性,具有进一步被开发为微生物菌剂以用于多环芳烃污染修复的巨大潜力,为海草床生态系统中多环芳烃污染的微生物修复研究提供了理论依据和可利用的微生物资源。张煜航 Manzoor Ahmad 董俊德 杨清松 周卫国 凌娟 2021微生物学通报2021,48,6:3

15Evaluation of biomarkers, genetic mutations, and epigenetic modifications in early diagnosis of pancreatic cancer显示文摘BACKGROUND Pancreatic cancer(PC)is one of the deadliest malignancies with an alarming mortality rate.Despite significant advancement in diagnostics and therapeutics,early diagnosis remains elusive causing poor prognosis,marred by mutations and epigenetic modifications in key genes which contribute to disease progression.AIM To evaluate the various biological tumor markers collectively for early diagnosis which could act as prognostic biomarkers and helps in future therapeutics of PC in Kashmir valley.METHODS A total of 50 confirmed PC cases were included in the study to evaluate the levels of carbohydrate antigen 19-9(CA 19-9),tissue polypeptide specific antigen(TPS),carcinoembryonic antigen(CEA),vascular endothelial growth factor-A(VEGF-A),and epidermal growth factor receptor(EGFR).Mutational analysis was performed to evaluate the mutations in Kirsten rat sarcoma(KRAS),Breast cancer type 2(BRCA-2),and deleted in pancreatic cancer-4(DPC-4)genes.However,epigenetic modifications(methylation of CpG islands)were performed in the promoter regions of cyclin-dependent kinase inhibitor 2A(p16;CDKN2A),MutL homolog 1(hMLH1),and Ras association domain-containing protein 1(RASSF1A)genes.RESULTS We found significantly elevated levels of biological markers CA 19-9(P≤0.05),TPS(P≤0.05),CEA(P≤0.001),and VEGF(P≤0.001).Molecular genetic analysis revealed that KRAS gene mutation is predominant in codon 12(16 subjects,P≤0.05),and 13(12 subjects,P≤0.05).However,we did not find a mutation in DPC-4(1203G>T)and BRCA-2(617delT)genes.Furthermore,epigenetic modification revealed that CpG methylation in 21(P≤0.05)and 4 subjects in the promoter regions of the p16 and hMLH1 gene,respectively.CONCLUSION In conclusion,CA 19-9,TPS,CEA,and VEGF levels were significantly elevated and collectively have potential as diagnostic and prognostic markers in PC.Global data of mutation in the KRAS gene commonly in codon 12 and rare in codon 13 could augment the predisposition towards PC.Additionally,methylation of the p16 gene could also modulate transcription of genes thereby increasing the predisposition and susceptibility towards PC.Bilal Rah Manzoor Ahmad Banday Gh Rasool Bhat Omar J Shah Humira Jeelani Fizalah Kawoosa Tahira Yousuf Dil Afroze 2021World Journal of Gastroenterology2021,27,36:2
16Rice molecular markers and genetic mapping:Current status and prospects显示文摘Dramatic changes in climatic conditions that supplement the biotic and abiotic stresses pose severe threat to the sustainable rice production and have made it a difficult task for rice molecular breeders to enhance production and productivity under these stress factors. The main focus of rice molecular breeders is to understand the fundamentals of molecular pathways involved in complex agronomic traits to increase the yield. The availability of complete rice genome sequence and recent improvements in rice genomics research has made it possible to detect and map accurately a large number of genes by using linkage to DNA markers. Linkage mapping is an effective approach to identify the genetic markers which are co-segregating with target traits within the family. The ideas of genetic diversity, quantitative trait locus(QTL) mapping, and marker-assisted selection(MAS) are evolving into more efficient concepts of linkage disequilibrium(LD) also called association mapping and genomic selection(GS), respectively. The use of cost-effective DNA markers derived from the fine mapped position of the genes for important agronomic traits will provide opportunities for breeders to develop high-yielding, stress-resistant, and better quality rice cultivars. We herein reported a 32 years old female operated for lower abdominal lump,which was diagnosed,as PNET later confirmed by immunohistochemistry (IHC). She was lost to follow-up and presented 10 months later with recurrence. 20Spatial Trend of Foot and Mouth Disease Virus (FMDV) Serotypes in Cattle and Buffaloes, Pakistan显示文摘The present study describes the frequency of Foot and Mouth Disease (FMD) virus serotypes (O, A and Asia-1) in major regions (all provinces) of Pakistan using Indirect Sandwich ELISA. Also, spatial distribution of various FMD serotypes and their comparison is discussed. A total of 590 samples (Epithelial tissue) have been analyzed during a period of five years (2005-2009). Out of 590 samples, 180 were found positive, giving an overall confirmation of FMDV about 33.2 %. Of the prevalent serotypes, FMDV 'O' serotype caused most outbreaks (20.7 %), followed by serotype A (6.6 %) and serotype Asia-1 (4.6 %) while there was no positive case of type 'C'. The study clearly showed that the disease was more frequent in the agro-climatic zones than in hilly areas. Based on the data of 590 samples (>50 outbreaks), the overall prevalence of FMDV in cattle and buffaloes in Pakistan was 33.2 %, while in cattle alone, it was 37.1 %, higher than in buffalo (28.7 %). There were eight cases of mixed serotypes infection, indicating the presence of endemic state of disease. Another significant feature was the change over time. In phase-I (2005-2007), there was an overall prevalence of 29.4 %, while the occurrence of the serotype O, A and Asia-1 was 20.4 %, 2.9 % and 4.7 %, respectively. During phase-II (2008-2009), the overall prevalence was 59.21 %, while those of serotype O, A and Asia-1 were 22.4 %, 31.6 % and 4.0 %, respectively.
