1Soil Microbiological Activity and Carbon Dynamics in the Current Climate Change Scenarios:A Review显示文摘Microbial activities are affected by a myriad of factors with end points involved in nutrient cycling and carbon sequestration issues.Because of their prominent role in the global carbon balance and their possible role in carbon sequestration, soil microbes are very important organisms in relation to global climate changes. This review focuses mainly on the responses of soil microbes to climate changes and subsequent effects on soil carbon dynamics. An overview table regarding extracellular enzyme activities(EAA) with all relevant literature data summarizes the effects of different ecosystems under various experimental treatments on EAA. Increasing temperature, altered soil moisture regimes, and elevated carbon dioxide significantly affect directly or indirectly soil microbial activities.High temperature regimes can increase the microbial activities which can provide positive feedback to climate change, whereas lower moisture condition in pedosystem can negate the increase, although the interactive effects still remain unanswered. Shifts in soil microbial community in response to climate change have been determined by gene probing, phospholipid fatty acid analysis(PLFA),terminal restriction length polymorphism(TRFLP), and denaturing gradient gel electrophoresis(DGGE), but in a recent investigations,omic technological interventions have enabled determination of the shift in soil microbe community at a taxa level, which can provide very important inputs for modeling C sequestration process. 4Effect of fluence and ambient environment on the surface and structural modification of femtosecond laser irradiated Ti显示文摘Under certain conditions, ultrafast pulsed laser interaction with matter leads to the formation of self-organized conical as well as periodic surface structures(commonly reffered to as, laser induced periodic surface structures, LIPSS). The purpose of the present investigations is to explore the effect of fsec laser fluence and ambient environments(Vacuum &O_2) on the formation of LIPSS and conical structures on the Ti surface. The surface morphology was investigated by scanning electron microscope(SEM). The ablation threshold with single and multiple(N = 100) shots and the existence of an incubation effect was demonstrated by SEM investigations for both the vacuum and the O_2 environment. The phase analysis and chemical composition of the exposed targets were performed by x-ray diffraction(XRD) and energy dispersive x-ray spectroscopy(EDS), respectively. SEM investigations reveal the formation of LIPSS(nano & micro). FFT d-spacing calculations illustrate the dependence of periodicity on the fluence and ambient environment. The periodicity of nano-scale LIPSS is higher in the case of irradiation under vacuum conditions as compared to O_2. Furthermore, the O2 environment reduces the ablation threshold. XRD data reveal that for the O_2 environment, new phases(oxides of Ti) are formed. EDS analysis exhibits that after irradiation under vacuum conditions, the percentage of impurity element(Al) is reduced. 7Role of PTPN2/22 polymorphisms in pathophysiology of Crohn's disease显示文摘AIM To establish the relationship of protein tyrosine phosphatase non-receptor type 2 and 22(PTPN2/22) polymorphisms and mycobacterial infections in Crohn's disease(CD). METHODS All 133 subjects' blood samples were genotyped for nine single nucleotide polymorphisms(SNPs) in PTPN2/22 using TaqMan^(?) genotyping, while the effect of the SNPs on PTPN2/22 and IFN-γ gene expression was determined using RT-PCR. Detection of Mycobacterium avium subspecies paratuberculosis(MAP) IS900 gene was done by nPCR after DNA extraction from the isolated leukocytes of each subjects' blood samples. T-cells isolated from the patient samples were tested for response to phytohematoagglutonin(PHA) mitogen or mycobacterial antigens by Brd U proliferation assays for T-cell activity. RESULTS Out of the nine SNPs examined, subjects with either heterozygous(TC)/minor(CC) alleles in PTPN2:rs478582 occurred in 83% of CD subjects compared to 61% healthy controls(P-values < 0.05; OR = 3.03). Subjects with either heterozygous(GA)/minor(AA) alleles in PTPN22:rs2476601 occurred in 16% of CD compared to 6% healthy controls(OR = 2.7). Gene expression in PTPN2/22 in CD subjects was significantly decreased by 2 folds compared to healthy controls(P-values < 0.05). IFN-γ expression levels were found to be significantly increased by approxiately 2 folds in subjects when either heterozygous or minor alleles in PTPN2:rs478582 and/or PTPN22:rs2476601 were found(P-values < 0.05). MAP DNA was detected in 61% of CD compared to only 8% of healthy controls(P-values < 0.05, OR = 17.52), where subjects with either heterozygous or minor alleles in PTPN2:rs478582 and/or PTPN22:rs2476601 had more MAPbacteremia presence than subjects without SNPs did. T h e average T-cell proliferation in CD treated with PHA or mycobacteria antigens was, respectively, 1.3 folds and 1.5 folds higher than healthy controls without any significant SNP. 17Critical medical ecology and SARS-COV-2 in the urban environment:a pragmatic,dynamic approach to explaining and planning for research and practice显示文摘Background:Practitioners and researchers in the midst of overwhelming coronavirus disease 2019(COVID-19)outbreaks are calling for new ways of looking at such pandemics,with an emphasis on human behavior and holistic considerations.Viral outbreaks are characterized by socio-behaviorally-oriented public health efforts aimed at reducing exposure and prevention of morbidity/mortality once infected.These efforts involve different points-ofview,generally,than do those aimed to understand the virus’natural history.Rampant spread of SARS-CoV-2 infection in cities clearly signals that urban areas contain conditions favorable for rapid transmission of the virus.Main text:The Critical Medical Ecology model is a multidimensional,multilevel way of viewing pandemics comprehensively,rooted simultaneously in microbiology and in anthropology,with shared priority for evolution,context,stressors,homeostasis,adaptation,and power relationships.Viewing COVID-19 with a Critical Medical Ecological lens suggests three important interpretations:1)COVID-19 is equally-if not more-a socially-driven disease as much as a biomedical disease,2)the present interventions available for primary prevention of transmission are social and behavioral interventions,and 3)wide variation in COVID-19 hospitalization/death rates is not expected to significantly be attributable to a more virulent and rapidly-evolving virus,but rather to differences in social and behavioral factors-and power dynamics-rather than(solely)biological and clinical factors.Cities especially are challenged due to logistics and volume of patients,and lack of access to sustaining products and services for many residents living in isolation.Conclusions:In the end,SARS-CoV-2 is acting upon dynamic social human beings,entangled within structures and relationships that include but extend far beyond their cells,and in fact beyond their own individual behavior.As a comprehensive way of thinking,the Critical Medical Ecology model helps identify these elements and dynamics in the context of ecological processes that create,shape,and sustain people in their multidimensional,intersecting environments.Timothy De Ver Dye Erin Muir Lorne Farovitch Shazia Siddiqi Saloni Sharma 2020Infectious Diseases of Poverty2020,9,3:1
