2SDF-1/CXCR4 axis modulates bone marrow mesenchymal stem cell apoptosis, migration and cytokine secretion显示文摘Bone marrow mesenchymal stem cells(MSCs)are considered as a promising cell source to treat the acute myocardial infarction.However,over 90%of the stem cells usually die in the first three days of transplantation.Survival potential,migration ability and paracrine capacity have been considered as the most important three factors for cell transplantation in the ischemic cardiac treatment.We hypothesized that stromal-derived factor-1(SDF-1)/CXCR4 axis plays a critical role in the regulation of these processes.In this study,apoptosis was induced by exposure of MSCs to H2O2 for 2 h.After re-oxygenation,the SDF-1 pretreated MSCs demonstrated a significant increase in survival and proliferation.SDF-1 pretreatment also enhanced the migration and increased the secretion of pro-survival and angiogenic cytokines including basic fibroblast growth factor and vascular endothelial growth factor.Western blot and RT-PCR demonstrated that SDF-1 pretreatment significantly activated the pro-survival Akt and Erk signaling pathways and up-regulated Bcl-2/Bax ratio.These protective effects were partially inhibited by AMD3100,an antagonist of CXCR4.We conclude that the SDF-1/CXCR4 axis is critical for MSC survival,migration and cytokine secretion.Xiaolei Liu Biyan Duan Zhaokang Cheng Xiaohua Jia Lina Mao Hao Fu Yongzhe Che Lailiang Ou Lin Liu Deling Kong 2011Protein & Cell2011,2,10:53

4Timing of the Wudangshan, Yaolinghe volcanic sequences and mafic sills in South Qinling: U-Pb zircon geochronology and tectonic implication显示文摘The Wudangshan, Yaolinghe volcanic-sedimentary sequences and doleritic-gabbroic sills comprise the largest exposed Precambrian basement in South Qinling. Zircons separated from 5 volcanic-pyroclas- tic samples of the Wudangshan Group, 2 volcanic samples of the Yaolinghe Group and one sample for the mafic sills were used for U-Pb dating by laser ablation-inductively coupled plasma mass spec- trometry (LA-ICPMS). The results reveal that the Wudangshan volcanic sequence was formed at (755±3) Ma (a weighted mean from the 5 samples, MSWD=0.47), whereas the Yaolinghe volcanic suite and the mafic sill were crystallized at (685±5) (2 samples, MSWD=0.36) and (679±3) Ma (MSWD=1.6), respectively, which are equal to each other within analysis errors. These ages are markedly younger than those previously documented for the rocks. The newly obtained ages for the Wudangshan and Yaolinghe Groups are identical to those of the bottom Liantuo and slightly older than those of the Nantuo Forma- tions, respectively, lower strata of the Nanhua (middle to late Neoproterozoic) stratotype section in eastern Three Gorges, Yangtze craton. A range of inherited magmatic zircons was recognized with ages of 830 to 780 Ma, which are typical of Neoprotzrozoic magmatisms recorded along the margins and interior of the Yangtze craton. Thus, there is Neoproterozoic basement comprising 830―780 Ma igne- ous suites in South Qinling; the inherited zircons were detrital sediments derived from the northern margin of the Yangtze craton. 5Prognostic factors and long-term outcomes of hilar cholangiocarcinoma:A single-institution experience in China显示文摘AIM: To evaluate the prognostic factors of hilar cholangiocarcinoma in a large series of patients in a single institution.METHODS: Eight hundred and fourteen patients with a diagnosis of hilar cholangiocarcinoma that were evaluated and treated between 1990 and 2014, of which 381 patients underwent curative surgery, were included in this study. Potential factors associated with overall survival(OS) and disease-free survival(DFS) were evaluated by univariate and multivariate analyses.RESULTS: Curative surgery provided the best long-term survival with a median OS of 26.3 mo. The median DFS was 18.1 mo. Multivariate analysis showed that patients with tumor size > 3 cm [hazard ratio(HR) = 1.482, 95%CI: 1.127-1.949; P = 0.005], positive nodal disease(HR = 1.701, 95%CI: 1.346-2.149; P < 0.001), poor differentiation(HR = 2.535, 95%CI: 1.839-3.493; P < 0.001), vascular invasion(HR = 1.542, 95%CI: 1.082-2.197; P = 0.017), and positive margins(HR = 1.798, 95%CI: 1.314-2.461; P < 0.001) had poor OS outcome. The independent factors for DFS were positive nodal disease(HR = 3.383, 95%CI: 2.633-4.348; P < 0.001), poor differentiation(HR = 2.774, 95%CI: 2.012-3.823; P < 0.001), vascular invasion(HR = 2.136, 95%CI: 1.658-3.236; P < 0.001), and positive margins(HR = 1.835, 95%CI: 1.256-2.679; P < 0.001). Multiple logistic regression analysis showed that caudate lobectomy [odds ratio(OR) = 9.771, 95%CI: 4.672-20.433; P < 0.001], tumor diameter(OR = 3.772, 95%CI: 1.914-7.434; P < 0.001), surgical procedures(OR = 10.236, 95%CI: 4.738-22.116; P < 0.001), American Joint Committee On Cancer T stage(OR = 2.010, 95%CI: 1.043-3.870; P = 0.037), and vascular invasion(OR = 2.278, 95%CI: 0.997-5.207; P = 0.051) were independently associated with tumorfree margin, and surgical procedures could indirectly affect survival outcome by influencing the tumor resection margin. CONCLUSION: Tumor margin, tumor differentiation, vascular invasion, and lymph node status were independent factors for OS and DFS. 7Multimodal treatment for spinal cord injury: a sword of neuroregeneration upon neuromodulation显示文摘Spinal cord injury is linked to the interruption of neural pathways,which results in irreversible neural dysfunction.Neural repair and neuroregeneration are critical goals and issues for rehabilitation in spinal cord injury,which require neural stem cell repair and multimodal neuromodulation techniques involving personalized rehabilitation strategies.Besides the involvement of endogenous stem cells in neurogenesis and neural repair,exogenous neural stem cell transplantation is an emerging effective method for repairing and replacing damaged tissues in central nervous system diseases.However,to ensure that endogenous or exogenous neural stem cells truly participate in neural repair following spinal cord injury,appropriate interventional measures(e.g.,neuromodulation)should be adopted.Neuromodulation techniques,such as noninvasive magnetic stimulation and electrical stimulation,have been safely applied in many neuropsychiatric diseases.There is increasing evidence to suggest that neuromagnetic/electrical modulation promotes neuroregeneration and neural repair by affecting signaling in the nervous system;namely,by exciting,inhibiting,or regulating neuronal and neural network activities to improve motor function and motor learning following spinal cord injury.Several studies have indicated that fine motor skill rehabilitation training makes use of residual nerve fibers for collateral growth,encourages the formation of new synaptic connections to promote neural plasticity,and improves motor function recovery in patients with spinal cord injury.With the development of biomaterial technology and biomechanical engineering,several emerging treatments have been developed,such as robots,brain-computer interfaces,and nanomaterials.These treatments have the potential to help millions of patients suffering from motor dysfunction caused by spinal cord injury.However,large-scale clinical trials need to be conducted to validate their efficacy.This review evaluated the efficacy of neural stem cells and magnetic or electrical stimulation combined with rehabilitation training and intelligent therapies for spinal cord injury according to existing evidence,to build up a multimodal treatment strategy of spinal cord injury to enhance nerve repair and regeneration.Ya Zheng Ye-Ran Mao Ti-Fei Yuan Dong-Sheng Xu Li-Ming Cheng 2020Neural Regeneration Research2020,15,8:29

9U-Pb dating of detrital zircons from the Wudangshan Group in the South Qinling and its geological significance显示文摘U-Pb dating was made by the LA-ICP-MS method for detrital zircons from the Wudangshan volcanic-sedimentary succession in the South Qinling. Samples comprise quartz sandstones of the Wudangshan Group collected from the base of the Yangping For-mation and an upper layer of the Shuangtai Formation overlain its volcanic sequence, and two river-sand collections from the drainage systems cutting across the two formations, respectively. The results show that the Yangping detrital zircons are domi-nated by 830–780 Ma grains with a minor population of ~1.0–0.84 Ga and sporadic grains of ~2.6, ~2.4 and 2.0 Ga, whereas the Shuangtai zircons yield an upper intercept age of 763±33 Ma, identical to the timing of the Wudangshan volcanism within error, with few concordant grains of 1.9 and 0.86 Ga. Age spectra for the two river-sand samples are similar to those of the Yangping and Shuangtai Formations, respectively. It thus suggests that the Wudangshan strata are less than 780 Ma, whereas their major detrital zircon populations of 1.0–0.85 Ga and 830–780 Ma are consistent with timing of the Hannan magmatic activities along the northwestern margin of the South China Block. This suggests a Hannan or adjacent area provenance for the Wudangshan strata. The Wudang area is characterized by rift-related igneous events at ~755 and ~680 Ma, respectively, pointing to a tectono-magmatic history different from the Hannan area. 12Effects of blueberry on hepatic fibrosis and transcription factor Nrf2 in rats显示文摘AIM:To investigate the effects of blueberry on hepatic fibrosis and NF-E2-related factor 2(Nrf2) transcription factor in rats.METHODS:Forty-five male Sprague-Dawley rats were randomly divided into control group(A);CCl4-induced hepatic fibrosis group(B);blueberry prevention group(C);Dan-shao-hua-xian capsule(DSHX) prevention group(D);and blueberry + DSHX prevention group(E).Liver fibrosis was induced in rats by subcutaneous injection of CCl4 and a high-lipid/low-protein diet for 8 wk(except the control group).The level of hyaluronic acid(HA) and alanine aminotransferase(ALT) in serum was examined.The activity of superoxide dismutase(SOD),glutathione-S-transferase(GST) and malondialdehyde(MDA) in liver homogenates was determined.The degree of hepatic fibrosis was evaluated by hematoxylin and eosin and Masson staining.Expression of Nrf2 and NADPH quinone oxidoreductase 1(Nqo1) was detected by real-time reversed transcribed-polymerase chain reaction,immunohistochemical techniques,and western blotting.RESULTS:Compared with group B,liver indices,levels of serum HA and ALT of groups C,D and E were reduced(liver indices:0.038 ± 0.008,0.036 ± 0.007,0.036 ± 0.005 vs 0.054 ± 0.009,P<0.05;HA:502.33 ± 110.57 ng/mL,524.25 ± 255.42 ng/mL,499.25 ± 198.10 ng/mL vs 828.50 ± 237.83 ng/mL,P<0.05;ALT:149.44 ± 16.51 U/L,136.88 ± 10.07 U/L,127.38 ± 11.03 U/L vs 203.25 ± 31.62 U/L,P<0.05),and SOD level was significantly higher,but MDA level was lower,in liver homogenates(SOD:1.36 ± 0.09 U/mg,1.42 ± 0.13 U/mg,1.50 ± 0.15 U/mg vs 1.08 ± 0.19 U/mg,P<0.05;MDA:0.294 ± 0.026 nmol/mg,0.285 ± 0.025 nmol/mg,0.284 ± 0.028 nmol/mg vs 0.335 ± 0.056 nmol/mg,P<0.05).Meanwhile,the stage of hepatic fibrosis was significantly weakened(P<0.05).Compared with group A,the activity of GST liver homogenates and expression levels of Nrf2 and Nqo1 in group B were elevated(P<0.05).The expression level of Nrf2 and Nqo1 in groups C,D,and E were increased as compared with group B,but the difference was not significant.CONCLUSION:Blueberry has preventive and protective effects on CCl4-induced hepatic fibrosis by reducing hepatocyte injury and lipid peroxidation.However,these effects may not be related to the activation of Nrf2 during long-term of CCl4.Wang, Yu-Ping Cheng, Ming-Liang Zhang, Bao-Fang Mu, Mao Wu, Jun 2010World Journal of Gastroenterology2010,16,21:19
13Effect of blueberry on hepatic and immunological functions in mice显示文摘BACKGROUND:Conventional drugs used in the treatment and prevention of liver diseases often have side effects,therefore research into natural substances are of significance.This study examined the effects of blueberry on liver protection and cellular immune functions. METHODS:To determine the effects of blueberry on liver protective function,male mice were orally administered blueberry(0.6 g/10 g)or normal saline for 21 days.Hepatic RNA was extracted by Trizol reagent,and the expression of Nrf2,HO-1,and Nqo1 was determined by real-time RT- PCR.Superoxide dismutase(SOD)and malondialdehyde (MDA)in liver homogenate were determined,and liver index was measured.To assess the effects of blueberry on cellular immune function,male mice received blueberry(0.4,0.6,or 0.8 g/10 g)for 35 days,and the percentages of CD3 + ,CD4 + ,and CD8 + T lymphocyte subgroups in peripheral blood were detected by flow cytometry,the index of the thymus and spleen was measured,and lymphocyte proliferation in the spleen was determined by MTT assay. RESULTS:Blueberry treatment significantly increased the expression of Nrf2,HO-1,and Nqo1,the important antioxidant components in the liver.Hepatic SOD in the blueberry group was higher and MDA was lower than that in the control group (P<0.05).Blueberry also increased the index of the spleen and enhanced the proliferation of lymphocytes of the spleen (P<0.05).The percentages of the CD3 + and CD4 + T lymphocyte subsets and the CD4 + /CD8 + ratio were also increased by blueberry(P<0.05). CONCLUSIONS:Blueberry induces expression of Nrf2,HO-1,and Nqo1,which can protect hepatocytes from oxidative stress. 14Preoperative assessment of hilar cholangiocarcinoma:combination of cholangiography and CT angiography显示文摘BACKGROUND:Hilar cholangiocarcinoma is one of the most difficult carcinomas to manage because of the location of the main tumor at the hepatic hilus and the complex anatomy of the biliary,arterial,and portal systems.To plan an operation,it is important to acquire accurate information about the relationship between hilar cholangiocarcinoma and adjacent vessels.This study aimed to evaluate the clinical value of cholangiography combined with spiral CT three-dimensional (3D)angiography for a preoperative assessment of hilar cholangiocarcinoma. METHODS:From March 2007 to August 2009,cholangiography was performed in 13 patients with hilar cholangiocarcinoma. Meanwhile,contrast-enhanced abdominal scanning was performed using 16-slice spiral CT,and the 3D images of the hepatic artery and portal vein were acquired.The level and range of invasion of the hepatic artery,the portal vein,and the bile duct,the preoperative Bismuth classification,and T-staging were recorded and compared with those after surgical exploration. RESULTS:The hepatic artery and portal vein were recon- structed successfully in all these patients.Percutaneous transhepatic cholangiography was performed in 9 patients,endoscopic retrograde cholangiopancreatography in 1,and magnetic resonance cholangiopancreatography in 3.The CT angiography records of invasion of the hepatic artery were consistent with the results of explorations in these patients. The data from 5 of the 13 patients were consistent with those on invasion of the portal vein.The results of the Bismuth classification and the T-staging system were consistent with those of surgical exploration in 12 of the 13 patients.Seven of 8 patients who were estimated to be suitable for operation based on images were curatively treated and 5 who were judged to be unsuitable for curative operation by cholangiography and CT angiography were confirmed intraoperatively and underwent palliative procedures. 17Deep-learning-based extraction of the animal migration patterns from weather radar images显示文摘Continental coverage and year-round operation of the weather radar networks provide an unprecedented opportunity for studying large-scale airborne migration.The broad and local-scale airborne information collected by these infrastructures can answer many ecological questions.However,extracting and interpreting the biological information from such massive weather radar data remains an intractable problem.Recently,many big-data problems have been solved using the deep learning technology.In this study,the biological information in the weather radar data is identified using the advanced deep learning method.The proposed method consists of two main parts,i.e.,a rendering and casting procedure and an image segmentation procedure based on a convolutional neural network.The biological data are automatically extracted by rendering and mapping,image segmentation,and result masking.By analyzing the typical radar data from single and multiple stations,we partly reveal the intensity and speed of the migration pattern.We present the first feasibility study of the extraction of local and large-scale biological phenomena from the Chinese weather radar network data.Kai CUI Cheng HU Rui WANG Yi SUI Huafeng MAO Huayu LI 2020Science China(Information Sciences)2020,63,4:11
18Metabonomic analysis of hepatitis B virus-induced liver failure:identification of potential diagnostic biomarkers by fuzzy support vector machine显示文摘Hepatitis B virus(HBV)-induced liver failure is an emergent liver disease leading to high mortality.The severity of liver failure may be reflected by the profile of some metabolites.This study assessed the potential of using metabolites as bio- markers for liver failure by identifying metabolites with good discriminative performance for its phenotype.The serum samples from 24 HBV-induced liver failure patients and 23 healthy volunteers were collected and analyzed by gas chromatography-mass spectrometry(GC-MS)to generate metabolite profiles.The 24 patients were further grouped into two classes according to the severity of liver failure.Twenty-five commensal peaks in all metabolite profiles were extracted,and the relative area values of these peaks were used as features for each sample.Three algorithms,F-test,k-nearest neighbor(KNN)and fuzzy support vector machine(FSVM)combined with exhaustive search(ES),were employed to identify a subset of metabolites(biomarkers)that best predict liver failure.Based on the achieved experimental dataset,93.62%predictive accuracy by 6 features was selected with FSVM-ES and three key metabolites,glyceric acid,cis-aconitic acid and citric acid,are identified as potential diagnostic bio- markers.Yong MAO Xin HUANG Ke YU Hai-bin QU Chang-xiao LIU Yi-yu CHENG 2008Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2008,9,6:11
19Effect of Danshao Huaxian capsule on Gremlin and bone morphogenetic protein-7 expression in hepatic fibrosis in rats显示文摘AIM:To observe the effect of Danshao Huaxian capsule(DHC)on the expression of Gremlin and bone morphogenetic protein-7(BMP-7)in the liver of hepatic fibrosis rats.METHODS:A total of 75 male Wistar rats were randomly divided into a normal control group(A),a CCl4-induced hepatic fibrosis model group(B),a natural recovery group(C),a low-dose DHC-treated group(D),and a high-dose DHC-treated group(E),with 15 rats in each group.Liver fibrosis was induced by subcutaneous injections of carbon tetrachloride(CCl4)and a highlipid/low-protein diet for 8 wk,except for the rats in group A.Then,the rats in the two DHC-treated groups were administered 0.5 and 1.0 g/kg DHC by gastrogavage once per day for 8 successive weeks,respectively.By the end of the experiment,the level of transforming growth factorβ1(TGF-β1)in the liver homogenate was determined by an enzyme-linked immunosorbent assay.The mRNA and protein expression of Gremlin and BMP-7 in the liver tissue was determined by reversetranscription polymerase chain reaction,an immunohistochemical assay,and Western blot analysis.RESULTS:Compared with group A,the level of TGF-β1and the mRNA and protein expression of Gremlin were significantly higher in group B(TGF-β1:736.30±24.40μg/g vs 284.20±18.32μg/g,P<0.01;mRNA of Gremlin:80.40±5.46 vs 49.83±4.20,P<0.01;positive protein expression rate of Gremlin:38.46%±1.70%vs 3.83%±0.88%,P<0.01;relative protein expression of Gremlin:2.81±0.24 vs 0.24±0.06,P<0.01),and the mRNA and protein expression of BMP-7was significantly lower in group B(mRNA:54.00±4.34vs 93.99±7.03,P<0.01;positive protein expression rate:28.97%±3.14%vs 58.29%±6.02,P<0.01;relative protein expression:0.48±0.31 vs 1.05±0.12,P<0.01).Compared with groups B and C,the degree of hepatic fibrosis was significantly improved,and the level of TGF-β1 and the mRNA and protein expression of Gremlin were significantly lowered in the two DHCtreated groups(TGF-β1:523.14±21.29μg/g,441.86±23.18μg/g vs 736.30±24.40μg/g,651.13±15.75μg/g,P<0.01;mRNA of Gremlin:64.86±2.83,55.82±5.39 vs 80.40±5.46,70.37±4.01,P<0.01;positive protein expression rate of Gremlin:20.78%±1.60%,17.43%±2.02%vs 38.46%±1.70%,29.50%±2.64%,P<0.01;relative protein expression of Gremlin:1.95±0.26,1.65±0.20 vs 2.81±0.24,2.22±0.63,P<0.01),and the mRNA and protein expression of BMP-7 was higher in the two DHC-treated groups(mRNA:73.52±4.56,81.78±5.38 vs 54.00±4.34,62.28±4.51,P<0.01;positive protein expression rate:41.44%±4.77%,47.49%±4.59%vs28.97%±3.14%,35.85%±3.50%,P<0.01;relative protein expression:0.71±0.06,0.81±0.07 vs 0.48±CONCLUSION:The therapeutic mechanism of DHC forhepatic fibrosis in rats may be associated with inhibitionof the expression of Gremlin and up-regulation of the expression of BMP-7.Xue-Ke Zhao Ming-Liang Cheng Rong-Min Wu Yu-Mei Yao Mao Mu Juan-Juan Zhu Bao-Fang Zhang Ming-Yu Zhou 2014World Journal of Gastroenterology2014,20,40:11
20Fine Mapping of qPAA8,a Gene Controlling Panicle Apical Development in Rice显示文摘In rice,one detrimental factor influencing single panicle yield is the frequent occurrence of panicle apical abortion(PAA) under unfavorable climatic conditions.Until now,no detailed genetic information has been available to avoid PAA in rice breeding.Here,we show that the occurrence of PAA is associated with the accumulation of excess hydrogen peroxide.Quantitative trait loci(QTLs) mapping for PAA in an F2 population derived from the cross of L-05261(PAA line) × IRAT129(non-PAA variety) identified seven QTLs over a logarithm of the odd(LOD) threshold of 2.5,explaining approximately 50.1% of phenotypic variance for PAA in total.Five of the QTLs with an increased effect from L-05261,were designated as qPAA3-1,qPAA3-2,qPAA4,qPAA5 and qPAA8,and accounted for 6.8%,5.9%,4.2%,13.0% and 12.2% of phenotypic variance,respectively.We found that the PAA in the early heading plants was mainly controlled by qPAA8.Subsequently,using the sub-populations specific for qPAA8 based on markerassisted selection,we further narrowed qPAA8 to a 37.6-kb interval delimited by markers RM22475 and 8-In112.These results are beneficial for PAA gene clone.Zhi-Jun Cheng Bi-Gang Mao Su-Wei Gao Ling Zhang Jiu-Lin Wang Cai-Lin Lei Xin Zhang Fu-Qing Wu Xiu-Ping Guo Jianmin Wan 2011Journal of Integrative Plant Biology2011,53,9:10
