| 2 | Downregulation of serum mi R-17 and mi R-106b levels in gastric cancer and benign gastric diseases显示文摘Altered micro RNA(mi RNA) associated with gastric cancer(GC) development and mi R-17 and mi R-106 b were differentially expressed in GC tissues. This study detected serum levels of mi R-17 and mi R-106 b expression in GC, benign gastric disease(BGD) and healthy controls to assess them as tumor markers for GC. Serum samples from 40 GC, 32 BGD(10 gastric ulcer, 14 gastric polyps, and 8 gastric ulcer with polyps) and 36 healthy individuals were subjected to quantitative reverse transcription polymerase chain reaction(q RT-PCR) analysis of mi R-17 and mi R-106 b expression. The data showed that the serum levels of mi R-17 and mi R-106 b were significantly reduced in healthy individuals and BGD patients compared to GC patients. There was a significant association of mi R-17 and mi R-106 b expression with age, but not with other clinicopathological features, such as gender, tumor differentiation, stage and lymphatic metastasis. Further analysis showed that, in discriminating GC patients from healthy controls, mi R-17 could yield a receiver-operating characteristic(ROC) area under the curve(AUC) of 0.879 with 80.6% sensitivity and 87.5% specificity and mi R-106 b could yield an AUC of 0.856 with 75.0% sensitivity and 92.5% specificity. The combined AUC of mi R-17 and mi R-106 b was 0.913 with 83.3% sensitivity and 87.5% specificity. | 4 | Role of calcium mobilization in the regulation of spontaneous transient outward currents in porcine coronary artery myocytes显示文摘The purpose of the present study was to further study the characteristics and regulation of spontaneous transient outward currents (STOCs) in freshly isolated porcine coronary artery smooth muscle cells (ASMCs). STOCs were recorded using the perforated whole-cell patch-clamp configuration. STOCs were voltage-dependent and superimposed stochastically onto whole-cell Ca2+-activated-K+ (BKCa) currents. Charybdotoxin (ChTX, 200 nmol/L), a selective blocker of BKCa channels, completely inhibited STOCs within 10 min. STOCs activity was greatly suppressed when extracellular Ca2+ concentration decreased from 1.8 mmol/L to 200 nmol/L, further removal of Ca2+ abolished STOCs activity. Ca2+ ionophore A23187 (10 μmol/L) increased STOCs activity significantly. Verapamil (20 μmol/L) and CdCl2 (200 μmol/L), two kinds of organic L-type voltage-dependent Ca2+ channels (L-VDCCs) antagonists, had little effect on STOCs. In addition, the ryanodine receptors (RyRs) agonist caffeine (5 mmol/L) significantly activated STOCs. Application of ryanodine (50 μmol/L) to block RyRs abolished STOCs, subsequent washout of ryanodine or application of caffeine failed to reproduce STOCs activity. Inhibition of inositol 1,4,5-trisphosphate receptors (IP3Rs) by 2APB (40 μmol/L) greatly suppressed the activity of STOCs, application of caffeine (5 mmol/L) in the presence of 2APB caused a burst of outward currents followed by inhibition of STOCs. These results suggest that STOCs in porcine coronary ASMCs are mediated by BKCa channels. Extracellular Ca2+ is essential for STOCs activity, while Ca2+ entry through L-VDCCs has little effect on STOCs. | 15 | THE METHOD OF SEPARATING HARMONIC SIGNALS FROM MULTIPLICATIVE AND ADDITIVE NOISES显示文摘This paper focuses on the extraction of a harmonic signal from multiplicative and additive noises. A method is proposed in two stages: (1) to square the original discrete time series, which in- cludes both signals and noises, and form a new time series. By this means, the multiplicative noise is converted to additive noise; and (2) to filter out the noise by using existing noise removal schemes. With a large amount of simulation, experimental results demonstrated the efficiency and effective- ness of this newly developed method in terms of Signal-to-Noise Ratio (SNR) and other criteria. From the experiment, it is also found that: the two kinds of noises affect the SNR differently. In gen- eral, the SNR is not influenced by multiplicative Gaussian noise regardless of its variance. However, if both kinds of noise exist, the SNR decreases with the incensement of the Variance of Additive Noise to Multiplicative Noise Ratio (VAMNR). | 16 | Self-sufficient nanoparticles with dual-enzyme activity trigger radical storms and activate cascade-amplified antitumor immunologic responses显示文摘Radiotherapy(RT) can potentially induce systemic immune responses by initiating immunogenic cell death(ICD) of tumor cells.However,RT-induced antitumor immunologic responses are sporadic and insufficient against cancer metastases.Herein,we construct multifunctional self-sufficient nanoparticles(MARS) with dual-enzyme activity(GOx and peroxidase-like) to trigger radical storms and activate the cascade-amplified systemic immune responses to suppress both local tumors and metastatic relapse.In addition to limiting the Warburg effect to actualize starvation therapy,MARS catalyzes glucose to produce hydrogen peroxide(H_(2)O_(2)),which is then used in the Cu^(+)-mediated Fenton-like reaction and RT sensitization.RT and chemodynamic therapy produce reactive oxygen species in the form of radical storms,which have a robust ICD impact on mobilizing the immune system.Thus,when MARS is combined with RT,potent systemic antitumor immunity can be generated by activating antigen-presenting cells,promoting dendritic cells maturation,increasing the infiltration of cytotoxic T lymphocytes,and reprogramming the immuno suppre ssive tumor microenvironment.Furthermore,the synergistic therapy of RT and MARS effectively suppresses local tumor growth,increases mouse longevity,and results in a 90% reduction in lung metastasis and postoperative recurrence.Overall,we provide a viable approach to treating cancer by inducing radical storms and activating cascade-amplified systemic immunity. | Liping Bai Jin Yang Siting Yu Zhongzheng Xiang Yuanyuan Zeng Meiling Shen Xiaorong Kou Qinjie Wu Changyang Gong | 2024 | Acta Pharmaceutica Sinica B2024,14,2: | 0 |
| 17 | High-entropy alloy catalysts:From bulk to nano toward highly efficient carbon and nitrogen catalysis显示文摘High-entropy alloys(HEAs)have attracted widespread attention as both structural and functional materials owing to their huge multielement composition space and unique high-entropy mixing structure.Recently,emerging HEAs,either in nano or highly porous bulk forms,are developed and utilized for various catalytic and clean energy applications with superior activity and remarkable durability.Being catalysts,HEAs possess some unique advantages,including(1)a multielement composition space for the discovery of new catalysts and fine-tuning of surface adsorption(i.e.,activity and selectivity),(2)diverse active sites derived from the random multielement mixing that are especially suitable for multistep catalysis,and(3)a high-entropy stabilized structure that improves the structural durability in harsh catalytic environments.Benefited from these inherent advantages,HEA catalysts have demonstrated superior catalytic performances and are promising for complex carbon(C)and nitrogen(N)cycle reactions featuring multistep reaction pathways and many different intermediates.However,the design,synthesis,characterization,and understanding of HEA catalysts for C-and N-involved reactions are extremely challenging because of both complex high-entropy materials and complex reactions.In this review,we present the recent development of HEA catalysts,particularly on their innovative and extensive syntheses,advanced(in situ)characterizations,and applications in complex C and N looping reactions,aiming to provide a focused view on how to utilize intrinsically complex catalysts for these important and complex reactions.In the end,remaining challenges and future directions are proposed to guide the development and application of HEA catalysts for highly efficient energy storage and chemical conversion toward carbon neutrality. | Lanlan Yu Kaizhu Zeng Chenghang Li Xiaorong Lin Hanwen Liu Wenhui Shi Hua-Jun Qiu Yifei Yuan Yonggang Yao | 2022 | Carbon Energy2022,4,5: | 0 |
