7The PIWI protein Aubergine recruits eIF3 to activate translation in the germ plasm显示文摘Piwi-interacting RNAs(piRNAs)and PIWI proteins are essential in germ cells to repress transposons and regulate mRNAs.In Drosophila,piRNAs bound to the PIWI protein Aubergine(Aub)are transferred maternally to the embryo and regulate maternal mRNA stability through two opposite roles.They target mRNAs by incomplete base pairing,leading to their destabilization in the soma and stabilization in the germ plasm.Here,we report a function of Aub in translation.Aub is required for translational activation of nanos mRNA,a key determinant of the germ plasm.Aub physically interacts with the poly(A)-binding protein(PABP)and the translation initiation factor elF3.Polysome gradient profiling reveals the role of Aub at the initiation step of translation.In the germ plasm,PABP and elF3d assemble in foci that surround Aub-containing germ granules,and Aub acts with elF3d to promote nanos translation.These results identify translational activation as a new mode of mRNA regulation by Aub,highlighting the versatility of PIWI proteins in mRNA regulation.Anne Ramat Maria-Rosa Garcia-Silva Camille Jahan Rima Nait-Saidi Jeremy Dufourt Celine Garret Aymeric Chartier Julie Cremaschi Vipul Patel Mathilde Decourcelle Amandine Bastide Francois Juge Martine Simonelig 2020Cell Research2020,30,5:1

13NumericalMethods for Balance Laws with Space Dependent Flux:Application to Radiotherapy Dose Calculation显示文摘The present work is concerned with the derivation of numerical methods to approximate the radiation dose in external beam radiotherapy.To address this issue,we consider a moment approximation of radiative transfer,closed by an entropy minimization principle.The model under consideration is governed by a system of hyperbolic equations in conservation form supplemented by source terms.The main difficulty coming from the numerical approximation of this system is an explicit space dependence in the flux function.Indeed,this dependence will be seen to be stiff and specific numerical strategiesmust be derived in order to obtain the needed accuracy.A first approach is developed considering the 1D case,where a judicious change of variables allows to eliminate the space dependence in the flux function.This is not possible in multi-D.We therefore reinterpret the 1D scheme as a scheme on two meshes,and generalize this to 2D by alternating transformations between separate meshes.We call this procedure projection method.Several numerical experiments,coming from medical physics,illustrate the potential applicability of the developed method.Christophe Berthon Martin Frank Celine Sarazin Rodolphe Turpault 2011Communications in Computational Physics2011,10,10:0
14Qualitative and quantitative analysis of Arnebiae Radix and Dictamni Cortex and efficacy study of herbal extracts on allergic contact dermatitis using 3D human reconstructed epidermis显示文摘Objective:To evaluate the quality of Arnebiae Radix(AR)and Dictamni Cortex(DC)and study the efficacy of herbal extracts of these two herbs on the treatment of allergic contact dermatitis(ACD).Methods:Qualitative and quantitative analysis of effective components was performed using High Performance Thin Layer Chromatography(HPTLC),High Performance Liquid Chromatography(HPLC),and HPLC-Quadrupole Time of Flight-Mass Spectrometry(HPLC-QTOF-MS).In vitro allergic ACD 3D model was established by incubating 3D reconstructed human epidermis(RHE)with skin sensitizer,potassium dichromate.A total of 65 gene expression that were associated with ACD,which included 24 antioxidant responsive element(ARE)and 41 SENS-IS genes were quantified by q RT-PCR.More than or equal to 10 ARE genes and 18 SENN-IS genes were induced by 1.3-fold,demonstrating the successful establishment of in vitro ACD model.Oil extracts of AR and DC were applied on the in vitro ACD model to study the efficacy.Results:Batch 3 of AR and batch 2 of DC showed presence of all active ingredients with the highest concentrations.Active ingredients of the herbs were extracted using a special oil and formulated into herbal oil extracts.The herbal oil extracts were able to down regulate the induced genes in the in-vitro ACD skin model,bringing the tissue back to homeostatic status.Conclusion:The oil extracts showed the potent efficacy of using AR and DC in ACD treatment.The combination study will be done to optimize the formulation ratio which will be developed into a topical cream.Huan Li Esther Lim Gladys Ang Zhi-qing Lim Martin Hui Cai Jo-anne Loh Celine Ng Peijia Seetoh Edmund Tian Lay Beng Goh 2021Chinese Herbal Medicines2021,13,4:0
