| 1 | Wnt and BMP signaling crosstalk in regulating dental stem cells:Implications in dental tissue engineering显示文摘Tooth is a complex hard tissue organ and consists of multiple cell types that are regulated by important signaling pathways such as Wnt and BMP signaling.Serious injuries and/or loss of tooth or periodontal tissues may significantly impact aesthetic appearance,essential oral functions and the quality of life.Regenerative dentistry holds great promise in treating oral/dental disorders.The past decade has witnessed a rapid expansion of our understanding of the biological features of dental stem cells,along with the signaling mechanisms governing stem cell self-renewal and differentiation.In this review,we first summarize the biological characteristics of seven types of dental stem cells,including dental pulp stem cells,stem cells from apical papilla,stem cells from human exfoliated deciduous teeth,dental follicle precursor cells,periodontal ligament stem cells,alveolar bone-derived mesenchymal stem cells(MSCs),and MSCs from gingiva.We then focus on how these stem cells are regulated by bone morphogenetic protein(BMP)and/or Wnt signaling by examining the interplays between these pathways.Lastly,we analyze the current status of dental tissue engineering strategies that utilize oral/dental stem cells by harnessing the interplays between BMP and Wnt pathways.We also highlight the challenges that must be addressed before the dental stem cells may reach any clinical applications.Thus,we can expect to witness significant progresses to be made in regenerative dentistry in the coming decade. | Fugui Zhang Jinlin Song Hongmei Zhang Enyi Huang Dongzhe Song Viktor Tollemar Jing Wang Jinhua Wang Maryam Mohammed Qiang Wei Jiaming Fan Junyi Liao Yulong Zou Feng Liu Xue Hu Xiangyang Qu Liqun Chen Xinyi Yu Hue H.Luu Michael J.Lee Tong-Chuan He Ping Ji | 2016 | Genes & Diseases2016,3,4: | 16 |
| 2 | NAT2 polymorphism in Omani gastric cancer patients-risk predisposition and clinicopathological associations显示文摘AIM: To study whether N-acetyltransferase 2 (NAT2) genotypes and phenotypes are associated with increased risk factor for gastric cancer in Omani patients and to study the clinico-pathological correlations and the prognostic signifi cance of NAT2.METHODS: Genomic DNA was extracted from peripheral blood of 100 gastric cancer patients and 100 control subjects. NAT2 genotyping was performed using DNA sequencing. The prognostic significance of NAT2 and other clinicopathological features was assessed by univariate and multivariate analyses.RESULTS: We observed no significant association between NAT2 genotypes and phenotypes and gastric cancer risk. The NAT2 phenotype polymorphisms and gastric cancer risk predisposition were not modified by concomitant H pylori infection and smoking. There was no significant association between NAT2 and clinicopathological features, and NAT2 had no independent prognostic signifi cance.CONCLUSION: In the current study, NAT2 genotypes and phenotypes are not associated with gastric cancer risk predisposition. | 10 | Surfen and oxalyl surfen decrease tau hyperphosphorylation and mitigate neuron deficits in vivo in a zebrafish model of tauopathy显示文摘Background:Tauopathies comprise a family of neurodegenerative disorders including Alzheimer’s disease for which there is an urgent and unmet need for disease-modifying treatments.Tauopathies are characterized by pathological tau hyperphosphorylation,which has been shown to correlate tightly with disease progression and memory loss in patients suffering from Alzheimer’s disease.We recently demonstrated an essential requirement for 3-O-sulfated heparan sulfate in pathological tau hyperphosphorylation in zebrafish,a prominent model organism for human drug discovery.Here,we investigated whether in vivo treatment with surfen or its derivatives oxalyl surfen and hemisurfen,small molecules with heparan sulfate antagonist properties,could mitigate tau hyperphosphorylation and neuronal deficits in a zebrafish model of tauopathies.Results:In vivo treatment of Tg[HuC::hTau^(P301L);DsRed]embryos for 2 days with surfen or oxalyl surfen significantly reduced the accumulation of the pThr181 tau phospho-epitope measured by ELISA by 30%and 51%,respectively.Western blot analysis also showed a significant decrease of pThr181 and pSer396/pSer404 in embryos treated with surfen or oxalyl surfen.Immunohistochemical analysis further confirmed that treatment with surfen or oxalyl surfen significantly decreased the AT8 tau epitope in spinal motoneurons.In addition,in vivo treatment of Tg[HuC::hTau^(P301L);DsRed]embryos with surfen or oxalyl surfen significantly rescued spinal motoneuron axon-branching defects and,as a likely consequence,the impaired stereotypical touch-evoked escape response.Importantly,treatment with hemisurfen,a surfen derivative devoid of heparan sulfate antagonist activity,does not affect tau hyperphosphorylation,nor neuronal or behavioural deficits in Tg[HuC::hTau^(P301L);DsRed]embryos.Conclusion:Our findings demonstrate for the first time that surfen,a well-tolerated molecule in clinical settings,and its derivative,oxalyl surfen,could mitigate or delay neuronal defects in tauopathies,including Alzheimer’s disease. | Seyedeh Maryam Alavi Naini Constantin Yanicostas Rahma Hassan-Abdi Sébastien Blondeel Mohamed Bennis Ryan J.Weiss Yitzhak Tor Jeffrey D.Esko Nadia Soussi-Yanicostas | 2018 | Translational Neurodegeneration2018,7,1: | 1 |

| 18 | Diagnosis of Clinical Cases of Infectious Bursal Disease Using a Modified Rapid Taq Man-MGB Real-Time RT-PCR Assay显示文摘Infectious bursal disease(IBD)is an important contagious viral infection of immune system of poultry.This infection possesses a permanent threat to the profitability of poultry industry worldwide.The aim of this work was to modify the Taq Man-MGB real-time reverse transcription-polymerase chain reaction(rRT-PCR)in one step involving two fluorogenic Taq Man labeled probe and using this protocol for detection of infectious bursal disease virus(IBDV)collected from suspected cases distributed in different regions of the country during the period 2013-2016.The intralaboratory validation of modified method was realized for specificity,linearity,repeatability,sensitivity and reproducibility.It allowed reducing the test running time by six folds.This method was applied on 102 pools of bursa of fabricius(BF)samples collected from affected broiler farms suspected to be infected by IBDV.Birds showing macroscopic lesions including muscle petechial hemorrhages,hypertrophy and hemorrhage of BF,were subjected to molecular analysis using modified protocol“Taq Man-MGB rRT-PCR”.The validation satisfied all criteria and the assay developed could be a useful tool for a very rapid diagnosis of IBDV and permit to detect and to discriminate in one-step very virulent(vv)from non-vv(classic and variant)IBDV strains.Out of 84 IBDV positive samples,a prevalence of 39%for vv strains and 61%for classical strains was noted.These results indicate that despite the vaccination against IBDV,the vv form of this pathologie continues to cause serious problems for Moroccan broiler chickens.The obtained results indicate the successfully detection of IBDV and differentiated all vvIBDV strains from non-vvIBDV strains;Avian infectious agent RNA viruses tested are negative,demonstrating great specificity of the assay.The results obtained indicate that this method is suitable as a routine laboratory test for the rapid detection and differentiation of IBDV strains in samples of avian origin. | Maryame Cheggag Khalil Zro Ghizlane Sebbar Naoufal Rahmatallah Mohamed Mouahid Mohammed EL Houadfi Faouzi Kichou | 2018 | Journal of Agricultural Science and Technology(A)2018,8,4: | 0 |
| 19 | Analysing user daylight preferences in heritage buildings using virtual reality显示文摘Technology has always been creating effective ways to support human decisions.Immersive virtual reality(IVR)has emerged to engage users in a simulated world,and this has gained the interest of a wide variety of users in the heritage industry.A historical case study built in the early 19th century is considered for an adaptive reuse exhibition.The palace is located in Cairo,Egypt,and named after Prince Omar Tosson.The current palace state incorporates a smashed top-lit zone,which is being studied and analyzed for daylighting adequacy.Three simulated distinct optimum skylight configurations are suggested for the redesign where the selection should not be based solely on simulation data,but should consider real-user preferences.Most daylight design criteria are previously based on simulation data that do not necessarily indicate the users'preferences.But utilizing user interactive tools such as IVR to test daylight redesign options,a whole new dimension is provided.In this study,the VR users’survey data is statistically analyzed using Statistical Package for Social Sciences(SPSS),where results revealed that the assessment attributes succeeded in reflecting the users'preferences;which,motivated designers to consider potential users’daylight preferences in reused spaces.The paper highlights the most significant emotional attributes that provide cost-effective and reliable information concerned with the performance of daylight in IVR before design implementation.This study compares and analyzes the effect of three skylight designs(Cases A,B&C)on the users'perception before design implementation using(IVR)post-survey.Forty-eight participants have contributed to the study,providing their feedback on six attributes namely:Pleasant,Contrasting,Brightness,Uniform Distribution,Visual Comfort,and Satisfaction.Those attributes are evaluated for the three cases in space using five scale rating values to reveal that the'Pleasant'attribute is most reliable in the study to reflect the users'preferences for design Case B. | Mohamed Marzouk Maryam ElSharkawy Ayman Mahmoud | 2022 | Building Simulation2022,15,9: | 0 |
