7Use of hybrid chitosan membranes and human mesenchymal stem cells from the Wharton jelly of umbilical cord for promoting nerve regeneration in an axonotmesis rat model显示文摘Many studies have been dedicated to the development of scaffolds for improving post-traumatic nerve regeneration. The goal of this study was to assess the effect on nerve regeneration, associating a hybrid chitosan membrane with non-differentiated human mesenchymal stem cells isolated from Wharton's jelly of umbilical cord, in peripheral nerve reconstruction after crush injury. Chromosome analysis on human mesenchymal stem cell line from Wharton's jelly was carried out and no structural alterations were found in metaphase. Chitosan membranes were previously tested in vitro, to assess their ability in supporting human mesenchymal stem cell survival, expansion, and differentiation. For the in vivo testing, Sasco Sprague adult rats were divided in 4 groups of 6 or 7 animals each:Group 1, sciatic axonotmesis injury without any other intervention (Group 1-Crush); Group 2, the axonotmesis lesion of 3 mm was infiltrated with a suspension of 1 250-1 500 human mesenchymal stem cells (total volume of 50 μL) (Group 2-CrushCell); Group 3, axonotmesis lesion of 3 mm was enwrapped with a chitosan type III membrane covered with a monolayer of non-differentiated human mesenchymal stem cells (Group 3-CrushChitIIICell) and Group 4, axonotmesis lesion of 3 mm was enwrapped with a chitosan type III membrane (Group 4-CrushChitIII). Motor and sensory functional recovery was evaluated throughout a healing period of 12 weeks using sciatic functional index, static sciatic index, extensor postural thrust, and withdrawal reflex latency. Stereological analysis was carried out on regenerated nerve fibers. Results showed that infiltration of human mesenchymal stem cells, or the combination of chitosan membrane enwrapment and human mesenchymal stem cell enrichment after nerve crush injury provide a slight advantage to post-traumatic nerve regeneration. Results obtained with chitosan type III membrane alone confirmed that they significantly improve post-traumatic axonal regrowth and may represent a very promising clinical tool in peripheral nerve reconstructive surgery. 11Esophagectomy for a traumatic esophageal perforation with delayed diagnosis显示文摘Esophageal perforations are rare,and traumatic perforations are even more infrequent.Due to the rarity of this condition and its nonspecific presentation,the diagnosis and treatment of this type of perforation are delayed in more than 50% of patients,which leads to a high mortality rate.An 18-year-old male patient was brought to the emergency room with a penetrating neck injury,caused by a gunshot wound.He was taken to the operating room and underwent surgical exploration of the neck and a chest tube was inserted to treat the hemo-and pneumothorax.During the procedure,a 2 cm lesion was detected in the esophagus,and the patient underwent a primary repair.A contrast leakage into his right hemithorax was noticed on the 4th postoperative day;he was submitted to new surgery,and a subtotal esophagectomy and jejunostomy were performed.He was discharged from the hospital in good condition 20 d after the last procedure.The discussion around this topic focuses on the importance of the timing of diagnosis and the subsequent treatment.In early diagnosed patients,more conservative therapeutics should be performed,such as primary repair,while in those with delayed diagnosis,the patient should be submitted to more aggressive and def initive treatment.Alexandre Zanchenko Fonseca Marcelo Augusto Fontenelle Ribeiro Jr Mariana Frazo Maurício Campanelli Costas Lanes Spinelli Orlando Contrucci 2009World Journal of Gastrointestinal Surgery2009,1,1:0
12Selection of High-quality Sperms by the Nanotechnological Method of Magnetic Activation in Brazilian Cervids显示文摘Cervids show a high degree of abnormalities in their sperm cells.Thus,this study aimed to select high-quality spermatozoa using magnetic-activated sperm sorting(MASS)compared to density gradient centrifugation(DGC)by assessing the post-selection cell quality.Semen from six Mazama deer was collected by electroejaculation after chemical restraint.The semen was analyzed in four samples:Fresh,DGC,SEMgood-non-apoptotic fraction,and SEMpoor-apoptotic fraction.The material was analyzed for motility and vigor(light microscopy),concentration(Neubauer chamber),semen morphology(phase contrast),and supravital staining test(eosin/nigrosine).The DGC method used 20 x 106 cells in 90%and 45%percoll®gradient.The MASS used 10 x 106 cells with 20μl of iron nanoparticles attached to Annexin V and filtration in a magnetic separation column.Both processing methods(DGC and MASS)were effective in producing high-quality sperm samples,with a marked reduction in abnormalities from 41.83±10.25(fresh)to 14.83±3.17(DGC)and 12±3.01(SEMgood),with 80.3%±2.06 livings cells.These findings suggest that this nanotechnological method,using nanoparticles,effectively produces high-quality semen samples in cervids for use in assisted reproduction.Teresinha Inês Assumpção AndréLuíz Quagliatto Santos JoséMaurício Barbanti Duarte Maurício Foschini Caroline Silva Vieira 2021Journal of Zoological Research2021,3,2:0
