| 1 | Mesenchymal stem cell-derived small extracellular vesicles mitigate oxidative stress-induced senescence in endothelial cells via regulation of miR-146a/Src显示文摘Senescent endothelial cells (ECs) could impair the integrity of the blood vessel endothelium, leading to vascular aging and a series of diseases, such as atherosclerosis, diabetes. Preventing or mitigating EC senescence might serve as a promising therapeutic paradigm for these diseases. Recent studies showed that small extracellular vesicles (sEV) have the potential to transfer bioactive molecules into recipient cells and induce phenotypic changes. Since mesenchymal stem cells (MSCs) have long been postulated as an important source cell in regenerative medicine, herein we investigated the role and mechanism of MSC-derived sEV (MSC-sEV) on EC senescence. In vitro results showed that MSC-sEV reduced senescent biomarkers, decreased senescence-associated secretory phenotype (SASP), rescued angiogenesis, migration and other dysfunctions in senescent EC induced by oxidative stress. In the In vivo natural aging and type-2 diabetes mouse wound-healing models (both of which have senescent ECs), MSC-sEV promoted wound closure and new blood vessel formation. Mechanically, miRNA microarray showed that miR-146a was highly expressed in MSC-sEV and also upregulated in EC after MSC-sEV treatment. miR-146a inhibitors abolished the stimulatory effects of MSC-sEV on senescence. Moreover, we found miR-146a could suppress Src phosphorylation and downstream targets VE-cadherin and Caveolin-1. | 2 | Degradation of 2,2′,4,4′-tetrabromodiphenyl ether by Pycnoporus sanguineus in the presence of copper ions显示文摘The degradation of 2,2′,4,4′-tetrabromodiphenyl ether(BDE-47) by Pycnoporus sanguineus was investigated in order to explore the impact of the heavy metal Cu2+ on BDE-47 decomposition and the subsequent formation of metabolites, as well as to further elucidate the degradation mechanism of BDE-47. An increase in degradation rate from 18.63% to49.76% in the first four days and its stabilization at(51.26 ± 0.08)% in the following days of BDE-47 incubation were observed. The presence of Cu2+ at 1 and 2 mg/L was found to promote the degradation rate to 56.41% and 60.79%, respectively, whereas higher level of Cu2+(≥ 5 mg/L) inhibited the removal of BDE-47. The similar concentration effects of Cu2+ was also found on contents of fungal protein and amounts of metabolites. Both intracellular and extracellular enzymes played certain roles in BDE-47 transportation with the best degradation rate at 27.90% and 27.67% on the fourth and third day, individually. During the degradation of BDE-47, four types of hydroxylated polybrominated diphenyl ethers(OH-PBDEs), i.e., 6′-OH-BDE-47, 5′-OH-BDE-47, 4′-OH-BDE-17, 2′-OH-BDE-28, and two bromophenols, i.e., 2,4-DBP and 4-BP were detected and considered as degradation products. | 3 | Study on drying and combustion process in grate-CFB incinerator显示文摘The drying and combustion process in the combined grate and circulating fluidized bed(grate-CFB) municipal solid waste(MSW) incinerator was investigated experimentally and mathematically.The drying grate bed was simulated by a muffle furnace,which could be controlled at a constant tempera-ture level.The kind of wastes,thickness of waste layer fed and temperature were chosen as the ad-justable parameters to study their effect on the drying process.The experimental results indicated that the hydrophilic wastes were more difficult to be dried than the hydrophobic wastes.The higher the temperature is the easier the waste is to be dried.The thinner waste layer is favorable to drying the waste.The pyrolysis experiment in the furnace showed that the higher temperature level could reduce the conversion rate of carbon to carbon monoxide.The semi-empirical mathematical model that in-cluded the bed material distribution submodel,volatile matter release submodel,carbon particle combustion submodel and so on was proposed.A 260 t/d grate-CFB incinerator was modeled and the model predicted bulk density agreed with the measured value from industrial field test.The predicted flue gas(e.g.CO2,CO) concentration deviated slightly from the industrial test data.The parameters and variables used in the model were determined by the experiments or practical field test.This model can be used to design the grate-CFB incinerator and guide its operation. | QingHai Li YanGuo Zhang MeiQian Chen AiHong Meng ChangHe Chen | 2009 | Science China(Technological Sciences)2009,52,5: | 3 |
