1Guiding T lymphopoiesis from pluripotent stem cells by defined transcription factors显示文摘Achievement of immunocompetent and therapeutic T lymphopoiesis from pluripotent stem cells(PSCs)is a central aim in T cell regenerative medicine.To date,preferentially reconstituting T lymphopoiesis in vivo from PSCs remains a practical challenge.Here we documented that synergistic and transient expression of Runx1 and Hoxa9 restricted in the time window of endothelial-to-hematopoietic transition and hematopoietic maturation stages in a PSC differentiation scheme(iR9-PSC)in vitro induced preferential generation of engraftable hematopoietic progenitors capable of homing to thymus and developing into mature T cells in primary and secondary immunodeficient recipients.Single-cell transcriptome and functional analyses illustrated the cellular trajectory of T lineage induction from PSCs,unveiling the T-lineage specification determined at as early as hemogenic endothelial cell stage and identifying the bona fide pre-thymic progenitors.The induced T cells distributed normally in central and peripheral lymphoid organs and exhibited abundant TCRαβrepertoire.The regenerative T lymphopoiesis restored immune surveillance in immunodeficient mice.Furthermore,gene-edited iR9-PSCs produced tumor-specific T cells in vivo that effectively eradicated tumor cells.This study provides insight into universal generation of functional and therapeutic T cells from the unlimited and editable PSC source.Rongqun Guo Fangxiao Hu Qitong Weng Cui Lv Hongling Wu Lijuan Liu Zongcheng Li Yang Zeng Zhijie Bai Mengyun Zhang Yuting Liu Xiaofei Liu Chengxiang Xia Tongjie Wang Peiqing Zhou Kaitao Wang Yong Dong Yuxuan Luo Xiangzhong Zhang Yuxian Guan Yang Geng Juan Du Yangqiu Li Yu Lan Jiekai Chen Bing Liu Jinyong Wang 2020Cell Research2020,30,1:7
2Plasma-grating-induced breakdown spectroscopy显示文摘.Laser-induced breakdown spectroscopy(LIBS)is a useful tool for determination of elements in solids,liquids,and gases.For nanosecond LIBS(ns-LIBS),the plasma shielding effect limits its reproducibility,repeatability,and signal-to-noise ratios.Although femtosecond laser filament induced breakdown spectroscopy(FIBS)has no plasma shielding effects,the power density clamping inside the filaments limits the measurement sensitivity.We propose and demonstrate plasma-grating-induced breakdown spectroscopy(GIBS).The technique relies on a plasma excitation source-a plasma grating generated by the interference of two noncollinear femtosecond filaments.We demonstrate that GIBS can overcome the limitations of standard techniques such as ns-LIBS and FIBS.Signal intensity enhancement with GIBS is observed to be greater than 3 times that of FIBS.The matrix effect is also significantly reduced with GIBS,by virtue of the high power and electron density of the plasma grating,demonstrating great potential for analyzing samples with complex matrix.Mengyun Hu Junsong Peng Sheng Niu Heping Zeng 2020Advanced Photonics2020,2,6:5
3Meteorin-β/Meteorin like/IL-41 attenuates airway inflammation in house dust mite-induced allergic asthma显示文摘We sought to examine the regulatory effect of Meteorin-β(Metrnβ)/Meteorin like(Metrnl)/IL-41 on lung inflammation in allergic asthma.We found that Metrnβwas elevated significantly in asthmatic patients and in mice with allergic asthma induced by house dust mite(HDM)extract.Upon exposure to HDM,Metrnβwas secreted predominantly by airway epithelial cells and inflammatory cells,including macrophages and eosinophils.The increased Metrnβeffectively blocked the development of airway hyperreactivity(AHR)and decreased inflammatory cell airway infiltration and type 2 cytokine production,which was associated with downregulated DC-mediated adaptive immune responses.Moreover,Metrnβimpaired the maturation and function of bone marrow-derived dendritic cells in vitro.Asthmatic mice adoptively transferred with dendritic cells isolated from Metrnβ-treated allergic mice displayed decreased AHR,airway inflammation,and lung injury.Metrnβalso displayed anti-inflammatory properties in immunodeficient SCID mice with allergic asthma and in in vitro 3D ALI airway models.Moreover,blockade of Metrnβby anti-Metrnβantibody treatment promoted the development of allergic asthma.These results revealed the unappreciated protective roles of Metrnβin alleviating DC-mediated Th2 inflammation in allergic asthma,providing the novel treatment strategy of therapeutic targeting of Metrnβin allergic asthma.Xun Gao Ting-Fan Leung Gary Wing-Kin Wong Wing-Hung Ko Mengyun Cai Ellie Jiayi He Ida Miu-Ting Chu Miranda Sin-Man Tsang Ben Chung-Lap Chan Jiawei Ling Xiao Fan Liwei Lu Christopher Wai-Kei Lam Chun-Kwok Wong 2022Cellular & Molecular Immunology2022,19,2:5
4Spreading and vanishing in a West Nile virus model with expanding fronts显示文摘We study a simplified version of a West Nile virus(WNv) model discussed by Lewis et al.(2006),which was considered as a first approximation for the spatial spread of WNv. The basic reproduction number R_0 for the non-spatial epidemic model is defined and a threshold parameter R_0~D for the corresponding problem with null Dirichlet boundary condition is introduced. We consider a free boundary problem with a coupled system, which describes the diffusion of birds by a PDE and the movement of mosquitoes by an ODE. The risk index R_0~F(t) associated with the disease in spatial setting is represented. Sufficient conditions for the WNv to eradicate or to spread are given. The asymptotic behavior of the solution to the system when the spreading occurs is considered. It is shown that the initial number of infected populations, the diffusion rate of birds and the length of initial habitat exhibit important impacts on the vanishing or spreading of the virus. 5Adhesive anastomosis for organ transplantation显示文摘The recent development of tough tissue adhesives has stimulated intense interests among material scientists and medical doctors.However,these adhesives have seldom been tested in clinically demanding surgeries.Here we demonstrate adhesive anastomosis in organ transplantation.Anastomosis is commonly conducted by dense sutures and takes a long time,during which all the vessels are occluded.Prolonged occlusion may damage organs and even cause death.We formulate a tough,biocompatible,bioabsorbable adhesive that can sustain tissue tension and pressurized flow.We expose the endothelial surface of vessels onto a gasket,press two endothelial surfaces to the adhesive using a pair of magnetic rings,and reopen the bloodstream immediately.The time for adhesive anastomosis is shortened compared to the time for sutured anastomosis.We have achieved adhesive anastomosis of a great vein in transplanting the liver of a pig.After the surgery,the adhesive is absorbed,the vein heals,and the pig lives for over one month.Kang Liu Hang Yang Gaobo Huang Aihua Shi Qiang Lu Shanpei Wang Wei Qiao Haohua Wang Mengyun Ke Hongfan Ding Tao Li Yanchao Zhang Jiawei Yu Bingyi Ren Rongfeng Wang Kailing Wang Hui Feng Zhigang Suo Jingda Tang Yi Lv 2022Bioactive Materials2022,7,7:3
6Cylindrical polymer brushes-anisotropic unimolecular micelle drug delivery system for enhancing the effectiveness of chemotherapy显示文摘Polymer systems can be designed into different structures and morphologies according to their physical and chemical performance requirements,and are considered as one of the most promising controlled delivery systems that can effectively improve the cancer therapeutic index.However,the majority of the polymer delivery systems are designed to be simple spherical nanostructures.To explore morphology/size-oriented delivery performance optimization,here,we synthesized three novel cylindrical polymer brushes(CPBs)by atom transfer radical polymerization(ATRP),which were cellulose-g-(CPT-b-OEGMA)(CCO)with different lengths(~86,~40,and~21 nm).The CPBs are composed of bio-degradable cellulose as the carrier,poly(ethylene glycol)methyl ether methacrylate(OEGMA)as hydrophily block,and glutathione(GSH)-responsive hydrophobic camptothecin(CPT)monomer as loaded anticancer drug.By controlling the chain length of the initiator,three kinds of polymeric prodrugs with different lengths(CCO-1,CCO-2,and CCO-3)could be self-organized into unimolecular micelles in water.We carried out comparative studies of three polymers,whose results verified that the shorter CPBs exhibited higher drug release efficiency,more cellular uptake,and enhanced tumor permeability,accompanied by shortened blood circulation time and lower tumor accumulation.As evidenced by in vivo experiments,the shorter CPBs exhibited higher anti-tumor efficiency,revealing that the size advantage has a higher priority than the anisotropic structure advantage.This provided vital information as to design an anisotropic polymer-based drug delivery system for cancer therapy.Shuang Bai Die Jia Xianbin Ma Mengyun Liang Peng Xue Yuejun Kang Zhigang Xu 2021Bioactive Materials2021,6,9:2
7Copper‑Based Catalysts for Electrochemical Carbon Dioxide Reduction to Multicarbon Products显示文摘Electrochemical conversion of carbon dioxide into fuel and chemicals with added value represents an appealing approach to reduce the greenhouse effect and realize a carbon-neutral cycle,which has great potential in mitigating global warming and effectively storing renewable energy.The electrochemical CO_(2) reduction reaction(CO_(2)RR)usually involves multiproton coupling and multielectron transfer in aqueous electrolytes to form multicarbon products(C_(2+) products),but it competes with the hydrogen evolution reaction(HER),which results in intrinsically sluggish kinetics and a complex reaction mechanism and places higher requirements on the design of catalysts.In this review,the advantages of electrochemical CO_(2) reduction are briefly introduced,and then,different categories of Cu-based catalysts,including monometallic Cu catalysts,bimetallic catalysts,metal-organic frameworks(MOFs)along with MOF-derived catalysts and other catalysts,are summarized in terms of their synthesis method and conversion of CO_(2) to C2+products in aqueous solution.The catalytic mechanisms of these catalysts are subsequently discussed for rational design of more efficient catalysts.In response to the mechanisms,several material strategies to enhance the catalytic behaviors are proposed,including surface facet engineering,interface engineering,utilization of strong metal-support interactions and surface modification.Based on the above strategies,challenges and prospects are proposed for the future development of CO_(2)RR catalysts for industrial applications.Fangfang Chang Meiling Xiao Ruifang Miao Yongpeng Liu Mengyun Ren Zhichao Jia Dandan Han Yang Yuan Zhengyu Bai Lin Yang 2022Electrochemical Energy Reviews2022,5,3:2

9Qingre Jianpi decoction(清热健脾汤)attenuates inflammatory responses by suppressing NOD-like receptor family pyrin domain-containing 3 inflammasome activation in dextran sulfate sodium-induced colitis mice显示文摘OBJECTIVE:To investigate the effects of Qingre Jianpi decoction(清热健脾汤,QRJPD)on dextran sulfate sodium(DSS)-induced colitis mice and explore its mechanism.METHODS:All mice were randomly divided into six groups.Weight changes,disease activity index values,and histological damage were detected.Inflammatory cytokines and immune cell infiltration were measured using enzyme-linked immunosorbent assays(ELISA)and immunohistochemistry(IHC)method.The key protein expression levels of the NOD-like receptor family pyrin domain-containing 3(NLRP3)inflammasome were detected by western blot analysis,IHC,and quantitative reverse transcription polymerase chain reaction.RESULTS:QRJPD played an anti-inflammatory role in the treatment of ulcerative colitis(UC),reduced the secretion of inflammatory cytokines,and inhibited the inflammatory infiltration of immune cells by suppressing DSS-induced activation of the NLRP3 inflammasome.CONCLUSION:QRJPD exerts protective effects by inhibiting DSS-induced NLRP3 inflammasome activation.ZHANG Jiayan KANG Xiuhong SUN Mengyun ZHANG Shengsheng 2021Journal of Traditional Chinese Medicine2021,41,1:2
10Synthesis of silica/metatitanic acid nanocomposite and evaluation of its catalytic performance for aquathermolysis reaction of extra-heavy crude oil显示文摘A lipophilic silica/metatitantic acid(denoted as Si O2/H2 TiO 3) nanocomposite was synthesized by hydrothermal reaction with surface-modified Si O2 as the lipophilic carrier. As-synthesized Si O2/H2 TiO 3nanocomposite was used as a catalyst to promote the aquathermolysis reaction of extra-heavy crude oil thereby facilitating the recovering from the deep reservoirs at lowered temperature. The catalytic performance of the as-synthesized Si O2/H2 TiO 3catalyst for the aquathermolysis reaction of the heavy oil at a moderate temperature of 150 °C was evaluated in relation to the structural characterizations by TEM,FTIR,XRD and FESEM as well as the determination of the specific surface area by N2adsorption–desorption method. Findings indicate that as-synthesized Si O2/H2 TiO 3nanocomposite exhibits an average size of about 20 nm as well as good lipophilicity and dispersibility in various organic solvents; and it shows good catalytic performance for the aquathermolysis reaction of the extra-heavy oil extracted from Shengli Oilfield of China. 12Sexual Dimorphism of the Jilin Clawed Salamander,Onychodactylus zhangyapingi,(Urodela:Hynobiidae:Onychodactylinae) from Jilin Province,China显示文摘Sexual dimorphism in size and shape is common in many organisms, and is a key evolutionary feature. In this study, we analyzed morphometric data of the Jilin clawed salamander Onychodactylus zhangyapingi, an endemic Chinese salamander, to examine sexual size and shape dimorphism. The morphometric data included 14 characteristics of 13 females and 11 males and was analyzed using univariate and multivariate methods. Our results showed that sexual dimorphism occurs not only in body size, but also in body shape. Males have a longer snout-vent length than females, a rarely reported pattern of male-biased sexual size dimorphism. Females have a larger space between the axilla and groin than males, while males have longer and larger tails compared to females. 13A ROS-responsive,self-immolative and self-reporting hydrogen sulfide donor with multiple biological activities for the treatment of myocardial infarction显示文摘Myocardial infarction(MI),as one of the leading causes of global death,urgently needs effective therapies.Recently,hydrogen sulfide(H2S)has been regarded as a promising therapeutic agent for MI,while its spatiotemporally controlled delivery remains a major issue limiting clinical translation.To address this limitation,we designed and synthesized a novel H2S donor(HSD-R)that can produce H2S and emit fluorescence in response to reactive oxygen species(ROS)highly expressed at diseased sites.HSD-R can specifically target mitochondria and provide red fluorescence to visualize and quantify H2S release in vitro and in vivo.Therapeutically,HSD-R significantly promoted the reconstruction of cardiac structure and function in a rat MI model.Mechanistically,myocardial protection is achieved by reducing cardiomyocyte apoptosis,attenuating local inflammation,and promoting angiogenesis.Furthermore,inhibition of typical pro-apoptotic genes(Bid,Apaf-1,and p53)played an important role in the anti-apoptotic effect of HSD-R to achieve cardioprotection,which were identified as new therapeutic targets of H2S against myocardial ischemia injury.This ROS-responsive,self-immolative,and fluorescent H2S donor can serve as a new theranostic agent for MI and other ischemic diseases.Mengyun Yao Yifei Lu Lin Shi Yong Huang Qing Zhang Jianglin Tan Ping Hu Jianxiang Zhang Gaoxing Luo Ning Zhang 2022Bioactive Materials2022,7,3:2
14Polyimides as Promising Materials for Lithium-Ion Batteries:A Review显示文摘Lithium-ion batteries(LIBs)have helped revolutionize the modern world and are now advancing the alternative energy field.Several technical challenges are associated with LIBs,such as increasing their energy density,improving their safety,and prolonging their lifespan.Pressed by these issues,researchers are striving to find effective solutions and new materials for next-generation LIBs.Polymers play a more and more important role in satisfying the ever-increasing requirements for LIBs.Polyimides(PIs),a special functional polymer,possess unparalleled advantages,such as excellent mechanical strength,extremely high thermal stability,and excellent chemical inertness;they are a promising material for LIBs.Herein,we discuss the current applications of PIs in LIBs,including coatings,separators,binders,solid-state polymer electrolytes,and active storage materials,to improve high-voltage performance,safety,cyclability,flexibility,and sustainability.Existing technical challenges are described,and strategies for solving current issues are proposed.Finally,potential directions for implementing PIs in LIBs are outlined.Mengyun Zhang Li Wang Hong Xu Youzhi Song Xiangming He 2023Nano-Micro Letters2023,15,9:1

19A binocular parallel rendering method for VR globes显示文摘The scene-rendering mechanism based on binocular vision is one of the key techniques for the VR globe to achieve immersion-type visualization of global 3D scenes.However,this special rendering mechanism also requires that the 3D scene is continuously drawn twice within one frame,which significantly affects the rendering efficiency of VR globes.Therefore,we propose a binocular parallel rendering method.This method first improves the current rendering process of VR globes by assigning the rendering tasks for the left and right camera of VR to be processed on different CPU cores,thereby achieving parallel rendering of binocular scenes.Second,due to the problem of inconsistent resolution of binocular scenes caused by different viewpoints for the left and right cameras,we propose a resolution synchronize algorithm.this algorithm conducts realtime synchronization on the resolution of scene in the rendering process and thus avoids the problem of erroneous binocular stereo matching.Finally,we validate the effectiveness of the method in this paper through experiments.The results of experiments indicate that while the method in this paper can ensure the consistency of binocular scene resolution,it can decrease the frame time of VR globes by approximately 27%on average.Wumeng Huang Jing Chen Mengyun Zhou 2020International Journal of Digital Earth2020,13,9:1
