| 2 | Semaphorin 3A controls allergic and inflammatory responses in experimental allergic conjunctivitis显示文摘AIM: To assess the efficacy of topical Semaphorin 3A(SEMA3A) in the treatment of allergic conjunctivitis.METHODS: Experimental allergic conjunctivitis(EAC)mice model induced by short ragweed pollen(SRW) in 4-week-old of BALB/c mice, mice were evaluated using haematoxylin and eosin(H&E) staining, immunofluor-escence and light microscope photographs. Early phase took the samples in 24 h after instillation and late phase took the samples between 4 to 14 d after the start of treatment. The study use of topical SEMA3A(10 U, 100 U,1000 U) eye drops and subconjunctival injection of SEMA3 A with same concentration. For comparison, five types of allergy eyedrops were quantified using clinical characteristics.· RESULTS: Clinical score of composite ocular symptoms of the mice treated with SEMA3 A were significantly decreased both in the immediate phase and the late phase compared to those treated with commercial ophthalmic formulations and non-treatment mice. SEMA3 A treatment attenuates infiltration of eosinophils entering into conjunctiva in EAC mice. The score of eosinophil infiltration in the conjunctiva of SEMA3 A 1000 U-treated group were significantly lower than low-concentration of SEMA3 A treated groups and non-treated group. SEMA3 A treatment also suppressed T-cell proliferation in vitro and decreased serum total Ig E levels in EAC mice. Moreover, treatment of SEMA3 A suppressed Th2-related cytokines(IL-5, IL-13 and IL-4)and pro-inflammatory cytokines(IFN-γ, IL-17 and TNF-α)release, but increased regulatory cytokine IL-10 concentration in the conjunctiva of EAC mice.CONCLUSION: SEMA3 A as a biological agent, showed the beneficial activity in ocular allergic processes with the less damage to the intraocular tissue. | 4 | Effect of Double Air Injection on Performance Characteristics of Centrifugal Compressor显示文摘In the operation of a centrifugal compressor of turbocharger, instability phenomena such as rotating stall and surge are induced at a lower flow rate close to the maximum pressure ratio. In this study, for the suppression of surge phenomenon resulting in the extension of the stable operating range of centrifugal compressor to lower flow rate, the compressed air at the compressor exit was re-circulated and injected into the impeller inlet by using the double injection nozzle system. The experiments were performed to find out the optimum circumferential position of the second nozzle relative to the fixed first one and the optimum inner diameter of the injection nozzles, which are able to most effectively reduce the flow rate of surge inception. | 5 | Treatment with dimethyl fumarate ameliorates liver ischemia/reperfusion injury显示文摘AIM To investigate the hypothesis that treatment with dimethyl fumarate(D MF) mayame liorate liver ischemia/reperfusion injury(I/RI).METHODS Rats were divided into 3 groups: sham, control(CTL), and DMF. DMF(25 mg/kg, twice/d) was orally administered for 2 d before the procedure. The CTL and DMF rats were subjected to ischemia for 1 h and reperfusion for 2 h. The serum alanine aminotransferase(ALT) and malondialdehyde(MDA) levels, adenosine triphosphate(ATP), NO × metabolites, anti-oxidant enzyme expression level, antiinflammatory effect, and anti-apoptotic effect were determined.RESULTS Histological tissue damage was significantly reduced in the DMF group(Suzuki scores: sham: 0 ± 0; CTL: 9.3± 0.5; DMF: 2.5 ± 1.2; sham vs CTL, P < 0.0001; CTL vs DMF, P < 0.0001). This effect was associated with significantly lower serum ALT(DMF 5026 ± 2305 U/L vs CTL 10592 ± 1152 U/L, P = 0.04) and MDA(DMF 18.2 ± 1.4 μmol/L vs CTL 26.0 ± 1.0 μmol/L, P = 0.0009). DMF effectively improved the ATP content(DMF 20.3 ± 0.4 nmol/mg vs CTL 18.3 ± 0.6 nmol/mg, P = 0.02), myeloperoxidase activity(DMF 7.8 ± 0.4 m U/m L vs CTL 6.0 ± 0.5 m U/m L, P = 0.01) and level of endothelial nitric oxide synthase expression(DMF 0.38 ± 0.05-fold vs 0.17 ± 0.06-fold, P = 0.02). The higher expression levels of anti-oxidant enzymes(catalase and glutamatecysteine ligase modifier subunit and lower levels of key inflammatory mediators(nuclear factor-kappa B and cyclooxygenase-2 were confirmed in the DMF group.CONCLUSION DMF improved the liver function and the anti-oxidant and inflammation status following I/RI. | 6 | Measurement and Numerical Simulation of a Small Centrifugal Compressor Characteristics at Small or Negative Flow Rate显示文摘For centrifugal compressors used in automotive turbochargers, the extension of the surge margin is demanded because of lower engine speed. In order to estimate the surge line exactly, it is required to acquire the compressor characteristics at small or negative flow rate. In this paper, measurement and numerical simulation of the characteristics at small or negative flow rate are carried out. In the measurement, an experimental facility with a valve immediately downstream of the compressor is used to suppress the surge. In the numerical work, a new boundary condition that specifies mass flow rate at the outlet boundary is used to simulate the characteristics around the zero flow rate region. Furthermore, flow field analyses at small or negative flow rate are performed with the numerical results.
