1Monitoring microbial community structure and succession of an A/O SBR during start-up period using PCR-DGGE显示文摘Polymerase chain reaction(PCR)-denaturing gradient gel electrophoresis(DGGE) protocol was employed for revealing microbial community structure and succession in a sequential anaerobic and aerobic reactor performing enhanced biological phosphorus removal(EBPR) during start-up period.High phosphorus removal was achieved after 15 d.On day 30,phosphorus removal effciency reached to 83.2% and the start-up was finished.DGGE profiles of periodical sludge samples showed that dominant microbial species were 19 OTUs(operational taxonomy units).Unweighted pair-group method using arithmetic averages(UPGMA) clustering analysis revealed that rapid community succession correlated to low phosphorus removal rate and high phosphorus removal effciency reflected on steady community structure.Sequencing results indicated that determined sequences(12 OTUs) belonged to Proteobacterium,Actinobacteria,Gemmatimonadales and unaffiliate group.Proteobacterium,Tetrasphaera elongate and Gemmatimonas aurantiaca may act important roles in phosphorus removal.With little amount as known glycogen accumulating organisms,Candidatus Competibacter phosphatis still at accumulating-phase had limited effect on microbial community structure.When climax community was obtained,dominant microbes were 14 OTUs.Microbes in a large amount were uncultured bacterium Thauera sp.,uncultured γ-Proteobacterium and Tetrasphaera elongata.WANG Xiuheng,ZHANG Kun,REN Nanqi,LI Nan,REN Lijiao State Key Laboratory of Urban Water Resource and Environment,Harbin Institute of Technology,Harbin 150090,China. 2009Journal of Environmental Sciences2009,21,2:17
2Levels and distribution of brominated flame retardants in the soil of Harbin in China显示文摘This study reports the presence of brominated flame retardants in the topsoil in and around Harbin, a city in northeastern China. Samples of soil were collected from 17 locations in 2006, and the levels of 9 polybrominated diphenylethers (PBDEs 17, 28, 47, 66, 99, 100, 153, 154, and 183) ranged from 2.45 to 55.9 pg/g dry weight (dw) with a mean of 26.3 pg/g dw. These levels are very low comparing with those for some cities in Europe and USA. BDE 209 and hexabromocyclododecane were the two dominant congeners, with mean concentrations of 520 pg/g dw and 1750 pg/g dw, respectively. The concentrations of the total nine PBDE congeners clearly decreased from urban areas to background, but the compositions of individual congeners differed. Proportions of heavier congeners decreased while those of lighter congeners increased, along urban-rural-background transect, providing evidence for an urban fractionation effect. Correlation analysis indicated similar sources for PBDEs, hexabromocyclododecane, and 1,2-bis(2,4,6-tribromophenoxy)-ethane from urban areas but pentabromoethylbenzene was probably present due to long-range atmospheric transport. 3Levels, distributions, and source identification of organochlorine pesticides in the topsoils in Northeastern China显示文摘Seventeen topsoil samples (9 urban, 4 suburban, 3 rural and 1 background) were collected in/around Harbin, a typical city in northeast of China, to measure concentration levels of organocholrine pesticides (OCPs) in topsoil of Northeastern China in 2006. Hexachlorohexanes (HCH), dichlorodiphenyltrichloroe thane (DDT), and hexachlorobenzene (HCB) were detected in soil samples with mean concentrations (in pg/g dry weight (dw)) of 7120, 5425, and 1039, respectively. The mean concentrations for other OCPs were very low, 4.8 pg/g dw for chlordane and 3.3 pg/g dw for endosulfan. Source identification analysis reveals that all OCPs found in soil samples were due to historical use of these chemicals or from other source regions through long- and short-range atmospheric transport. DDT was mainly used in the rural sites, whereas the sources of HCB, chlordane and endosulfan were mainly in the urban area. HCH was found almost equally in both urban and rural area. Soil concentrations of all detected OCPs, except HCHs, in and around Harbin were much lower than those in the southeast of China, which is expected since the use of these OCPs in the former was much lower than that in the latter, however higher HCH concentrations in and around Harbin than those found in most places of the Southeast China is not expected. 4Monitoring of microbial community structure and succession in the biohydrogen production reactor by denaturing gradient gel electrophoresis(DGGE)显示文摘To study the structure of microbial communities in the biological hydrogen produc- tion reactor and determine the ecological function of hydrogen producing bacteria, anaerobic sludge was obtained from the continuous stirred tank reactor (CSTR) in different periods of time, and the diversity and dynamics of microbial communities were investigated by denaturing gra- dient gel electrophoresis (DGGE). The results of DGGE demonstrated that an obvious shift of microbial population happened from the beginning of star-up to the 28th day, and the ethanol type fermentation was established. After 28 days the structure of microbial community became stable, and the climax community was formed. Comparative analysis of 16S rDNA sequences from reamplifying and sequencing the prominent bands indicated that the dominant population belonged to low G+C Gram-positive bacteria (Clostridium sp. and Ethanologenbacterium sp.), β- proteobacteria (Acidovorax sp.), γ-proteobacteria (Kluyvera sp.), Bacteroides (uncultured bacte- rium SJA-168), and Spirochaetes (uncultured eubacterium E1-K13), respectively. The hydrogen production rate increased obviously with the increase of Ethanologenbacterium sp., Clostridium sp. and uncultured Spirochaetes after 21 days, meanwhile the succession of ethanol type fer- mentation was formed. Throughout the succession the microbial diversity increased however it decreased after 21 days. Some types of Clostridium sp. Acidovorax sp., Kluyvera sp., and Bac- teroides were dominant populations during all periods of time. These special populations were essential for the construction of climax community. Hydrogen production efficiency was de- pendent on both hydrogen producing bacteria and other populations. 5Towards a better hydraulic cleaning strategy for ultrafiltration membrane fouling by humic acid: Effect of backwash water composition显示文摘As a routine measurement to alleviate membrane fouling, hydraulic cleaning is of great significance for the steady operation of ultrafiltration(UF) systems in water treatment processes. In this work, a comparative study was performed to investigate the effects of the composition of backwash water on the hydraulic cleaning performance of UF membranes fouled by humic acid(HA). Various types of backwash water, including UF permeate, Milli-Q water, Na Cl solution, CaCl_2 solution and HA solution, were compared in terms of hydraulically irreversible fouling index, total surface tension and residual HA. The results indicated that Milli-Q water backwash was superior to UF permeate backwash in cleaning HA-fouled membranes, and the backwash water containing Na+or HA outperformed Milli-Q water in alleviating HA fouling. On the contrary, the presence of Ca^(2+) in backwash water significantly decreased the backwash efficiency. Moreover, Ca^(2+) played an important role in foulant removal, and the residual HA content closely related to the residual Ca^(2+) content.Mechanism analysis suggested that the backwash process may involve fouling layer swelling, ion exchange, electric double layer release and competitive complexation. 6Short-term effect of temperature variation on the competition between PAOs and GAOs during acclimation period of an EBPR system显示文摘Sequencing batch reactor(SBR)for enhanced biological phosphorus removal(EBPR)processes was used to investigate the impact of the temperature shock on the competition between phosphorus-accumulating organisms(PAOs)and glycogen accumulating organisms(GAOs)in start-up stage.During the 34 days operation,SBR was set with temperature variation(0-5 d,22±1℃;6-13 d,29±1℃;14-34 d,14±1℃).PAOs and GAOs were analyzed by fluorescent in situ hybridization(FISH),and intracellular polyphosphate granules were stained by Neisser-stain.The results showed that the influence of temperature shock on PAOs’abundance was more serious than that on GAOs in the enriching process.Under sudden and substantially temperature variation,from 22±1℃ to 29±1℃ and then to 14±1℃,the domination of PAOs was deteriorated.After temperature shock,PAOs’competitive advantages at low temperature that concluded in other study did not appear in our study.As mesophilic,GAOs(indicated by Alphaproteobacteria and Gammaproteobacteria)were more temperature adaptive and better grew and took the domination at 14±1℃ in the end.In the competition process,organisms of tetrad forming organisms(TFOs)-like shape which were considered as typical GAOs,were observed.With the evidence of poly-P granules containing by Neisser-straining and result of FISH,these organisms of TFOs-like shape were better to be assumed as adaption state or a special self-protecting shape of PAOs.Nanqi REN Han KANG Xiuheng WANG Nan LI 2011Frontiers of Environmental Science & Engineering2011,5,2:4
7Activation of anterior thalamic reticular nucleus GABAergic neurons promotes arousal from propofol anesthesia in mice显示文摘Propofol is widely used for the induction and maintenance of anesthesia,which causes a rapid loss of consciousness.However,the mechanisms underlying the hypnosis effect of propofol are still not fully understood.The thalamic reticular nucleus(TRN)is crucial for regulating wakefulness,sleep rhythm generation,and sleep stability,while the role of TRN in the process of propofol-induced anesthesia is still unknown.Here,we investigated the function of the anterior TRN in propofol general anesthesia.Our results demonstrated that the neural activity of anterior TRN is suppressed during propofol anesthesia,whereas it is robustly activated from anesthesia by recording the calcium signals using fiber photometry technology.The results showed that the activation of anterior TRN neurons by chemogenetic and optogenetic methods shortens the emergency time without changing the induction time.Conversely,chemogenetic or optogenetic inhibition of the TRN neurons leads to a delay in the recovery time.Our study showed that anterior TRN is crucial for behavioral arousal without affecting the induction time of propofol anesthesia.Yanjun Liu Bing Chen Yirong Cai Yuan Han Ying Xia Nanqi Li Bingqian Fan Tianjie Yuan Junli Jiang Po Gao Weifeng Yu Yingfu Jiao Wenxian Li 2021Acta Biochimica et Biophysica Sinica2021,53,7:3
8Co-application of energy uncoupling and ultrafiltration in sludge treatment:Evaluations of sludge reduction,supernatant recovery and membrane fouling control显示文摘Energy uncoupling is often used for sludge reduction because it is easy to operate and does not require a significant amount of extra equipments(i.e.no additional tank required).However,over time the supernatant extracted using this method can deteriorate,ultimately requiring further treatment.The purpose of this study was to determine the effect of using a low-pressure ultrafiltration membrane process for sludge water recovery after the sludge had undergone an energy uncoupling treatment(using 3,3’,4’,5-tetrachlorosalicylanilide(TCS)).Energy uncoupling was found to break apart sludge floe by reducing extracellular polymeric substances(EPS)and adenosine triphosphate(ATP)content.Analysis of supernatant indicated that when energy uncoupling and membrane filtration were coapplied and the TCS dosage was below 30 mg/L,there was no significant deterioration in organic component removal.However,ammonia and phosphate concentrations were found to increase as the concentration of TCS added increased.Additionally,due to low sludge concentrations and EPS contents,addition of 30-60 mg/L TCS during sludge reduction increased the permeate flux(two times higher than the control)and decreased the hydraulic reversible and cake layer resistances.In contrast,high dosage of TCS aggravated membrane fouling by forming compact fouling layers.In general,this study found that the co-application of energy uncoupling and membrane filtration processes represents an effective alternative method for simultaneous sludge reduction and sludge supernatant recovery.An Ding Yingxue Zhao Zhongsen Yan Langming Bai Haiyang Yang Heng Liang Guibai Li Nanqi Ren 2020Frontiers of Environmental Science & Engineering2020,14,4:3

11Characterization and biodegradation kinetics of a new cold-adapted carbamazepine-degrading bacterium, Pseudomonas sp. CBZ-4显示文摘Carbamazepine is frequently detected in waters and hardly eliminated during conventional wastewater treatment processes due to its complicated chemical structure and resistance to biodegradation. A carbamazepine-degrading bacterium named CBZ-4 was isolated at a low temperature(10°C) from activated sludge in a municipal wastewater treatment plant. Strain CBZ-4, which can use carbamazepine as its sole source of carbon and energy, was identified as Pseudomonas sp. by the 16S rRNA gene sequence. The composition and percentage of fatty acids, which can reveal the cold-adaptation mechanism of strain CBZ-4, were determined. Strain CBZ-4 can efectively degrade carbamazepine at optimal conditions: pH 7.0, 10°C, 150 r/min rotation speed, and 13% inoculation volume. The average removal rate of carbamazepine was 46.6% after 144 hr of incubation. The biodegradation kinetics of carbamazepine by CBZ-4 was fitted via the Monod model.
