2The ATF/CREB site is the key element for transcription of the human RNA methyltransferase like 1 (RNMTL1) gene, a newly discovered 17p13.3 gene显示文摘The human RNA methyltransferase like i gene (RNMTL1) is one of thirteen newly discovered geneswithin a 116 Kb segment of the chromosome 17p13.3 that suffers from a high frequent loss of heterozygosityin human hepatocellular carcinoma in China[1-5]. To understand the molecular mechanisms underlyingtranscription control of the RNMTL1 gene in human cancers, we decline using of the conventional approachwhere the cis-elements bound by the known transcription factors are primary targets, and carried out thesystematic analyses to dissect the promoter structure and identify/characterize the key cis-elements thatare responsible for its strong expression in cell. The molecular approaches applied included 1, the primerextension for mapping of the transcription starts; 2, the transient transfection/reporter assays on a largenumber of deletion and site-specific mutants of the promoter segment for defining the minimal promoterand the crucial elements within; and 3, the electrophoresis mobility shift assay with specific antibodies forreconfirming the nature of the transcription factors and their cognate cis-elements. We have shown that theinteraction of an ATF/CREB element (-38 to -31) and its cognate transcription factors play a predominantrole in the promoter activity of the RNMTL1 gene. The secondary DNA structures of the ATF/CREBelement play a more vital role in the protein-DNA interaction. 3Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs. 6Application of transient burning rate model of solid propellant in electrothermal-chemical launch simulation显示文摘A 30 mm electrothermal-chemical(ETC) gun experimental system is employed to research the burning rate characteristics of 4/7 high-nitrogen solid propellant. Enhanced gas generation rates(EGGR) of propellants during and after electrical discharges are verified in the experiments. A modified 0D internal ballistic model is established to simulate the ETC launch. According to the measured pressure and electrical parameters, a transient burning rate law including the influence of EGGR coefficient by electric power and pressure gradient(dp/dt) is added into the model. The EGGR coefficient of 4/7 high-nitrogen solid propellant is equal to 0.005 MW-1. Both simulated breech pressure and projectile muzzle velocity accord with the experimental results well. Compared with Woodley's modified burning rate law, the breech pressure curves acquired by the transient burning rate law are more consistent with test results. Based on the parameters calculated in the model, the relationship among propellant burning rate, pressure gradient(dp/dt) and electric power is analyzed. 7Reduced function and disassembled microtubules of cultured cardiomyocytes in spaceflight显示文摘Lack of gravity during spaceflight has profound effects on cardiovascular system, but little is known about how the cardiomyocytes respond to microgravity. In the present study, the effects of spaceflight on the structure and function of cultured cardiomyocytes were reported. The primary cultures of neo- natal rat cardiomyocytes were carried on Shenzhou-6 spacecraft and activated at 4 h in orbit. 8 samples were fixed respectively at 4, 48 and 96 h after launching for immunofluorescence of cytoskeleton, and 2 samples remained unfixed to analyze contractile and secretory functions of the cultures. Ground sam- ples were treated in our laboratory in parallel. After 115 h spaceflight, video recordings displayed that the number of spontaneous beating sites in flown samples decreased significantly, and the cells in the beating aggregate contracted in fast frequency without synchrony. Radioimmunoassay of the medium showed that the atrial natriuretic peptide secreted from flown cells reduced by 59.6%. Confocal images demonstrated the time-dependant disassembly of mirotubules versus unchanged distribution and or- ganization of microfilaments. 8Calibration procedures for the CHASE/HIS science data显示文摘The Hα line is an important optical line in solar observations containing the information from the photosphere to the chromosphere. To study the mechanisms of solar eruptions and the plasma dynamics in the lower atmosphere, the Chinese Hα Solar Explorer(CHASE) was launched into a Sun-synchronous orbit on October 14, 2021. The scientific payload of the CHASE satellite is the Hα Imaging Spectrograph(HIS). The CHASE/HIS acquires, for the first time, seeing-free Hα spectroscopic observations with high spectral and temporal resolutions. It consists of two observational modes. The raster scanning mode provides full-Sun or region-of-interest spectra at Hα(6559.7-6565.9 ?) and Fe I(6567.8-6570.6 ?) wavebands. The continuum imaging mode obtains full-Sun photospheric images at around 6689 ?. In this paper, we present detailed calibration procedures for the CHASE/HIS science data, including the dark-field and flat-field correction, slit image curvature correction, wavelength and intensity calibration, and coordinate transformation. 9Exogenous strigolactones promote lateral root growth by reducing the endogenous auxin level in rapeseed显示文摘Strigolactones(SLs)are newly discovered plant hormones which regulate the normal development of different plant organs,especially root architecture.Lateral root formation of rapeseed seedlings before winter has great effects on the plant growth and seed yield.Here,we treated the seedlings of Zhongshuang 11(ZS11),an elite conventional rapeseed cultivar,with different concentrations of GR24(a synthetic analogue of strigolactones),and found that a low concentration(0.18μmol L–1)of GR24 could significantly increase the lateral root growth,shoot growth,and root/shoot ratio of seedlings.RNA-Seq analysis of lateral roots at 12 h,1 d,4 d,and 7 d after GR24 treatment showed that 2301,4626,1595,and 783 genes were significantly differentially expressed,respectively.Function enrichment analysis revealed that the plant hormone transduction pathway,tryptophan metabolism,and the phenylpropanoid biosynthesis pathway were over-represented.Moreover,transcription factors,including AP2/ERF,AUX/IAA,NAC,MYB,and WRKY,were up-regulated at 1 d after GR24 treatment.Metabolomics profiling further demonstrated that the amounts of various metabolites,such as indole-3-acetic acid(IAA)and cis-zeatin were drastically altered.In particular,the concentrations of endogenous IAA significantly decreased by 52.4 and 75.8%at 12 h and 1 d after GR24 treatment,respectively.Our study indicated that low concentrations of exogenous SLs could promote the lateral root growth of rapeseed through interaction with other phytohormones,which provides useful clues for the effects of SLs on root architecture and crop productivity.MA Ni WAN Lin ZHAO Wei LIU Hong-fang LI Jun ZHANG Chun-lei 2020Journal of Integrative Agriculture2020,19,2:3

12The Influence of Diabetes, Hypertension, and Hyperlipidemia on the Onset of Age-Related Macular Degeneration in North China: The Kailuan Eye Study显示文摘Objective To analyze the prevalence of dry and wet age-related macular degeneration(AMD)in patients with diabetes,hypertension and hyperlipidemia,and to analyze the risk factors for AMD.Methods A population-based cross-sectional epidemiologic study was conducted involving 14,440 individuals.We assessed the prevalence of dry and wet AMD in diabetic and non-diabetic subjects and analyzed the risk factors for AMD.Results The prevalence of wet AMD in diabetic and non-diabetic patients was 0.3%and 0.5%,respectively,and the prevalence of dry AMD was 17%and 16.4%,respectively.The prevalence of wet AMD in healthy,hypertensive,hyperlipidemic,and hypertensive/hyperlipidemic populations was 0.5%,0.3%,0.2%,and 0.7%,respectively.The prevalence of dry AMD in healthy,hypertensive,hyperlipidemic,and hypertensive/hyperlipidemic populations was 16.6%,16.2%,15.2%,and 17.2%,respectively.Age,sex,body mass index,and use of hypoglycemic drugs or lowering blood pressure drugs were corrected in the risk factor analysis of AMD.Diabetes,diabetes/hypertension,diabetes/hyperlipidemia,and diabetes/hypertension/hyperlipidemia were analyzed.None of the factors analyzed in the current study increased the risk for the onset of AMD.Conclusion There was no significant difference in the prevalence of wet and dry AMD among diabetic and non-diabetic subjects.Similarly,there was no significant difference in the prevalence of wet and dry AMD among subjects with hypertension and hyperlipidemia.Diabetes co-existing with hypertension and hyperlipidemia were not shown to be risk factors for the onset of dry AMD.ZHANG Yong Peng WANG Ya Xing ZHOU Jin Qiong WANG Qian YAN Yan Ni YANG Xuan YANG Jing Yan ZHOU Wen Jia WANG Ping SHEN Chang YANG Ming LUAN Ya Nan WANG Jin Yuan WU Shou Ling CHEN Shuo Hua WANG Hai Wei FANG Li Jian WAN Qian Qian ZHU Jing Yuan NIE Zi Han CHEN Yu Ning XIE Ying JONAS JB WEI Wen Bin 2022Biomedical and Environmental Sciences2022,35,7:2

14Triangulation of red sprites observed above a mesoscale convective system in North China显示文摘The triangulation of red sprites was obtained, based on concurrent observations over a mesoscale convective system(MCS) in North China from two stations separated by about 450 km. In addition, broadband sferics from the sprite-producing lightning were measured at five ground stations, making it possible to locate and identify the individual causative lightning discharges for different elements in this dancing sprite event. The results of our analyses indicate that the sprites were produced above the trailing stratiform region of the MCS, and their parent strokes were located mainly in the peripheral area of the stratiform. The lateral offset between sprites and causative strokes ranges from a few km to more than 50 km. 15Nonlinear oscillation of pathological vocal folds during vocalization显示文摘The extended two-mass model is adopted to analyze the nonlinear oscillation of pathological vocal folds during vocalization. Redundant tissue or area in laryngeal patients is modeled as a massless rigid connected to the upper mass of the vocal folds, and a parameter Q is introduced to represent the change of glottal configurations and tension imbalance between the left and right sides of vocal folds. Numerical simulations demonstrate that the pathological vocal-fold decreases the threshold of Q to generate nonlinear vocal oscillation, indicating the improvement of the sensitivity of vocal folds to asymmetries and enhancing the coupling between two sides. Furthermore, the pathological vocal-fold can lower the fundamental frequency and eliminate high-order harmonics, For example, the fundamental frequency decreases from 119.94 Hz to 84.95 Hz when Q=0.58 and the sub-glottal pressure 1450 Pa.
