| 2 | Impact of antiviral therapy on post-hepatectomy outcome for hepatitis B-related hepatocellular carcinoma显示文摘The outcome after curative resection for hepatocellular carcinoma(HCC)remains unsatisfactory due to the high recurrence rate after surgery.In patients with hepatitis B virus(HBV)-related HCC,which is the majority of patients with HCC in Asia,a high viral load is a strong risk factor for HCC recurrence.It is logical to believe that antiviral therapy may improve the postoperative outcome by promoting viral clearance and hepatocyte regeneration,as well as improving residual liver volume in HCC patients with hepatitis B.However,the effect of antiviral therapy on clinical outcomes after liver resection in patients with HBV-related HCC remains to be established.There are two main groups of antiviral treatment for HBV-oral nucleos(t)ide analogues and interferon.Interferon treatment reduces the overall incidence of HBV-related HCC in sustained re-sponders.However,side effects may limit its long-term clinical application.Nucleos(t)ide analogues carry fewer side effects and are potent in terms of viral suppression when compared to interferon and are typically implemented for patients with more advanced liver diseases.They may also improve the outcome after curative resection for HBV-related HCC.There are increasing evidence to suggest that antiviral therapy could suppress HBV,decrease the perioperative reactivation of viral replication,reduce liver injury,preserve the liver function before and after operation,and may lower the risk of HCC recurrence.After all,antiviral therapy may improve the survival after liver resection by reducing recurrence and delaying the liver damage by the virus,resulting in a higher chance of receiving aggressive salvage therapy during HCC recurrence. | Charing Ching Ning Chong Grace Lai Hung Wong Paul Bo San Lai | 2014 | World Journal of Gastroenterology2014,20,20: | 15 |
| 3 | Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs. | 4 | FLIC: Fast linear iterative clustering with active search显示文摘In this paper, we reconsider the clustering problem for image over-segmentation from a new perspective. We propose a novel search algorithm called'active search' which explicitly considers neighbor continuity. Based on this search method, we design a back-and-forth traversal strategy and a joint assignment and update step to speed up the algorithm. Compared to earlier methods, such as simple linear iterative clustering(SLIC) and its variants, which use fixed search regions and perform the assignment and the update steps separately, our novel scheme reduces the number of iterations required for convergence,and also provides better boundaries in the oversegmentation results. Extensive evaluation using the Berkeley segmentation benchmark verifies that our method outperforms competing methods under various evaluation metrics. In particular, our method is fastest,achieving approximately 30 fps for a 481 × 321 image on a single CPU core. | 6 | Latest Wuchiapingian to Earliest Triassic Conodont Zones and δ13 Ccarb Isotope Excursions from Deep-Water Sections in Western Hubei Province, South China显示文摘Deep-water facies sections have advantages of recording complete information across the Permian-Triassic boundary(PTB). Here we present a detailed study on the conodont biostratigraphy and carbon isotope profile ranges from the Wuchiapingian-Changhsingian boundary(WCB) to the PTB of two deep-water facies sections at Zhuqiao and Shiligou in the Middle Yangtze region, western Hubei, South China. Fifteen species and three genera are identified. Eight conodont zones are recognized which in ascending order are the Clarkina orientalis, C. wangi, C. subcarinata, C. changxingensis, C. yini, C. meishanensis, Hindeodus parvus and Isarcicella isarcica zones. The onset of deposition of the deep-water siliceous strata of Dalong Formation in western Hubei began in the Late Wuchiapingian and persisted to the Late Changhsingian. Carbon isotope negative excursions occur near both the WCB and PTB in both sections. The WCB δ13 Ccarb negative excursion is in the C. orientalis and C. wangi zones. The PTB δ13 Ccarb negative excursion began in the C. yini Zone and extended to the I. isarcica Zone. | 8 | Integrative taxonomy of the Russet Bush Warbler Locustella mandelli complex reveals a new species from central China显示文摘Background: The Russet Bush Warbler Locustella(previously Bradypterus) mandelli complex occurs in mountains in the eastern Himalayas, southern China, Vietnam, the Philippines, and Indonesia. The taxonomy has been debated,with one(L. seebohmi) to four(L. seebohmi, L. mandelli, L. montis and L. timorensis) species having been recognised.Methods: We used an integrative approach, incorporating analyses of morphology, vocalizations and a molecular marker, to re-evaluate species limits in the L. mandelli complex.Results: We found that central Chinese L. mandelli differed from those from India through northern Southeast Asia to southeast China in plumage, morphometrics and song. All were easily classified by song, and(wing + culmen)/tail ratio overlapped only marginally. Both groups were reciprocally monophyletic in a mitochondrial cytochrome b(cytb) gene tree, with a mean divergence of 1.0 ± 0.2%. They were sympatric and mostly altitudinally segregated in the breeding season in southern Sichuan province. We found that the Mt Victoria(western Myanmar) population differed vocally from other L. mandelli, but no specimens are available. Taiwan Bush Warbler L. alishanensis was sister to the L. mandelli complex, with the most divergent song. Plumage, vocal and cytb evidence supported the distinctness of the south Vietnamese L. mandelli idonea. The Timor Bush Warbler L. timorensis, Javan Bush Warbler L.montis and Benguet Bush Warbler L. seebohmi differed distinctly in plumage, but among-population song variation in L. montis exceeded the differences between some populations of these taxa, and mean pairwise cytb divergences were only 0.5–0.9%. We also found that some L. montis populations differed morphologically.Conclusions: We conclude that the central Chinese population of Russet Bush Warbler represents a new species,which we describe herein, breeding at mid elevations in Sichuan, Shaanxi, Hubei, Hunan and Guizhou provinces.The taxonomic status of the other allopatric populations is less clear. However, as they differ to a degree comparable with that of the sympatric L. mandelli and the new species, we elevate L. idonea to species status, and retain L. seebohmi and L. montis as separate species, the latter with timorensis as a subspecies. | 9 | Mapping Quantitative Trait Loci Associated with Aluminum Toxin Tolerance in NJRIKY Recombinant Inbred Line Population of Soybean (Glycine max)显示文摘To investigate the genetic mechanism of Al-tolerance in soybean, a recombinant inbred line population (RIL) with 184 F2:7:11 lines derived from the cross of Kefeng No.1 × Nannong 1138-2 (Al-tolerant × Al-sensitive) were tested in pot experiment with sand culture medium in net room in Nanjing. Four traits, i.e. plant height, number of leaves, shoot dry weight and root dry weight at seedling stage, were evaluated and used to calculate the average membership index (FAi) as the indicator of Al-tolerance. The composite interval mapping (CIM) under WinQTL Cartographer v. 2.5 detected five QTLs (i.e. qFAi-1, qFAi- 2, qFAi-3, qFAi-4 and qFAi-5), explaining 5.20%-9.07% of the total phenotypic variation individually. While with the multiple interval mapping (MIM) of the same software, five QTLs (qFAi-1, qFAi-5, qFAi-6, qFAi-7, and qFAi-8) explaining 5.7%-24.60% of the total phenotypic variation individually were mapped. Here qFAi-1 and qFAi-5 were detected by both CIM and MIM with the locations in a same ?anking marker region, GMKF046-GMKF080 on B1 and satt278-sat_95 on L, respectively. While qFAi-2 under CIM and qFAi-6 under MIM both on D1b2 were located in neighboring regions with their confidence intervals overlapped and might be the same locus. Segregation analysis under major gene plus polygene inheritance model showed that Al-tolerance was controlled by two major genes (h2mg = 33.05%) plus polygenes (h2pg = 52.73%). | 11 | Ticagrelor inhibits the NLRP3 inflammasome to protect against inflammatory disease independent of the P2Y_(12 ) signaling pathway显示文摘Ticagrelor is the first reversibly binding oralP2Y_(12) receptor antagonist to inhibit platelet activation and has been approved by the Food and Drug Administration for the treatment of coronary artery disease. At present, the other pharmacological functions of ticagrelor remain poorly understood. The NOD-, LRR-, and pyrin domain-containing protein 3 (NLRP3) inflammasome plays a critical role in the innate immune system, but its excessive activation also contributes to the pathogenesis of complex diseases. In this study, we systematically examined the effects of ticagrelor on the NLRP3 inflammasome and found that ticagrelor inhibits NLRP3 inflammasome activation in macrophages independent of its classic inhibitory effect on theP2Y_(12) signaling pathway. Further mechanistic studies demonstrate that ticagrelor attenuates the oligomerization of apoptosis-associated speck-like protein containing a CARD (ASC) by blocking chloride efflux, an effect achieved through the degradation of chloride intracellular channel proteins (CLICs) and blockade of the translocation of CLICs to the plasma membrane. Moreover, experiments on lipopolysaccharide-induced sepsis and alum-induced peritonitis in mice confirmed that ticagrelor mitigates the severity of systemic inflammation independent ofP2Y_(12) receptor antagonism. Importantly, oral administration of ticagrelor rapidly and strongly inhibited NLRP3 inflammasome activation in peripheral blood mononuclear cells from patients with acute coronary syndrome. | 13 | Ages and Hf isotopes of detrital zircons from the Permian strata in the engbatu area(Inner Mongolia) and tectonic implications显示文摘The Central Asian Orogenic Belt(CAOB) was built up through protracted accretion and collision of a variety of terranes/micro-continents during Neoproterozoice Mesozoic time. To understand potential links among Paleozoic subduction and accretionary processes that were operative during the development of the southeastern CAOB, we conducted a combined U-Pb and Hf-isotope analysis of detrital zircons from previously defined Devonian, Carboniferous and Early Permian strata in the Bengbatu area,Inner Mongolia. Detrital zircons from(meta-) sandstones in these strata commonly yield major Paleozoic age populations at ca. 300-261 Ma, 351-300 Ma and 517-419 Ma, and also give several Precambrian ages that range from 2687 Ma to 544 Ma. The youngest ages redefine the deposition of all these strata to be in the Middle Permian(Wordiane Capitanian) or later, much younger than previously considered.These ages, coupled with regional magmatic records, support an interpretation of most surrounding areas as possible detritus sources, including the Mongolian arcs to the north, the Northern Accretionary Orogen to the south, and the intervening Erenhote Hegenshan Ophiolite Belt. Zircons with magmatic ages of ca. 500-350 Ma and ca. 300-261 Ma display a large range of εHf(t) values(-13.97 to +15.31),whereas ca. 350-300 Ma zircons are dominated by positive εHf(t) values(+0.14 to +16.00). These results support the occurrence of two significant shifts of the zircon εHf(t) values, which has tectonic implications for the understanding of the Carboniferouse Permian evolution of the southeastern CAOB. A marked shift from mixed to positive zircon εHf(t) values at 350 -330 Ma likely manifests the incipient opening of the Hegenshan Ocean, due to the slab rollback of the subducting Paleo-Asian Oceanic lithosphere. | 18 | Development of FBMINC model for particle diffusion in fluids显示文摘An accurate particle tracking method using FBMINC (new fractional Brownian motion) is outlined. It generates non-Fickian diffusion rather than Fickian diffusion as traditional particle tracking model does. The FBMINC model is based on fractional Brownian motion (fBm) which is generalization of regular Brownian motion. The two models of fBms (FBM model and FBMINC model) were explored and the differences of the two models are compared from the three aspects: the standard deviation of each step, the small memory and the effect of the number of particles in the cloud. The results show the FBMINC model is a better model as it produces more accurate statistics. The effect of simple shear dispersion for both Brownian and fBm was investigated. The power law scaling of fBm shear dispersion was correctly identified. In addition, a scaling coefficient was found numerically. The FBMINC model is then used for producing both superdiffusive and subdiffusive particle paths, therefore, the non-Fickian diffusion of soil particle clouds can be modelled. The particle clouds represent soil contaminant are released in an idealised coastal bay and the fBm particle tracking method is used for simulation the particle cloud spreading in the bay. There is a noticeable increase in the spreading rate of the cloud. In addition, owning to the spreading rate of the cloud, a noticeable part of it has escaped the bay area and transported downstream. The variation of the Hurst exponent can lead to an area of the flow being affected by a contaminant cloud which is not picked up by the regular Brownian motion models. The purpose of this paper is to bring soil transport engineers a new angle on soil particle transport research in fluids.
