| 1 | Extracellular vesicle activities regulating macrophage- and tissue-mediated injury and repair responses显示文摘Macrophages are typically identified as classically activated(M1) macrophages and alternatively activated(M2) macrophages,which respectively exhibit pro-and anti-inflammatory phenotypes,and the balance between these two subtypes plays a critical role in the regulation of tissue inflammation,injury,and repair processes.Recent studies indicate that tissue cells and macrophages interact via the release of small extracellular vesicles(EVs) in processes where EVs released by stressed tissue cells can promote the activation and polarization of adjacent macrophages which can in turn release EVs and factors that can promote cell stress and tissue inflammation and injury and vice versa.This review discusses the roles of such EVs in resulating such interactions to influence tissue inflammation and injury in a number of acute and chronic inflammatory disease conditions,and the potential applications,advantage and concerns for using EV-based therapeutic approaches to treat such conditions,including their potential role of drug carriers for the treatment of infectious diseases. | Qian Hu Christopher J.Lyon Jesse K.Fletcher Wenfu Tang Meihua Wan Tony Y.Hu | 2021 | Acta Pharmaceutica Sinica B2021,11,6: | 22 |

| 4 | On generation source sites of internal waves in the Luzon Strait显示文摘This effort aims to determine the generation source sites in the Luzon Strait for energetic,long-crest,transbasin internal waves(IW)observed in the northern South China Sea(NSCS).The roles of islands distributed on eastern side of the strait,Kuroshio,submarine ridges,shoaling thermocline,and strait configuration played in the IW generation are examined using the cruise data analysis,satellite data interpretation,and dynamical analysis.The islands and channels on eastern side of the strait are excluded from a list of possible IW source sites owing to their unmatched horizontal dimensions to the scale of IW crest line length,and the relative low Reynolds number.The Kuroshio has a potential to be a radiator for the long-crest IW disturbances,meanwhile,the Kuroshio west(east)wing absorbs the eastward(westward)propagating IW disturbance.Namely,the Kuroshio blockades the outside west-east propagating IW disturbances.The 3-D configuration of the Luzon Strait is characterized by a sudden,more than one order widening of the cross-section areas at the outlets on both sides,providing a favorable condition for IW type initial disturbance formation.In the Luzon Strait,the thermocline is featured by a westward shoaling all the year around,providing the dynamical conditions for the amplitude growth(declination)to the westward(eastward)propagating IW type disturbance.Thus,the west slope of western submarine ridge at the western outlet of the Luzon Strait is a high possibility source sites for energetic,long-crest,transbasin IWs in the NSCS.The interpretation results of satellite SAR images during a 13 a period from 1995 to 2007 provide the convincing evidence for the conclusions. | ZHENG Quanan SONG Y Tony LIN Hui HU Xiaomin MENG Junmin WANG Dan | 2008 | Acta Oceanologica Sinica2008,27,3: | 5 |

| 6 | Nanotrap-enabled quantification of KRAS-induced peptide hydroxylation in blood for cancer early detection显示文摘Circulating peptide is a potential source of biomarkers for cancer detection.However,the existence of large molecular weight proteins in plasma have a disastrous effect on circulating peptides isolating and detecting.Herein,nanotrap fractionation following by mass spectrometry have been applied to quantify the levels of bradykinin (BK) and hydroxylated bradykinin (Hyp-BK) as a relative measure of KRAS-regulated prolyl-4-hydroxylase alpha-1 (P4HA1) which may serve as early diagnosis marker for pancreatic ductal adenocarcinoma (PDAC).We found that P4HA1 can be upregulated by KRASG12v,which is a PDAC driver mutation,using HPNE/KRAS and HPNE cells.And we revealed that P4HA1 is overexpressed in PDAC tumors,compared to normal and inflamed pancreatic tissues.RNA interference revealed that P4HA1 activity was primarily responsible for Hyp-BK production.Mass spectrometry analysis revealed that plasma Hyp-BK/BK ratio was higher in PDAC than pancreatitis patients and healthy controls,while the area under the receiver operating characteristic (ROC) curve (AUC) is 0.8209 (95%Cl,0.7269-0.9149).The Hyp-BK/BK association with PDAC was reproduced in another cohort,where this ratio was found to increase with advancing tumor stage.These novel findings paved the way for wider applications of Nanotrap coupled mass spectrometry as a powerful tool for revealing biosignatures from plasma. | Zaian Deng Zhen Zhao Bo Ning Jeffery Basilio Karen Mann Jie Fu Yajun Gu Yuanqing Ye Xifeng Wu Jia Fan Paul Chiao Tony Hu | 2019 | Nano Research2019,12,6: | 2 |

| 16 | Exploring the impact of a coordinated variable speed limit control on congestion distribution in freeway显示文摘Over the past few decades,urban freeway congestion has been highly recognized as a serious and worsening traffic problem in the world.To relieve freeway congestion,several active traffic and demand management(ATDM) methods have been developed.Among them,variable speed limit(VSL) aims at regulating freeway mainline flow upstream to meet existing capacity and to harmonize vehicle speed.However,congestion may still be inevitable even with VSL implemented due to extremely high demand in actual practice.This study modified an existing VSL strategy by adding a new local constraint to suggest an achievable speed limit during the control period.As a queue is a product of the congestion phenomenon in freeway,the incentives of a queue build-up in the applied coordinated VSL control situation were analyzed.Considering a congestion occurrence(a queue build-up)characterized by a sudden and sharp speed drop,speed contours were utilized to demonstrate the congestion distribution over a whole freeway network in various scenarios.Finally,congestion distributions found in both VSL control and non-VS control situations for various scenarios were investigated to explore the impact of the applied coordinated VSL control on the congestion distribution.An authentic stretch of Whitemud Drive(WMD),an urban freeway corridor in Edmonton,Alberta,Canada,was employed to implement this modified coordinated VSL control strategy;and a calibrated micro-simulation VISSIM model(model functions) was applied as the substitute of the real-world traffic system to test the above mentioned performance.The exploration task in this study can lay the groundwork for future research on how to improve the presented VSL control strategy for achieving the congestion mitigation effect on freeway. | Jing Cao Dawei Hu Ying Luo Tony Z.Qiu Zhuanglin Ma | 2015 | Journal of Traffic and Transportation Engineering(English Edition)2015,2,3: | 0 |

| 19 | Development and validation of a rapid UPLC/MS method for the simultaneous determination of I3C, DIM, and related metabolites and its application to pharmacokinetics studies显示文摘Indole-3-carbinol(I3C) and diindolylmethane(DIM) are naturally derived dietary phytochemicals with promising anti-cancer properties that have been demonstrated both in vitro and in vivo. Using reversed-phase ultra-performance liquid chromatography(UPLC) coupled with mass spectrometry(MS), a rapid, specific, and high throughput method was developed and validated for the quantification and identification of I3 C, DIM, and other I3 C metabolites in plasma. Samples containing I3 C or DIM and the internal standard 4-methoxy indole(IS) were extracted using a liquid-liquid extraction technique. The mean recovery was 96.21% for I3 C and 108.5% for DIM. Separation was achieved using a Waters Acquity UPLC HSS T3, 1.8 μm, 2.1 mm×150 mm column and acetonitrile–water gradient elution. The flow rate was 0.3 m L/min and the run time was 9 min. The limits of detection and quantification for I3 C and DIM were 15 ng/m L and 25 ng/m L, respectively. Calibration curves for I3 C and DIM were linear(r2>0.99) over a concentration range of 0.025–20 μg/m L. Precision, accuracy, and stability analysis fulfilled the CDER guidelines criteria. The method was successfully applied to the determination of the pharmacokinetic parameters of I3 C or DIM after oral, intravenous, or intraperitoneal administration to Sprague Dawley rats.
