| 1 | Electrochemical single nucleotide polymorphisms genotyping on surface immobilized three-dimensional branched DNA nanostructure显示文摘An electrochemical assay for single nucleotide polymorphisms (SNPs) genotyping is reported. Although electrochemical method is sensitive for DNA detection on surfaces, the ability of surface assay to precisely recognize DNA hybridization event is sacrificed to some extent due to the crowded confined surfaces environments that disfavor DNA hybridization. In the present study, we employed branched tetrahedron structure probes (TSPs) to replace regular linear single stranded DNA capture probes that were immobilized on solid surfaces. This three-dimensional DNA nanostructure lowers the density of immobilized DNA probes on confined surfaces, providing a hybridization environment that is similar to homogenous solution. | 2 | Effects of grass contour hedgerow systems on controlling soil erosion in red soil hilly areas,Southeast China显示文摘Soil erosion by water is a well-recognized serious environmental problem in the world.While contour hedgerow systems are an effective method for soil water conservation,there are a few studies on its effect in the red soil hilly areas in Southeast China.With a fixed field experiment,we constructed a runoff plot at hilly area in Zhuji County,Zhejiang province,to evaluate the effect of the grass hedgerows in soil water conservation,and to determine the optimized hedgerow patterns.Hemerocallis citrine(HC)and Ophiopogon japonicas(OJ) were selected to build the hedgerows in patterns of one row and two rows.The REE method was used to trace the source of the sediment for a better understanding of the characteristic and mechanism of erosion with hedgerows control.Our results showed that(1) hedgerows reduced erosion and surface runoff by 31.99-67.22%and 15.44-45.11%,respectively;(2) hedgerows delayed the development of rills;(3) hedgerows reduced the soil nutrients loss;(4) hedgerows reshaped the soil physical properties,especially in increasing > 0.25 mm water-stable aggregates.Taken together,our results suggest that two-row OJ is the optimized contour hedgerow pattern in the experiment condition,and downward sloping land should have the highest priority to take measures for soil water conservation.This research comprehensively studied the effects and mechanism of contour hedgerows in controlling soil and water loss in red soil hilly areas,Southeast China,so that the practice of soil and water conservation can be implemented more effectively in these areas. | Ji Fan Lijiao Yan Pei Zhang Ge Zhang | 2015 | International Journal of Sediment Research2015,30,2: | 13 |

| 4 | Intensive phototherapy vs.exchange transfusion for the treatment of neonatal hyperbilirubinemia:a multicenter retrospective cohort study显示文摘Background:Intensive phototherapy(IPT)and exchange transfusion(ET)are the main treatments for extreme hyperbilirubinemia.However,there is no reliable evidence on determining the thresholds for these treatments.This multicenter study compared the effectiveness and complications of IPT and ET in the treatment of extreme hyperbilirubinemia.Methods:This retrospective cohort study was conducted in seven centers from January 2015 to January 2018.Patients with extreme hyperbilirubinemia that met the criteria of ET were included.Patients were divided into three subgroups(low-,medium-,and high-risk)according to gestational week and risk factors.Propensity score matching(PSM)was performed to balance the data before treatment.Study outcomes included the development of bilirubin encephalopathy,duration of hospitalization,expenses,and complications.Mortality,auditory complications,seizures,enamel dysplasia,ocular motility disorders,athetosis,motor,and language development were evaluated during follow-up at age of 3 years.Results:A total of 1164 patients were included in this study.After PSM,296 patients in the IPT only group and 296 patients in the IPT plus ET group were further divided into the low-,medium-,and high-risk subgroups with 188,364,and 40 matched patients,respectively.No significant differences were found between the IPT only and IPT plus ET groups in terms of morbidity,complications,and sequelae.Hospitalization duration and expenses were lower in the low-and medium-risk subgroups in the IPT only group.Conclusions:In this study,our results suggest that IPT is a safe and effective treatment for extreme hyperbilirubinemia.The indication of ET for patients with hyperbilirubinemia could be stricter.However,it is necessary to have a contingency plan for emergency ET as soon as IPT is commenced especially for infants with risk factors.If IPT can be guaranteed and proved to be therapeutic,ET should be avoided as much as possible. | Meng Zhang Yang He Jun Tang Wenbin Dong Yong Zhang Benjin Zhang Hong Wan Quanmin Deng Lirong Guan Bin Xia Zhong Chen Min Ge Jing Zhao Wenxing Li Jingjun Pei Yi Qu Dezhi Mu | 2022 | Chinese Medical Journal2022,,5: | 7 |

| 6 | Mitochondrial aldehyde dehydrogenase(ALDH2)rescues cardiac contractile dysfunction in an APP/PS1 murine model of Alzheimer’s disease via inhibition of ACSL4-dependent ferroptosis显示文摘Alzheimer’s disease(AD)is associated with high incidence of cardiovascular events but the mechanism remains elusive.Our previous study reveals a tight correlation between cardiac dysfunction and low mitochondrial aldehyde dehydrogenase(ALDH2)activity in elderly AD patients.In the present study we investigated the effect of ALDH2 overexpression on cardiac function in APP/PS1 mouse model of AD.Global ALDH2 transgenic mice were crossed with APP/PS1 mutant mice to generate the ALDH2-APP/PS1 mutant mice.Cognitive function,cardiac contractile,and morphological properties were assessed.We showed that APP/PS1 mice displayed significant cognitive deficit in Morris water maze test,myocardial ultrastructural,geometric(cardiac atrophy,interstitial fibrosis)and functional(reduced fractional shortening and cardiomyocyte contraction)anomalies along with oxidative stress,apoptosis,and inflammation in myocardium.ALDH2 transgene significantly attenuated or mitigated these anomalies.We also noted the markedly elevated levels of lipid peroxidation,the essential lipid peroxidation enzyme acyl-CoA synthetase long-chain family member 4(ACSL4),the transcriptional regulator for ACLS4 special protein 1(SP1)and ferroptosis,evidenced by elevated NCOA4,decreased GPx4,and SLC7A11 in myocardium of APP/PS1 mutant mice;these effects were nullified by ALDH2 transgene.In cardiomyocytes isolated from WT mice and in H9C2 myoblasts in vitro,application of Aβ(20μM)decreased cell survival,compromised cardiomyocyte contractile function,and induced lipid peroxidation;ALDH2 transgene or activator Alda-1 rescued Aβ-induced deteriorating effects.ALDH2-induced protection against Aβ-induced lipid peroxidation was mimicked by the SP1 inhibitor tolfenamic acid(TA)or the ACSL4 inhibitor triacsin C(TC),and mitigated by the lipid peroxidation inducer 5-hydroxyeicosatetraenoic acid(5-HETE)or the ferroptosis inducer erastin.These results demonstrate an essential role for ALDH2 in AD-induced cardiac anomalies through regulation of lipid peroxidation and ferroptosis. | Zhi-yun Zhu Yan-dong Liu Yan Gong Wei Jin Elena Topchiy Subat Turdi Yue-feng Gao Bruce Culver Shu-yi Wang Wei Ge Wen-liang Zha Jun Ren Zhao-hui Pei Xing Qin | 2022 | Acta Pharmacologica Sinica2022,43,1: | 6 |
| 7 | Gene expression profiling and endothelin in acute experimental pancreatitis显示文摘AIM:To analyze gene expression profiles in an experimental pancreatitis and provide functional reversal of hypersensitivity with candidate gene endothelin-1 antagonists.METHODS:Dibutyltin dichloride(DBTC) is a chemical used as a polyvinyl carbonate stabilizer/catalyzer,biocide in agriculture,antifouling agent in paint and fabric.DBTC induces an acute pancreatitis flare through generation of reactive oxygen species.Lewis-inbred rats received a single i.v.injection with either DBTC or vehicle.Spinal cord and dorsal root ganglia(DRG) were taken at the peak of inflammation and processed for transcriptional profiling with a cDNA microarray biased for rat brain-specific genes.In a second study,groups of animals with DBTC-induced pancreatitis were treated with endothelin(ET) receptor antagonists [ET-A(BQ123) and ET-B BQ788)].Spontaneous pain related mechanical and thermal hypersensitivity were measured.Immunohistochemical analysis was performed using anti-ET-A and ET-B antibodies on sections from pancreatic tissues and DRG of the T10-12 spinal segments.RESULTS:Animals developed acute pancreatic inflammation persisting 7-10 d as confirmed by pathological studies(edema in parenchyma,loss of pancreatic architecture and islets,infiltration of inflammatory cells,neutrophil and mononuclear cells,degeneration,vacuolization and necrosis of acinar cells) and the painrelated behaviors(cutaneous secondary mechanical and thermal hypersensitivity).Gene expression profile was different in the spinal cord from animals with pancreatitis compared to the vehicle control group.Over 260 up-regulated and 60 down-regulated unique genes could be classified into 8 functional gene families:circulatory/acute phase/immunomodulatory;extracellular matrix;structural;channel/receptor/transporter;signaling transduction;transcription/translation-related;antioxidants/chaperones/heat shock;pancreatic and other enzymes.ET-1 was among the 52 candidate genes upregulated greater than 2-fold in animals with pancreatic inflammation and visceral pain-related behavior.Treatments with the ET-A(BQ123) and ET-B(BQ-788) antagonists revealed significant protection against inflammatory pain related mechanical and thermal hypersensitivity behaviors in animals with pancreatitis(P < 0.05).Open field spontaneous behavioral activity(at baseline,day 6 and 30 min after drug treatments(BQ123,BQ788) showed overall stable activity levels indicating that the drugs produced no undesirable effects on normal exploratory behaviors,except for a trend toward reduction of the active time and increase in resting time at the highest dose(300 μmol/L).Immunocytochemical localization revealed that expression of ET-A and ET-B receptors increased in DRG from animals with pancreatitis.Endothelin receptor localization was combined in dual staining with neuronal marker NeuN,and glia marker,glial fibrillary acidic protein.ET-A was expressed in the cell bodies and occasional nuclei of DRG neurons in na ve animals.However,phenotypic expression of ET-A receptor was greatly increased in neurons of all sizes in animals with pancreatitis.Similarly,ET-B receptor was localized in neurons and in the satellite glia,as well as in the Schwann cell glial myelin sheaths surrounding the axons passing through the DRG.CONCLUSION:Endothelin-receptor antagonists protect against inflammatory pain responses without interfering with normal exploratory behaviors.Candidate genes can serve as future biomarkers for diagnosis and/or targeted gene therapy. | Helieh S Oz Ying Lu Louis P Vera-Portocarrero Pei Ge Ada Silos-Santiago Karin N Westlund | 2012 | World Journal of Gastroenterology2012,18,32: | 6 |
| 8 | Serum microRNA-204 levels are associated with long-term cardiovascular disease risk based on the Framingham risk score in patients with type 2 diabetes: results from an observational study显示文摘Background Previous studies have demonstrated that micro RNA-204(mi R-204) is involved in atherosclerosis and vascular calcification. However, the value of mi R-204 as the predictive biomarker for cardiovascular disease(CVD) remains unclear. We aimed to evaluate the association between the circulating mi R-204 level and ten-year CVD risk based on the Framingham risk score(FRS). Methods In this retrospective study, we enrolled 194 consecutive patients with type 2 diabetes mellitus(T2DM) without CVD in Beijing Anzhen Hospital between January 2015 and September 2016. We used the FRS to evaluate the risk of CVD for each patient. Circulating mi R-204 levels were measured by quantitative real-time polymerase chain reaction. Results Circulating mi R-204 levels were significantly lower in the group of patients(0.49 ± 0.13) at high risk of CVD(FRS > 20%) than in the low(FRS < 10%) and intermediate(FRS: 10%–20%) risk groups(0.87 ± 0.19 and 0.75 ± 0.25, respectively;P < 0.001). FRS was negatively correlated with mi R-204 levels(r =-0.421, P < 0.001). According to multivariate logistic analyses, reduced mi R-204 level was independently associated with an increased risk of CVD after adjusting for conventional risk factors(OR = 0.876, 95% CI: 0.807–0.950, P = 0.001). Receiver-operating characteristic curve analysis showed that the circulating mi R-204 level can predict the high risk of CVD with higher specificity than the traditional risk factor of high systolic blood pressure or the protective factor of high-density lipoprotein cholesterol. Conclusions Our study demonstrated that patients with lower circulating mi R-204 levels were at high risk for CVD. Absorption spectra and fluorescence spectra were measured at room temperature. The values of field splitting parameter Dq and Racah parameter B were obtained by the peak values of absorption spectra. The value 10Dq/B=23.14 manifests that in β-Ga2O3 crystals Cr3+ ions are influenced by low energy crystal field. After high temperature annealing in air, the Cr3+ intrinsic emission was enhanced and the green lumines-cence disappeared. The strong and broad 691 nm emission was obtained at 420 nm excitation due to the electron transition occurred from 4T2 to 4A2. | 11 | Comparison of Achievable Rates of OFDM and Single Carrier Communication Systems显示文摘This paper presents a theoretical comparison of a single carrier system and a multi-carrier system through an analysis of the achievable rate under frequency selective fading with channel state information at the receiver. A scheme was designed to compare the achievable rates of a single carrier system and an Or-thogonal Frequency Division Multiplexing (OFDM) system. A thorough theoretical analysis of the two-path channel was conducted, and simulations were also used to analyze practical stochastic channels. Analysis and simulation results show that the achievable rates of the two approaches are comparable when the channel is flat fading. However, when the channel is frequency selective fading, the single carrier system outperforms the OFDM system.
