1Isolation and Identification of Cancer Stem-Like Cells from Murine Melanoma Cell Lines显示文摘In current study,cancer stem-like cells in the murine melanoma B16F10 cells were investigated.CD phenotypes of the B16F10 cells were analyzed by flow cytometry,and the specific CD phenotype cells from the B16F10 cells were isolated by MACS.Then we used colony formation assay in soft agar media,the cell growth assay in serum-free culture media as well as the tumorigenicity investigation of the specific CD phenotype cells in C57BL/6 mice, respectively,to identify cancer stem-like cells in the B16F10 cells.The results showed that the B16F10 cells could form spherical clones in serum-free culture media,and the rate of clonegenesis of CD133 + ,CD44 + and CD44 + CD133 + cells was higher than that of CD133 - ,CD44 - and CD44 + CD133 - cells in soft agar media,respectively. The tumorigenic potential of CD133 + ,CD44 + ,CD44 + CD133 + cells and CD44 + CD133 + CD24 + cells was stronger than that of CD133 - ,CD44 - ,CD44 + CD133 - cells and CD44 + CD133 + CD24 - cells in mice,respectively.In conclusion,the CD44 + CD133 + CD24 + cells have some biological properties of cancer stem-like cells or are highly similar to the characteristics of cancer stem cells(CSC).These results provide an important method for identifying cancer stem-like cells in B16F10 cells and for further cancer target therapy.Jun Dou Meng Pan Ping Wen Yating Li Quan Tang Lili Chu Fengshu Zhao Chuilian Jiang Weihua Hu Kai Hu Ning Gu 2007Cellular & Molecular Immunology2007,4,6:25

5Beclin 1 cleavage by caspase-3 inactivates autophagy and promotes apoptosis显示文摘Autophagy and apoptosis are both highly regulated biological processes that play essential roles in tissue homeostasis,development and diseases.Autophagy is also described as a mechanism of death pathways,however,the precise mechanism of how autophagy links to cell death remains to be fully understood.Beclin 1 is a dual regulator for both autophagy and apoptosis.In this study we found that Beclin 1 was a substrate of caspase-3 with two cleavage sites at positions 124 and 149,respectively.Furthermore,the autophagosome formation occurred,followed by the appearance of morphological hallmarks of apoptosis after staurosporine treatment.The cleavage products of Beclin 1 reduced autophagy and promoted apoptosis in HeLa cells and the cells in which Beclin 1 was stably knocked down by specific shRNA.In addition,the cleavage of Beclin 1 resulted in abrogating the interaction between Bcl-2 with Beclin 1,which could be blocked by z-VAD-fmk.Thus,our results suggest that the cleavage of Beclin 1 by caspase-3 may contribute to inactivate autophagy leading towards augmented apoptosis.Yushan Zhu Lixia Zhao Lei Liu Ping Gao Weili Tian Xiaohui Wang Haijing Jin Haidong Xu Quan Chen 2010Protein & Cell2010,1,5:17
6Dissection of No.13 lymph node in radical gastrectomy for gastric carcinoma显示文摘AIM: To evaluate the feasibility and safety of No. 13 lymphadenectomy in radical gastrectomy for gastric carcinoma. METHODS: Medical records of the patients undergone No. 13 lymph node dissection during D2 gastrectomy for gastric carcinoma, were reviewed from March 2003 to May 2007. RESULTS: One hundred and fifty-eight patients underwent No. 13 lymph node dissection for D2 gastric carcinoma, of them, 4 (2.53%) were found to have metastasis in No. 13 lymph node. Metastasis to No. 12 lymph node was detected in 6 patients and 4 of them had positive No. 13 lymph node. The operative morbidity except for wound infection was 15.19% (24/158), and hospital death rate was 1.27% (2/158). No obstructive jaundice caused by No. 13 lymph node metastasis after No. 13 lymph node dissection in radical gastrectomy for gastric carcinoma was detected during the follow-up study to end of January 2007. CONCLUSION: Dissection of No. 13 lymph node in D2 gastrectomy for gastric carcinoma is safe with a low morbidity and mortality rate. 7Impact of Heat Wave in 2005 on Mortality in Guangzhou, China显示文摘Objective To assess the impact of the heat wave in 2005 on mortality among the residents in Guangzhou and to identify susceptible subpopulations in Guangzhou, China. Methods The data of daily number of deaths and meteorological measures from 2003 to 2006 in Guangzhou were used in this study. Heat wave was defined as ≥7 consecutive days with daily maximum temperature above 35.0 ℃ and daily mean temperature above the 97th percentile during the study period. The excess deaths and rate ratio (RR) of mortality in the case period compared with the reference period in the same summer were calculated. Results During the study period, only one heat wave in 2005 was identified and the total number of excess deaths was 145 with an average of 12 deaths per day. The effect of the heat wave on non-accidental mortality (RR=1.23, 95% CI: 1.11-1.37) was found with statistically significant difference. Also, greater effects were observed for cardiovascular mortality (RR=1.34, 95% CI: 1.13-1.59) and respiratory mortality (RR=1.31, 95% CI: 1.02-1.69). Females, the elderly and people with lower socioeconomic status were at significantly higher risk of heat wave-associatedmortality. Conclusion The 2005 heat wave had a substantial impact on mortality among the residents in Guangzhou, particularly among some susceptible subpopulations. 8MicroRNA-185-5p mediates regulation of SREBP2 expression by hepatitis C virus core protein显示文摘AIM: To investigate the molecular mechanism for regulation of cholesterol metabolism by hepatitis C virus(HCV) core protein in Hep G2 cells.METHODS: HCV genotype 1b core protein was cloned and expressed in Hep G2 cells. The cholesterol content was determined after transfection. The expression of sterol regulatory element binding protein 2(SREBP2) and the rate-limiting enzyme in cholesterol synthesis(HMGCR) was measured by quantitative real-time PCR and immunoblotting after transfection. The effects of core protein on the SREBP2 promoter and 3'-untranslated region were analyzed by luciferase assay. We used different target predictive algorithms, micro RNA(mi RNA) mimics/inhibitors, and site-directed mutation to identify a putative target of a particular mi RNA.RESULTS: HCV core protein expression in Hep G2 cells increased the total intracellular cholesterol level(4.05 ± 0.17 vs 6.47 ± 0.68, P = 0.001), and this increase corresponded to an increase in SREBP2 and HMGCR m RNA levels(P = 0.009 and 0.037, respectively) and protein expression. The molecular mechanism studyrevealed that the HCV core protein increased the expression of SREBP2 by enhancing its promoter activity(P = 0.004). In addition, mi R-185-5p expression was tightly regulated by the HCV core protein(P = 0.041). Moreover, overexpression of mi R-185-5p repressed the SREBP2 m RNA level(P = 0.022) and protein expression. In contrast, inhibition of mi R-185-5p caused upregulation of SREBP2 protein expression. mi R-185-5p was involved in the regulation of SREBP2 expression by HCV core protein. 10Dental implant treatment for renal failure patients on dialysis:a clinical guideline显示文摘Chronic kidney disease(CKD) is a worldwide public health problem that is growing in prevalence and is associated with severe complications. During the progression of the disease, a majority of CKD patients suffer oral complications. Dental implants are currently the most reliable and successful treatment for missing teeth. However, due to complications of CKD such as infections,bone lesions, bleeding risks, and altered drug metabolism, dental implant treatment for renal failure patients on dialysis is more challenging. In this review, we have summarized the characteristics of CKD and previous publications regarding dental treatments for renal failure patients. 12Spatial and temporal distributions of nitrogen,phosphorus and heavy metals in the intertidal sediment of the Chang jiang River Estuary in China显示文摘Spatial and temporal distributions of nutrients(N and P) and heavy metals(Cu,Zn,Pb and Cd) in the intertidal sediments at Dongtan wetland of the Changjiang River(Yangtze River) Estuary were examined by laying one transect running seaward to landward during January,April,June,August and October of 2005. Total nitrogen(TN) and heavy metal concentrations in the intertidal sediment showed an increasing trend from open bare flat to the high marsh,with the maximum concentrations in the high marsh and the minimum values in open bare flat. In contrast,sediment total phosphorus(TP) concentrations changed little among sampling sites. One-way ANOVA found that there were significantly temporal variations of nutrients and heavy metals concentrations in the intertidal sediment(P <0.05) . The concentrations of heavy metals generally showed an increasing trend from January to October. 14Ca2+/calmodulin-dependent protein kinase II regulates colon cancer proliferation and migration via ERK1/2 and p38 pathways显示文摘AIM To investigate the role of calmodulin-dependent protein kinase Ⅱ(Ca MKⅡ) in colon cancer growth,migration and invasion.METHODS Ca MKⅡ expression in colon cancer and paracancerous tissues was evaluated via immunochemistry. Transcriptional and posttranscriptional levels of Ca MKⅡin tissue samples and MMP2,MMP9 and TIMP-1 expression in the human colon cancer cell line HCT116 were assessed by q RTPCR and western blot. Cell proliferation was detected with the MTT assay. Cancer cell migration and invasion were investigated with the Transwell culture system and woundhealing assay.RESULTS We first demonstrated that CaMK Ⅱ was ove rexpressed in human colon cancers and was associated with cancer differentiation. In the human colon cancer cell line HCT116,the Ca MKII-specific inhibitor KN93,but not its inactive analogue KN92,decreased cancer cell proliferation. Furthermore,KN93 also significantly prohibited HCT116 cell migration and invasion. 15Food Frequency Questionnaire for Chinese Children Aged 12-17 Years: Validity and Reliability显示文摘Objective The primary objective of this study was to examine the validity and reliability of a semi-quantitative food frequency questionnaire(FFQ) among Chinese children aged 12-17 years. Methods A semi-quantitative 72-food item FFQ was developed for children aged 12-17 years. The reliability and validity of this FFQ were evaluated against 24-h dietary recalls(24 h DRs) to measure the consumption of foods and nutrients. We administered two FFQs and three DRs to children(N = 160) over a period of 1 month to evaluate the reliability and validity. Reliability was examined by quartile agreement and intraclass correlation coefficients(ICCs), and validity was examined by quartile agreement, Bland-Altman plots and correlation with DRs. Results For reliability, the ICCs between the two FFQs ranged from 0.21 to 0.76 for foods and nutrients, and the quartile agreement ranged from 70.0% to 95.0% in the same or adjacent quartiles. Spearman’s correlation coefficients of foods and nutrients between the second FFQ and the 24 h DRs ranged from-0.04 to 0.59. The Bland-Altman plots demonstrated good agreement across the range of intakes among nutrients. The quartile agreement ranged from 50.0% to 100.0%, with infrequent misclassification. 17Alterations of brain local functional connectivity in amnestic mild cognitive impairment显示文摘Background:Resting-state functional magnetic resonance imaging studies using a regional homogeneity(ReHo)method have reported that amnestic mild cognitive impairment(aMCI)was associated with abnormalities in local functional connectivity.However,their results were not conclusive.Methods:Seed-based d Mapping was used to conduct a coordinate-based meta-analysis to identify consistent ReHo alterations in aMCI.Results:We identified 10 studies with 11 datasets suitable for inclusion,including 378 patients with aMCI and 435 healthy controls.This meta-analysis identified significant ReHo alterations in patients with aMCI relative to healthy controls,mainly within the default mode network(DMN)(bilateral posterior cingulate cortex[PCC],right angular gyrus,bilateral middle temporal gyri,and left parahippocampal gyrus/hippocampus),executive control network(right superior parietal lobule and dorsolateral prefrontal cortex),visual network(right lingual gyrus and left middle occipital gyrus),and sensorimotor network(right paracentral lobule/supplementary motor area,right postcentral gyrus and left posterior insula).Significant heterogeneity of ReHo alterations in the bilateral PCC,left parahippocampal gyrus/hippocampus,and right superior parietal lobule/angular gyrus was observed.Exploratory meta-regression analyses indicated that general cognitive function,gender distribution,age,and education level partially contributed to this heterogeneity.Conclusions:This study provides provisional evidence that aMCI is associated with abnormal ReHo within the DMN,executive control network,visual network,and sensorimotor network.These local functional connectivity alterations suggest coexistence of functional deficits and compensation in these networks.These findings contribute to the modeling of brain functional connectomes and to a better understanding of the neural substrates of aMCI.Confounding factors merit much attention and warrant future investigations.Dan Zheng Wei Xia Zhong Quan Yi Pan Wen Zhao Jian Guo Zhong Hai Cun Shi Hua Liang Li Zhen Yu Dai Ping Lei Pan 2018Translational Neurodegeneration2018,7,1:5
18The Impact of Relative Humidity and Atmospheric Pressure on Mortality in Guangzhou, China显示文摘Objective Although many studies have examined the effects of ambient temperatures on mortality, little evidence is on health impacts of atmospheric pressure and relative humidity. This study aimed to assess the impacts of atmospheric pressure and relative humidity on mortality in Guangzhou, China. Methods This study included 213,737 registered deaths during 2003-2011 in Guangzhou, China. A quasi-Poisson regression with a distributed lag non-linear model was used to assess the effects of atmospheric pressure/relative humidity. Results We found significant effect of low atmospheric pressure/relative humidity on mortality. There was a 1.79%(95% confidence interval: 0.38%-3.22%) increase in non-accidental mortality and a 2.27%(0.07%-4.51%) increase in cardiovascular mortality comparing the 5th and 25 th percentile of atmospheric pressure. A 3.97%(0.67%-7.39%) increase in cardiovascular mortality was also observed comparing the 5th and 25 th percentile of relative humidity. Women were more vulnerable to decrease in atmospheric pressure and relative humidity than men. Age and education attainment were also potential effect modifiers. Furthermore, low atmospheric pressure and relative humidity increased temperature-related mortality. Conclusion Both low atmospheric pressure and relative humidity are important risk factors of mortality. 20Nighttime sap flow of Acacia mangium and its implications for nighttime transpiration and stem water storage显示文摘Aims Nighttime sap flow of trees may indicate transpiration and/or recharge of stem water storage at night.This paper deals with the water use of Acacia mangium at night in the hilly lands of subtropical South China.Our primary goal was to reveal and understand the nature of nighttime sap flow and its functional significance.Methods Granier’s thermal dissipation method was used to determine the nighttime sap flux of A.mangium.Gas exchange system was used to estimate nighttime leaf transpiration and stomatal conductance of studied trees.Important Findings Nighttimesap flowwas substantial and showed seasonal variation similar to the patterns of daytime sap flowin A.mangium.Mean nighttime sap flow was higher in the less precipitation year of 2004(1122.4 mm)than in the more precipitation year of 2005(1342.5 mm)since more daytime transpiration and low soil water availability in the relatively dry 2004 can be the cause of more nighttime sap flow.Although vapor pressure deficit and air temperature were significantly correlated with nighttime sap flow,they could only explain a small fraction of the variance in nighttime sap flow.The total accumulated water loss(E_(L))by transpiration of canopy leaves was only;2.6–8.5%of the total nighttime sap flow(E_(t))during the nights of July 17–18 and 18–19,2006.Therefore,it is likely that the nighttime sap flow was mainly used for refillingwater in the trunk.The stem diameter at breast height,basal area and sapwood area explained much more variance of nighttime water recharge than environmental factors and other tree form features,such as tree height,stem length below the branch,and canopy size.The contribution of nighttime water recharge to the total transpiration ranged from 14.7 to 30.3%depending on different DBH class and was considerably higher in the dry season compared to the wet season.Hua Wang Ping Zhao Dirk Hölscher Quan Wang Ping Lu Xi A.Cai Xiao P.Zeng 2012Journal of Plant Ecology2012,5,3:5
