Alteration in the expression of microRNA-21 regulated target genes:Role in breast cancer显示文摘Breast cancer, also recognized as the principal cause of cancer-related deaths among women,is the second most familiar and prevalent form of cancer.New diagnostic and prognostic biomarkers that are highly specific are urgently needed for its early prognosis.MicroRNAs(miRNAs),a class of non-coding RNAs,are known to control the biological processes involving transcription,post-transcriptional and covalent modifications, splicing, translation, cell differentiation, proliferation, apoptosis, cancer progression, and invasion.Any dysregulation in miRNA expression, demonstrating their oncogenic and tumor-suppressive functions, contributes to cancer progression.MicroRNA-21(miR-21),an‘onco-miR’in breast cancer,is involved in tumor progression and metastasis by suppressing the activity of the target gene via its interaction with the 3’UTR of the target gene. The upregulation of miR-21 is observed in many instances of breast cancer. Our review aims to summarize the current understanding of miR-21 in the regulation of important cellular functions via regulation of its target genes. We discuss its biosynthesis, oncogenic function in breast cancer, and different methods used for its detection. This will increase the current understanding of the role of miR-21 in breast cancer tumorigenesis, which will offer a perception of using miR-21 as an early detection molecular prognostic and diagnostic biomarker and as a therapeutic target in breast cancer care.PRIYANKA THAKUR REENA V.SAINI ANIL K.CHHILLAR NEERAJ K.SAINI VIJAY KUMAR THAKUR SAMARJEET SINGH SIWAL ADESH K.SAINI 2022BIOCELL2022,46,2:1

Activation of RAS/MAPK pathway confers MCL-1 mediated acquired resistance to BCL-2 inhibitor venetoclax in acute myeloid leukemia显示文摘Despite high initial response rates, acute myeloid leukemia(AML)treated with the BCL-2-selective inhibitor venetoclax(VEN)alone or in combinations commonly acquires resistance. We performed gene/protein expression, metabolomic and methylation analyses of isogenic AML cell lines sensitive or resistant to VEN,and identified the activation of RAS/MAPK pathway, leading to increased stability and higher levels of MCL-1 protein,as a major acquired mechanism of VEN resistance.Qi Zhang Bridget Riley-Gillis Lina Han Yanan Jia Alessia Lodi Haijiao Zhang Saravanan Ganesan Rongqing Pan Sergej N.Konoplev Shannon R.Sweeney Jeremy A.Ryan Yulia Jitkova Kenneth Dunner Jr Shaun E.Grosskurth Priyanka Vijay Sujana Ghosh Charles Lu Wencai Ma Stephen Kurtz Vivian R.Ruvolo Helen Ma Connie CWeng Cassandra LRamage Natalia Baran Ce Shi Tianyu Cai Richard Eric Davis Venkata L.Battula Yingchang Mi Jing Wang Courtney D.DiNardo Michael Andreeff Jeffery W.Tyner Aaron Schimmer Anthony Letai Rose Ann Padua Carlos E.Bueso-Ramos Stefano Tiziani Joel Leverson Relja Popovic Marina Konopleva 2022Signal Transduction and Targeted Therapy2022,7,3:1
Conservation Strategy for African Medicinal Species: In Vitro Biotechnological Approach显示文摘The use of medicinal plants for different therapeutic values is well documented in African continent.African diverse biodiversity hotspots provide a wide range of endemic species, which ensures a potential medicinal value. The feasible conservation approach and sustainable harvesting for the medicinal species remains a huge challenge. However, conservation approach through different biotechnological tools such as micropropagation, somatic embryogenesis, synthetic seed production, hairy root culture, molecular markers based study and cryopreservation of endemic African medicinal species is much crucial. In this review,an attempt has been made to provide different in vitro biotechnological approaches for the conservation of African medicinal species. The present review will be helpful in further technology development and deciding the priorities at decision-making levels for in vitro conservation and sustainable use of African medicinal species.Priyanka Jha Swati Chahal Devendra Kumar Pandey Joginder Singh Ram Prasad Vijay Kumar 2020Phyton-International Journal of Experimental Botany2020,89,4:1

Does a biological link exist between periodontitis and rheumatoid arthritis?显示文摘Periodontitis or Periodontal disease(PD) and Rheumatoid arthritis(RA) are two the most common chronic inflammatory diseases. Periodontitis is a biofilm associated destructive inflammatory disease of the periodontium caused by specific microorganisms. Rheumatoid arthritis is an autoimmune condition and is identified by elevated serum autoantibody titre directed against citrullinated peptides or rheumatoid factor. Periodontitis may involve some elements of autoimmunity. Recent studies have established that PD and RA show a common pathway and could be closely associated through a common dysregulation and dysfunction in inflammatory mechanism. The enzyme peptidyl arginine deiminase(PAD), expressed by Porphyromonas gingivalis(P. gingivalis) is responsible for the enzymatic deimination of arginine residuals to citrulline resulting in protein citrullination and its increased accumulation in RA.Citrullination by PAD may act as a putative biologic link between PD and RA. Association of Human leukocytic antigen-DR4 antigen has been established both with RA and PD. Several interleukins and inflammatory mediators(ILs) and Nuclear factor kappa beta ligand are linked to these common chronic inflammatory diseases. Antibodies directed against heat shock protein(hsp 70 ab) of P. gingivalis, P. melanogenicus and P. intermedia are raised in PD as well as RA. Both the conditions share many pathological and immunological similarities. Bacterial infection, genetic susceptibility, altered immune reaction and inflammatory mediators considered responsible for RA are also associated with PD. So it is plausible that a biological link may exist between PD and RA. Therapies aimed at modifying the expression and effect of inflammatory mediators and effector molecules such as matrix metalloproteinases, proinflammatory cytokines and autoantibodies of structural proteins may probably reduce the severity of both RA and PD.Rosamma Joseph MG Jose Raj Shobha Sundareswaran Priyanka Chand Kaushik Amol Vijay Nagrale Susan Jose Sreeraj Rajappan 2014World Journal of Rheumatology2014,4,3:0
Antimalarial drug artesunate affords protection against carrageenan induced acute inflammation in rat显示文摘Artesunate, an antimalarial drug, has been shown to inhibit the release of inflammatory mediators in various disease conditions. The present study was carried out to evaluate the anti-edematogenic effect of arte- sunate in the rat paw edema model. Inflammation was induced in the hind paw of rat by sub-plantar injection of 0.1 mL of 0.5% carrageenan and the paw volume was measured up to a fixed mark just before the injection and then after 3 h. The difference in two volumes gave a measure of edema formation. At 3h the level of TNF-α, PGE_(2) and myeloperoxidase were estimated in the inflamed paw tissue. Treatment of rats with single dose of artesunate at 50 and 150 mg/kg produced a dose-dependent inhibition in paw inflammation where a significant reduction in edema volume and mediator release was observed. Our study shows that by inhibiting the release of inflam- matory mediators artesunate affords protection against acute inflammation induced in the rat paw and suggests that it has a potential to be used in the treatment of inflammatory disease conditions.Vijay L.KUMAR B.GURUPRASAD Priyanka CHAUDHARY 2014BIOCELL2014,38,1:0
