| 1 | High expression of ACE2 receptor of 2019-nCoV on the epithelial cells of oral mucosa显示文摘It has been reported that ACE2 is the main host cell receptor of 2019-nCoV and plays a crucial role in the entry of virus into the cell to cause the final infection. To investigate the potential route of 2019-n Cov infection on the mucosa of oral cavity, bulk RNA-seq profiles from two public databases including The Cancer Genome Atlas(TCGA) and Functional Annotation of The Mammalian Genome Cap Analysis of Gene Expression(FANTOM5 CAGE) dataset were collected. RNA-seq profiling data of 13 organ types with para-carcinoma normal tissues from TCGA and 14 organ types with normal tissues from FANTOM5 CAGE were analyzed in order to explore and validate the expression of ACE2 on the mucosa of oral cavity. Further, single-cell transcriptomes from an independent data generated in-house were used to identify and confirm the ACE2-expressing cell composition and proportion in oral cavity. The results demonstrated that the ACE2 expressed on the mucosa of oral cavity. Interestingly, this receptor was highly enriched in epithelial cells of tongue. | 5 | Aberrant translation regulated by METTL1/WDR4-mediated tRNA N7-methylguanosine modification drives head and neck squamous cell carcinoma progression显示文摘Background:Cancer cells selectively promote the translation of oncogenic tran-scripts to stimulate cancer progression.Although growing evidence has revealed that tRNA modifications and related genes participate in this process,their roles in head and neck squamous cell carcinoma(HNSCC)remain largely unchar-acterized.Here,we sought to investigate the function and mechanisms of the transfer RNA(tRNA)N7-methylguanosine(m'G)modification in regulating the occurrence and development of HNSCC.Methods:Cell lost of-function and gain-of function assays,xenograft models,conditional knockout and knockin mouse models were used to study the physi-ological functions of tRNA m'G modification in HNSCC tumorigenesis.tRNA modification and expression profiling,mRNA translation profiling and res-cue assays were performed to uncover the underlying molecular mechanisms.Single-cell RNA sequencing(scRNA seq)was conducted to explore the tumor microenvironment changes.Results:The tRNA.m7G methyltransferase complex components Methyltransferase-like 1(METTL1)/WD repeat domain 4(WDR4)were upregulated in HNSCC and associated with a poor prognosis.Functionally,METTL1/WDR4 promoted HNSCC progression and metastasis in cell-based and transgenic mouse models.Mechanistically,ablation of METTL1 reduced the m'G levels of 16 tRNAS,inhibiting the translation of a subset of oncogenic transcripts,including genes related to the phosphatidylinositol-3-kinase/protein kinase B/mammalian target of rapamycin(PI3K/AKT/mTOR)signaling pathway.In addition,chemical modulators of the PI3K/Akt/mTOR signaling pathway reversed the effects of Mettll in mouse HNSCC.Furthermore,scRNA-seq results revealed that Mettll knockout in mouse tumor cells altered the immune landscape and cell-cell interaction between the tumor and stromal compartment.Conclusions:The tRNA m?G methyltransferase METTLI was found to promote the development and malignancy of HNSCC through regulating global mRNA translation,including the PI3K/AKT/mTOR signaling pathway,and found to alter immune landscape.METTLI could be a promising treatment target for HNSCC patients. | Jie Chen Kang Li Jianwen Chen Xiaochen Wang Rongsong Ling Maosheng Cheng Zhi Chen Fangfang Chen Qianting He Shuai Li Caihua Zhang Yizhou Jiang Qianming Chen Anxun Wang Demeng Chen | 2022 | Cancer Communications2022,42,3: | 6 |
| 6 | Carrier dynamic overlapping switching frequency optimal pulse width modulation method for modular multilevel converters显示文摘Modular multilevel converters(MMCs) operate in the low modulation index region in many applications.However, when utilized at the low modulation index region, large harmonics appear in the output voltage, which degrade the performance of the MMC. To improve the harmonic characteristic in the low modulation index, the carrier dynamic overlapping switching frequency optimal pulse width modulation(CDOSFOPWM) method is proposed for the MMC here. The whole modulation index region is divided into three regions: high modulation index region, middle modulation index region, and low modulation index region. The carrier amplitude, carrier overlap ratio, and frequency of triangular carriers are chosen dynamically according to the modulation index region of the modulation signals, to achieve the optimal harmonic characteristic in the whole modulation index region and maintain the switching loss. The number of on-state submodules(SMs) can be calculated by CDOSFOPWM, and the selection of SMs is performed by a reducing switching frequency voltage balancing algorithm. Alkali metal insertion of MoS_2 had been proved an effective method to cause phase transition early in 1970s, and has been gaining renewed interest recently, due to the possible application of MoS_2 in energy storage. The alkali metal intercalation of MoS_2 has been studied by various techniques, among which in-situ transmission electron microscopy (TEM) provides unique capability of real time resolving the structural evolution of the materials at high spatial resolutions. Here by in-situ TEM technique we investigated the structural evolution of MoS_2 upon lithium and sodium intercalation, along with transformation of the nanosheet and variation of the electron diffraction patterns. The intercalation process is accompanied by emergence of superstructures, which exist in several forms. The ion intercalation results in phase transition of MoS_2 from 2H to 1T, and the driving mechanism of the phase transition are discussed. | 11 | Electrochemical solid-state amorphization in the immiscible Cu-Li system显示文摘As a typical immiscible binary system, copper (Cu) and lithium (Li) show no alloying and chemical intermixing under normal circumstances. Here we show that, when decreasing Cu nanoparticle sizes into ultrasmall range, the nanoscale size effect can play a subtle yet critical role in mediating the chemical activity of Cu and therefore its miscibility with Li, such that the electrochemical alloying and solidstate amorphization will occur in such an immiscible system. This unusual observation was accomplished by performing in-situ studies of the electrochemical lithiation processes of individual CuO nanowires inside a transmission electron microscopy (TEM). Upon lithiation, CuO nanowires are first electrochemically reduced to form discrete ultrasmall Cu nanocrystals that, unexpectedly, can in turn undergo further electrochemical lithiation to form amorphous Cu Lixnanoalloys. Real-time TEM imaging unveils that there is a critical grain size (ca. 6 nm), below which the nanocrystalline Cu particles can be continuously lithiated and amorphized. | 12 | Comparison of fatty acid profile of three adipose tissues in Ningxiang pigs显示文摘The present study is conducted to determinate fatty acids(FA) composition in 3 adipose tissues. Subcutaneous and perirenal adipose tissues were prepared from 24 Ningxiang castrated boars and 24 castrated gilts fattened by a traditional diet for 56 d, respectively. The results showed that the FA profile in the 3 adipose tissues(dorsal subcutaneous adipose [DSA], abdominal subcutaneous adipose [ASA], and perirenal adipose [PA]) differed greatly. In boars, the proportions of oleic acid(c18:1 n9 c)(P < 0.05), cis-11-20 c acid(c20:1)(P < 0.05), and a-linolenic acid(c18:3 n3)(P < 0.05) in DSA were the highest among 3 adipose tissues, whereas palmitic acid(c16:0)(P < 0.05) and stearic acid(c18:0)(P < 0.05) in DSA had the lowest proportion. In gilts, cis-11-20 c acid(c20:1)(P < 0.05) in DSA was the highest, while stearic acid(c18:0)(P < 0.05) in subcutaneous adipose was the lowest among these deposits. | 17 | OsMADS32 interacts with PI-like proteins and regulates rice flower development显示文摘OsMADS32 is a monocot specific MIKC^c type MADS-box gene that plays an important role in regulating rice floral meristem and organs identity, a crucial process for reproductive success and rice yield. However, its underlying mechanism of action remains to be clari fied. Here, we characterized a hypomorphic mutant allele of OsMADS32/CFO1, cfo1-3 and identi fied its function in controlling rice flower development by bioinformatics and protein-protein interaction analysis. The cfo1-3 mutant produces defective flowers,including loss of lodicule identity, formation of ectopic lodicule or hull-like organs and decreased stamen number,mimicking phenotypes related to the mutation of B class genes. Molecular characterization indicated that mis-splicing of OsMADS32 transcripts in the cfo1-3 mutant resulted in an extra eight amino acids in the K-domain of OsMADS32 protein.By yeast two hybrid and bimolecular fluorescence complementation assays, we revealed that the insertion of eight amino acids or deletion of the internal region in the K1 subdomain of Os MADS32 affects the interaction between OsMADS32 with PISTILLATA(PI)-like proteins OsMADS2 and OsMADS4.
