1Integrative single-cell analysis of transcriptome,DNA methylome and chromatin accessibility in mouse oocytes显示文摘Oocyte growth is a key step in forming mature eggs that are ready to be fertilized.The states and modifications of chromatin represent critical sources of information for this process.However,the dynamics and interrelations of these chromatin characteristics remain elusive.In this study,we developed an improved scCOOL-seq technique (iscCOOL-seq),which is a multiomics,single-cell and single-base resolution method with high mapping rates,and explored the chromatin accessibility landscape and its relationship to DNA methylation in growing mouse oocytes.The most dramatic change in chromatin accessibility occurs during oocyte growth initiation,accompanied with prominent transcriptome alterations and an elevated variation in DNA methylation levels among individual oocytes.Unlike CpG islands (CGIs),partially methylated domains (PMDs)are associated with a low density of nucleosome-depleted regions (NDRs)during the whole maturation period.Surprisingly,highly expressed genes are usually associated with NDRs at their transcriptional end sites (TESs).In addition,genes with de novo methylated gene bodies during oocyte maturation are already open at their promoters before oocyte growth initiation.Furthermore,epigenetic and transcription factors that might be involved in oocyte maturation are identified.Our work paves the way for dissecting the complex, yet highly coordinated,epigenetic alterations during mouse oocyte growth and the establishment of totipotency.Chan Gu Shanling Liu Qihong Wu Lin Zhang Fan Guo 2019Cell Research2019,29,2:14
2RNA-binding protein LIN28B inhibits apoptosis through regulation of the AKT2/FOXO3A/BIM axis in ovarian cancer cells显示文摘LIN28B is an evolutionarily conserved RNA-binding protein that regulates mRNA translation and miRNA let-7 maturation in embryonic stem cells and developing tissues.Increasing evidence demonstrates that LIN28B is activated in cancer and serves as a critical oncogene.However,the underlying molecular mechanisms of LIN28B function in tumorigenesis are still largely unknown.Here we report that LIN28B was expressed in over half of the patients with epithelial ovarian cancer who were examined(n=584).Functional experiments demonstrated that LIN28B inhibited ovarian cancer cell apoptosis.Furthermore,we showed that the proapoptotic factor BIM played an essential role in the antiapoptotic function of LIN28B.RNA-IP microarray analysis suggested that LIN28B binds to mRNAs that are associated with the DNA damage pathway,such as AKT2,in ovarian cancer cells.By binding to AKT2 mRNA and enhancing its protein expression,LIN28B regulated FOXO3A protein phosphorylation and decreased the transcriptional level of BIM,which antagonized the antiapoptosis activity of LIN28B.Taken together,these results mechanistically linked LIN28B and the AKT2/FOXO3A/BIM axis to the apoptosis pathway.The findings may have important implications in the diagnosis and therapeutics of ovarian cancer.Xiaojuan Lin Jianfeng Shen Dan Peng Xinhong He Congjian Xu Xiaojun Chen Janos LTanyi Kathleen Montone Yi Fan Qihong Huang Lin Zhang Xiaomin Zhong 2018Signal Transduction and Targeted Therapy2018,3,1:8

4Anal swab findings in an infant with COVID-19显示文摘Introduction: The transmission pathways of coronavirus disease 2019 (COVID-19) remain not completely clear. In this case study the test for the novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in pharyngeal swab and anal swab were compared.Case presentation: A 3-month-old girl was admitted to our hospital with COVID-19. Her parents had both been diagnosed with COVID-19. The results of pharyngeal swab and anal swab of the little girl were recorded and compared during the course of the disease. The oropharyngeal specimen showed negative result for SARS-CoV-2 on the 14th day after onset of the illness. However, the anal swab was still positive for SARS-CoV-2 on the 28th day after the onset of the illness.Conclusion: The possibility of fecal-oral transmission of COVID-19 should be assessed. 5Apolipoprotein E gene polymorphism in cerebrovascular diseases of the Chinese Naxi populations from Yunnan province显示文摘Currently it is not well known whether apolipoprotein E(ApoE) is a genetic susceptibility factor for cerebrovascular diseases in the Chinese Naxi population.The present study detected and sequenced ApoE polymorphisms of 90 patients with cerebrovascular diseases(58 cases of cerebral infarction and 32 cases of intracerebral hemorrhage),and 50 normal people of Naxi nationality from Yunnan province,China.The populations were used to analyze the relationship of ApoE polymorphisms with cerebral infarction and intracerebral hemorrhage.Results showed an association between ApoE gene polymorphism and the onset of cerebral infarction,and a possibility that the ε4 allele is a susceptibility locus for the risk of cerebral infarction.However,there was no evidence of a relationship between the ApoE gene polymorphism and cerebral hemorrhage.Hong Xu Qihong Yuan Xijun Fan Guoqiang He 2011Neural Regeneration Research2011,6,9:1

8Comparisons of two serious air pollution episodes in winter and summer in Beijing显示文摘Characteristics of two serious air pollution episodes(9–15 January, as the winter case; and30 June to 1 July, as the summer case), which occurred in Beijing in 2013 were investigated and compared using multi-method observations and numerical simulations. During these two air pollution episodes, PM_(2.5) concentrations varied significantly within Beijing, with PM_(2.5) concentrations in southern parts of Beijing being significantly higher than in northern areas. Typically, heavy air pollution episodes begin in the southern parts and disperse towards the northern parts of Beijing. Clearly, synoptic patterns and the stability of atmospheric circulation patterns were the main factors controlling air pollution in Beijing.During the winter case, a warm center above 900 h Pa occurred over Beijing. Meanwhile, in the summer case, although there was only a weak inversion, the convective inhibition energy was strong(over 200 J/k G). This clearly influenced the duration of the air pollution event. Except for the local accumulation and secondary atmospheric reactions in both cases, regional straw burnings contributed a lot to the PM_(2.5) concentrations in summer case.Using the CAMxmodel, we established that regional transport contributed almost 59% to the PM_(2.5) averaged concentration in Beijing in the winter case, but only 31% in the summer case. Thus, the winter case was a typical regional air pollution episode, while the summer case resulted from local accumulation straw burnings transportation and strong secondary atmospheric reactions. 9Recurrent kawasaki disease with intussusception:A case report and a review of the literature显示文摘Objective:Kawasaki disease(KD)is a multisystemic vasculitic disease that occasionally presents with gastrointestinal tract symptoms.Case presentation:A 2-year-old girl diagnosed with KD 4 months prior was referred to our hospital with complaints of vomiting.She was subsequently diagnosed with intussusception,which was efficiently alleviated through air insufflation.However,despite her improved clinical status,she had a fever unresponsive to antibiotic therapy.Four days later,the patient exhibited typical symptoms of Kawasaki disease.Thus,intravenous immunoglobulin and high-dose aspirin were administrated,and the outcome was satisfactory.Conclusions:In young children who present with crying,vomiting,and passage of bloody stools,color ultrasonography should be performed promptly to avoid missed diagnosis and misdiagnosis.Yan Pan Qihong Fan 2022Discussion of Clinical Cases2022,9,2:0
10Epidemiological characteristics and clinical manifestations of pediatric patients with COVID-19 in China: A multicenter retrospective study显示文摘Importance:The Coronavirus disease 2019(COVID-19)global pandemic poses a considerable challenge for pediatricians.Objective:This study aimed to identify the epidemiological characteristics and clinical features of pediatric patients with COVID-19 in China.Methods:This multicenter retrospective study included pediatric patients from 46 hospitals in China,covering 12 provinces and two municipalities.Epidemiological,demographic,clinical,laboratory,treatment,and outcome data were analyzed.Results:In total,211 pediatric patients with COVID-19 were included in this study.The median age was 7.0 years(range:22 days to 18 years).Approximately 16.3%of the patients exhibited asymptomatic infections,23.0%had upper respiratory tract infections,and 60.7%had pneumonia,including two with severe pneumonia and one with critical illness.Approximately 78.7%of the pediatric patients occurred in familial clusters.The most three common symptoms or signs at onset in children with COVID-19 were fever(54.5%),cough(49.3%),and pharyngeal congestion(20.8%).Only 17.6%of the patients presented with decreased lymphocyte count,whereas 13.6%had increased lymphocyte count.Among the patients with pneumonia who exhibited abnormal chest computed tomography findings,18.2%(23/127)of the patients had no other symptoms.Generally,the chest radiographs showed abnormalities that affected both lungs(49.6%);ground-glass opacity(47.2%)was the most common manifestation.The cure and improvement rates were 86.7%(183/211)and 13.3%(28/211),respectively.Only one patient with an underlying condition received invasive mechanical ventilation;none of the patients died.Interpretation:Similar to adults,children of all age groups are susceptible to COVID-19.Fortunately,most pediatric patients have mild symptoms or remain asymptomatic,despite the high incidence of pneumonia.Decreased proportions of white blood cells and lymphocytes are less frequent in children than in adults.Yan Bai Liwei Gao Xianfeng Wang Lili Zhong Jingfeng Li Shenggang Ding Yuejie Zheng Jun Liu Yuxia Cui Lina Wang Mingfeng Han Jun Liu Xinping Jiang Min Jiang Junhua Li Ning Chen Yunxiao Shang Hourong Zhou Yi Xu Gen Lu Xing Chen Jizhi Xu Qihong Fan Yu Tang Jiang Wu Cen Li Xiaoxiang Yang Chunxi Chen Yonghong Yang Gary Wing-Kin Wong Adong Shen Tianyou Wang Runming Jin Baoping Xu Kunling Shen 2021Pediatric Investigation2021,5,3:0

12Serum erythropoietin and transferrin in children with idiopathic nephrotic syndrome显示文摘Idiopathic nephrotic syndrome(INS)is char-acterized by marked urinary excretion of albumin and other intermediate-sized plasma proteins,such as trans-ferrin and vitamin D-binding protein.Some cases even develop anemia.The aim of this study was to investigate the changes in serum iron,transferrin,and erythropoietin,and the relationships between serum and urine transferrin and erythropoietin.Thirty-seven children with INS and 35 age-and sex-matched healthy children were investi-gated.The indexes related to iron metabolism,including serum iron,ferritin,transferrin,total iron-binding capa-city(TIBC),transferrin saturation,and hematological parameters[hemoglobin(Hb),mean corpuscular volume(MCV),mean corpuscular hemoglobin(MCH)],and urin-ary transferrin and erythropoietin were measured in 37 children with INS before treatment and at the remission stage.Thirty-five age-and sex-matched healthy children served as controls.Serum iron levels(18.8±3.8)mmol/L in INS patients before treatment were significantly lower than those of the healthy controls(22.2±3.8)mmol/L and those measured at the remission stage(21.0±3.5)mmol/L(all P<0.01).Serum transferrin levels in INS patients before therapy(1.9±0.3)g/L also decreased compared with the healthy controls(3.1±0.5)g/L and the measures at the remission stage(2.9±0.6)g/L(all P<0.01).In contrast,serum TIBC and transferrin saturation were sig-nificantly higher in INS patients before treatment than in the healthy controls[TIBC(56.4±9.2)mmol/L vs(50.7±6.8)mmol/L,P<0.01;transferrin saturation(55.7±9.2)%vs(46.4±8.2)%,P<0.01]and they were also higher than the measures at remission stage[(51.9±7.7)mmol/L and(47.4±13.3)mmol/L](all P<0.01).Serum transferrin was positively correlated with serum albumin(r= 50.609,P<0.01)and negatively correlated with urinary transferrin(r= 520.550,P<0.01)in INS patients before treatment.We conclude that serum iron,transferrin and erythropoietin levels are markedly decreased in INS patients,which may be partially related to the urinary loss of these indexes.Hongzhu LU Lin WANG Qihong FAN Dan LIU Wanming ZHANG Yuesha YUAN Hongyan KUANG 2008Frontiers of Medicine2008,2,3:0
