2Indoor Allergen Levels and Household Distributions in Nine Cities Across China显示文摘Objective Chinese allergic subjects have high levels of sensitization to house dust mite(HDM) and other indoor allergens. This study quantifies common indoor allergen levels in Chinese households. Methods Dust samples were collected from nine cities. Major allergens Der p 1 and Der f 1 from Dermatophagoides pteronyssinus and D. farinae, and specific antigens of Blomia tropicalis, Tyrophagus putrescentiae, Acarus siro, and cockroach species Blattella germanica and Periplaneta americana were measured by ELISA. Results HDM allergens were found in dust samples from bedding in 95% of the Chinese households. The median levels varied from <0.006 to 9.2 μg/g of dust, depending on the city. The percentages of households having HDM allergen levels associated with the risk of developing allergy sensitization and asthma were 65% and 25%, respectively. Specific antigens of the storage mite and cockroach were only found in samples from the southern and tropical regions of China. Levels of mite allergens were generally higher in samples from bedding compared to samples from the living room, even for storage mites, whereas levels of cockroach antigens were higher in the living room samples. Conclusion HDM allergens are present in bedding dust samples from most Chinese households. Cities in southern and central China have relatively high levels of HDM major allergens compared to cities in northern and western China. 5Effects of Temperature and Moisture on Soil Organic Matter Decomposition Along Elevation Gradients on the Changbai Mountains, Northeast China显示文摘Decomposition of soil organic matter(SOM) is of importance for CO_2 exchange between soil and atmosphere and soil temperature and moisture are considered as two important factors controlling SOM decomposition. In this study, soil samples were collected at 5 elevations ranging from 753 to 2 357 m on the Changbai Mountains in Northeast China, and incubated under different temperatures(5, 10, 15, 20, 25, and 30?C) and soil moisture levels(30%, 60%, and 90% of saturated soil moisture) to investigate the effects of both on SOM decomposition and its temperature sensitivity at different elevations. The results showed that incubation temperature(F = 1 425.10, P < 0.001), soil moisture(F = 1 327.65, P < 0.001), and elevation(F = 1 937.54, P < 0.001) all had significant influences on the decomposition rate of SOM. The significant effect of the interaction of incubation temperature and soil moisture on the SOM decomposition rate was observed at all the 5 sampling elevations(P < 0.001). A two-factor model that used temperature and moisture as variables fitted the SOM decomposition rate well(P < 0.001) and could explain 80%–93% of the variation of SOM decomposition rate at the 5 elevations. Temperature sensitivity of SOM decomposition, expressed as the change of SOM decomposition rate in response to a 10?C increase in temperature(Q_(10)), was significantly different among the different elevations(P < 0.01), but no apparent trend with elevation was discernible. In addition, soil moisture and incubation temperature both had great impacts on the Q_(10) value(P < 0.01), which increased significantly with increasing soil moisture or incubation temperature. Furthermore, the SOM decomposition rate was significantly related to soil total Gram-positive bacteria(R^2= 0.33, P < 0.01) and total Gram-negative bacteria(R^2= 0.58, P < 0.001). 6Transcriptional landscape of rice roots at the single-cell resolution显示文摘There are two main types of root systems in flowering plants,namely taproot systems of dicots and fibrous root systems found in monocots.Despite this fundamental split,our current knowledge of cellular and molecular mechanism driving root development is mainly based on studies of the dicot model Arabidopsis.However,the world major crops are monocots and little is known about the transcriptional programs underlying cell-type specification in this clade.Here,we report the transcriptomes of more than 20000 single cells derived from root tips of two agronomically important rice cultivars.Using combined computational and experimental analyses we were able to robustly identify most of the major cell types and define novel cell-type-specific marker genes for both cultivars.Importantly,we found divergent cell types associated with specific regulatory programs,including phytohormone biosynthesis,signaling,and response,which were well conserved between the two rice cultivars.In addition,we detected substantial differences between the cell-type transcript profiles of Arabidopsis and rice.These species-specific features emphasize the importance of analyzing tissues across diverse model species,including rice.Taken together,our study provides insight into the transcriptomic landscape of major cell types of rice root tip at single-cell resolution and opens new avenues to study cell-type specification,function,and evolution in plants.Qing Liu Zhe Liang Dan Feng Sanjie Jiang Yifan Wang Zhuoying Du Ruoxi Li Guihua Hu Pingxian Zhang Yanfei Ma Jan ULohmann Xiaofeng Gu 2021Molecular Plant2021,14,3:9
7Relationship between Maternal PBMC HBV cccDNA and HBV Serological Markers and its Effect on HBV Intrauterine Transmission显示文摘Objective To investigate the relationship between maternal peripheral blood mononuclear cells (PBMC) hepatitis B virus(HBV) covalenty closed circular deoxyribonucleic acid(cccDNA) and other HBV serological markers and its effects on HBV intrauterine transmission. Methods We enrolled 290 newborns and their hepatitis B surface antigen(HBsAg) positive mothers. HBV cccDNA in PBMC and HBV DNA in serum were detected by a real‐time PCR‐TaqM an probe while HBV serological markers were detected with an electrochemiluminescence immunoassay. Results There was a positive correlation between the levels of PBMC HBV cccD NA and serum HBV DNA and HBeA g(r = 0.436 and 0.403, P < 0.001). The detection rate of pattern A [‘HBsA g(+), HBeA g(+), and anti‐HBc(+)’] was significantly higher in the PBMC HBV cccD NA positive group than in the control group(χ^2 = 48.48, P < 0.001). There was a significant association between HBV intrauterine transmission and PBMC HBV cccD NA(χ^2 = 9.28, P = 0.002). In the presence of serum HBV DNA, HBeA g, and PBMC HBV cccD NA, the risk of HBV intrauterine transmission was three times higher(OR = 3.69, 95% CI: 1.30‐10.42) than that observed in their absence. The risk of HBV intrauterine transmission was the greatest(OR = 5.89, 95% CI: 2.35‐14.72) when both PBMC HBV cccD NA and pattern A were present. A Bayesian network model showed that maternal PBMC HBV cccD NA was directly related to HBV intrauterine transmission. Conclusion PBMC HBV cccDNA may be a direct risk factor for HBV intrauterine transmission. 9Tumor microenvironments self-activated nanoscale metal-organic frameworks for ferroptosis based cancer chemodynamic/photothermal/chemo therapy显示文摘Ferroptosis,as a newly discovered cell death form,has become an attractive target for precision cancer therapy.Several ferroptosis therapy strategies based on nanotechnology have been reported by either increasing intracellular iron levels or by inhibition of glutathione(GSH)-dependent lipid hydroperoxidase glutathione peroxidase 4(GPX4).However,the strategy by simultaneous iron delivery and GPX4 inhibition has rarely been reported.Herein,novel tumor microenvironments(TME)-activated metal-organic frameworks involving Fe&Cu ions bridged by disulfide bonds with PEGylation(FCSP MOFs)were developed,which would be degraded specifically under the redox TME,simultaneously achieving GSH-depletion induced GPX4 inactivation and releasing Fe ions to produce ROS via Fenton reaction,therefore causing ferroptosis.More ROS could be generated by the acceleration of Fenton reaction due to the released Cu ions and the intrinsic photothermal capability of FCSP MOFs.The overexpressed GSH and H2O2 in TME could ensure the specific TME self-activated therapy.Better tumor therapeutic efficiency could be achieved by doxorubicin(DOX)loading since it can not only cause apoptosis,but also indirectly produce H2O2 to amplify Fenton reaction.Remarkable anti-tumor effect of obtained FCSP@DOX MOFs was verified via both in vitro and in vivo assays.Yu Liang Li Zhang Chao Peng Shiyu Zhang Siwen Chen Xin Qian Wanxian Luo Qing Dan Yongyan Ren Yingjia Li Bingxia Zhao 2021Acta Pharmaceutica Sinica B2021,11,10:7
10Neonatal ketamine exposure-induced hippocampal neuroapoptosis in the developing brain impairs adult spatial learning ability显示文摘Ketamine exposure can lead to selective neuroapoptosis in the developing brain.p66ShcA,the cellular adapter protein expressed selectively in immature neurons,is a known pro-apoptotic molecule that triggers neuroapoptosis when activated.Sprague-Dawley rats at postnatal day 7 were subcutaneously injected in the neck with ketamine 20 mg/kg,six times at 2-hour intervals.At 0,1,3,and 6 hours after final injection,western blot assay was used to detect the expression of cleaved caspase-3,p66ShcA,and phosphorylated p66ShcA.We found that the expression of activated p66ShcA and caspase-3 increased after ketamine exposure and peaked at 3 hours.The same procedure was performed on a different group of rats.At the age of 4 weeks,spatial learning and memory abilities were tested with the Morris water maze.Latency to find the hidden platform for these rats was longer than it was for control rats,although the residence time in the target quadrant was similar.These findings indicate that ketamine exposure resulted in p66ShcA being activated in the course of an apoptotic cascade during the neonatal period.This may have contributed to the deficit in spatial learning and memory that persisted into adulthood.The experimental protocol was approved by the Institutional Animal Care and Use Committee at the University of Texas at Arlington,USA (approval No.A13.008) on January 22,2013.Dan Lyu Ning Tang Andrew W. 13Neuroimaging mechanisms of high-frequency repetitive transcranial magnetic stimulation for treatment of amnestic mild cognitive impairment:a double-blind randomized sham-controlled trial显示文摘Individuals with amnestic mild cognitive impairment(aMCI)have a high risk of developing Alzheimer’s disease.Although repetitive transcranial magnetic stimulation(rTMS)is considered a potentially effective treatment for cognitive impairment in patients with aMCI,the neuroimaging mechanisms are poorly understood.Therefore,we performed a double-blind randomized sham-controlled trial in which rTMS was applied to the left dorsolateral prefrontal cortex of aMCI patients recruited from a community near the Third Hospital Affiliated to Sun Yat-sen University,China.Twenty-four patients with aMCI were randomly assigned to receive true rTMS(treatment group,n=12,6 men and 6 women;age 65.08±4.89 years)or sham stimulation(sham group,n=12,5 men and 7 women;age 64.67±4.77 years).rTMS parameters included a stimulation frequency of 10 Hz,stimulation duration of 2 seconds,stimulation interval of 8 seconds,20 repetitions at 80%of the motor threshold,and 400 pulses per session.rTMS/sham stimulation was performed five times per week over a period of 4 consecutive weeks.Our results showed that compared with baseline,Montreal Cognitive Assessment scores were significantly increased and the value of the amplitude of low-frequency fluctuation(ALFF)was significantly increased at the end of treatment and 1 month after treatment.Compared with the sham group,the ALFF values in the right inferior frontal gyrus,triangular part of the inferior frontal gyrus,right precuneus,left angular gyrus,and right supramarginal gyrus were significantly increased,and the ALFF values in the right superior frontal gyrus were significantly decreased in the treatment group.These findings suggest that high-frequency rTMS can effectively improve cognitive function in aMCI patients and alter spontaneous brain activity in cognitive-related brain areas.This study was approved by the Ethics Committee of Shenzhen Baoan Hospital of Southern Medical University,China(approval No.BYL20190901)on September 3,2019 and registered in the Chinese Clinical Trials Registry(registration No.ChiCTR1900028180)on December 14,2019.Li-Qiong Yuan Qing Zeng Dan Wang Xiu-Yun Wen Yu Shi Fen Zhu Shang-Jie Chen Guo-Zhi Huang 2021Neural Regeneration Research2021,16,4:6
14Comparison of Histological, Microbiological, and Molecular Methods in Diagnosis of Patients with TBLN Having Different Anti-TB Treatment Background显示文摘Objective The influence of anti-tuberculosis(TB) treatment history on tuberculous lymphadenitis(TBLN) diagnosis is unclear. Therefore, this study aims to evaluate the diagnostic methods, including histology, microbiology, and molecular tests, used for TBLN. Methods In this study, suspected patients with TBLN and having different anti-TB treatment background were enrolled. All the samples were tested simultaneously by histology, Ziehl-Neelsen(ZN) staining, mycobacterial culture(culture), Xpert MTB/RIF(xpert), real-time PCR, and high-resolution melting curve PCR(HRM). Thereafter, the performance of these methods on samples with different anti-TB treatment background was assessed. Results In our study, 89 patients were prospectively included 82 patients with TBLN and 7 with other diseases. The overall sensitivities of Xpert, real-time PCR, histology, ZN staining, and culture were 86.6%, 69.5%, 58.5%, 43.9%, and 22.0%, respectively. The anti-TB treatment history revealed dramatic influences on the sensitivity of culture(P < 0.0001). In fact, the treatment that lasted over 3 months also influenced the sensitivity of Xpert(P < 0.05). However, the treatment history did not affect the performance of remaining tests(P > 0.05). For rifampicin drug susceptibility test(DST), the anti-TB treatment showed only significant influence on the success rate of culture DST(P = 0.001), but not on those of Xpert and HRM tests(P > 0.05). 18Transmissive-detected laser speckle contrast imaging for blood flow monitoring in thick tissue: from Monte Carlo simulation to experimental demonstration显示文摘Laser speckle contrast imaging(LSCI)is a powerful tool to monitor blood flow distribution and has been widely used in studies of microcirculation,both for animal and clinical applications.Conventionally,LSCI usually works on reflectivedetected mode.However,it could provide promising temporal and spatial resolution for in vivo applications only with the assistance of various tissue windows,otherwise,the overlarge superficial static speckle would extremely limit its contrast and resolution.Here,we systematically investigated the capability of transmissive-detected LSCI(TR-LSCI)for blood flow monitoring in thick tissue.Using Monte Carlo simulation,we theoretically compared the performance of transmissive and reflective detection.It was found that the reflective-detected mode was better when the target layer was at the very surface,but the imaging quality would rapidly decrease with imaging depth,while the transmissivedetected mode could obtain a much stronger signal-to-background ratio(SBR)for thick tissue.We further proved by tissue phantom,animal,and human experiments that in a certain thickness of tissue,TR-LSCI showed remarkably better performance for thick-tissue imaging,and the imaging quality would be further improved if the use of longer wavelengths of near-infrared light.Therefore,both theoretical and experimental results demonstrate that TR-LSCI is capable of obtaining thick-tissue blood flow information and holds great potential in the field of microcirculation research.Dong-Yu Li Qing Xia Ting-Ting Yu Jing-Tan Zhu Dan Zhu 2021Light(Science & Applications)2021,10,12:5
19Least Squares Evaluations for Form and Profile Errors of Ellipse Using Coordinate Data显示文摘To improve the measurement and evaluation of form error of an elliptic section, an evaluation method based on least squares fitting is investigated to analyze the form and profile errors of an ellipse using coordinate data. Two error indicators for defining ellipticity are discussed, namely the form error and the profile error, and the difference between both is considered as the main parameter for evaluating machining quality of surface and profile. Because the form error and the profile error rely on different evaluation benchmarks, the major axis and the foci rather than the centre of an ellipse are used as the evaluation benchmarks and can accurately evaluate a tolerance range with the separated form error and profile error of workpiece. Additionally, an evaluation program based on the LS model is developed to extract the form error and the profile error of the elliptic section, which is well suited for separating the two errors by a standard program. Finally, the evaluation method about the form and profile errors of the ellipse is applied to the measurement of skirt line of the piston, and results indicate the effectiveness of the evaluation. 20Tau-Induced Ca^(2+)/Calmodulin-Dependent Protein Kinase-IV Activation Aggravates Nuclear Tau Hyperphosphorylation显示文摘Hyperphosphorylated tau is the major protein component of neurofibrillary tangles in the brains of patients with Alzheimer's disease(AD). However, the mechanism underlying tau hyperphosphorylation is not fully understood. Here, we demonstrated that exogenously expressed wild-type human tau40 was detectable in the phosphorylated form at multiple AD-associated sites in cytoplasmic and nuclear fractions from HEK293 cells.Among these sites, tau phosphorylated at Thr205 and Ser214 was almost exclusively found in the nuclear fraction at the conditions used in the present study. With the intracellular tau accumulation, the Ca^(2+)concentration was significantly increased in both cytoplasmic and nuclear fractions. Further studies using site-specific mutagenesis and pharmacological treatment demonstrated that phosphorylation of tau at Thr205 increased nuclear Ca^(2+)concentration with a simultaneous increase in the phosphorylation of Ca^(2+)/calmodulin-dependent protein kinase IV(Ca MKIV) at Ser196. On the other hand, phosphorylation of tau at Ser214 did not significantly change the nuclear Ca^(2+)/Ca MKIV signaling. Finally, expressing calmodulin-binding protein-4 that disrupts formation ofthe Ca^(2+)/calmodulin complex abolished the okadaic acidinduced tau hyperphosphorylation in the nuclear fraction.We conclude that the intracellular accumulation of phosphorylated tau, as detected in the brains of AD patients, can trigger nuclear Ca^(2+)/Ca MKIV signaling, which in turn aggravates tau hyperphosphorylation.
