1On the origin and continuing evolution of SARS-CoV-2显示文摘The SARS-Co V-2 epidemic started in late December 2019 in Wuhan, China, and has since impacted a large portion of China and raised major global concern. Herein, we investigated the extent of molecular divergence between SARS-CoV-2 and other related coronaviruses. Although we found only 4% variability in genomic nucleotides between SARS-CoV-2 and a bat SARS-related coronavirus(SARSr-CoV;Ra TG13), the difference at neutral sites was 17%, suggesting the divergence between the two viruses is much larger than previously estimated. Our results suggest that the development of new variations in functional sites in the receptor-binding domain(RBD) of the spike seen in SARS-Co V-2 and viruses from pangolin SARSr-Co Vs are likely caused by natural selection besides recombination. Population genetic analyses of 103 SARS-CoV-2 genomes indicated that these viruses had two major lineages(designated L and S), that are well defined by two different SNPs that show nearly complete linkage across the viral strains sequenced to date. We found that L lineage was more prevalent than the S lineage within the limited patient samples we examined. 2Phytoplankton community structure and environmental parameters in aquaculture areas of Daya Bay,South China Sea显示文摘Environmental characteristics and phytoplankton community structure were investigated in two aquaculture areas in Dapeng Cove of Daya Bay,South China Sea,between April 2005 and June 2006.Phytoplankton abundance ranged between 5.0 and 8877.5 cells/mL,with an average of 751.8 cells/mL.The seasonal cycle of phytoplankton were demonstrated by frequent oscillations,with recurrent high abundances from late spring to autumn and a peak stage in late winter.Diatoms were the predominant phytoplankton group,accounting for 93.21%of the total abundance.The next most abundant group was the dinoflagellates,which made up only 1.24%of total abundance. High concentrations of Alexandrium tamarense(Lebour) Balech with a maximum of 603.0 cells/mL were firstly recorded in this area known for high rates of paralytic shellfish poisoning(PSP) contamination.Temperatures and salinities were within the suitable values for the growth of phytoplankton,and were important in phytoplankton seasonal fluctuations.The operation of the Daya Bay Nuclear Power Station(DNPS) exerts influences on the phytoplankton community and resulted in the high abundances of toxic dinoflagellate species during the winter months.Dissolved inorganic nitrogen(DIN) and dissolved silicate(DSi) were suffcient,and rarely limited for the growth of phytoplankton.Dissolved inorganic phosphorus(DIP) was the most necessary element for phytoplankton growth. 7Stability of roof structure and its control in steeply inclined coal seams显示文摘To improve the effectiveness of control of surrounding rock and the stability of supports on longwall topcoal caving faces in steeply inclined coal seams, the stability of the roof structure and hydraulic supports was studied with physical simulation and theoretical analysis. The results show that roof strata in the vicinity of the tail gate subside extensively with small cutting height, while roof subsidence near the main gate is relatively assuasive. With increase of the mining space, the caving angle of the roof strata above the main gate increases. The characteristics of the vertical and horizontal displacement of the roof strata demonstrate that caved blocks rotate around the lower hinged point of the roof structure, which may lead to sliding instability. Large dip angle of the coal seam makes sliding instability of the roof structure easier.A three-hinged arch can be easily formed above both the tail and main gates in steeply inclined coal seams. 17Improving Plant Genome Editing with High-Fidelity xCas9 and Non-canonical PAM-Targeting Cas9-NG显示文摘Two recently engineered SpCas9 variants, namely xCas9 and Cas9-NG, show promising potential in improving targeting specificity and broadening the targeting range. In this study, we evaluated these Cas9 variants in the model and crop plant, rice. We first tested xCas9-3.7, the most effective xCas9 variant in mammalian cells, for targeted mutagenesis at 16 possible NGN PAM (protospacer adjacent motif) combinations in duplicates. xCas9 exhibited nearly equivalent editing efficiency to wild-type Cas9 (Cas9-WT) at most canonical NGG PAM sites tested, whereas it showed limited activity at non-canonical NGH (H = A, C, T) PAM sites. High editing efficiency of xCas9 at NGG PAMs was further demonstrated with C to T base editing by both rAPOBECI and PmCDAI cytidine deaminases. With mismatched sgRNAs, we found that xCas9 had improved targeting specificity over the Cas9-WT. Furthermore, we tested two Cas9-NG variants, Cas9-NGv1 and Cas9-NG, for targeting NGN PAMs. Both Cas9-NG variants showed higher editing efficiency at most non-canonical NG PAM sites tested, and enabled much more efficient editing than xCas9 at AT-rich PAM sites such as GAT, GAA, and CAA. Nevertheless, we found that Cas9-NG variants showed significant reduced activity at the canonical NGG PAM sites. In stable transgenic rice lines, we demonstrated that Cas9-NG had much higher editing efficiency than Cas9-NGv1 and xCas9 at NG PAM sites. To expand the base-editing scope, we developed an efficient C to T base-editing system by making fusion of Cas9-NG nickase (D10A version), PmCDAI, and UGI. 18Integrated(de)multiplexer for orbital angular momentum fiber communication显示文摘The quickly increasing data transfer load requires an urgent revolution in current optical communication. Orbital angular momentum(OAM) multiplexing is a potential candidate with its ability to considerably enhance the capacity of communication. However, the lack of a compact, efficient, and integrated OAM(de)multiplexer prevents it from being widely applied. By attaching vortex gratings onto the facets of a few-mode fiber, we demonstrate an integrated fiber-based OAM(de)multiplexer. A vortex grating fabricated on the fiber facet enables the direct multiplexing of OAM states at one port and the demultiplexing of OAM states at the other port. The measured bit error rate of the carrier signal after propagating through a 5-km few-mode fiber confirms the validity and effectiveness of the proposed approach. 19Controllable Synthesis of Fluorescent Carbon Dots and Their Detection Application as Nanoprobes显示文摘Carbon dots(CDs), as a new member of carbon nanomaterial family, have aroused great interest since their discovery in 2004. Because of their outstanding water solubility, high sensitivity and selectivity to target analytes, low toxicity, favorable biocompatibility, and excellent photostability, researchers from diverse disciplines have come together to further develop the fundamental properties of CDs. Many methods for the production of CDs have been reported, therein, hydrothermal and solvothermal technology needs simple equipments, and microwave synthesis needs less reaction time, hence these methods become current common synthesis methods, in which many precursors have been applied to produce CDs. Due to their excellent fluorescence, CDs have made impressive strides in sensitivity and selectivity to a diverse array of salt ions,organic/biological molecules and target gases. The development of CDs as nanoprobes is still in its infancy, but continued progress may lead to their integration into environmental and biological applications. 20Chalcogenide glass photonic integration for improved 2μm optical interconnection显示文摘In this work,on-chip chalcogenide glass photonic integrations with several fundamental photonic building blocks are designed and fabricated based on the As2 S3 platform for improved 2μm optical interconnection,achieving a broadened wavelength bandwidth and improved fabrication tolerance.A 600 nm thick As2 S3 strip waveguide has low propagation loss of 1.447 dB/cm at 2μm.Broadband vertical coupling is realized by a grating coupler with 4.3 dB coupling loss.A Bragg grating filter,power splitter,Mach–Zander interferometer,and mode converter for on-chip mode division multiplexing(MDM)are also reported at 2μm with reliable performances.Finally,a record high MDM optical interconnection capacity of 3×80 Gbps at 2μm is experimentally demonstrated based on the proposed As2 S3 chip,drawing promising prospects for future photonic integration and high-speed interconnection at the 2μm waveband.WEIHONG SHEN PINGYANG ZENG ZELIN YANG DI XIA JIANGBING DU BIN ZHANG KE XU ZUYUAN HE AND ZHAOHUI LI 2020Photonics Research2020,8,9:10
