| 1 | Development of cell-based pseudovirus entry assay to identify potential viral entry inhibitors and neutralizing antibodies against SARS-CoV-2显示文摘Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)is the causative virus of the coronavirus disease 2019(COVID-19)pandemic.To establish a safe and convenient assay system for studying entry inhibitors and neutralizing antibodies against SARS-CoV-2,we constructed a codon-optimized,full-length C-terminal mutant spike(S)gene of SARS-CoV-2.We generated a luciferase(Luc)-expressing pseudovirus containing the wild-type or mutant S protein of SARS-CoV-2 in the envelope-defective HIV-1 backbone.The key parameters for this pseudovirus-based assay,including the S mutants and virus incubation time,were optimized.This pseudovirus contains a Luc reporter gene that enabled us to easily quantify virus entry into angiotensin-converting enzyme 2(ACE2)-expressing 293T cells.Cathepsin(Cat)B/L inhibitor E64d could significantly block SARS-CoV-2 pseudovirus infection in 293T-ACE2 cells.Furthermore,the SARS-CoV-2 spike pseudotyped virus could be neutralized by sera from convalescent COVID-19 patients or recombinant ACE2 with the fused Fc region of human IgG1.Thus,we developed a pseudovirus-based assay for SARS-CoV-2,which will be valuable for evaluating viral entry inhibitors and neutralizing antibodies against this highly pathogenic virus. | Jie Hu Qingzhu Gao Changlong He Ailong Huang Ni Tang Kai Wang | 2020 | Genes & Diseases2020,7,4: | 15 |
| 2 | Grassland degradation in Northern Tibet based on remote sensing data显示文摘This study selected vegetation cover as the main evaluation index, calculated the grassland degradation index (GDI) and established the remote sensing monitoring and evaluation system for grassland degradation in Northern Tibet, according to the National Standard (GB19377-2003), based on the remote sensing data such as NDVI data derived from NOAA/AVHRR with a spatial resolution of 8 km of 1981-2000, from SPOT/VGT with a spatial resolution of 1 km of 2001 and from MODIS with a spatial resolution of 0.25 km of 2002-2004 respectively in this area, in combination with the actual condition of grassland degradation. The grassland degradation processes and their responses to climate change during 1981-2004 were discussed and analyzed in this paper. The result indicated that grassland degradation in Northern Tibet is very serious, and the mean value of GDI in recent 20 years is 2.54 which belongs to the serious degradation grade. From 1981 to 2004, the GDI fluctuated distinctly with great interannual variations in the proportion of degradation degree and GDI but the general tendency turned to severe-grade during this period with the grassland degradation grade changed from light degraded to serious degraded in Northern Tibet. The extremely serious degraded and serious degraded grassland occupied 1.7% and 8.0% of the study area, the moderate and light degraded grassland accounted for 13.2% and 27.9% respectively, and un-degraded grassland occupied 49.2% of the total grassland area in 2004. The grassland degradation was serious, especially in the conjunctive area of Naqu, Biru and Jiali counties, the headstream of the Yangtze River lying in the Galadandong snow mountain and glaciers, the area along the Qinghai-Tibet highway and railway, and areas around the Tanggula and Nianqingtanggula snow mountains and glaciers. | 5 | Overexpression of the neuroglobin gene delivered by ultrasound-targeted microbubble destruction protects SH-SY5Y cells against cobalt chloride induced hypoxia显示文摘In this study, we examined the effects of neuroglobin gene (Ngb) transfection into SH-SY5Y cells, using ultrasound-targeted microbubble destruction (UTMD), on cobalt chloride-induced hypoxia. With an ultrasound intensity of 0.8 W/cm2, a 60-second exposure duration, 50% duty cycle, and 20% microbubble concentration, pAcGFP1-C1-Ngb-transfected cells exhibited the highest cell viability and transfection efficiency. The efficiency of plasmid delivery was significantly higher with UTMD than transfection with plasmid alone, transfection with plasmid using microbubbles, or transfection of plasmid by ultrasound. In addition, during cobalt chloride-induced hypoxia, caspase-3 activity in pAcGFP1-C1-Ngb-transfected cells was significantly lower than in untransfected cells. Ngb protein and mRNA expression were significantly higher in cells transfected by UTMD than in cells transfected with the other methods. The mechanism for the atomic H initiated reactions with complete series of nineteen fluorophenol congeners was studies using the density functional theory. At the MPWB1K/6-31+G(d,p) level, the geometries and frequencies of reactants, transition states, and products were obtained, and the accurate energetic values were acquired at the MPWB1K/6-311+G(3df,2p) level. The rate constants were evaluated by the canonical variational transition-state theory with the small curvature tunneling contribution over a wide temperature range of 600–1000 K. The study shows that the intramolecular hydrogen-bond in the ortho-substituted FPs as well as the inductive efect of the electron-withdrawing fluorine and steric repulsion of multiple substitutions may ultimately be responsible for the relative strength of the O–H bonds in FPs. Several energetically favorable reaction pathways were revealed. The formation mechanisms of secondary pollutants were presented and discussed. The rate constants were deduced over the temperature range of 273–333 K using canonical variational transition-state(CVT) theory with the small curvature tunneling(SCT) method. Our study shows that H abstraction from the alkyl group and NO3 addition to the C3 atom of the benzene ring are the dominant reaction pathways. The rate-temperature formula of the overall rate constants is k(T)(DDT+NO3) =(7.21 × 10.15)exp(–153.81/T) cm3/(mol·sec) over the possible atmospheric temperature range of 273–333 K.
