1Particle size distributions, PM2.5 concentrations and water- soluble inorganic ions in different public indoor environments: a case study in Jinan, China显示文摘In this study, we collected particles with aerodynamic diameter≤2.5μm (PM2.5) from three different public indoor places (a supermarket, a commercial office, and a university dining hall) in Jinan, a medium-sized city located in northern China. Water- soluble inorganic ions of PM2.5 and particle size distributions were also measured. Both indoor and outdoor PM2.5 levels (102.3-143.8 μg·m- 3 and 160.2-301.3 μg·m-3, respectively) were substantially higher than the value recommended by the World Health Organization (25 I.μg·m-3), and outdoor sources were found to be the major contributors to indoor pollutants. Diurnal particle number size distributions were different, while the maximum volume concentrations all appeared to be approximately 300nm in the three indoor locations. Concentrations of indoor and outdoor PM2.5 were shown to exhibit the same variation trends for the supermarket and dining hall. For the office, PM2.5 concentrations during nighttime were observed to decrease sharply. Among others, SO4^2-, NH4+ and NO3- were found to be the dominant water-soluble ions of both indoor and outdoor particles. Concentrations of NO3- in the supermarket and office during the daytime were observed to decrease sharply, which might be attributed to the fact that the indoor temperature was much higher than the outdoor temperature. In addition, domestic activities such as cleaning, water usage, cooking, and smoking also played roles in degraded indoor air quality. 21,25-Dihydroxyvitamin D_3 modulates calcium transport in goat mammary epithelial cells in a dose- and energydependent manner显示文摘Background: Calcium is a vital mineral and an indispensable component of milk for ruminants. The regulation of transcellular calcium transport by 1,25-dihydroxyvitamin D3(1,25-(OH)2D3, the active form of vitamin D) has been confirmed in humans and rodents, and regulators, including vitamin D receptor(VDR), calcium binding protein D9k(calbindin-D9k), plasma membrane Ca2+-ATPase 1b(PMCA1b), PMAC2 b and Orai1, are involved in this process.However, it is still unclear whether 1,25-(OH)2D3could stimulate calcium transport in the ruminant mammary gland.The present trials were conducted to study the effect of 1,25-(OH)2D3supplementation and energy availability on the expression of genes and proteins related to calcium secretion in goat mammary epithelial cells.Methods: An in vitro culture method for goat secreting mammary epithelial cells was successfully established. The cells were treated with different doses of 1,25-(OH)2D3(0, 0.1, 1.0, 10.0 and 100.0 nmol/L) for calcium transport research, followed by a 3-bromopyruvate(3-Br PA, an inhibitor of glucose metabolism) treatment to determine its dependence on glucose availability. Cell proliferation ratios, glucose consumption and enzyme activities were measured with commercial kits, and real-time quantitative polymerase chain reaction(RT-q PCR), and western blots were used to determine the expression of genes and proteins associated with mammary calcium transport in dairy goats, respectively.Results: 1,25-(OH)2D3promoted cell proliferation and the expression of genes involved in calcium transport in a dose-dependent manner when the concentration did not exceed 10.0 nmol/L. In addition, 100.0 nmol/L 1,25-(OH)2D3inhibited cell proliferation and the expression of associated genes compared with the 10.0 nmol/L treatment.The inhibition of hexokinase 2(HK2), a rate-limiting enzyme in glucose metabolism, decreased the expression of PMCA1 b and PMCA2 b at the m RNA and protein levels as well as the transcription of Orai1, indicating that glucose availability was required for goat mammary calcium transport. The optimal concentration of 1,25-(OH)2D3that facilitated calcium transport in this study was 10.0 nmol/L.Conclusions: Supplementation with 1,25-(OH)2D3influenced cell proliferation and regulated the expression of calcium transport modulators in a dose- and energy-dependent manner, thereby highlighting the role of 1,25-(OH)2D3as an efficacious regulatory agent that produces calcium-enriched milk in ruminants when a suitable energy status was guaranteed.Feifei Sun Yangchun Cao Chao Yu Xiaoshi Wei Junhu Yao 2016Journal of Animal Science and Biotechnology2016,7,4:2
3Voice conversion using structured Gaussian mixture model in cepstrum eigenspace显示文摘A new methodology of voice conversion in cepstrum eigenspace based on structured Gaussian mixture model is proposed for non-parallel corpora without joint training.For each speaker,the cepstrum features of speech are extracted,and mapped to the eigenspace which is formed by eigenvectors of its scatter matrix,thereby the Structured Gaussian Mixture Model in the EigenSpace(SGMM-ES)is trained.The source and target speaker's SGMM-ES are matched based on Acoustic Universal Structure(AUS)principle to achieve spectrum transform function.Experimental results show the speaker identification rate of conversion speech achieves95.25%,and the value of average cepstrum distortion is 1.25 which is 0.8%and 7.3%higher than the performance of SGMM method respectively.ABX and MOS evaluations indicate the conversion performance is quite close to the traditional method under the parallel corpora condition.The results show the eigenspace based structured Gaussian mixture model for voice conversion under the non-parallel corpora is effective.LI Yangchun YU Yibiao 2015Chinese Journal of Acoustics2015,34,3:2

6Yak rumen microbiome elevates fiber degradation ability and alters rumen fermentation pattern to increase feed efficiency显示文摘Rumen microbes play an important role in ruminant energy supply and animal performance.Previous studies showed that yak(Bos grunniens)rumen microbiome and fermentation differ from other ruminants.However,little is understood about the features of the rumen microbiome that make yak adapted to their unique environmental and dietary conditions.This study was to investigate the rumen microbiome and metabolome to understand how yak adapt to the coarse forage and harsh environment in the Qinghai-Tibetan plateau.Nine female Qaidam yellow cattle(Bos taurus),9 dzomo(hybrids of cattle and yak)and 9 female plateau yak(B.grunniens),about 5 to 6 years old,were used in this study.Rumen fermentation parameters,fibrolytic enzyme activities,and rumen metataxonomic were determined.Then 18(6 samples per group)were selected for rumen metagenomic and metabolome analysis.Metataxonomic analysis revealed that the rumen microbiota was significantly different among plateau yak,Qaidam yellow cattle,and dzomo(P<0.05).Metagenomic analysis displayed a larger gene pool encoding a richer repertoire of carbohydrate-active enzymes in the rumen microbiome of plateau yak and dzomo than Qaidam yellow cattle(P<0.05).Some of the genes encoding glycoside hydrolases that mediate the digestion of cellulose and hemicellulose were significantly enriched in the rumen of plateau yak than Qaidam yellow cattle,but glycoside hydrolase 57 that primarily includes amylases was abundant in Qaidam yellow cattle(P<0.05).The rumen fermentation profile differed also,Qaidam yellow cattle having a higher molar proportion of acetate but a lower molar proportion of propionate than dzomo and plateau yak(P<0.05).Based on metabolomic analysis,rumen microbial metabolic pathways and metabolites were different.Differential metabolites are mainly amino acids,carboxylic acids,sugars,and bile acids.Changes in rumen microbial composition could explain the above results.The present study showed that the rumen microbiome of plateau yak helps its host to adapt to the Qinghai-Tibetan plateau.In particular,the plateau yak rumen microbiome has more enzymes genes involved in cellulase and hemicellulase than that of cattle,resulting higher fibrolytic enzyme activities inyak,further providing stronger fiber degradation function.Congcong Zhao Lamei Wang Shanlin Ke Xinhua Chen Akos Kenez Wei Xu Dangdang Wang Fan Zhang Yong Li Zhanhong Cui Yu Qiao Jing Wang Wenjuan Sun Jianhua Zhao Junhu Yao Zhongtang Yu Yangchun Cao 2022Animal Nutrition2022,,4:1

9The relative importance of soil moisture in predicting bacterial wilt disease occurrence显示文摘Soil-borne plant diseases cause major economic losses globally.This is partly because their epidemiology is difficult to predict in agricultural fields,where multiple environmental factors could determine disease outcomes.Here we used a combination of field sampling and direct experimentation to identify key abiotic and biotic soil properties that can predict the occurrence of bacterial wilt caused by pathogenic Ralstonia solanacearum.By analyzing 139 tomato rhizosphere soils samples isolated from six provinces in China,we first show a clear link between soil properties,pathogen density and plant health.Specifically,disease outcomes were positively associated with soil moisture,bacterial abundance and bacterial community composition.Based on soil properties alone,random forest machine learning algorithm could predict disease outcomes correctly in 75%of cases with soil moisture being the most significant predictor.The importance of soil moisture was validated causally in a controlled greenhouse experiment,where the highest disease incidence was observed at 60%of maximum water holding capacity.Together,our results show that local soil properties can predict disease occurrence across a wider agricultural landscape,and that management of soil moisture could potentially offer a straightforward method for reducing crop losses to R.solanacearum.Gaofei Jiang Ningqi Wang Yaoyu Zhang Zhen Wang Yuling Zhang Jiabao Yu Yong Zhang Zhong Wei Yangchun Xu Stefan Geisen Ville-Petri Friman Qirong Shen 2021Soil Ecology Letters2021,3,4:1

12Effect of Rare Earths on Microstructure and Properties of Sn2.0Ag0.7CuRE Solder Alloy for Surface Mount Technology显示文摘A new type of lead-free solder alloy Sn20Ag0.7CuRE was fabricated under vacuum condition. The microstructure and properties of the material, such as tensile strength, elongation, melting range, conductance and spreading area were all investigated. Result shows that when the content of RE≤0.1%(mass fraction), RE distribute uniformly in the solder alloy, and the tensile strength and conductance of Sn20Ag0.7CuRE solder alloy are better than those of traditional Sn37Pb solder. Its elongation and spreading area are almost equal to those of Sn37Pb solder. When the content of RE reaches 0.5%, RE compounds can be easily found around the boundaries of grains and phases, and the tensile strength and elongation and spreading area of Sn2.0Ag0.7CuRE solder alloy all decrease sharply. 14Characteristics and Clinical Implication of UGT1A1 Heterozygous Mutation in Tumor显示文摘Background:The literature recommends that reduced dosage of CPT-11 should be applied in patients with UGT1 A1 homozygous mutations,but the impact of UGT1 A1 heterozygous mutations on the adverse reactions of CPT-11 is still not fully clear.Methods:A total of 107 patients with UGT1 A1 heterozygous mutation or wild-type,who were treated with CPT-11 from January 2018 to September 2021 in Peking University Third Hospital,were retrospectively enrolled.The adverse reaction spectra of patients with UGT1 A1*6 and UGT1 A1*28 mutations were analyzed.Adverse reactions were evaluated according to National Cancer Institute Common Terminology Criteria for Adverse Events(NCI-CTCAE) 5.0.The efficacy was evaluated according to Response Evaluation Criteria in Solid Tumors(RECIST) 1.1.The genotypes of UGT1 A1*6 and UGT1 A1*28 were detected by digital fluorescence molecular hybridization.Results:There were 43 patients with UGT1 A1*6 heterozygous mutation,26 patients with UGT1 A1*28 heterozygous mutation,8 patients with UGT1 A1*6 and UGT1 A1*28 double heterozygous mutations,61 patients with heterozygous mutation at any gene locus of UGT1 A1*6 and UGT1 A1*28.Logistic regression analysis showed that the presence or absence of vomiting(P=0.013) and mucositis(P=0.005) was significantly correlated with heterozygous mutation of UGT1 A1*28,and the severity of vomiting(P<0.001) and neutropenia(P=0.021) were significantly correlated with heterozygous mutation of UGT1 A1*6.In colorectal cancer,UGT1 A1*6 was significantly correlated to diarrhea(P=0.005),and the other adverse reactions spectrum was similar to that of the whole patient cohort,and efficacy and prognosis were similar between patients with different genotypes and patients treated with reduced CPT-11 dosage or not.Conclusion:In clinical use,heterozygous mutations of UGT1 A1*6 and UGT1 A1*28 are related to the risk and severity of vomiting,diarrhea,neutropenia and mucositis in patients with Pan-tumor and colorectal cancer post CPT-11 therpy.In colorectal cancer,UGT1 A1*6 is significantly related to diarrhea post CPT-11 use,efficacy and prognosis is not affected by various genotypes or CPT-11 dosage reduction.Qian LI Tao SUN Hua ZHANG Wei LIU Yu XIAO Hongqi SUN Wencheng YIN Yanhong YAO Yangchun GU Yan'e LIU Fumei YI Qiqi WANG Jinyu YU Baoshan CAO Li LIANG 2022中国肺癌杂志2022,25,3:0
15Rumen bacterial cluster identification and its influence on rumen metabolites and growth performance of young goats显示文摘Enterotypes,which are defined as bacterial clusters in the gut microbiome,have been found to have a close relationship to host metabolism and health.However,this concept has never been used in the rumen,and little is known about the complex biological relationships between ruminants and their rumen bacterial clusters.In this study,we used young goats(n=99)as a model,fed them the same diet,and analyzed their rumen microbiome and corresponding bacterial clusters.The relationships between the bacterial clusters and rumen fermentation and growth performance in the goats were further investigated.Two bacterial clusters were identified in all goats:the P-cluster(dominated by genus Prevotella,n=38)and R-cluster(dominated by Ruminococcus,n=61).Compared with P-cluster goats,Rcluster goats had greater growth rates,concentrations of propionate,butyrate,and 18 free amino acids)and proportion of unsaturated fatty acids,but lower acetate molar percentage,acetate to propionate ratio,and several odd and branched chain and saturated fatty acids in rumen fluid(P<0.05).Several members of Firmicutes,including Ruminococcus,Oscillospiraceae NK4A214 group,and Christensenellaceae R-7 group were significantly higher in the R-cluster,whereas Prevotellaceae members,such as Prevotella and Prevotellaceae UCG-003,were significantly higher in P-cluster(P<0.01).Co-occurrence networks showed that R-cluster enriched bacteria had significant negative correlations with P-cluster enriched bacteria(P<0.05).Moreover,we found the concentrations of propionate,butyrate and free amino acids,and the proportions of unsaturated fatty acids were positively correlated with R-cluster enriched bacteria(P<0.05).The concentrations of acetate,acetate to propionate ratio,and the proportion of odd and branched chain and saturated fatty acids were positively correlated with P-cluster enriched bacteria(P<0.05).Overall,our results indicated that rumen bacterial clusters can influence rumen fermentation and growth performance of young goats,which may shed light on modulating the rumen microbiome in early life to improve the growth performance of ruminant animals.Dangdang Wang Guangfu Tang Yannan Wang Junjian Yu Luyu Chen Jie Chen Yanbo Wu Yuanjie Zhang Yangchun Cao Junhu Yao 2023Animal Nutrition2023,,4:0
