1Preparation and characteristics of DNA-nanoparticles targeting to hepatocarcinoma cells显示文摘AIM:To prepare thymidine kinase gene (TK gene) nanopartides and to investigate the expression of TK gene.METHODS: Poly(D,L-lactic-co-glycolic acid) (PLGA), a biodegradable and biocompatible polymer, was used to prepare recombinant plasmid p^EGFP-AFP nanoparticles by a double-emulsion evaporation technique. Characteristics of the nanoparticles were investigated in this study, including morphology, entrapment efficiency, and tissue distribution.The expression of TK gene was also investigated by MTT assay, by which the viable cells were determined alter the addition of ganciclovir (GCV).The enhanced green fluorescent protein (EGFP) expression in human hepatocellular carcinoma SMMC-7721 cells and normal parenchymal Chang liver cellswere assessed by flow cytometry.RESULTS: The prepared plasmid-nanoparticles had regular spherical surface and narrow particle size span with a mean diameter of 72±12nm.The mean entrapment efficiency was 91.25%. A total of 80.14% DNA was found to be localized in the livers after 1-h injection with ^32P-DNA-PLGA nanoparticles in mouse caudal vein. The expression of DNA encapsulated in nanopartides was much higher than that in naked DNA, and human hepatocellular carcinoma SMMC-7721 cells were more sensitive to GCV than human normal parenchymal Chang liver cells.CONCLUSION:The enhanced transfection efficiency and stronger ability to protect plasmid DNA from being degraded by nucleases are due to nanoparticles encapsulation.QinHe JiLiu XunSun Zhi-RongZhang 2004World Journal of Gastroenterology2004,10,5:4

3Metal-Organic Framework-based Wood Aerogel for Effective Removal of Micro/Nano Plastics显示文摘The large amount of microplastic pollution in the aquatic environment has been regarded as a major challenge facing the world. Although some traditional water treatment processes can effectively remove microplastics in the millimeter range, research is still needed to solve the problem of removing smaller microplastics. Here, we made a durable zinc metal-organic framework-based composite material ZIF-8@Aerogel by in situ growing ZIF-8 on wood aerogel fibers, which can successfully remove microplastics in simulated water and seawater. The removal efficiencies on micro/nano plastics including poly(1,1-difluoroethylene)(60-110 nm) and polystyrene(90-140 nm) reached 91.4% and 85.8%, respectively. 4Kinectin 1 promotes the growth of triple-negative breast cancer via directly co-activating NF-kappaB/p65 and enhancing its transcriptional activity显示文摘Triple-negative breast cancer(TNBC)is the most challenging subtype of breast cancer.Various endeavor has been made to explore the molecular biology basis of TNBC.Herein,we reported a novel function of factor Kinectin 1(KTN1)as a carcinogenic promoter in TNBC.KTN1 expression in TNBC was increased compared with adjacent tissues or luminal or Her2 subtypes of breast cancer,and TNBC patients with high KTN1 expression have poor prognosis.In functional studies,knockdown of KTN1 inhibited the proliferation and invasiveness of TNBC both in vitro and in vivo,while overexpression of KTN1 promoted cancer cell proliferation and invasiveness.RNA-seq analysis revealed that the interaction of cytokine-cytokine receptor,particularly CXCL8 gene,was upregulated by KTN1,which was supported by the further experiments.CXCL8 depletion inhibited the tumorigenesis and progression of TNBC.Additionally,rescue experiments validated that KTN1-mediated cell growth acceleration in TNBC was dependent on CXCL8 both in vitro and in vivo.Furthermore,it was found that KTN1 enhanced the phosphorylation of NF-κB/p65 protein at Ser536 site,and specifically bound to NF-κB/p65 protein in the nucleus and cytoplasm of cells.Moreover,the transcription of CXCL8 gene was directly upregulated by the complex of KTN1 and NF-κB/p65 protein.Taken together,our results elucidated a novel mechanism of KTN1 gene in TNBC tumorigenesis and progression.KTN1 may be a potential molecular target for the development of TNBC treatment.Lin Gao Shanze Chen Malin Hong Wenbin Zhou Bilan Wang Junying Qiu Jinquan Xia Pan Zhao Li Fu Jigang Wang Yong Dai Ni Xie Qinhe Yang Hsien-Da Huang Xiang Gao Chang Zou 2021Signal Transduction and Targeted Therapy2021,6,8:2

7Discussion on Petroleum Migration in Bamianhe Oilfield of Dongying Depression, East China显示文摘Petroleum migration process of Bamianhe oilfield of Dongying depression, East China is evaluatedon the basis of our detailed work on geochemistry of the crude oil and the source. Molecular tracers, especially nitrogen compounds, are employed in assessing style of oil migration and accumulation. Additionally, absolute quantitative results of heteroaromatics including dibenzofuran, dibenzothiophene and fluorene with similar basic frames to pyrrolic nitrogenic compounds are also used to evaluate the quantity of petroleum migration. According to apparent migration effects of the pyrrolic nitrogen compounds, it is indicated that most of Bamianhe oil was derived from potential source kitchen-Niu-zhuang sag, migrated and accumulated at Bamianhe fault belt through selective paths according to a main filling point displayed. Generally, upward migration trend from the main point along the south slope is noted. There are observable migration effects from Guangli subsag to the north Bamianhe and Yangjiaogou oilfields showing Guangli subsag was a secondary kitchen accounting for the oils in the north portion. Significant vertical migration effects shown in several cross sections demonstrate that faults well developed in the area play an important role in oil migration and accumulation. Unformality, sandstone reservoirs distributed locally also suggest important fairways in connecting sources with traps. The tectonic and a combination of tectonic and lithological mechanism are identified as the critical mechanisms in entrapping hydrocarbons. Migration trend suggested by nitrogenic compounds agrees well with analysis of lateral compositional variety and thermal maturity gradient. This study also confirms well with our previous studies. It is the location of source rocks, tectonic and stratigraphic characteristics that have a comprehensive control on the model of hydrocarbon migration and accumulation in Bamianhe oilfield. 11Clinical Pathological Analysis of Synovial Sarcoma显示文摘OBJECTIVE To investigate the clinical diagnosis and differential diagno- sis of synovial sarcoma (SS). METHODS A total of 41 paraffin-embedded synovial sarcoma samples were examined by H&E staining, immunohistochemistry staining and the re- verse transcriptase polymerase chain reaction (RT-PCR), in order to provide a scientific bases for diagnosis and differential diagnosis. RESULTS Twelve cases were a biphasic type, 22 cases were a mono- phasic fibrous type, and 7 cases were a poorly differentiated type. Thirty-six cases were both CK (and/or EMA) and Vim positive. Five cases were only Vim positive. A SYT-SSX fusion gene was detected in 18 cases by RT-PCR.
