1Exhaustion and senescence: two crucial dysfunctional states of T cells in the tumor microenvironment显示文摘The failure of a massive influx of tumor-infiltrating T lymphocytes to eradicate tumor cells in the tumor microenvironment is mainly due to the dysfunction of T cells hyporesponsive to tumors.T-cell exhaustion and senescence induced by malignant tumors are two important dysfunctional states that coexist in cancer patients,hindering effective antitumor immunity and immunotherapy and sustaining the suppressive tumor microenvironment.Although exhausted and senescent T cells share a similar dysfunctional role in antitumor immunity,they are distinctly different in terms of generation,development,and metabolic and molecular regulation during tumor progression.Here,we discuss the unique phenotypic and functional characteristics of these two types of dysfunctional T cells and their roles in tumor development and progression.In addition,we further discuss the potential molecular and metabolic signaling pathways responsible for the control of T-cell exhaustion and senescence in the suppressive tumor microenvironment.Understanding these critical and fundamental features should facilitate rethinking the unresponsiveness to current immunotherapies in clinical patients and lead to further development of novel and effective strategies that target different types of dysfunctional T cells to enhance cancer immunotherapy.Yangjing Zhao Qixiang Shao Guangyong Peng 2020Cellular & Molecular Immunology2020,17,1:11
2Intestinal dysbiosis exacerbates the pathogenesis of psoriasis-like phenotype through changes in fatty acid metabolism显示文摘The intestinal microbiota has been associated with host immunity as well as psoriasis;however,the mechanism of intestinal microbiota regulating psoriasis needs to be demonstrated systematically.Here,we sought to examine its role and mechanism of action in the pathogenesis of psoriasis.We found that the severity of psoriasis-like skin phenotype was accompanied by changes in the composition of the intestinal microbiota.We performed co-housing and fecal microbial transplantation(FMT)experiments using the K14-VEGF transgenic mouse model of psoriasis and demonstrated that the transfer of intestinal microbiota from mice with severe psoriasis-like skin phenotype exacerbated psoriasiform skin inflammation in mice with mild symptoms,including increasing the infiltration and differentiation of Th17,and increased the abundance of Prevotella,while decreasing that of Parabacteroides distasonis,in the colon.These alterations affected fatty acid metabolism,increasing the abundance of oleic and stearic acids.Meanwhile,gentamicin treatment significantly reduced the abundance of Prevotella and alleviated the psoriasis-like symptoms in both K14-VEGF mice and imiquimod(IMQ)-induced psoriasis-like mice.Indeed,administration of oleic and stearic acids exacerbated psoriasis-like symptoms and increased Th17 and monocyte-derived dendritic cell infiltration in the skin lesion areas in vivo,as well as increased the secretion of IL-23 by stimulating DCs in vitro.At last,we found that,treatment of PDE-4 inhibitor alleviated psoriasis-like phenotype of K14-VEGF mice accompanied by the recovery of intestinal microbiota,including the decrease of Prevotella and increase of Parabacteroides distasonis.Overall,our findings reveal that the intestinal microbiota modulates host metabolism and psoriasis-like skin inflammation in mice,suggesting a new target for the clinical diagnosis and treatment of psoriasis.Qixiang Zhao Jiadong Yu Hong Zhou Xiaoyan Wang Chen Zhang Jing Hu Yawen Hu Huaping Zheng Fanlian Zeng Chengcheng Yue Linna Gu Zhen Wang Fulei Zhao Pei Zhou Haozhou Zhang Nongyu Huang Wenling Wu Yifan Zhou Jiong Li 2023Signal Transduction and Targeted Therapy2023,8,2:2

11Novel numerical model to simulate water seepage through segmental gasketed joints of underwater shield tunnels considering the superimposed seepage squeezing effect显示文摘The water leakage through segmental joint gaskets has become a major concern that adversely affects the normal serviceability of underwater shield tunnels throughout the construction and operational periods.Therefore,it is of great significance to investigate the sealing performances of the joint gaskets,which directly helps evaluate the waterproof capacity of underwater shield tunnels.To date,the numerical modeling plays an irreplaceable role in the analysis on the waterproof capacity of the joint gaskets.Nevertheless,conventional methods tend to ignore the self-sealing effect induced by the water seepage pressurization,thus failing to reveal the progressive evolution of the water infiltration process through the joint gasket.To remedy this defect,this paper proposed a novel numerical model to simulate the penetration process of the sealing gasket based on the Python language-enabled secondary programming in the ABAQUS software,which could fully consider the superimposed seepage squeezing effect.Based on the proposed model,the waterproof failure process and the dynamic contact stress of the gasket’s water seepage path subject to excessive hydraulic pressure were thoroughly investigated.Moreover,indoor tests on the waterproof capacity of the gasket were also performed to validate the proposed model.It is found that the numerical results from the developed model are consistent with the experimental results.This research will contribute to better understanding of the gaskets’hydraulic penetration process and more accurate prediction of the maximum waterproof capacity in underwater shield tunnels.Qixiang Yan Haojia Zhong Chuan Zhang Zechang Zhao Yanxin Wen Ping Wang 2023Underground Space2023,,6:0
12Facile preparation and efficient MnxCoy porous nanosheets for the sustainable catalytic process of soot显示文摘The pursuit of high-performance is worth considerable effort in catalysis for energy efficiency and environmental sustainability. To develop redox catalysts with superior performance for soot combustion, a series of Mn_(x)Co_(y) oxides were synthesized using MgO template substitution.This method greatly improves the preparation and catalytic efficiency and is more in line with the current theme of green catalysts and sustainable development. The resulting Mn_(1)Co_(2.3) has a strong activation capability of gaseous oxygen due to a high concentration of Co^(3+) and Mn^(3+). The Mn doping enhanced the intrinsic activity by prompting oxygen vacancy formation and gaseous oxygen adsorption. The nanosheet morphology with abundant mesoporous significantly increased the solid–solid contact efficiency and improved the adsorption capability of gaseous reactants. 13Identifying Qingkailing(清开灵)ingredients-dependent mesenchymal-epithelial transition factor-axiation“π”structuring module with angiogenesis and neurogenesis effects显示文摘OBJECTIVE:To explore the functional role of the drugdependent mesenchymal-epithelial transition(Met)-axiation'π'structural module of neurogenesis after processing by three components of Qingkailing injection in neurogenesis and angiogenesis in cerebral ischemia.METHODS:We used a Glutathione S-transferase(GST)-pull down assay,isothermal titration calorimetry assay,and other related methods to identify the relationships among Met,inositol polyphosphate phosphatase like 1(Inppl1),and death associated protein kinase 3(Dapk3)in this allosteric module.The biological effects of the modules of neurons generation composed of Met,Inppl1,and Dapk3 were measured through Western blot,apoptosis analysis,and double immunofluorescence labeling.RESULTS:The GST-pull down assay revealed that proline-serine-threonine rich domain of Met binds to the Src homology domain of Inppl1 to form a protein-protein complex;Dapk3 with a C-terminal domain interacts weakly with the protein kinase C domain of Met in the intracellular region.Thus,we obtained a“π”structuring module considered a neural regeneration module.The biological effects of angiogenesis and neurogenesis modules composed of Met,Inppl1,and Dapk3 were also verified.CONCLUSION:The study suggested that understanding the functional modules that contribute to pharmaceutics might provide novel signatures that can be used as endpoints to define disease processes under stroke or cerebral ischemia conditions.CHENG Kunming YUAN Jianan LIU Jun ZHANG Shengpeng XU Qixiang XIE Yong ZHAO Jingfeng ZHANG Xiaoxu TANG Xudong ZHENG Yongqiu WANG Zhong 2024Journal of Traditional Chinese Medicine2024,44,1:0
14Gut microbiota,immunity,and bile acid metabolism:decoding metabolic disease interactions显示文摘In recent decades,the global prevalence of metabolic syndrome has surged,posing a significant public health challenge.Metabolic disorders,encompassing diabetes,obesity,nonalcoholic fatty liver disease,and polycystic ovarian syndrome,have been linked to alterations in the gut microbiota.Nonetheless,the connection between gut microbiota and host metabolic diseases warrants further investigation.In this review,we delve into the associations between various metabolic disorders and the gut microbiota,focusing on immune responses and bile acid(BA)metabolism.Notably,T helper cells,innate lymphoid cells,macrophages,and dendritic cells have been shown to modulate host metabolism through interactions with intestinal microorganisms and the release of cytokines.Furthermore,secondary BA metabolites,derived from the microbiota,are involved in the pathogenesis of metabolic diseases via the farnesoid X receptor and Takeda G protein-coupled receptor 5.By covering both aspects of this immune system-microorganism axis,we present a comprehensive overview of the roles played by the gut microbiota,microbiota-derived BA metabolites,and immune responses in metabolic diseases,as well as the interplay between these systems.Qixiang Zhao Jiayu Wu Yong Ding Yanli Pang Changtao Jiang 2023Life Metabolism2023,2,6:0
